Changes in Blood Cellular Components, Serum Chemical Values and Serum Enzyme Activities in Korean Native Cattle infested with Fasciola hepatica (간질감염(肝蛭感染)이 한우혈액(韓牛血液)의 구성성분(構成成分)에 미치는 영향(影響))

  • Some hematological and biochemical indices were assayed in Korean native cattle naturally harbouring Fasciola hepatica infection and compared with uninfected controls. Affected animals revealed reduction in total erythrocyte count and hemoglobin content, and increase in total leukocyte count. Infected cattle had significantly lower levels of serum total protein, serum albumin and magnesium. Significantly higher values were obtained for the serum enzymes aspartate aminotransferase and alanine aminotransferase.

  • The effects of glycyrrhizic acid (GLZ) on protein binding of diltiazem, verapamil, and nifedipine were investigated. Protein binding studies (human serum, human serum albumin (HSA) and (X1-acid glycoprotein (AAG)) were conducted using the equilibrium dialysis method with and without addition of GLZ. The binding parameters, such as the number of moles of bound drug per mole of protein, the number of binding sites per protein molecule, and the association con-stant, were estimated using the Scatchard plot. The serum binding of nifedipine, verapamil, and diltiazem was displaced with addition of GLZ, and the decreases of Ks for serum were observed. GLZ decreased the association constants of three drugs for HSA and AAG, while the binding capacity remained similar with addition of GLZ. Although the characteristics of interaction were not clear, GLZ seemed to mainly affect HSA binding of nifedipine rather than AAG binding, while GLZ seemed to affect both AAG- and HSA-bindings of verapamil and dilt-iazem resulting in a serum binding displacement.

Correlation of Hemoglobin and RBC count with Adiponectin Level

  • Adiponectin is secreted specifically by adipose tissue and regulates lipid and glucose metabolism. In addition, adiponectin has been found to inhibit inflammatory process and possibly atherogenesis. This study was done to compare hematological parameters, serum lipids with serum adiponectin level according to body mass index (BMI) and gender in eighty eight Korean adults aged 40 to 68. In this study, the association between serum adiponectin and other variables including RBC counts, hemoglobin, hematocrit and albumin were investigated. RBC, hemoglobin status, was inversely associated with serum adiponectin levels in normal adult women. Adiponectin levels were significantly lower in men than women (p<0.01). In men, serum adiponectin levels were negatively correlated with body mass index (BMI) (p<0.05). However, no correlations were found in women. These results might imply that the regulation of key adipokines such as adiponectin might be a strategy for the prevention or treatment of obesity-associated diseases.

  • The effect of the extracellular protease of Salmonella schottmulleri on human serum constituents such as immunoglobulins, hemoglobin and lysozyme and tissue constituents such as fibronectin and collagens was investigated. This protease degraded collagens (type I and III), fibronectin and serum proteins such as human hemoglobin and lysozyme. Bovine serum albumin was degraded slightly. Thus, the present study suggested the possibility that this protease is not only played an important role in invasion of S. schottmulleri by degrading the constituent proteins such as collagens and fibronectin but also induced complications observed in septicemia and chronic infections by degrading the serum proteins. This protease is also capable of degrading defence-oriented humoral proteins, immunoglobulins (IgG and IgM). Furthermore, it is toxic to HEp-2 cells. These findings clarified the possible role of Salmonella protease as a virulence factor in the pathogenesis of Salmonella infections.

Grazing system and floor type effects on blood biochemistry, growth and carcass characteristics of Nguni goats

  • Objective: Purpose was to determine the effects of grazing system and floor type on concentrations of blood metabolites, activity of creatine kinase, body weight and carcass characteristics of castrated Nguni goats. Methods: Forty eight, 7 month old goats were randomly allocated to herding and tethering treatments from 0800 to 1300 hours and accommodated on slatted and earth floors daily. Blood samples were collected by jugular venipuncture every fifteenth day for metabolite analysis. Slaughter was done at a commercial abattoir following 5 months of monitoring. Results: Tethered goats had significantly higher concentrations of urea (5.19 mmol/L) (p<0.001), creatinine ($55.87{\mu}mol/L$) (p<0.05), total protein (64.60 g/L) (p<0.01), and globulin (49.79 g/L) (p<0.001), whereas herded goats had higher glucose (3.38 mmol/L) (p<0.001), albumin (15.33 g/L) (p<0.05), albumin/globulin ratio (0.34) (p<0.01), and body weight (24.87 kg) (p<0.001). Slatted floors caused higher (p<0.01) albumin at 15.37 g/L. The interaction of grazing system and floor type affected creatinine, total protein, globulin at (p<0.01) and albumen/globulin ratio at (p<0.01). The least creatinine concentration and albumin/globulin ratio was in herded and tethered goats that were accommodated on earth floors, respectively. The highest total protein and globulin concentrations were in serum of tethered goats that were accommodated on earth floors. The highest (p<0.05) dressing percentage (45.26%) was in herded goats accommodated on slatted floors. Conclusion: Herding of goats lowered globulin concentration, improved estimated feed intake, blood glucose and albumin concentrations, albumin globulin ratio, increased body weights and weight related carcass characteristics. Floor type had very little effects on metabolites where earth floors only reduced albumin concentration. Tethering and housing goats on earth floors resulted in double stress that increased chronic infections.

  • 표면에 여러 가지 기능성기를 가지는 microspheres는 immunoassay, drug delivery system, cell separation등 의용공학분야에 응용이 기대되고 있다. 이들 분야의 응용을 위하여 유화제를 사용하지 않으면서, 기존의 회분식, 반회분식, seed 중합법등의 문제점을 극복한 two stage shot growth technique올 개발하여 여러 가지 기능성기가 표면에 도입된 microspheres를 제조하였으며, 응용의 전단계로서 이들 microspheres에 대한 모델 단백질(BSA)의 흡착실험을 pH, 기능 성기의 종류와 양, BSA농도를 변수로 행하여 최대 흡착량을 보이는 조건을 결정하였다.

The Effect of Ginseng Saponins on the Activity of Lipoprotein Lipase in Vitro (Lipoprotein Lipase의 활성에 미치는 인삼 Saponin의 영향)

  • In order to investigate the effect of ginseng saponins on the activity of lipoprotein lipase, it was attempted to conform the enzymatic hydrolysis of chylomicron with post-heparin induced plasma lipoprotein lipase of normal rabbit in vitro. And the activity of lipoprotein llipase was determined by the quantitative determination of liberated free fatty acids on the hydrolysis of chylomicron. As the result, it was observed that the ginseng saponins accelerated the hydrolysis of chylomicron by post-heparin plasma in vitro. And the optimum concentration of ginseng saponins for the activity of the lipoprotein lipase in the 2% bovine serum albumin was $10^{-4}%$. But ginseng saponins on the hydrolysis of chylomicron was influenced by the presence and the absence of albumin. And the optimum concentration of albumin and Na-cholate on the activity of lipoprotein lipase was each of the $10^{-6}%$ albumin and 5mM Na-cholate. From these results, it seems that ginseng saponins might stimulate the intravascular hydrolysis of chylomicron.

Covalent Interactions of Reactive Pentachlorophenol Metabolites with Cellular Macromolecules (Pentachlorophenol 대사물과 세포내 거대분자물의 반응에 관한 연구)

  • Pentachlorophenol(PCP) which ks widely used in wood preservation, pulp and paper mills, has led to a substantial envirortmental contamination. To get the reliable data for the effective health risk assessment with PCP, covalent binding potential of PCP to cellular macromolecules and glutathione(GSH) was investigated after intraperitoneal administration of $^{14}C-PCP$ to rats. PCP metabolites were able to bind covalently to serum albumin and hepatic protein in a dose- and time-dependent manner. Hepatic protein adducts of PCP metabolites were increased as a function of cytochrome P-450 activities, whereas, albumin adducts significantly decreased. Covalent binding of PCP metabolites with DNA or hemoglobin was not observed. GSH levels in liver tissue decreased over 12hrs, however, the level was recovered after 48hrs. Tetrachloro-1,4-benzoquinone (1,4-TCBQ), one of the most reactive PCP metabolites, conjugated with GSH very rapidly. Base on our results, we could conclude that PCP metabolized to reactive electrophilic metabolites by cytochrome P-450 isoenzymes and conjugated rapidly with neighboring protein or nonprotein sulfhydryl before reacting with DNA or hemoglobin. We propose that albumin adducts and mercapturic acids of PCP metabolites can be used good biomarker of recent PCP exposure.

Effect of Diet Contents on Serum Composition and Urinary Excretion in Female Students According to Sasang Constitution (사상 체질별로 분류한 여대생의 식사내용이 혈청과 뇨중 성분치에 미치는 영향에 대 한 연구)

  • This study was conducted to compare the serum composition and urinary excretion in four institutional groups and the effects of intakes of good foods and harmful foods according to the Sasang constitiution for a short time on serum and ruine compositon. The contitutions of 33 female stucdents were classified by O-ring test and muscular strength test according to the Sasang constitution, The number of each institutional group was So-yang-in 11, So-em-in 10 and Tae-em-in 12. No subject belonged to Tae-yang-in There were no significant differences among contitutional groups in the values of most serum composition and urinary excretion Serum levels of total cholesterol, glucose, calcium and phosphorus in Tae-em-in was the highest among groups and the level of albumin and total protein in So-em-in was highest. The values of serum compositions changed after the 33 subjects took good foods and then harmful foods according to The Sasang constitution for a short time(5 days). Dietary behavior of the subjects had some effects among the constitutional groups in the values of most serum composition and urinary excretion. But the effects of dietary behavior on the serum composition and urinary excretion showed various trends in the four institutional groups.

Investigation of the incidence rate of second grade milk in dairy farms on the central-southern region of Korea (우리나라 중남부지역 젖소목장에서 이등유 발생 조사)

  • The incidence of second-grade milk production in 9 dairy farms of South Korea was investigated from May 2011 to March 2012, and the serum composition of cows producing first- and second-grade milk in 14 farms including the 9 farms was analyzed. The incidence rate of second-grade milk production of 402 cows in nine dairy farms located in the central and southwestern regions of Korea was 15.4% with the highest rate being 34.4%. Seasonal morbidity was higher during late winter (February) and early summer (June) with the highest rate observed in February (32.6%) followed by November (33.3%). Second-grade milk was most frequently found within one month postpartum (34.1%) while only 3.5% was found during the first 60~90 days of lactating period (n=785, 5 herds). The morbidity increased thereafter (P<0.05) with the highest observed between 270~300 days of lactation (36.1%). The acidity was not significantly different between second-grade ($0.159{\pm}0.026%$) and first-grade milk ($0.158{\pm}0.027%$). Blood serum analysis of 371 cows in the 14 dairy farms indicated that aspartate aminotransferase (AST) level was significantly higher (P<0.001) in cows producing second-grade milk while albumin was significantly lower (P<0.001) than cows producing first-grade milk. Total protein and triglyceride was also significantly low along with glucose, non-esterified fatty acid and blood urea nitrogen in cows producing second-grade milk. Statistical analysis including sensitivity, specificity and positive/negative prediction values showed that lactating cows with high AST, low albumin, total protein and triglyceride levels in the serum tended to produce second-grade milk. It was concluded that serological parameters, especially live functional and metabolic-related serum compositions (AST, albumin, total protein and triglyceride), were significantly influenced in cows producing second-grade milk.
