재발성 삼출성 중이염과 혈청 면역글로불린 농도의 저하 (Decreased Serum Immunoglobulin in Recurrent Otitis Media with Effusion)

  • Background: Defective or immature antibody responses to pathogens in children may explain the increased susceptibility to acute otitis media. However, there is no study in Korea patients whether a correlation exists between otitis media with effusion and the levels of serum immunoglobulins, IgG subclasses, IgA, IgM and IgE. Methods: 45 children with otitis media with effusion more than 4 episodes in 12 months or 3 episodes in 6 months, 62 children with otitis media with effusion less than 3 episodes in 12 months and 102 children for control group took part in the study at the Department of Otorhinolaryngology of the KyungHee University from May 2004 to Feburary 2007. Serum immunoglobulin levels were determined by nephelometry. And then the relationship between otitis media with effusion and serum immunoglobulin level was evaluated. Results: In otitis media prone group, serum IgG1, IgG2, IgG4, and IgA level was lower than those level of control group, it was significantly decreased (p<0.05). In otitis media group, serum IgA, IgE, and IgG4 level was lower than those level of control. But it was not statistically significant (p>0.05). Conclusion: Lower immunoglobulins in children with otitis media with effusion suggest a generalized decreased antibody responses. Lower levels of serum IgG1, IgG2, IgG4, and IgA may be related with chronicity or intractability of otitis media with effusion.

Chamomile German 오일도포가 아토피성 피부염을 가진 NC/Nga 생쥐모델의 혈청 IgE와 IgG1양 변동에 미치는 영향 (Effect of Chamomile German Oil Application of IgG1 and IgE 1 to Atopic Dermatitis in NC/Nga Mice Model)

  • Atopic dermatitis is one of the most common skin disease in children and a characteristic chronically recurrent form of dermatitis with a hereditary tendency, affecting infants and may extend to the childhood or th the adult age. Environmental factors, stress, and foodstuffs such as milk, egg may cause atopic dermatitis. Nc/Nga mice were used as an animal model for human atopic dermatitis. The divided by 4group such as normal group, BMAC group, FK506 group, MR group for this study raised in conventional conditions. To investigate effect of Chamomile German on atopic dermatitis in NC/Nga mice, the serum IgE and IgG1 level were measured while the severity degree of the skin lesion was examined by the naked eyes of two volunteers who were unaware of the treatment status. The results were the followings. 1. The score on the severity degree of skin dermatitis in FK 506 and MR group was lower than that in control group. 2. The serum IgE level in control group was higher 25% than that in normal group. 3. The serum level of IgE in FK506 and MR group compares to control group was decreased. 4. The serum IgG1 level was decreased more than 3.5 times in FK506 compared to control group while MR group had significantly less the serum IgG1 than control group. From the above results, treatment of Chamomile German oil had the effect on atopic dermatitis in NC/Nga mice. If scientific researches on aroma oil are performed in various way, aroma oil will be used to cure skin dermatitis as a alternative therapy in the future.

  • Numerous studies have been focused on the immunologic aspects of inflamed pulp and periapical tissues. The purpose of this study was to evaluate levels of serum IgG and IgM in patients of acute pulpitis and acute apical abscess using Enzyme-Linked Immunosorbent Assay. Streptococcus mutans, Streptococcus sanguis, Bacteroides intermedius and Bacteroides gingivalis were grown for use as antigen and they were harvested by centrifugation. The patients were divided into 3 groups; patients of acute apical abecess, acute pulpitis and normal control 5 patients of each group were selected and their blood was obtained via intravenous puncture. Sera were prepared by centrifugation of each blood samples. Then serum antibodies were measured by modified ELISA. The following results were obtained; 1. Serum IgM levels of patients with acute pulpitis and acute apical abscess seemed to be slightly higher than those of normal control 2. Serum IgG levels of patients with acute apical abscess were slightly higher than those of normal control 3. Serum IgG and IgM levels of acute apical abscess patients and serum IgM levels of acute pulpitis were highest to Bacteroides gingivalis.

육용계의 혈청중 면역글로불린(IgA, IgG, IgM) 농도의 발육시기별 변화상 I. 혈청 IgG의 분리 및 발육시기별 농도 수준 (Developmental Changes of Serum IgA, IgG and IgM Concentrations in Broiler Chicks 1. Isolation of IgG and Developmental Changes of Serum IgG Levels)

  • 본 실험은 닭의 혈청으로부터 IgG를 순수분리하고 이에 대한 항혈청을 대량 생산하여 IgG의 양적 측정에 필요한 물질의 생산을 자체 정립하고 아울러 육용계의 전사육기간 동안의 혈청중 IgG농도의 변화상을 조사하여 닭의 면역반응에 대한 기초적인 자료를 제공하고자 실시하였다. 1. 닭의 혈청으로부터 순수한 IgG를 분리하였으며 이를 항원으로 면역시켜 IgG에서만 특이적으로 반응하는 항혈청을 생산하였다. 2. 표준항원(IgG)에 대한 항체(anti-IgG)와의 diffusion상태를 측정하여 표준곡선을 작성한 바, diffusion소요시간이 16시간에서 가장 양흐한 회귀직선방정식을 얻었다($R^2$=0.999). 3. 혈청중 IgG의 농도의 변화상은 1일령에서(4.25mg/$m\ell$) 가장 높았으며 그 이후 급격히 감소하여 11일령에 최저수준(0.81mg/$m\ell$ ; 1일령의 19%수준)에 도달하였다. 그 이후에는 서서히 증가하여 7주령시에는 2.48 mg/$m\ell$의 수준을 보였다. 4. 성별에 따른 IgG 농도간에는 유의적인 차이는 없었으며 3주령까지의 IgG의 농도는 수컷에 비하여 암컷이 약간 높은 수준을 보였다. 한편 수컷은 11일령에서 최저 수준에 도달한 반면에 암컷은 14일령에 가장 낮은 수준에 도달하여 수컷의 능동면역 체계가 암컷보다 조기에 발달되는 것으로 추정된다.

Usefulness of IgG4 subclass antibodies for diagnosis of human clonorchiasis

  • The present study analyzed serum IgG subclass antibody reaction to major antigenic bands of Clonorchis sinensis to investigate improvement of its serodiagnosis. Of the four subclass antibodies, IgG1 and IgG2 antibodies were produced but not specific, IgG3 antibody was least produced, and IgG4 antibody was prominent and specific. The serum IgG antibody reaction to any of 43-50, 34-37, 26-28, and 8 kDa bands was found in 65.5% of 168 egg positive cases while IgG4 antibody reaction was found in 22.0% of them. The positive rates of IgG and IgG4 antibodies were directly correlated with the intensity of infection. All of the sera from heavily infected cases over EPG 5,000 showed positive reaction for specific IgG and IgG4 antibodies. The specific serum IgG4 antibody disappeared within 6 months after treatment. The bands of 35 kDa and 67 kDa cross-reacted with IgG antibodies but not with IgG4 antibodies in sera of other trematode infections. The present findings suggest that serum IgG4 antibody reaction to 8 kDa band is specific but not sensitive. Any method to increase its sensitivity is required for improved serodiagnosis.

  • An experiment was conducted to establish a large scale production method of anti-serum against chicken IgA and to profile the developmental changes of serum IgA levels during the feeding period(from hatching to 7 weeks of age) in broiler chicks. Blood samples were taken from Hubbard chicken at the age of hatching, 3 days of age, and weekly thereafter till to 7 weeks of age. The pure IgA was isolated from ammonium sulfate treated chicken bile juice by gel filtration chromatography ( Sepharose GL-6B) - The quantitative assay of serum IgA were carried by RID method. Developmental changes of serum IgA concentrations were 0.42 mg /mL at hatching, thereafter dicreased gradually, lowest at 1 week of age(0.17 mg /mL), and gradually increased to 7 weeks of age(2.73 mg /mL). There was no sexual difference in serum IgA level, but female chicks showed higher IgA levels than male chicks during the experimental period.

Evaluation of serum immunoglobulin G4 concentrations in canine pancreatitis

  • The goal of this study was to measure immunoglobulin G4 (IgG4) concentrations and to evaluate the significance of these values in the management of canine pancreatitis. The medical records of 24 dogs that visited the Veterinary Medical Teaching Hospital between December 2016 and June 2018 were retrospectively reviewed to identify dogs that had been diagnosed with pancreatitis. The serum C-reactive protein and serum IgG4 concentration in the affected dogs were highly increased compared to the healthy group. Particularly, serum IgG4 measured significantly higher in dogs with pancreatitis and concurrent immune-mediated disease (p < 0.05). In conclusion, increased serum IgG4 concentrations are a characteristic finding in dogs with pancreatitis. The results of this research indicate that an elevation in IgG4 has the potential of being used as a tool for the diagnosis of pancreatitis and concurrent immune-mediated disease.

질트리코모나스 환자에서 효소표식 면역검사법을 이용한 혈청 내 항-질트리코모나스 IgG 및 IgM 항체가의 측정 (Detection of IgG and IgM antibodies with ELISA technique in human trichomoniasis)

  • 질트리코모나스(Trichomonas vaginalis)감염 환자 30명과 비감염 건강 대조군 30명을 패 상으로 혈청 내 항-질트리코모나스 IgG 및 IgM 항체가를 효소표식 면역검사법(enzyme-linked immunosorbent assay: ELISA)으로 측정하여 이 질환 진단법으로서 ELISA의 이용 가능성을 검토 하고, 현펑 내 IgG 및 IsM 항체가의 상관성을 알아보았다. 질트리코모나스 환자의 혈청 내 IgG 항체가는 0.37±0.134로 건강 대조군의 0.21±0.054 보다 유 의하게 높았다(p<0.005). 또 혈청 내 IgG 항체가 0.32 이상을 양성으로 하였을 때 ELISA 진단법 의 감수성은 70.0%이었고 특이성은 96.7%이었다. 질트리코모나스 환자의 혈청 내 IsM 항체가는 0.33±0.177로 건강 대조군의 0.11±0.051 보다 유의하게 높았다(p<0.005). 또 혈청 내 Is변 항체 가 0.21 이상을 양성 반응으로 하였을 때 감수성은 70.0%이었고 특이성은 96.7%이었다. 질트리코 모나스 환자의 혈청 내 IgG 와 IsM 항체가의 상관 관계는 상관계수 r=0.77로 유의한 상관관계가 인 정 되 었 다(p<0.005). 이상의 결과로 보아 질트리코모나스 환자에서 혈청 내 19G 및 IsM 항체가를 ELISA법으로 측정 함으로써 질트리코모나스 감염을 진단하거나 역학적 조사에 이용할 수 있을 것으로 생각된다.

흰쥐에서 식이 단백질 수준이 수유를 통한 수동면역에 미치는 영향 (Effect of Dietary Protein Level on Immune Substances in Milk and its Transfer to Pups in Rats)

  • This study was performed to determine the effect of maternal protein intake on 1) the concentration of immune substances in milk 2) degree of passive immunity to pups via lactation, and 3) specific antibody production to a specific antigen, $\beta$-lactoglobulin(BLG). 4) the effect of passive immunity that pups received from mother during lactation on the production of antibodies when the pups were challenged to the same antigen. Part of the female rats were immunized with BLG before and during pregnancy. The pregnant rats were placed into either 25% or 10% isolated soy protein diet throughout gestation and lactation. After weaning, pups from each group continued to be fed the same diet. At 18 weeks of age, all the pups were challenged with BLG. Total IgA and IgG, lysozyme, BLG-specific IgA and IgG were measured in dam's serum, dam's milk, and pup's serum. Total IgG, and lysozyme in dam's serum and milk were higher in high protein group. Total IgA and IgG in pup's serum remained higher in high protein group from 5 to 18 weeks of age. BLG-specific antibodies were found in the milk and serum of immunized dams, and in serum of pups born to immunized dams but not in the non-immunized group. BLG-specific IgA and IgG were again higher in high protein group and declined with time. The concentration decreased faster in the low proetein group than in the high protein groups. After immunization the pups with LBG, serum BLG-specific antibodies were not differ between rats born to immunized dams and those born to non-immunized dams. Therefore passive immunity rats received via milk as a pup had no effect on the BLG-specific antibody production later in life. This study shows the importance of protein status of mother and strongly support to the endorsement of breast feeding.

Effect of Leptin and IGFBP-3 Gene Polymorphisms on Serum IgG Level of Cattle Calves

  • Leptin and IGFBP-3 are two proteins that play an important role in growth and metabolism of the animals. They are also involved in the immune function of animals and, thus, are candidate genes for the study of association with immune functions. Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) of these two genes was done to screen 64 crossbred (Holstein Friesian${\times}$Hariana) female calves of one year of age. From each RFLPs (fragments) three genotypes were observed. In all the RFLPs the mutant homozygotes were very less in numbers and, hence, were excluded from the least squares analysis. The serum IgG level was estimated using SRID assay. The mean level of serum IgG was $28.83{\pm}2.73mg/ml$. The effect of these identified genotypes on serum IgG level of calves at one year of age was analysed using least squares analysis. The HaeIII RFLP-AB genotype had significantly (p<0.05) higher serum IgG level ($31.86{\pm}3.05$) than the HaeIII RFLP-AA ($25.62{\pm}2.96$) genotype. There was no significant effect of leptin genotypes on the IgG level. The present results indicated a role of the IGFBP-3 gene on serum IgG level of cattle calves.
