A Study on the Heavy Metal Contamination of paddy Soil in the Vicinity of the Seosung Pb-Zn Mine (서성 연-아연광산 주변 농경지 토양의 중금속 오염 연구)

  • Fifty seven soil samples were collected from the paddy soil filled with tailings in the vicinity of the Seosung Pb-Zn mine. Those samples were analyzed for As, Cd, Co, Cr, Cu, Pb, and Zn in order to investigate heavy metal pollution levels in the paddy soil. Analyses of the soil samples were carried out using Inductively Coupled Plasma Atomic Emission Spectrometry(ICP-AES) . Paddy soils show pH range from 6.55 to 8.26. X-ray diffraction analyses of the paddy soil indicate that the soils consist predominantly ankerite, siderite, quartz, mica, and clay minerals with minor amounts of amphibole and chlorite. The mineral composition of the waste rocks consists of massive galena, sphalerite, and minor amounts of pyrite, arsenopyrite, chalcopyrite, calcite, siderite, Pb-sulfosalt, and marcasite. The paddy soils were significantly contaminated by heavy metals(average concentrations, As: 334.4 ppm, Cd: 37.6 ppm, Co: 15.7 ppm, Cu: 214.1 ppm, Pb: 4,612 ppm, and Zn: 4,468 ppm).

Development and Verification of a Simultaneous Analytical Method for Whole Blood Metals and Metalloids for Biomonitoring Programs (바이오모니터링 프로그램을 위한 혈중 금속류 동시분석법 개발 및 확인 평가)

  • Objective: Biological monitoring of trace elements in human blood samples has become an important indicator of the health environment. The purpose of this study was to detect and evaluate multiple metal items in blood samples based on ICP-MS, to perform comparative evaluation with the existing analysis method, and to develop and verify a new method. Methods: 100 μL of whole blood from 80 healthy subjects was used to analyze ten metals (Sb, tAs, Cd, Pb, Mn, Hg, Mo, Ni, Se, Tl) using ICP-MS. Verification of the analysis method included calculation of linearity, accuracy, precision and detection limits. In addition, a comparative test with the conventional graphite furnace atomic absorption spectroscopy (GF-AAS) method was performed. In the case of Pb, Cd, and Hg in whole blood, cross-analysis between Pb, Cd, and Hg analysis methods was performed to confirm the difference between the existing method and the new method (ICP-MS). Results: The coefficient of determination (R2) was 0.999 or higher in seven items and 0.995 or higher in three items. The Pb result showed that Pearson's correlation coefficient was very high at 0.983, and the intraclass correlation coefficient was 0.966. The Cd result showed that Pearson's correlation coefficient was 0.917 between the existing method and the new analysis concentration value. Its intraclass correlation coefficient was 0.960, and there was no significant difference between the two groups. Hg had a low correlation at 0.687, and the intraclass correlation coefficient was 0.761, which was lower than that of Pb and Cd. The intra-day and inter-day accuracy of Pd and Cd were satisfactory, but Hg did not meet the criteria for both accuracy and precision when compared with the conventional analysis method. Conclusion: This study can be meaningful in that it proposes a more efficient and feasible analysis method by verifying a blood heavy metal concentration experiment using multiple simultaneous analyses. All samples were processed and analyzed using the new ICP-MS. It was confirmed that the agreement between the two methods was very high, with the agreement between the current and new methods being 0.769 to 0.998. This study proposes an efficient simultaneous methodology capable of analyzing multiple elements with small samples. In the future, studies of various applications and the reliability of ICP-MS analysis methods are required, and research on the verification of accurate, precise, and continuous analysis methods is required.

Application of Toxicity Identification Evaluation Procedures for Toxic Effluents from the Aluminum Rolling Industry (알루미늄 가공 공장 배출 방류수의 독성 원인물질 탐색)

  • Objectives: The objective of this study is to identify toxicants causing acute toxicity in effluents from the aluminum rolling industry that violate the discharge limits in Korea. Methods: Whole effluent toxicity tests (WET) were conducted on effluent discharged from the aluminum rolling industry following the US EPA WET test methods. We collected effluent samples three times and evaluated acute toxicity by using Daphnia magna. We employed toxicity identification evaluation (TIE) procedures to identify toxicants causing toxicity in the effluent. Results: No specific chemical groups were identified in the seven different manipulations applied to the of wastewater effluent samples showing 1.3 toxic units (TU) according to the TIE phase I procedures. Water quality parameters for water hardness, electric conductivity and heavy metals (Mn) were 4,322 mg/l as $CaCO_3$, 11.39 mS/cm, and $5,551{\mu}g/l$, respectively. Considering water hardness and reference toxicity, high concentrations of Mn can be disqualified from the causative toxicants. Consequently, high ionic concentrations of $Na^+$(1,648 mg/l), $Ca^{2+}$(1,048 mg/l), $Mg^{2+}$(1,428 mg/l) and $SO_4{^{2-}}$(7,472 mg/l) were identified to be causative toxicants. Water hardness and electric conductivity exceed the $EC_{50}$ value obtained by biological toxicity tests using Daphnia magna. Conclusion: According to TIE procedures, high salt concentration is determined to be a major toxicant in the effluent of agro-industrial wastewater treatment plants receiving wastewater from the aluminum rolling industry.

Use of Paper Mill Sludge and Briquet Ash as Root Zone Soil Mixtures for Thrfgrass Culture (잔디상토로서의 제지스럿지와 연탄재 이용에 관한 연구)

  • To determine the use of waste materials as root zone soil mixtures for turfgrass culture, the effects of paper mill sludge and briquet ash on physical and chemical properties of soil and growth of turfgrasses were examined. Three turfgrass species of zoysiagrass(Zoysia japonicaSteud.). kentycky bluegrass(Poa pratensis L. 'Ram I') and creeping bentgrass(Agrostis panistris Huds 'Persucross') were cultured in 32cm diameter plastic pots containing various soil mixtures. The basic ingredients used for mixtures included sand(SD), field soil(SL), paper mill sludge(PS), sphagnum peat moss(PM) and briquet ash(BA). Seven combinations using these ingreients were mixed in different percentage by volume as follows: SD+SL+PM(80:10.10), SH+SL+PS(80:10:10), SD-PM(80:20), SD+PS (80:20), SD+BA(80:20), SD+BA+PM(60:20:20) and SD+BA+PS(60:20:20). 1. Paper mill sludge showed pH of 6.6, more than 30% of organic matter content, and higher concentrations of total N, P, k, Ca, Mg and CEC. Bulk density, fild moisture capacity and electrical conductivity of soil mixtures were increased by the comimation of 10~20% PS by volume. 2. Briquet ash showed pH of 8.0, and higher levels of P, k, Ca and Mg than those of field soiks. Bulk density, field moisture capacity and hardenss of soil mixtures were increased but vertical water flow rate and electrical conductivity were decreased by the combination of 20% BA by volume. 3. Phytotoxic effects of PS and BA on growth of turfgrasses were not found. Shoot growth of all three species was higher in soil combination of SD+BA+PS than that of SD+SL+PM added with fertilizer. However, root growth was better in soil mixtures combined with PM. Soil mixtureomposed of 60% SD, 20% BA and 20% PS by volume was most effective on growth of all three species. 4. Paper mill sludge resulted in higher N level in the leaf tissue. The contents of heavy metals such as Cd and Ph did not vary significantly among soil mixtures and species. However, the Mn level was 2~3 times higher in plants growh in mixtures containing PM compared with others, and especially it was higher in creeping bentgrass than other species.

Characteristics of Biosurfactant Producing Pseudomonas sp. G314 (생물 계면활성제를 생산하는 Pseudomonas sp. G314의 특성)

  • Three hundred thirty two bacterial colonies which were able to degrade crude oil were isolated from soil samples that were contaminated with oil in Daejon area. Among them, one bacterial strain was selected for this study based on its low surface tension ability, and this selected bacterial strain was identified as Pseudomonas sp. G314 through physiological-biochemical tests and analysis of its 16S rRNA sequence. Pseudomonas sp. G314 showed a high resistance to antibiotics such as ampicillin, chloramphenicol, spectinomycin, and streptomycin, and heavy metals such as Li, Cr, and Mn. It was found that the optimal pH and temperature for biosurfactant production of Pseudomonas sp. G314 were pH 7.0 and $30^{\circ}C$, respectively. After seven hours of inoculated, the biosurfactant activity reached the maximum, and surface tension of the culture broth was decreased from 72 to 25 dyne/cm. The crude biosurfactant was obtained from the culture broth by acid precipitation, followed by solvent extraction, evaporation and then freeze drying. The CMC (critical micelle concentration) value of the crude biosurfactant was 20 mg/L.

Characterization of Fe (III)-Reducing Bacteria Isolated from the Sediment of Chunho Reservoir (천호지 저질토에서 분리한 철환원세균의 특성)

  • Microbial Fe (III) reduction is important for the biogeochemical cycle in the sediment of freshwater system. Also, the Fe (III) reducing mechanism make a model of oxidizing organic compounds and reducing toxic heavy metals, such as chrome or uranium. Thirty-seven strains which have Fe (III) reducing activity were isolated from sediments in lake Soyang and Chunho reservoir. The initial concentration of Fe (II) was the highest in sediments of lake Soyang. However, the highest Fe (III) reducing activity was shown in Chunho reservoir. All isolates were tested for Fe (III) reducing activity. Strains C2 and C3, which were isolated from sediments of Chunho reservoir, showed the highest activity. These strains were tested to see if they utilize various electron donors such as glucose, yeast extract, acetate, ethanol and toluene. Significantly, glucose and yeast extract were used as electron donors. Also these strains were conformed to use humid acid and nitrate as electron accepters. The 16S rRNA sequences of strains C2 and C3 were closely related to Aeromonas hydrophila with 95% similarity.

Inspections on the Food Safety of Pheasant and Mallard as a Meat Resource (식육자원(食肉資源)으로서의 꿩과 청둥오리 고기의 안전성(安全性) 검사(檢査))

  • Studies on the food-safety of pheasant and mallard, which belong to wild fowl as new meat resources. were carried out. The results were summarized as follows : 1. Food poisoning bacteria including Salmonella spp, was not detected from the inspections of small intestine, cecum, and rectum. 2. Parasite inspection tests on blood, feces, digestive organ, and thoracic organs were negative. 3. Antibiotic residues from the carcass muscle by simplified disk methods were not detected. 4. Seven different pesticide residue tests, such as DDT and BHT, on the muscle and liver were negative. 5. Four different kinds of toxic heavy metals such as Cd were much lower than the permissible concentration. Studies on the food safety tests and inspections from the pheasant and mallard were revealed that from taking this new food resources, the toxicities would be very low for the human health by the direct influences.

Geochemical characteristics and benthic faunal facies in the sediments around the Oenaro Island, southern part of Korea (외나로도 주변해역 퇴적물의 지화학적 특성과 저서 생물상)

  • In order to evaluate the relationship between geochemical characteristics and benthic facies of the sediments from the Oenaro Island where red tide proliferation is first observed every year including this year, surface and short multiple core sediment samples were analyzed in terms of geochemical and benthic facies variation. The contents of organic carbon, carbonate, and sulfide gas were relatively low. The variation in C/N ratios, which indicate nature of organic carbon, suggested that the organic carbon recorded in the study area is composed of mixtures of marine and terrigenous organic matters. The concentration of minor elements found at the surface and multiple core sediment samples were also low as well as the enrichment factors(Ef) for the seven heavy metals indicated that the sediment of this area is not polluted significantly. The macrobenthic faunal community comprised 61 species, and their mean density was 708 ind./m2. Polychaete worms were major taxa of this benthic community. A crustacean amphipod, Melita sp. was the most abundant species accounted for 20.7% of total abundance, and the small polychaetes such as Heteromastus filiformis, Paralacydonia poradoxa, Magelona japonica and Sigambra tentaculata were the next dominant species. The macrobenthos around the Oenaro Island were more diverse and abundant than that in Gamak Bay. The benthic communities in the study area sustained somewhat different species composition based on the cluster analysis and the MDS ordination. The benthic community health condition at three stations seemed to be unbalanced, and slightly polluted based on the biological index such as BPI and BC. There was no clear relationship between the geochemistry characteristic and the benthic faunal facies attributed by the micro-algal blooms in this coastal area.

A Study on the Water-Purification Characteristics of Bio-Composite Planting Blocks (바이오 복합 식생블록의 수질정화 특성 연구)

  • This study was performed to evaluate the water purification properties of bio-composites planting blocks using oyster shell and effective microorganism that have high absorption ability of heavy metals and organics to develop environmentally friendly river embankment technique contained various factors such as oyster shells, effective microorganism, porous concrete and planting embankment block. To maximize greening effect, the seeds were arbitrarily sown. In addition, in order to analyze the effect of water quality purification after the planting, the samples were collected from each designated zone 1, 7 and 30 days after steeping in water. Then, the samples were analyzed in terms of seven test items such as SS, BOD, COD, T-N, T-P, pH, etc. on the basis of the test method for water pollution. The following conclusions were reached from the test result. As a result of analysis for water quality purification for the concrete block containing the effective microorganism, it was found that the values for SS, BOD, T-N and T-P for the sample taken after 30 days were lower than the initial values, which indicated that the water purification effect had been created. The result of the water quality purification analysis for the concrete block containing oyster shell showed that the values for SS, BOD, COD and T-P for the sample taken after 30 days were lower than the initial values which also indicated that it had been effective in water quality purification.

Establishment of analytical methods for HPHC list of mainstream cigarette smoke (담배 주류연 중 7개 그룹의 유해성분(HPHC) 분석법 확립 및 유효성 평가)

  • Harmful and potentially harmful constituents (HPHCs) are chemical compounds in tobacco smoke that cause harm to smokers and non-smokers. This study established and validated methods for the analysis of HPHCs from mainstream cigarette smoke. The analyzed HPHCs were categorized into seven groups: aromatic amines, volatile organic compounds (VOCs), heavy metals, tobacco specific nitrosamines (TSNAs), benzo[a]pyrene (B[a]P), ammonia, and carbonyl compounds. The methods were validated by specificity, linearity, limit of detection (LOD), accuracy, precision, and recovery. These validated methods were then applied to the reference cigarettes (1R5F, 3R4F). The correlation coefficients (r2) for the calibration curves of the seven groups were over 0.995. The LODs showed values of 0.01-0.04 ng/cig cig for aromatic amines, 0.01-0.16 μg/cig for VOCs, 0.01-1.27 ng/cig for heavy metals, 0.06-0.28 ng/cig for TSNAs, 0.04 ng/cig for benzo[a]pyrene, 0.08 μg/cig for ammonia, and 0.78-1.77 μg/cig for carbonyl compounds. The precisions obtained from the intra and inter-day batches were less than 15%. The accuracy and the recovery range were less than 15% and 79.2-117.5%, respectively. The proposed methods can therefore be applied for determining HPHCs in tobacco mainstream smoke.
