산국으로부터 분리한 Sesquiterpene Lactones의 흰쥐 복수암에 대한 효과 (Effects of Sesquiterpene Lactones Isolated from Chrysanthemum boreale M. against Sarcoma180 Implanted in ICR Mice)

  • 산국의 꽃으로부터 2종의 sesquiterpene lactones을 분리정제하였으며, 각각 Compound I 과 II로 명명하였으며, 이 두 물질을 Sarcoma180에 감염된 ICR mouse에 대하여 항암효과실험을 수행한 결과 Compound I과 II의 투여농도가 10mg/kg일 때 수명 연장효과가 각 각 143%, 134%로 나타났다. 또한 Compound I 과 II 를 1mg/kg 씩을 섞어서 투여하였을 때에 158%의 수명 연장효과가 나타났다

  • 지칭개, 구절초 및 산국에서 분리한 9종류의 sesquiterpene lactones에 대한 생리활성 실험의 일환으로, 10종의 세균에 대한 항세균활성과 6종의 식물병원균에 대한 항진균활성을 조사하였다. 항세균 활성실험에서는 지칭개에서 분리한 hemistepsin A와 B 그리고 산국에서 분리한 tulipinolide가 시험균주 전반에 대하여 강한 활성을 보였으며, 항진균 활성실험에서는 hemistepsin B, angeloylcumambrin B, tigloylcumambrin B, 그리고 costunolide가 광범위한 활성을 보였다. 또한, 항세균 활성실험과 항진균 활성실험 모두 비교적 뚜렷한 균주특이성을 나타내었다.

  • The inhibitory effect of Inula sesquiterpene lactones on the growth of Phaseolus vulgaris was tested and the abnormality of the stem organization caused by the lactones was also examined. The longitudinal growth of the young stem and the expansion of the young leaf were stopped by the application of the lactones. However, this inhibitory effect was appeared and strictly restricted within the treated area. So the young shoot was observed for possible bending as a result of the unilateral application of the lactones. When the application of the lactones into the medium, the growth of the plant was entirely repressed. However, the growth of shoot and re-initiation of root were started after the plant was transfered to the lactone free medium. And partial reversal of inhibition of the stem growth was achieved by the additions of gibberelline and the lactones.

산국의 잎과 줄기에서 분리한 Sesquiterpene Lactone들의 구조규명 및 생리활성 (Structural Analysis and Biological Activities of Sesquiterpene Lactones Isolated from the Leaves and Stems of Chrysanthemum boreale Makino)

  • Chrysanthemum boreale Makino is widely distributed in Korea, China, Japan and Southeast Asian countries. C. boreale is one of the herbs used for treating various inflammatory diseases in oriental medicine. The present study was conducted to identify biologically active compounds from the leaves and stems of C. boreale. We isolated two sesquiterpene sactones from the leaves and stems of C. boreale using silica gel column chromatography and recyclic high perfomance liquid chromatography. The lactones were characterized by their spectroscopic data (NMR, IR, MASS). These compounds were subjected to Farnesyl Protein Transferase (FPTase) inhibition, Nitric Oxide (NO) release inhibition and apoptosis inhibition. The structur of the following isolated compound were elucidated 8,10-${\small{O}$-Acetyl-2-methoxy-10-hydroxy-3,11(13)-guaiadiene-12,6-olide and 4,10-dihydroxy-8-${\small{O}$-Acetyl-2,11(13)-guaiadiene-12,6-olide. In the NO release inhibition assay, compound 2 showed strong activities, with an $IC_{50}$ value of $7{\mu}g/mL$, whereas compound 1 did not exhibit significant activity with an $IC_{50}$ value of over $14{\mu}g/mL$ against murine macrophage.

돼지감자로부터 분리된 Sesquiterpene Lactone의 세포독성 (Cytotoxity of Sesquiterpene Lactones from Leaves of Helianthus tuberosus L.)

  • The $CH_2Cl_2$ soluble part of the leaves of Helianthus tuberosus L. (Compositae) exhibited a potent cytotoxic activity against the cultured human tumor cell lines including A-549, SK-OV-3, SK-MEL-2, XF498 and HCT-15 in vitro. Bioassaydirected fractionation of the $CH_2Cl_2$ soluble part of this plant led to the isolation of four cytotoxic sesquiterpene lactones having ${\alpha}$-methylene-${\gamma}$-lactone ring in the molecule. On the basis of physical and spectral evidences, their structures were characterized as ${\Delta}^{4,15}$-isoatripliciolide tiglate (1), ${\Delta}^{4,15}$-isoatripliciolide methacrylate (2), budlein A isobutylate (3) and budlein A tiglate (4). The ${\Delta}^{4,15}$-isoatripliciolide tiglate (1) showed the most potent cytotoxic activity ($0.26{\mu}M<ED_{50}<2.16{\mu}M$) against all of the cell lines tested.

A Sesquiterpene, Dehydrocostus Lactone, Inhibits the Expression of inducible Nitric Oxide Synthase and TNF$\alpha$ in LPS- Activated Macrophages

  • An enhanced formation of nitric oxide (NO) is an important mediator of hypotention, peripheral vasodilation and vascular hyporeactivity to vasoconstrictor agents in endotoxaemia. And tumor necrosis factor (TNF${\alpha}$), as a primary mediator of circulatory shock has been known to induce inducible nitric oxide synthase (i-NOS), leading to excessive production of NO. We isolated two sesquiterpene lactone compounds from Saussurea lappa and their structures were elucidated as dehydrocostus lactone and costunolide. These compounds inhibit the production of both NO and TNF${\alpha}$ by LPS (1 $\mu\textrm{g}$/$m\ell$)-activated Raw 264.7 cells. NO was measured spectropho-tometrically as nitrite by the Griess reagent and TNF${\alpha}$ by ELISA. Dehydrocostus lactone (IC$\sub$50/ : 3.0 ${\mu}$M) and costunolide (IC$\sub$50/ : 4.5 ${\mu}$M) inhibited the production of NO in LPS-activated Raw 264.7 cells by suppressing inducible nitric oxide synthase enzyme expression. These compounds also decreased the TNF${\alpha}$ levels in LPS-activated system in vitro and in vivo.

수종의 국화과 식물에서 분리한 Sesquiterpene Lactone들의 생리활성(제1보) - 암세포주에 대한 세포독성 - (Biological Activities of Sesquiterpene Lactones isolated from Several Compositae Plants Part 1 - Cytotoxicity against Cancer Cell Lines -)

  • A diverse panel of human tumor cell lines and a mouse melanoma cell line (B16-F1) were used for the cytotoxicity test of the nine sesquiterpene lactones with ${\beta}-methylene-{\gamma}-lactone$ group isolated from Hemisteptia lyrata, Chrysanthemum zawadskii and Chrysanthemum boreale. In the cell adhesion inhibitory activity test against B16-F1 mouse melanoma cell, hemistepcin B, cumambrin B, costunolide and tulipinolide were shown significant activities with $IC_{50}$ range of 2.2, 4.1, 0.9 and $0.3\;{\mu}g/ml$, respectively. In the cytotoxicity test against human tumor cells, the most active compound was costunolide having $IC_{50}$ values of below $0.3\;{\mu}g/ml$ against all the tested cell lines except for UACC62. Cumambrin A, hendelin and costunolide exhibited more strong activity against HCT15 and UO-31 cell lines than a positive control, adriamycin. All tested compounds showed an $IC_{50}$ values of below $5.0\;{\mu}g/ml$ against all the tested cell lines.

  • Continuation of a phytochemical study of Cichorium intybus L. (Astraceae) growing in Egypt, resulted in the isolation and identification of a new sesquiterpene lactone 3,4-dihydrolactucin, in addition to the eight known compounds; kaempferol, isoscutellarin, cichoriin, umbelliferone, lupeol, lupeol acetate, ${\beta}-sitosterol$, and ${\beta}-sitosterol-3-O-glucoside$. Chemical structures of the isolated compounds were assigned based on different physical, chemical, and spectroscopic techniques including IR, UV, MS, 1D- and 2D-NMR spectra. Furthermore, the antimicrobial, and spasmogenic activities of some fractions and isolates were also assessed.

  • 쑥갓(Chrysanthemum coronarium L.) 꽃으로부터 암 예방활성 성분을 가진 2종의 화합물을 분리하여 다양한 spectral data를 종합한 결과, sesquiterpene lactone계인 eudesmanolide의 일종으로 밝혀졌으며 , compound 1과 2는 각각 douglanine과 reynosin인 것으로 동정되었다. 이 화합물들은 쓱갓에서는 처음으로 분리 보고되는 화합물이며, 세포독성을 측정한 결과, 두 화합물 모두 3종의 인체암 세포주에 대하여 비교적 강한 활성을 나타내었다.
