Actinobacillus actinomycetemcomitans의 혈청형별 제한절편장 다변화에 관한 연구 (DNA fingerprinting patterns of 5 serotypes of Actinobacillus actinomycetemcomitans)

  • 5 serotypes(a, b, c, d, e) of Actinobacillus actinomycetemcomitans showed distinct hybridization patterns(DNA fingerprinting patterns) when the bacterial DNA were hybridized with randomly cloned 4.7-Kb sized DNA probe. The sizes of hybridized bands in each serotypes were different among serotypes and represented unique patterns of hybridization with the probe used. The serotype a showed two bands of fingerprinting patterns: 23.1 kb and 2.5 kb respectively. Serotype b and c showed single band: 6.6 kb and 9.5 kb, respectively. Serotype d and e showed two bands of hybridization: 23.1 kb and 2.8 kb, and 23.7 kb and 2.1 kb, respectively. The results indicate that this standard fingerpriting patterns of DNA hybridization with 4.7 kb probe can be further used for genotyping clinical isolates of Actinobacillus 8ctinomycetemcomitansand its relevance with periodontal disease activity.

  • Colistin is one of the last-resort antibiotics used to treat multidrug-resistant Gram-negative bacterial infection in both human and animals. Here, we report the complete genome sequence of colistin-resistant Salmonella enterica serotype Enteritidis strain CRSE-01 isolated from poultry carcass in South Korea. The assembled genome consists of a 4,783,907-bp circular chromosome containing numerous antimicrobial resistance genes and a 59,372-bp plasmid.

  • Clostridium perfringens A형이 생산하는 장독소를 검색해본 결과, RPLA법에 있어서는 2배로 희석한 용액으로부터 64배로 희석한 용액 (NCTC 8239 Hobbs serotype 3 CPE$^+$)에서까지 양성반응을 보였으며 PCR 기법에 있어서는 10 pg 희석 용액까지 364 bp의 장독소 DNA fragment(NCTC 8238 Hobbs serotype 2 CPE$^+$)를 확인 할수 있었다. 그러므로 장독소를 검색하기 위해서는 PCR기법이 RPLA법에 비하여 훨씬 감도가 높음을 확인할 수 있었다.

  • 폐구균(Streptococcus pneumoniae)은 소아, 특히 어린 영아에서 심한 침습성 질병을 일으키는 중요한 원인균이다. 2003년부터 우리나라에서 접종하기 시작한 7가 폐구균백신(PCV7)은 소아에서 침습성 폐구균성 폐렴과 수막염을 예방하는 것으로 알려져 왔다. 그러나 최근 7가 폐구균백신을 접종한 이후부터 비7가 폐구균 혈청형(non-PCV7 serotype) 감염 발생률이 증가하고 있다. 14개월 된 여아가 내원 3일 전부터 열과 구토가 있었으나 증상 치료만 하고 있던 중 갑작스런 의식불명과 기면(lethargy)으로 응급실로 내원하였다. 입원 후 즉각적인 진단과 치료를 시행했음에도 불구하고 환자는 빠르게 혼수와 뇌사 상태로 진행하였다. 환자는 이전에 3회의 7가 폐구균백신을 접종하였으나 최종 진단은 비7가 폐구균 혈청형인 폐구균 19A 수막염이었고, 입원 20일째 사망하였다. 이는 한국에서 문헌상 보고된 적이 없는, 폐구균 19A 혈청형 수막염으로 사망한 첫번째 증례이므로 이에 보고하는 바이다.

  • Previous studies have demonstrated that not all A. actinomycetemcomitans produced significant level of leukotoxic factor and its leukotoxicity have associated with serotype and genetic variation. Our aim was to investigate on the interrelationship between serotype and leukotoxicity of an A. actinomycetemcomitans consisting of 13 clinically well characterized. Korean isolates and to evaluate if particular virulent clonal types of A. actinomycetemcomitans are associated with periodontal disease. For this study, 13 strains of A. actinomycetemcomitans from 6 patients with periodontal disease were isolated and identified by using a selective medium(tryptic soy agar supplemented with 10% serum, $75{\mu}g$ of bacitracin and $5{\mu}g$ of vancomycin per ml) in 10% C02 incubator for 3days with routine Gram staining, colony morphology and biochemical test..For serotyping, antisera were prepared from reference strains of 5 serotypes. (ATCC 29523,Y4, SUNY aB 67, IDH 781, IDH 1705) and then ammonium sulfate precipitation, immunoabsorption and indirect immunofluoroscent procedures were done. For analysis of leukotoxicity, sonic extract of A. actinomycetemcomitans exposed to PMN, and trypan blue was stained for counting the cell viability. Finally Southern blot analyses of genomic DNA digested with the restriction enzyme Tag I was done and the Southern blots were hybridized with the 530bp fragment, termed delta 530, originating from the ltx promoter of strain 652 and deleted from strain JP2. Also ltxA-3.1 and SC2 probe from strain JP2 were hybridized with genomic DNA fragments. Results reveal that strains isolated showed approximately equal proportions of 3 serotypes(b, d, e) and serotype b was not detected. 2 patients harbored 2 different serotypes in the same disease site. The prevalence of leukotoxic strain was 23% and there was no relationship between serotype, leukotoxicity and clinical observations. Especially virulent clonal types of Actinobacillus actinomycetemcomitan (JP2 strain) could not found. Further studies are necessary on the genetic polymorphism of leukotoxin and its relations to clinical status.

제주지역 돼지 흉막폐렴에 대한 병리학적 연구 및 원인체의 혈청형 (Pathologic studies for porcine pleuropneumonia and serotypes of the agents in Jeju)

  • Actinobacillus (A.) pleuropneumoniae is the etiological agent of a porcine pleuropneumonia and have great economic importance to the global swine industry. For recent 5 years, a total of 50 pleuropneumonia cases of 24 pig farms were selected from pig lungs submitted to the College of Veterinary Medicine, Jeju National University using polymerase chain reaction (PCR) analysis. Collected lungs were fixed in 10% neutral phosphate-buffered formalin and processed for histological examination. Serotypes of A. pleuropneumoniae in pneumonic lesions were analyzed by PCR methods. And the antimicrobial susceptibility of A. pleuropneumoniae isolates was determined by a disc diffusion test. Grossly, unilateral distribution of hemorrhagic or necrotic pneumonic lesions was more common than bilateral distribution in lungs. In peracute or acute cases, histopathologic changes were characterized by necrosis, hemorrhage, neutrophils infiltration, vascular thrombosis, widespread edema and fibrinous exudates. Following the acute response, macrophage infiltration, marked fibrosis around zonal necrotic areas, and marked fibrous pleuritis were characteristic in chronic cases. A total of 50 pleuropneumonia were associated with A. pleuropneumoniae serotype 5 in 46 cases (92%), serotype 2 in 3 cases (6%), and both 2 and 5 in 1 case (2%). More than 90% of collected isolates showed high sensitivity to ceftiofur, amoxicillin, and colistin. However, ampicillin, penicillin, and tylosin showed low susceptibility. The results of this study demonstrated that A. pleuropneumoniae serotype 5 was predominant at porcine pleuropneumonia cases in Jeju.

Differential Gene Expression in the Pathogenic Strains of Actinobacillus pleuropneumoniae Serotypes 1 and 3

  • The limited information on differential gene expression in the different serotypes of Actinobacillus pleuropneumoniae has significantly hampered the research on the pathogenic mechanisms of this organism and the development of multivalent vaccines against A. pleuropneumoniae infection. To compare the gene expressions in the A. pleuropneumoniae strains CVCC259 (serotype 1) and CVCC261 (serotype 3), we screened the differentially expressed genes in the two strains by performing representational difference analysis (RDA). Northern blot analyses were used to confirm the results of RDA. We identified 22 differentially expressed genes in the CVCC259 strain and 20 differentially expressed genes in the CVCC261 strain, and these genes were classified into 11 groups: (1) genes encoding APX toxins; (2) genes encoding transferrin-binding protein; (3) genes involved in lipopolysaccharide (LPS) biosynthesis; (4) genes encoding autotransporter adhesin; (5) genes involved in metabolism; (6) genes involved in the ATP-binding cassette (ABC) transporter system; (7) genes encoding molecular chaperones; (8) genes involved in bacterial transcription and nucleic acid metabolism; (9) a gene encoding protease; (10) genes encoding lipoprotein/membrane protein; and (11) genes encoding various hypothetical proteins. This is the first report on the systematic application of RDA for the analysis of differential gene expression in A. pleuropneumoniae serotypes 1 and 3. The determination of these differentially expressed genes will serve as an indicator for future research on the pathogenic mechanisms of A. pleuropneumoniae and the development of a multivalent vaccine against A. pleuropneumoniae infection.

치근단 병소가 있는 환자에서 Porphyromonas endodontalis 항원에 대한 혈청 특이 항체의 면역 반응 연구 (IMMUNE REACTION OF SPECIFIC SERUM ANTIBODIES TO PORPHYROMONAS ENDODONTALIS ANTIGEN IN PATIENTS WITH PERIAPICAL LESION)

  • Porphyromonas endodontalis is a black-pigmented anaerobic Gram-negative rod which is associated with endodontal infections and this microorganism possesses a potential for pathogenicity. The purpose of this study was to compare the membrane components of Porphyromonas endodontalis and Porphyromonas gingivalis and to study the immune reaction patterns of Porphyromonas endodontalis with patients with periapical lesion. Porphyromonas endodontalis (ATCC 35406), Porphyromonas gingivals serotypea (381), serotype b(W50), serotype c(A7A1-28) were cultured in anaerobic condition. Rabbit antisera were prepared by intravenous injection of formalized whole cells and human sera were obtained from patients and dental students. Indirect immunofluorescence method was used to study on the cross reaction between Porphyromonas endodontalis and Porphyromonas gingivalis serotype a, b, c antigen. Total membrane protein profiles of Porphyromonas endodontalis antigen were studied by sodium dodecyl sulfate polyacrylamide gel electrophoresis and the reactivity of antigenic components of Porphyromonas endodontalis against sera of patients and rabbit anti-Porphyromonas endodontalis antisera were assessed by Immunoblotting method. The following results were obtained : 1. Antigens of Porphyromonas endodontalis has multiple antigenic components, and both patients with periapical lesion and normal healthy individual showed immune response to this. 2. Patients group and healthy individual group showed a diversity of immune reaction pattern but they showed immune response against 43kd protein. 3. Patients with periapical lesion showed more diverse immune response than healthy individual and in some patients, much more bands appeared to lower molecular weight protein. 4. According to indirect immunofluorescence and Immunoblotting study, Porphyromonas endodontalis did not share common antigen with Porphyromonas gingivalis serotype a, b, c.

15년간(1991~2005년) 우리나라 소아에서 분리된 폐구균의 혈청형 분포의 변화 (Changes of Serotype Distribution of Streptococcus pneumoniae Isolated from Children in Korea over a 15 Year-period(1991~2005))

  • 목 적 : 폐구균 단백 결합 백신이 개발되었지만 이에 포함되는 혈청형은 7가지로 한정되어 있어, 지역별 주요 폐구균 혈청형의 분포에 따라 백신의 유용성에 차이가 있을 수 있다. 본 연구에서는 우리나라 소아에서 분리된 폐구균의 혈청형의 분포를 분석하고 지난 15년간 시기에 따른 변화 양상을 분석하고자 하였다. 방 법 : 1991년 1월부터 2005년 5월까지 서울대학교 어린이병원 소아 환자의 각종 임상 검체에서 분리된 폐구균 465균주의 혈청형을 quellung reaction으로 결정하였고, penicillin 감수성은 oxacillin 디스크 확산법으로 검사하였다. 연구기간은 1991~1995년(I분기), 1995~2000년(II분기), 2001~2005년(III분기) 등 3개의 분기로 나누어 분석하였다. 결 과 : 465균주의 주된 혈청형은 19F(22%), 23F(18%), 19A(10%), 6B(8%), 14(7%), 6A(7%), 9V(5%) 등이었고, 전체 균주의 79%이었다. 60개월 미만 연령군의 침습성 감염 검체에서 분리된 101균주에서의 주된 혈청형은 혈청형 23F(21%), 14(17%), 19F(15%), 19A(11%), 6B(10%), 6A(7%), 9V(7%) 등으로 총 70%이었다. 전체 균주에서 I, II, III의 각 분기별 혈청형 분포는 백신 혈청형에 포함된 혈청형 19F, 14가 감소하는 추세이었고, 교차 반응 혈청형에 포함된 혈청형 6A, 19A가 증가하는 추세이었다. 침습성 감염 균주에서 폐니실린 내성률은 백신 혈청형에서 92.6%, 교차 반응 혈청형에서 75.0%로 높았으나, 비백신 혈청형에서는 34.4%로 낮았다. 결 론 : 최근 15년간 소아의 임상 검체에서 분리된 폐구균의 혈청형은 주로 19F, 23F, 19A, 6B, 14, 6A, 9V 등이 가장 흔하였고, 최근에 혈청형 19A, 6A가 증가하는 추세이고, 혈청형 19F와 14가 감소하였다. 향후 백신 사용과 관련하여 국내 폐구균 혈청형의 분포에 관한 지속적인 관찰이 필요하다고 생각된다.

도축돈의 폐렴병소에서 분리한 Pasteurella multocida 에 대한 연구 (Studies on Pasteurella multocida isolated from pneumonic lungs of slaughter pigs)

  • 이 실험은 도축돈의 폐에서 Pasteurella multocida 감염의 발생빈도를 조사하고 분리된 Pasteurella multocida의 항생제에 대한 약제감수성, 생화학적특성, 협막혈청형의 분류 및 독소생성능에 대해서 조사하였다. 실험재료로는 영남지방의 돼지 450두가 사용되었으며, 재료채취는 1992년 4월부터 1993년 3월 사이에 시행되었다. P multocida는 도축돈의 폐 450예에서 80주가 분리되어 17.7%의 분리율을 나타내었다. 분리균 대부분의 생화학적 및 배양성상은 reference strain의 것과 일치하였으며, 분리균 80주의 협막혈청형을 동정한 결과 77주가 type A, 나머지 3주가 type D로 나타나 각각 96.3%, 3.8%의 분포비를 나타내었다. 항생제에 대한 감수성검사에서 모든 분리균주는 ampicillin, ceftiofur, cephalothin, ciprofloxacin, penicillin-G 등에 대해서는 매우 감수성이 높았으며 이들 분리균중 일부는 sulfadimethoxine과 streptomycin에 내성을 나타내었다. 분리균 80주중 61주가 독소생성능이 있는 것으로 나타났으며, type A 77, type D 3주중 76.6%가 각각 독소생성능이 있는 것으로 나타나 협막혈청형간에 독소생성능의 뚜렷한 차이가 인정되지 않았다.
