흰쥐에 마늘 투여로 혈액의 지질량과 그외 혈액성분 변화 고찰 (Effects of Garlic on the Blood Lipids and Other Serum Components in Rats)

  • In present study possible hypolipidemic effects of garlic were investigated in an experimental model which consisted of 4 groups of Wistar rats(three weeks old): CO group received a basal diet(as a standard diet) contained 6.3% of corn oil. LO group was taken the experimental diet in which only 6.3% corn oil in basal diet was substituted by 12.6% of lard oil. So the experimental diet was somewhat high saturated fat diet. LG1 group was treated orally with garlic juice as 1% of raw garlic in the diet together with the same diet as the diet for LO group. LG2 group was taken 2% of garlic under the same dietary condition as that of LG1 group. The rat body weights prior and posterior to the experimental period were measured and the amount of the experimental dietary intake was determined at every 3 day interval. After the experimental dietary period of 30 days the blood obtained from all the sacrificed rats were analyzed for the biochemical parameters. Over the one month period of experiment there was no abnormality or apparent change in appearance and activity or diet consumption in all experimental rat groups. LO group fed a diet rich in lard showed significantly increased body weight gain rate by 30% and also elevated levels of triglyceride, total cholesterol and LDL cholesterol by 29.9%, 27.4% and 62.3% respectively and no significant difference in the levels of HDL cholesterol, GOT, GPT, blood glucose and blood urea nitrogen as compared to those of control group. So it seemed that the high calorie diet in LO group mainly contributed to the significant increase in body weight gain rate and other biochemical parameters. In spite of 1% garlic treatment LG1 group had significantly increased body weight gain rate by 25.2%, levels of triglyceride, total choesterol and LDL cholesterol by 25.2%, 24.6% and 50.5% respectively as compared to control and LO group, and no increase in the levels of HDL cholesterol, blood glucose and blood urea nitrogen. At garlic 1% diet there was no hypolipidemic efficacy in rat serum. Under the treatment with garlic 2% in diet LG2 group showed significantly increased body weight gain rate by 19.7% as compared to control and elevated levels of GOT, GPT and BUN by 85.8~96.4%, 127~148% and 88% respectively as compared to control, but LG2 group had significantly reduced levels of triglyceride, total cholesterol and LDL cholesterol as compared to LOand LG1 group. In present study there were no significant change in HDL cholesterol and blood glucose value. In LG2 group GOT, GPT and blood urea nitrogen values were slightly increased presumably due to the inhibiting effects of garlic on the hepatic or renal function of rats. Nevertheless in this study garlic may have some demonstrable hypolipidemic effects in rat.

다시마 첨가 패티가 경계역 고콜레스테롤혈증 성인의 식후 혈청 지질 및 혈당 농도에 미치는 영향 (Effects of Sea Tangle-added Patty on Postprandial Serum Lipid Profiles and Glucose in Borderline Hypercholesterolemic Adults)

  • 본 연구는 생리활성이 높은 다시마를 첨가한 패티가 정상 지질혈증을 보이는 사람에서 섭취 후 혈청 지질 양상과 혈당을 개선한 선행연구의 후속으로 경계역 고콜레스테롤혈증을 지닌 사람을 대상으로 하여 동일한 효과를 나타내는지 규명하고자 수행되었다. 대표적 패스트푸드인 패티가 정상 지질혈증자는 물론 고콜레스테롤혈증자에서도 섭취 후 혈청 지질 농도나 혈당을 강하시킨다면 건강지향적인 패티 개발의 가능성이 더 커질 수 있을 것이다. 실험패티는 표준패티 재료 중 육류의 2.5%를 다시마 분말로 대체하여 제조하였다. 실험패티는 표준패티에 비해 열량, 조단백, 조지방 및 콜레스테롤 함량이 유의하게 낮았고, 수분, 조회분, 나트륨 및 조섬유 함량은 유의성 있게 높았으며, 탄수화물 함량은 차이가 없었다. 경계역 고콜레스테롤혈증을 지닌 11명의 성인에게 공복혈액을 채취한 직후 표준패티 또는 실험패티 200 g을 200 mL의 물과 함께 10분 사이에 섭취하도록 하였고, 이후 30분, 60분, 120분 후에 채혈하여 혈청 포도당과 C-peptide 농도를 추적하였으며, 혈중 지질 농도는 240분까지 채혈하여 분석하였다. 패티 섭취 후 240분까지 혈청 TG를 비롯해 T-chol, LDL-C 및 HDL-C의 시간대별 농도와 각 지질 성분의 ${\Delta}-AUC$는 모두 두 패티 간에 차이가 나타나지 않았다. 반면에 혈당 농도는 패티 섭취 후 120분까지 실험패티가 표준패티에 비해 모든 시간대에서 유의하게 낮았으며 ${\Delta}-AUC$도 유의성 있게 낮았다. 혈청 C-peptide 농도는 패티 섭취 후 120분까지 비록 모든 시간대에서 두 패티 사이에 차이를 보이지 않았으나 ${\Delta}-AUC$는 유의적으로 낮았다. 이러한 결과는 실험패티에 함유된 다시마 분말이 혈당을 낮추고 또한 인슐린 분비를 감소시켰다는 것을 시사한다. 본 연구의 제한점으로 각각의 패티 200 g을 통한 탄수화물과 지방 및 콜레스테롤 섭취량이 비교적 적었던 점과 패티 섭취시험 대상자의 수가 많지 않았던 점 등을 들 수 있다. 그러나 패티 재료 중 육류의 2.5%를 다시마 분말로 대체함으로써 패티의 영양성분 조성이 개선된다는 점과 이러한 영양소 조성의 변화와 함께 패티 100 g에 함유된 2.25 g의 다시마가 경계역 고콜레스테롤혈증 성인에서 섭취 후 비록 혈청 지질 양상을 개선시키지는 못했지만 혈당을 저하시켰고 인슐린 분비를 감소시킨 본 연구결과는 다시마를 건강기능성 패티 제조에 활용할 가치가 있음을 시사한다.

공복혈당장애 성인에서 엽산 또는 아스코르브산의 보충급여가 혈장 호모시스테인 수준과 산화 스트레스에 미치는 영향 (Effects of Folic Acid or Ascorbate Supplementation on Plasma Homocysteine Levels and Oxidative Stress in Korean Adults with Impaired Fasting Glucose)

  • Impaired fasting glucose (IFG) is one of significant risk factors of developing diabetes. The persons with IFG are, thus, an important target group for primary prevention of diabetes. It is well known that plasma homocysteine concentration may be increased in poor folate nutritional status. Elevated level of plasma homocysteine is considered as a marker of enhanced oxidative stress. In addition, the protective effect against oxidative stress may be diminished in poor antioxidative nutrient status as vitamin C. It is, therefore, important to maintain adequate nutritional status of folate and vitamin C in the patients with type 2 diabetes or IFG. This study was performed to determine the effects of supplementation of folic acid or vitamin C on plasma concentrations of homocysteine, oxidized LDL, and lipids and on the activity of plasma anti-oxidative enzyme in patients with IFG. A total of 97 patients with IFG were participated voluntarily with written consents. They were divided into one of the four experimental groups; Control (C), Folatesupplemented (F), Ascorbate-supplemented (A), and Folate plus ascorbate-supplemented (FA). The subjects in C were taken placebo, those in F were supplemented 1 mg of folate, those in A were received 1,000 mg of vitamin C, and those in FA were given 1 mg of folate plus 1,000 mg of vitamin C daily for 4 weeks. No change in plasma concentrations of vitamin C, lipids, and oxidized LDL and the activity of GSH-Px were observed in vitamin C-supplemented group (A + FA) and folate-supplemented group (F + FA) compared to the placebo group (C + A). Only the folate-supplemented group (F + FA) had significantly increased average serum folate concentration and lowered plasma homocysteine concentration compared to the placebo group (C + A). Thus, it should be recommended the patients with IFG to increase folate intake through diets and, if it is not sufficient, to take folic acid supplements to prevent the development of complications induced by hyperhomocysteinemia as well as oxidative stress.

Gene Expression Profile in the Liver Tissue of High Fat Diet-Induced Obese Mice

  • The purpose of this study was to investigate the gene profiles that were up- or down-regulated in the livers of high-fat diet-induced obese mice and $db_-/db_-$ mice with deficient leptin receptor. C57/BL6 normal mice and $db_-/db_-$ mice, respectively, were divided into two groups and fed a standard or high-fat diet for four weeks. Liver weight was unchanged in the normal mice but the high-fat diet led to a 10% weight increase in the $db_-/db_-$mice. Adipose tissue mass increased by about 88% in the normal mice that were fed a high-fat diet and by about 17% in the $db_-/db_-$mice on the high-fat diet. In terms of serum lipids, total cholesterol significantly increased in mice on the high-fat diet. Microarray analysis was carried out using total RNA isolated from the livers of standard or high-fat diet-fed mice of the normal and $db_-/db_-$ strains. The change of gene expression was confirmed by RT-PCR. About 1.6% and 6.8% of total genes, respectively, showed different expression patterns in the normal mice fed the high-fat diet and $db_-/db_-$ mice. As a result of microarray, many genes involved in metabolism and signal pathways were shown to have different expression patterns. Expression of Mgst3 gene increased in the livers of normal and $db_-/db_-$ mice that were fed a high-fat diet. Wnt7b and Ptk9l were down-regulated in the livers of the normal mice and $db_-/db_-$ mice that were fed a high-fat diet. In conclusion, a high-fat diet induced obesity and affected gene expression involved in metabolism and signal pathway.

걷기운동 프로그램이 농촌노인의 대사증후군 관련인자에 미치는 효과 (Effects of a Walking Program on Metabolic Syndrome Risk Factors in Older People in Rural Areas)

  • Purpose: The purpose of this study was to examine the effects of a 12 week walking program on the risk factors associated with metabolic syndrome in older people in rural areas. Method: A total of 31 people among the people who live within the area served by E County Community Health Practice were observed from December 5, 2006 to February 23, 2007. This group included 18 people who were identified as having metabolic syndrome on the basis of the Asia-Pacific fatness basic standard (the experimental group) and a control group of 13 people who did not have metabolic syndrome. Results: The results of the study are as follows: In the experimental group, there was no significant difference in body composition between pre and post measurement. The small change in values that were observed in the experimental group did not reach statistical significance. However, body composition values in the control group increased. In the experimental group, there were significant differences in waist circumference, blood pressure, HDL-C, and fasting blood sugar between pre and post measurement. But there were no changes in the control group. There was no difference in TG in either group. As to changes in risk-factors related metabolic syndrome after the 12 week walking program, the number of risk-factors were effectively reduced in the experimental group, but not in the control group. Conclusion: According to the present study, a 12 week walking program led to significant improvement in body weight, BMI as the factors composing the body and in waist circumference, HDL-C, blood pressure, and fasting blood sugar as risk-factors related to metabolic syndrome in older people in rural areas.

알코올 의존 환자의 적혈구막 지질과산화 정도와 적혈구 평균 용적 및 간 효소 지표와의 관련성 (The Relationship between Lipid Peroxidation of Red Blood Cell Membrane, and Mean Corpuscular Volume and Liver Enzyme Markers in Alcohol Dependence Patients)

  • Objectives : Alcohol-induced oxidative stress has been known to injure various tissues or organs. This stress is related with free radicals which are produced as the result of long-term alcohol consumption. Malonyldialdehyde(MDA) is produced by the interaction of free radicals and cell membrane lipids, and indicates the degree of lipid peroxidation indirectly. The purpose of this study was to investigate the relationship between red blood cell(RBC) membrane lipid peroxidation by free radicals, and associated hepatic injuries and hematologic changes. Methods : Thirty-three subjects diagnosed as alcohol dependence according to DSM-IV diagnostic criteria were evaluated within 72 hours after discontinuing alcohol drinking. Clinical characteristics were evaluated by CAGE questionnaire and Korean Michigan Alcoholism Screening Test(MAST). RBC membrane MDA level was measured as the marker of RBC membrane lipid peroxidation. Aspartate aminotransferase(AST), alanine aminotransferase(ALT) and gamma-glutamyltransferase(GGT) were used as the biochemical markers of liver damage due to alcohol ingestion. The alcohol-induced hematologic change was assessed by mean corpuscular volume(MCV). Results : The results were as follows. Clinical characteristics were not different between two groups having normal and abnormal levels of AST, ALT, GGT or MCV. The levels of MDA were not correlated with the clinical characteristics and serum levels of AST, ALT and GGT. However, there was a significant correlation between the levels of MDA and the value of MCV(p=0.017). Conclusions : These findings suggest that oxidative stress in alcohol dependence may not be reflected in liver enzyme markers such as AST, ALT and GGT, but may be reflected in MCV.

작약 약침액이 tert-butyl hydroperoxide 로 유도된 흰쥐 배양 간세포의 지질과산화반응 및 항산화효소 활성에 미치는 영향 (Effects of Paeoniae Radix Aqua-Acupuncture Solution on Tert-Butyl Hydroperoxide Induced Lipid Peroxidation and Antioxidative Enzymes in Cultured Rat Liver Cells)

  • Objectives : This study was purposed to investigate the antioxidative effects of Paeoniae radix aqua-acupuncture solution(PR) on culture liver cell system, lipid peroxidation and antioxidative enzyme activities in tert-butyl hydroperoxide(t-BHP) treatmented conditions. Methods : Cultured normal rat liver cell(Ac2F) were prepared and incubated with or without PR(at 2% volume in culture medium). After 16~18hr, cells placed in DMEM medium without serum, and then incubated with 1mM t-BHP for 2hr. Viable cells were detected by MTT assay, and the levels of lipid peroxide(LPO) were measured by TBA method. And catalase activity was measured as the decrease in hydrogen peroxide absorbance at 240nm on spectrophotometer using 30mM hydrogen peroxide. Superoxide dismutase(SOD) were assayed by recording the inhibition of nitro blue tetrazolium reduction with xanthine and xanthine oxidase. Glutathione peroxidase(GPX) activity was determined by the modified coupled assay developed by Paglia and Lawrence. The reaction was started by addition of 2.2mM hydrogen peroxide as substrate. The change in absorbance at 340nm was measured for 1min on spectrophotometer. Glutathione-S-transferase(GST) activity was assayed with CDNB as substrate and enzyme activity of GST towards the glutathione conjugation of CDNB. Results : Cell killing was significantly enhanced by addition of t-BHP compared to those of untreated group. PR pretreated cell resisted the toxic effects of t-BHP. LPO levels of t-BHP treatment group were significantly higher than other groups. This increased level was significandy reduced by PR pretreatment. The t-BHP treatment resulted in a decrease of catalase, GPX and GST activities. By contrast, PR pretreatment markedly increased compare to those of untreated groups. Conclusions : T-BHP which can produce intracellular free radical was used for inducer of the peroxidation of cellular lipids. PR protected the cell death induced by t-BHP and significantly increased cell viabiliry in the normal rat liver cell, and showed effective inhibition of lipid peroxidation, and elevations of catalase, GPX and GST activities. These results suggested that PR might play a protective role in lipid peroxidation by free radicals.

간세포 배양에서 Linoleic Acid와 혈청알부민의 첩가가 지질대사에 미치는 영향 (Effects of Linoleic Acid and Serum Albumin Concentrations on Lipid Metabolism in HepG2 Cells)

  • 사람 간세포 유래의 배양 세포인 HepG2 세포의 지질 합성과 분비에 미치는 linoleic acid(LA, 18 : 2 n-6) 및 우혈청알부민 (bovine serum albumin: BSA) 첨가 농도의 영향에 대하여 검토하였다. 간세포는 DMEM배지(기본 배지)에 0.2 mM LA을 첨가한 배지 및 LA와 BSA(0.2-1.0%)를 첨가한 배지(LA+BSA 배지)에서 배양하였다. 각 지질의 동위원소 표적에는 $[^{14}C]acetate$를 이용하여 6시간 배양후의 지질합성과 분비를 측정하였다. 그 결과, 기본 배지중에 LA의 첨가는 콜레스테롤의 $[^{14}C]acetate$ 표적량을 저하 시켰다. 한편, LA배지에 BSA의 첨가에 의해, 총 콜레스테롤의 $[^{14}C]acetate$ 표적량은 증가하는 경향을 나타내었다. LA+BSA 배지에서 간세포의 총 콜레스테롤 변동은 유리형 콜레스테롤에의 표적량 증가에 기인하는 것으로 나타났다. LA배지에 BSA를 첨가 하였을때 콜레스테롤 분비가 증가 하였는데, 이는 지질의 분비과정에 BSA가 관여하는 것을 시사하는 것이다. 간세포내 총 지질에의 $[^{14}C]acetate$ 표적량은 각 군간의 유의치는 인정되지 않았다. 그러나, LA 배지에의 BSA 첨가는 $[^{14}C]acetate$표식 지방산의 중성지질 획분에의 표적량은 증가하고, 인지질 획분에 표적량은 감소하여, 양지질의 합성에는 상반되는 결과를 나타내었다. $[^{14}C]acetate$ 표적 중성지질, 인지질 및 유리지방산의 분비는 LA배지에 비교해 LA+BSA 배지에서 현저하게 증가하였다. 이상의 결과에서, 사람 간배양 세포에서 LA와 BSA는 각각 지질대사에 다른 영향을 미치고, BSA 농도는 리포단백질 분비에 영향을 미치는 것으로 시사된다.

흑마늘과 쑥 추출물이 구속스트레스를 가한 흰쥐의 체내 지질 성분에 미치는 영향 (Effect of Black Garlic and Mugwort Extracts on Lipids Profile during Restraint Stress)

  • In vitro에서 흑마늘과 쑥 추출분말 및 이들의 비율별 혼합물 3종(MPA, 95:5; MPB, 90:10; MPC, 85:15, w/w%)의 생리활성을 비교하였다. 또한 7 brix 흑마늘 추출물(BE), 0.7 brix 쑥 추출물(ME), 이들을 부피비로 95:5(MA), 90:10(MB) 및 90.25:4.75 혼합물에 사과 추출물과 자일리톨을 각각 2%와 3%를 첨가한 음료(MC)를 흰쥐에게 4주간 급이한 후 24시간 구속스트레스를 부과하여 생체내 지질 성분 변화에 미치는 영향을 평가하였다. In vitro에서 흑마늘 추출물과 쑥 추출물의 혼합물(MPA, MPB, MPC)은 쑥 추출물의 첨가량이 증가됨에 따라 DPPH 및 ABTS 라디칼 소거활성이 유의적으로 증가하였다. 흑마늘 추출물(RS1), 쑥 추출물(RS2), 혼합물 MA(RS3), MB(RS4) 및 MC(RS5)를 각각 1%씩 흰쥐의 식이에 첨가 급이한 결과 식이섭취량은 RS1군에서 가장 높았으며, 체중 증가량과 식이효율은 실험군 간에 유의차가 없었다. 혈청의 총 콜레스테롤의 함량은 구속 대조군(RSC군)에 비해 모든 실험군에서 유의적으로 낮은 함량이었으며, RS3~RS5군 간에는 유의차를 보이지 않았다. 혈중 중성지질의 함량은 RS3~RS5군이 RS1과 RS2군보다 유의적으로 높은 수준이었다. HDL-C 및 LDL-C 함량, 동맥경 화지수(AI), 심혈관질환 위험지수(CRF)는 RS3~RS5군 간에 유의차가 없었다. AST 및 ALP 활성은 RSC군에 비해 실험군들에서 유의적으로 낮았다. 간 조직의 총 지질과 총 콜레스테롤 함량은 RSC군에 비해 모든 실험군에서 유의적으로 낮았고, 중성지방 함량은 RS1군만이 유의적으로 낮은 함량이었다. 간 글리코겐 함량은 RSC군에 비해 RS2와 RS3군이 유의적으로 높았고, 그 외 실험군에서는 유의차가 적었다. 지질과산화물 함량은 RSC군에 비해 실험군에서 유의적으로 낮았으며, RS3과 RS5군이 유사한 수준이었다. 본 연구결과에서 흑마늘 추출물, 쑥 추출물, 혼합물 및 개발 음료의 급이는 스트레스 부과로 증가될 수 있는 체내 지질 수준의 감소에 효과적이었다. 또한 각 물질의 단독 급이보다 혼합물의 급이시 더 효과적인 것으로 나타나 혼합물을 이용한 개발음료는 현대인의 복잡하고 다양한 스트레스 발생 시 생체대사 조절에 도움이 될 것으로 사료된다.

Alcohol과 쌀 곰팡이의 Toxicity가 간(肝)의 지방(脂肪) 축적(蓄積)에 미치는 영향(影響) (Effects of Alcohol Intoxication and Moldy Rice Consumption on the Liver Lipid Content in Rat)

  • For the elucidation of the metabolic effects due to alcoholic drink and moldy rice intake author designed two experiments as follows. In the first experiment, sixty male and female rats, divided into six groups, 10 rats each in both sexes, were given 7.5% sucrose-30% alcohol solution (Jinro-Soju) and diets varied of fat content (2%, 7%, 30% of diet) and protein quality (casein, bean, anchovy) for 6 weeks. In the Second experiment, six groups of rats, 10 male rats each, were fed the diets containing zero, 5%, 10%, 25%, 50%, and 100% moldy rice contaminated Penicillium Islandicum and the experimental periods were 4 weeks and 6 weeks. The results of these studies due to the toxicity of alcohol consumption and moldy rice ingestion of the fatty liver production and the other effects are as follows. 1. Food intake of alcohol consumed groups decreased to 50%-70% of that of standard group. In the second study, there was no significant differences on food intake due to the different contents of moldy rice and experimental period. 2. On the view paint of body weight gain, the body weight of alcohol consumed groups gained much less than that of standard group, because food consumption was decreased due to alcohol ingestion. 3. In comparison of F.E.R. and P.E.R. between standard and experimental groups, there was no significant differences in this study. 4. As a matter of fact, there was no significance on the nitrogen balance in both studies. 5. From the result of hematology, R.B.C. counts, W.B.C. counts, hemoglobin, and hematocrit showed normal values in all groups including in this study. So we might conclude that the toxicity of alcohol and moldy rice do not effect significantly on blood picture. 6. The larger organs shrank on the range of 20%-70% of that against standard group in this study. The major reason for the shrinkage of organs might be account of decreased food consumption due to alcoholic drink. There was no great changes on the organ weight due to the ingestion of moldy rice. 7. The nitrogen content in various organs in both experiments was revealed at the normal level for the worst condition in terms of the least food intake. In other word, it was noteworthy that the concentration of nitrogen in various organs was kept at the normal ratio as standard groups under the circumstances of this study. 8. The lipid content in the liver of rats fed alcohol and diets containing either various lipid contents or protein quality did not increase. Hepatic lipids accumulation due to the dietary fat content was observed, but there wvas no significances among the compared groups. In the second experiment, the difference of hepatic lipid content between the moldy rice groups and standard group was not showed. In addition to the result of total lipid, hepatic free cholesterol, free fatty acid, and triglyceride did not change in both studies, we might conclude that the toxicity of alcohol and moldy rice could not effect on the hepatic lipid contents. 9. There was no significant differences on the serum glucose level between alcohol groups and standard group.
