Characteristics and Pathogenicity for Japnaes Eel Anguilla japonica of Vibrio vulnificus Isolated from Oyster, Sediment and Seawater in the Korea Coast

  • Biotyping of Vibrio vulnificus strains isolated from marine environments along the south coast of Korea showed that the majority of the isolates (94.7%) belonged to biotype 1 and the remaining isolates (5.3%) belonged to biotype 2. Analysis of 16S rRNA V. vulnificus strains isolated from marine environments using a multiplex polymerase chain reaction (PCR) revealed that 78.7% were type A and 21.3% were type B. Random amplified polymorphic DNA (RAPD) was used to analyze the genomic differences in V. vulnificus among the biotype 2 strains isolated from marine environments (newly isolated strains group) and reference strains obtained from infected eels (reference strains group). The two groups had distinctly different profiles of the amplicons produced from RAPD. Additionally, biochemical comparison of these strains revealed that all four strains isolated from marine environments differed from the strains isolated from eels in their ability to promote D-mannitol fermentation. Two (NH 1 and NH 2) out of four isolates of biotype 2 from marine environments showed pathogenicity in eels Anguilla japonica in a challenge test. These isolates did not agglutinate with antisera against V. vulnificus NCIMB 2137 (serovar E), ATCC 27562 (non-serovar E), and ATCC 33816 (atypical serovar E).

Protective effects of mix-crude outer membrane protein Salmonella vaccine against salmonellosis in chickens and pigs (살모넬라 세포외막단백질 혼합백신을 이용한 돼지 및 닭에서의 살모넬라균감염증 방어효과)

  • We investigated the safety, immunogenicity and protectivity of mix-crude outer membrane protein (cOMP) vaccine against salmonellosis in animals. The mix-cOMP vaccine was extracted from Salmonella enterica serovar Typhimurium (ST) and Salmonella enterica serovar Enteritidis (SE) and Salmonella enterica serovar Braenderup (SB) isolated from pigs. The mix-cOMP vaccine gave significantly higher antibody response than ST-bacterin and ST-cOMP vaccine in guinea pigs. The survival rates of mix-cOMP vaccinated groups showed significantly higher (100%) than those (0-20%) of unvaccinated control group, challenged with 3 species of Salmonella (ST, SE and SB) in mice. Vaccinated groups in pigs showed reduction of clinical signs, increase of average weight gains, decrease of bacterial recovery rates, compared with unvaccinated groups. Especially, the survival rates (100%) of vaccinated groups in chickens showed higher than that (0%) of unvaccinated group. Based on these results, we suggest that the mix-cOMP Salmonella vaccine developed in this study will be effective for the protection against Salmonellosis caused by the various serotypes Salmonella species in animals.

  • The lethality to Pieris raepi larvae and the safety tests to mice of the endotoxin crystals and the pesticide of B thuringiensis serovar kurstaki were carried out. The LD50 of the endotoxin (3.9$\times$10$^9$ spores/$m{\ell}$) appeared to be 1$\mu$g and that of the pesticide (2.6$\times$10$^9$ spores/$m{\ell}$) was about 9 $\mu$g. When the endotoxin solution and the pesticide were injected to the mice's intraperitoneal cavities, 10 to 20% of the mite were dead. Also in the case of intracerebral injection 20% of the mice were dead at the doses of 7.8$\times$10$^3$ spores and 100% of the mice were dead at the concentration of 7.8$\times$10$^8$ spores. but the mice in the oral, subcutaneous and inhalation treatment groups were safe and healthy. When the pesticide applied to the cabbage field for raepi larvae, 52 to 65% of the larvae in the field were killed in 4 days postapplication.

  • Bacillus thuringiensis serovar israelensis 4Q1 contains 8 different covalently closed-circular (CCC) plasmids of molecular weight 204, 267, 109, 103, 16, 7.6, 6.4, and 5.0kb. The three smallest plasmids, designated pBti6, and pBti8 may prove to be useful as cloning vectors because of thier size and ease of isolation. The three plasmids were incubated separately with 9 different restriction enzymes and 7 of the enzymes tested cleaved one or more of the plasmids. Plasmid pBti6 has a single site for Bg1 II, Pst I and Pvu II, two sites for Bc1 I and Eco RI, and five sites for Hind III. Plasmid pBti7 has a single site for Bam HI and Pst I, two sites for Hind III, and three sites for PvuII. Plasmic pBti8 has a single site for Bam HI, BelI and Hind III, two sites for Eco RI, and three sites for Bgl II and Pvu II. Composite restriction enzyme maps for pBti6, pBti7 and pBti8 were constructed. The sites of restriction enzyme cleavage were determined by single, double and partial digests of the plasmid DNA. All the restriction sites were aligned relative to the single Bgl II(pBti6), Pst I(pBti7) or Hind III(pBti8) site, respectively.

Serovars distribution and antimicrobial resistance patterns of Salmonella spp. isolated from the swine farms and slaughter houses

  • Salmonella spp. is an important pathogen to both public and swine industry. The aim of this study was to investigate the distribution of Salmonella serovar and antibiotics susceptibility of the isolates from Korean swine producing systems. A total of 63 (5.28%) Salmonella spp. was isolated from 1,194 samples (977 fecal materials and 67 organ samples). The predominant Salmonella (S.) enterica serotype and serovar was group B (69.8%) and S. Typhimurium (47.6%), S. Derby (20.6%) and S. Heidelberg (1.6%). But S. Cholerasuis which is characterized host specific by septicemia and enteritis to pigs was not isolated. Antimicrobial susceptibility of the isolates varies as follows: Norfloxacine (75%), Ciprofloxacin (67.5%), Amikacin (60%), Colistin (60%), Enrofloxacin (55%). All of isolates were resistant to Erythromycin, Penicillin, Tetracycline and Lincomycin. The results of this study provided useful information regarding antimicrobial susceptibility and resistance patterns to treat salmonellosis and to prevent emergence of multidrug resistance Salmonella.

Antimutagenic Effects against N-methyl-N`-nitro-N-nitrosoguandine and 4-nitroquinoline-1-oxide on Cultrue Conditions of Leuconostoc mesenteroides subsp. cremoris DLAB19 isolated from Dongchimi (동치미에서 분리한 Leuconostoc mesenteroides subsp. cremoris DLAB19의 배양 조건에 따른 N-methyl-N`-nitro-N-nitrosoguandine과 4-nitroquinoline-1-oxide에 대한 항돌연변이 효과)

  • Leuconostoc mesenteroides subsp. cremoris DLAB19 were investgated under various culture conditions to maximize the production of antimutagenic substance(s) against N-methyl-N\`-nitro-N-nitrosoguandine(MNNG) on Salmonella enterica serovar typhimurium TA100 and 4-nitroquinoline-1-oxide(4-NQO) on S. enterica serovar typhimurium TA98. The MRS medium containing glucose (2%) as a carbon source and yeasty extract (1%) as a nitrogen source resulted in the highest production of the antimutagenic substance(s) against both mutagens in the culture supernatant of Leu. mesenteroides subsp. cremoris DLAB19. Optimal pH of the culture medium, culture temperature and shaking speed for the antimutagenic substance(s) production were pH 7.0, 3$0^{\circ}C$ and 150 rpm, respectively. Under the optimal condition, the antimutagenic effects of Leu. mesenteroides subsp. cremoris DLAB19 culture supernatant were 96.4% against MNNG on S.enterica serovar typhimurium TA100 and 53.8% against 4-NQO on S. enterica serovar typhimurium TA98.

Structural Analysis of Class I Integron Gene Cassette and Assessment of Genetic Relationships by PFGE of Salmonella enterica Serovar Typhimurium Isolated in Gyeongbuk Area (경북지역에서 분리된 Salmonella enterica Serovar Typhimurium의 Class I Integron Gene Cassette 특성과 PEGE 유형분석)

  • Thirty five Salmonella enterica serovar Typhimurium strains were isolated from diarrheic patients and pigs in Gyeongbuk area from 2003 to 2004. All 35 strains (17 strains from diarrheic patients and 18 from pigs) were resistant to more than one drug and most of strains isolated from pigs were resistant to ampicillin, ohloram-phenicol, streptomycin, sulfamethoxazole-trimethoprime, tetracyclin and nalidixic acid. Each isolate was also screened or the presence of class I, II and III integron gene cassettes. Among 35 strains,3 out of 17 strains isolated from diarrheic patients, carried dhfrX-orfF-aadA2 integron gene cassette and among 18 strains isolated from diseased pigs, 11 strains carried dhfrX-orfF-aadA2 integron gene cassette and 1 strain carried aadA2 integron only. But any class II and class II integron gene cassette were not detected in 35 strains. Thirty five strains were divided by five pulsotypes. Thirty one strains out of thirty five were pulsotype A. Among the remaining 4 strains, one each strain belonged to pulsotype B, C, D and pulsotype E. This data of pulsotypes showed that the widespread of pulsotype A, Salmonella enterica serovar Typhimurium in human and pigs in Gyeongbuk area may have been caused by the dissemination of a few epidemic strains in this area. Thirteen strains contain dhfrX-orfF-aadA2 integron gene cassette showed pulsotype A and one strain contains dhfrX-orfF-aadA2 integron gene cassette showed pulsotype B. One strain contains aadA2 integron showed pulsotype E. But fifteen strains do not contain any integron showed pulsotype A.

Pulsed-Field Gel Electrophoresis and Monoclonal Antibody Analysis of Leptospira interrogans Isolated in Korea (국내 분리 렙토스피라균의 단클론 항체 및 Genomic DNA의 Pulsed-Field Gel Electrophoresis 분석)

  • A total of 22 Leptospiua inlermgans field isolates from the ~ a t s captured in 5 provinces of Korea in 1996, and 6 antigenically closely related relerence serovars of lai, yeonchon, birkini. gem, mwogolo. and canicola were analysed. When the antigenic characteristics were analysed by reactivity with 7 monoclonal antibodies prepared with sh.ains belongng to serogroup Icterohaemorrhagiae. all 22 isolates showed the same reaction pattern with that of serovar lai. Large restriction fragment patterns obtained after cleavage of geno~nic DNAs with infrequently cuttimg restriction enzymes were analyzed by pulsed-field pel electrophoresis(PFGE). Identification of leptospira strains by PFGE with Nor I, Asc I or Iise I digests correlated with their antigenically typed serovars, silh a few exceptions. PFGE of isolates, except for JR89, digested wjth Nor I showed identical pattern w~th serovar lai, showing 13 Cragments between 940 kb and 63 kb. When PFGE pallerns of JR89 were compared with those of serovar lai, Not I digest showed additional two hands of 1000 kb and 460 kb, while Asc I digest showed 650 kb fragment and Fse I digest did not show the fragment of 280 kb. Whereas serovar yeonchon. which was isolated in Korea and identified as a new serovar previously. could be differentiated from serovar lai in antigenic reactivities with monoclonal antibodres. it showed the similar PFGE pattern with serovar lai includin~ reference and field isolates. It was suggested that Korean leptospiral field isolates are closely related in DNA level.

Characterization of Multidrug-resistant Salmonella enterica Serovar Typhimurium Isolated from Swine Sources

  • A total of 28 Salmonella enterica serovar Typhimurium isolated from diseased pigs and swine carcasses between 2001 and 2003 were characterized by the antimicrobial resistance profiles, PCR for detection of S. Typhimurium DT104 and pulsed-field gel electrophoresis (PFGE) with the restriction enzyme XbaI. All but one isolate presented multidrug resistance (MDR) to more than two antibiotics tested. A total of 11 resistance profiles were observed, and two phenotypes, ST and ASSuTG, were the most common among them. Two isolates were found to be S. Typhimurium DT104 isolates by PCR, and their resistance profile did not show the DT104 typical resistance type ACSSuT, but ACSSuTGK instead. PFGE identified 11 banding patterns in dendrogram, and three main clusters (designated A to C) were represented. Interestingly, sixteen of 19S. Typhimurium isolates belonging to cluster B showed an identical band pattern.

  • Crystals of six new Bacillus thuringiensis serovarieties [coreanensis (H25), konkukian (H34), leesis (H33), seoulensis (H35), sooncheon (H41), and yosoo (HI8a18c)] with different H-antigens, which are toxic to Bombyx mori and/or mosquito larvae, were serologically quite distinct from each other.
