Liver Targetability and Pharmacokinetics of $[^3H]$-Methotrexate-Lactosaminated Bovine Serum Albumin Conjugates ($[^3H]$-메토트렉세이트-락토오스아미노화한 소 혈청 알부민 공유결합체의 간표적성 및 체내동태)

  • The organ distribution of $[^3H]$-methotrexate-lactosaminated bovine serum albumin conjugates ($[^3H]$-MTX-LBSA) was investigated to examine their role as a liver-specific anticancer drug. Synthesis of lactosaminated bovine serum albumin(LBSA) with BSA, lactose and sodium cyanoborohydride through reductive amination was followed by its conjugation with methotrexate (MTX) and 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC), thereby synthesizing [$[^3H]$-MTX-LBSA conjugates. Organ distribution and plasma elimination profiles were studied in male Wistar rats after intravenous injection of [$[^3H]$-MTX-LBSA conjugates. The fates of $[^3H]$-MTX and the $[^3H]$-MTX-BSA conjugates´fates were also investigated for comparison. The results showed that the plasma level of $[^3H]$-MTX-LBSA conjugates declined more rapidly than those of $[^3H]$-MTX-BSA and their liver concentration was significantly higher than those of other treatment (p<0.01). In addition, their uptake compared to the amount taken up by the liver (1 : 33.1 at 10 min, 1 : 24.1 at 120 min). All these suggested that MTX-LBSA conjugate is one of the drug delivery system (DDS) that is advanced in concentrating MTX in the liver and minimizing the renal toxicity of MTX.

Effect of Cholesterol and Serum Albumin on Sperm Ability and Lipid Peroxidation during the Storage of Miniature Pig Sperm (미니돼지정액의 보존 시 콜레스테롤과 혈청 알부민이 정자 성상과 지질 과산화에 미치는 영향)

  • This study was undertaken to find out the effect of cholesterol and serum albumin on sperm ability and lipid peroxidation levels period to the liquid storage of miniature pig sperm. Ejaculated semen from miniature pigs was collected by gloved-hand method into a pre-warmed ($37^{\circ}C$) thermos bottle, and extended with Modena solution {with and without BSA, methyl-beta-cyclodextrin (-cholesterol) and cholesterol loaded cyclodextrin (+cholesterol)}. Each semen was assessed for viability (SYBR-14/PI staining) and acrosome intactness, intensity and capacitation status by chlorotetracycline (CTC) staining at 1, 3, 5, 7 and 10 days of storage. At for the effects of cholesterol and serum albumin on lipid peroxidation, semen were incubated with $H_2O_2$ ($10\;{\mu}M$), and lipid peroxidation level were measured by flow cytometry using the lipid peroxidation reporter probe $C_{11}-BODIPY^{581/591}$. The result, lipid peroxidation level in sperm added with cholesterol were lower in $10\;{\mu}M$ $H_2O_2$ compared to the added sperm with serum albumin. Also, added cholesterol to sperm had significant (p<0.05) higher viability when storage for 7 and 10 days and lower when 10 days of storage percentage of acrosome-reacted sperm (AR pattern) in acrosome state as say result compared to other treated groups. In conclusion, role of cholesterol during lipid storage in miniature pig spermatozoa was protected boar spermatozoa from lipid peroxidation prior to lipid storage. Addition serum albumin during lipid storage in sperm may be induce sperm membrane damage by lipid peroxidation. Therefore, addition of cholesterol to miniature pig sperm will be lead to extension of liquid storage periods.

Comparison of Protein Electrophoresis Fractions in the Leptospirosis Patient Serum

  • The mechanisms by which leptospires caused disease are not well understood. A number of putative virulence factors have been suggested, but with few exceptions their role in pathogenesis remains unclear. In these days, many cases of leptospirosis are diagnosed by serological immunoassay. Leptospirosis is characterized by the histopathology of liver, kidney, heart, and lung, but the electrophoresis fractions of serum protein are not compared. We analyzed total protein, albumin, aspartic aminotranferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), creatinine and urea nitrogen (UN) in sera of patients diagnosed leptospirosis. Total protein and albumin were decreased in 18.5% and 31.2% of patients, respectively. AST, ALT, ALP, UN and creatinine were increased in 90.4%, 66.9%, 28.0%, 15.9% and 10.8% of patients, respectively. We performed cellulose acetate membrane electrophoresis (EP) on sera of patients increased both of AST and ALT, and of patients increased both of creatinine and UN. In patients increased both of AST and ALT, and in patients increased both of AST and ALT, the relative percentage of albumin to total protein in patient serum was dcreased in 89.1 % of patients. α₁-globulin, α₂-globulin, β-globulin and γ-globulin were increased in 85.1 %, 75.2%, 33.6% and 98.0% of patients, respectively. In patients increased both of creatinine and UN, the relative percentage of albumin to total protein in patient serum was dcreased in 93.8% of patients. α₁-globulin, α₂-globulin, β-globulin and γ-globulin were increased in 87.5%, 100%, 31.2% and 93.8% of patients, respectively. These data indicate that infection of Leptospira causes severe liver damage to most of leptospirosis patients, but doesn't cause renal damage to most of them. The relative rate of serum protein electrophoresis fractions to total protein are not identical with them of typical hepatitis patient.

Improvement of the regeneration and protoplasts fusion of Candida pseudotropicalis by bovine serum albumin, myoinositol and ergosterol (Bovine serum albumin, Myoinositol과 Ergosterol에 의한 Candida pseudotropicalis의 원형질체 재생 및 융합증진)

  • The effects of bovine serum albumin, myoinositol and ergosterol on protoplast formation, regeneration and fusion from auxotrophic mutants of Candida pseudotropicalis were examined. Frequency of protoplast formation ranged from 48 to 98% depending on auxotrophic types. When myoinositol (0.5mg/ml) and ergosterol (0.1mg/ml) were supplemented in the medium of cell growth, and bovine serum albumin (4mg/ml)was added to protoplasting buffer, 50-100% of cells were converted to protoplasts. Such a treatment of three additives improved 2.2-3.0 fold of regeneration rate of protoplasts. The fusion frequencies between complementary auxotrophs ranged from $7.0\times 10^{-4}$ to $1.5\times 10^{-3}$ in the optimal conditions. These values showed 1.9-2.3 fold increase when compared with fusion frequencies obtained without the treatment of additives. These results suggested that these comsion frequencies obtained without the treatment of additives. These results suggested that these xompounds may improve protoplast regeneration and fusion between complementary auxotrophs used in this study.

  • The study was performed to investigate the inhibitory action on the growth rate of Candida albicans under the various immunological conditions, Candida albicans was grown in broth media containing 0.1ml. (in total broth volume 4.0ml.) of normal human serum, diabetic patient serum, albumin solution, artifical bulla content and nutrient solution as control subject. The inhibitory action of Candida albicans was indicated by mesurement of transparency rate with use of the spectrophotometer. The results are as follows: 1. Normal human serum shows inhibitory effect on the growth of the candida distinctly. 2. Albumin solution reveals almost similar to that of the effect of normal human serum. 3. Artificial bulla content which obtained by irradiation of ultraviolot ray after application of 1% 8-Methoxy-psolaren cream and the diabetic patient serum shows reduction of inhibitory effect as compared with that of the normal human serum. 4. It is estimated that the titer of negative effect of diabetic patient serum is not related with the variation of immunoglobulin titer in patient serum.

  • In order to study the effect of Dokwhalgisaentang on the blood in normal rats with Dokwhalgisaentang extract solution was adminstrated in rats. The rats carried on every day for 5days, l0days and 15days respecitively on the corresponding blood in rats, and thereafter the contents of serum BUN, creatinine, creatine, total protein, albumin, total bilirubin, cholesterol were measured in rats. The following results have been obtained : 1. Dokwhalgisaentang extract decreased the serum BUN significantly in rats. 2. Dokwhalgisaentang extract decreased the serum creatine significantly in rats. 3. Dokwhalgisaentang extract decreased the serum creatinine significantly in rats. 4. Dokwhalgisaentang extract decreased the serum total protein significantly in rats. 5. Dokwhalgisaentang extract decreased the serum albumin significantly in rats. 6. Dokwhalgisaentang extract increased the serum total bilirubin significantly in rats. 7. Dokwhalgisaentang extract decreased the serum cholresterol significantly in rats.

  • Simple coacervation of bovine serum albumin was studied to prepare biodegradable microacapsule. Albumin microcapsules were prepared by using acrylonitrilestyrene polymer (M-80) resin beads and sulfamethoxydiazine as the core materials. The albumin to beads ratio was found to be 1:2.3 and the albumin to sulfamethoxydiazine ratio to be 1:2.9. The 50% release times (T$_{50%}$) of sulfamethoxydiazine and microencapsulated sulfamethoxydiazine were 6 min. and 73 min., respectively. The surface appearance of the microcapsule collected after release experiment was not different from those of the original microcapsule. In addition to slowing release of drug the microencapsulation process masked characteristic bitter taste of sulfamethoxydiazine.

  • Pork is an excellent source of essential nutrients such as protein. However, pork can trigger hypersensitivity and serum albumin of pork is known as major allergen. In this study, to evaluate the effect of gamma irradiation on the allergenicity of porcine serum albumin (PSA), PSA solution was irradiated at 3, 5, 7, 10, 15, and 20 kGy. The changes in the ability of PSA to bind IgG and patient's serum caused by gamma irradiation were observed by ci-ELISA and immunoblotting. SDS-PAGE was used for measuring the conformational change of gamma-irradiated PSA. The ability of 3-kGy-irradiated PSA to bind p-IgG and patient's serum was decreased to 30% and 15%, respectively. The binding ability showed no significant differences among all irradiated samples. SDS-PAGE showed that the irradiated PSA bands were degraded and aggregated. Immunoblotting of irradiated PSA revealed that IgG and patient's serum were rarely recognized at 3 kGy. Therefore, gamma irradiation could be applied to less-allergenic pork products.

  • In order to examine the interaction of albumin with the sperm during capacitation in mouse, proteins of cauda epididymal sperm were extracted under various conditions and analyzed with SDS-PAGE. Sperm surface labeling patterms were also examined using fluorochroin~conjugated wheat germ agglutinin (WGA) and bovine serum albumin (BSA). Albumin was detached from the sperm surface during the incubation and seemed to be constituted the major protein components of the conditioned media in which sperm incubated for 90 mm. Detachment of albumin from the sperm was not affected by the Ca2+ in the medium. WGA-FITC labeling confirmed that Triton X-100 permeabilired plasma membrane overlaying the apical segment of sperm head and detached plasma membrane associated proteins having negatively charged glycoconjugates. BSA-FITC labeling of epididymal sperm occurred on the apical segment of periacrosoinal region and postacrosomal region of the head. BSA-FITC labeling was not observed in periacrosoinal region of the sperm treated with Ca2+-ionophore ~3187 (10 MM)~ whereas the postacrosome region of acrosome-reacted sperm was still labeled after the AR. These results suggest that albumin bound to the surface of epididymal sperm is detached during the capacitation process, and it might be involved In physiological change of sperm plasma membrane accompanying the capacitation.

  • This work was carried out to get some informations about blood types and their researches, involved blood stock and genetic identification. Horses examined were total 55 heads of sire, mare and their progeny in Korean Horse Affairs Association. 1. Albumin phenotypes of 26 mare were examined. The appearance of phenotype AA, BB, AB, was 1, 18, 7 respectively. The gene frequency of albumin A was 0.17 and albumin B was 0.76. 2. The appearance of phenotype AA, BB, AB in 29 progeny was 1, 16, 12 respectively. The gene frequency of albumin A was 0.24 and albumin B was 0.76. The gene frequency of gene A was higher than their parents. 3. Identification of the relationship between parents and their progeny was also examined. 4 of type AB between AA & BB, 4 of type BB between BB & BB, 13 of type AB between BB & AB were borned. In third case, all of progeny was type AB. This results suggest positive relationship between them.
