Single-chain variable fragment (scFv) is a fusion protein of the variable regions of the heavy (VH) and light (VL) chains of immunoglobulin, connected with a short linker peptide of 10 to about 20 amino acids. In this study, the scFv of a monoclonal antibody against the third domain of human CD4 was cloned from OKT4 hybridoma cells using the phage display technique and produced in E. coli. The expression, production, and purification of anti-CD4 scFv were tested using SDS-PAGE and Western blot, and the specificity of anti-CD4 scFv was examined using ELISA. A 31 kDa recombinant anti-CD4 scFv was expressed and produced in bacteria, which was confirmed by SDS-PAGE and Western blot assays. Sequence analysis proved the ScFv structure of the construct. It was able to bind to CD4 in quality ELISA assay. The canonical structure of anti-CD4 scFv antibody was obtained using the SWISS_MODEL bioinformatics tool for comparing with the scFv general structure. To the best of our knowledge, this is the first report for generating scFv against human CD4 antigen. Engineered anti-CD4 scFv could be used in immunological studies, including fluorochrome conjugation, bispecific antibody production, bifunctional protein synthesis, and other genetic engineering manipulations. Since the binding site of our product is domain 3 (D3) of the CD4 molecule and different from the CD4 immunological main domain, including D1 and D2, further studies are needed to evaluate the anti-CD4 scFv potential for diagnostic and therapeutic applications.
The purpose of this study is to empirically investigate critical success factors for effective implementation of web-based distance learning system. First of all, four critical success factors are theoretically derived from reviewing previous research. They are: (1) learner-related factor including the variables such as teaming ability, learning attitude, and attending motivation, (2) environmental factor including the variables of physical and mental support for learners, (3) instructional design factor represented by one variable, the degree of appropriateness of learning contents, and (4) the factor concerning the level of self-directed learning readiness embracing the variables such as curiosity for learning, openness towards challenge of learning and affection for learning. Subsequently, the relationships between these four critical success factors and the degree of learning satisfaction are empirically investigated. The data for empirical analysis of the research are collected from 1,020 respondents who have already passed the web-based distance learning courses which have been implemented in Information and Communication Officials Training Institute. Out of 1,020 responded questionnaires, 875 data were available for statistical analyses. The main results of this study are as follows. Firstly, the most important factor for successful implementation of the web-based distance learning system is shown to be the instructional design factor, and in the next place, the self-directed learning readiness factor, the environmental factor and the learner-related one in sequence. Secondly, additional analysis of the variables included in the instructional design factor shows that availability of practical information and knowledge is the most influencing variable, and next, interesting composition of contents, reasonable learning amount, optimal level of instruction, and understandable explanation are significantly important in the descending order. Lastly, among learning motivators, strong intention of acquiring business knowledges and skills is found to be the most important satisfier in the web-based distance learning. The theoretical contribution of this study is to derive a comprehensive model of critical success factors for implementing the web-based distance learning system. And, the practical implication of this study is to propose efficient and effective guidelines for developing and operating the web-based distance learning system in the various kinds of organizations.

This paper discusses a far-end crosstalk (FEXT) canceller for twisted-pair transmission. Many twisted-pair systems such as fiber-to-the-curb (FTTC), very high-speed digital subscriber line (VDSL), and high-speed LAN systems, use frequency-division duplexing (FDD) for duplex transmission. It is shown that the maximum reach of FDD twisted-pair system is limited by the performance of its upstream channel, which is located at higher frequencies than the downstream channel. In order to improve the performance of such FDD transceiver, FEXT cancellation is introduced for the channel at higher frequencies. A system arrangement and its blind start-up procedure are studied when the FEXT canceller and equalizer are jointly adapted to combat channel intersymbol interference (ISI), FEXT, and other additive noise. The initial convergence and the steady-state behavior of the proposed twisted-pair system without requiring transmission of an ideal training sequence are investigated. Measured characteristics as well as analytical model of the FEXT channel are used to estimate the time span needed for the FEXT canceller. It is also shown that the memory span for the FEXT canceller is almost independent of the channel, thus making our results useful for the twisted-pair system over all different channels.
It is difficult to identify reusable and independent components in component-based development(CBD) process. Therefore existing methodologies have dealt the problem of component identification based on only developer's intuition and heuristics. As a result, it is difficult to identify the business components by common developers. Therefore, in this paper, we propose a new baseline and technique to identify the business components based on domain model such as use case diagrams, class diagrams, and sequence diagrams. proposed method identifies components through two phases; system component identification and business component identification. Especially, we consider structural characteristics as well as dependency characteristics according to methods call types and directions in identifying components. We also present a case study and comparative analysis and assessment to prove the practical use of our technique.
The detection of lymph node metastasis is an important step in tumor staging and is significant for therapy planning. It has been challenged to yield an appropriate image with diagnostic methods such as Magnetic Resonance (MR) and Computed Tomography (CT). Though CT has been used widely and accessed easily to show internal organs, it can hardly provide difference between lymph node and adjacent vessel or fat tissue. It has been well established that MR can reveal the subtle discrepancy within soft tissue. This study investigated the suitability of MR lymph node imaging without contrast enhancement by comparison of T1-weighted image (T1WI) and T2- weighted image (T2WI) in ten normal rabbits. According to the pulse sequence optimized from preliminary study, T1-weighted spin-echo (repetition time/echo time=400/12 ms) and T-2 weighted fast spin-echo (repetition time/echo time=3500/84 ms) images covering the hind limbs and pelvic region were acquired at 1.5 T. Two radiologists scrupulously evaluated the MR images in consensus. And signal intensity of lymph nodes was compared with that of adjacent fat. Statistical analysis showed that T1-weighted coronal image visualized the lymph nodes (iliac, superficial inguinal and popliteal lymph nodes) quickly and consistently rather than T2-weighted one. Conclusively, T1WI for evaluation of lymph nodes is moderately better than T2WI and appears to have potential for quick and sufficient mapping of the lymph nodes. In addition, this normal MR image of lymph nodes could be applied to further study for the evaluation of lymphatic system in abscess and tumor bearing animal model.

We monitored the occurrence of human enteric viruses in urban rivers by cell culture-PCR and RT-nested PCR. Water samples were collected monthly or semimonthly between May 2002 and March 2003 in four urban tributaries. Enteric viruses were detected by RT-nested PCR and cell culture-PCR based on a combination of Buffalo Green monkey kidney (BGMK) and A549 cell lines, followed by phylogenetic analysis of amplicons. By RT-nested PCR analysis, 45 (77.6%), 32 (55.2%), 32 (55.2%), 26 (44.8%), 12 (20.7%), 2 (3.4%), 4 (6.9%), and 4 (6.9%) of 58 samples showed positive results with adenoviruses, enteroviruses, noroviruses (NV) genogroup I (GI) and II (GII), reoviruses, hepatitis A viruses, rotaviruses and sapoviruses, respectively. Adenoviruses were most often detected and only eight (13.8%) samples were negative for adenoviruses and positive for other enteric viruses in the studied sites. Thirty-one (77.5%) of the 40 samples were positive for infectious adenoviruses and/or enteroviruses based on cell culture-PCR, and the frequency of positive samples grown on A549 and BGMK (65.0%) was higher than that grown on BGMK alone (47.5%). The occurrence of each enteric virus, except reoviruses and hepatitis A viruses was not statistically correlated with the water temperature and levels of fecal coliforms according to Binary logistic regression model. By sequence analysis, most strains of adenoviruses and enteroviruses detected in this study are similar to the causative agent of viral diseases in Korea and most NV GI- and GII-grouped strains were closely related to the reference strains from China and Japan, and GII/4-related strains had similar sequences to strains recognized as a worldwide epidemic outbreak. Our results suggested that monitoring human enteric viruses is necessary to improve microbial quality and cell culture-PCR using the combination of A549 and BGMK cells and the adenovirus detection by PCR could be useful for monitoring viral contamination in the aquatic environment.
The exploratory test was introduced as a desirable test method due to its fast development cycle, but it is not actively adopted because documentation and analysis of the test range are required for application. On the other hand, keyword-based testing has been introduced as a way to save resources and facilitate maintenance, but it is difficult to plan tests in advance due to the large number of variables such as data, settings, interactions, sequence and timing. However, in keyword-based testing, you can create a test case based on keywords by presenting clear criteria and methods for creating keywords and applying the exploration testing process. In this paper, we propose a model that automates exploratory tests based on keywords. To verify the effectiveness, we compared the general keyword-based test(KBT) and keyword-based exploratory test(KBET), and compared with the exploratory normal test case(ETC) and keyword-based exploratory test(KBET).

In this paper, we propose a methodology to estimate the number of users in e-Commerce systems. There have been a lot of previous work under the closed-LAN system environment. But the study on the number of acceptable users in real network environment is hard to find in the literature. Our research applies a Hybrid Simulation by using QoS results for end-to-end high-speed Internet service, and experiments are performed with regard to LAN and WAN, network equipments, and various network bandwidth. As result of the experiments we observed that the response time of high-speed Internet service media(Wireless LAN, ADSL, Cable, VDSL) depends heavily on the sequence and depth of transactions and on the ratio of transactional and non-transactional interactions. That is, as the network and application get more loads, the number of acceptable users decreases. By adding a cache server and an L4 switch to the simulation model, we analysed the changes in the number of users and client response time.

The wired and wireless Internet is a useful window to easily acquire various types of media data. On the other hand, the public can easily get the media data including the object to which the personal information is exposed, which is a social problem. In this paper, we propose a method to robustly detect a target object that has exposed personal information using a learning algorithm and effectively block the detected target object area. In the proposed method, only the target object containing the personal information is detected using a neural network-based learning algorithm. Then, a grid-like mosaic is created and overlapped on the target object area detected in the previous step, thereby effectively blocking the object area containing the personal information. Experimental results show that the proposed algorithm robustly detects the object area in which personal information is exposed and effectively blocks the detected area through mosaic processing. The object blocking method presented in this paper is expected to be useful in many applications related to computer vision.

In the case of aircraft-launched guided weapons, various interface tests such as MIL-STD-1760 based power source, discrete signal, MUX communication as well as BIT of missile can verify system safety and reliability. The purpose of this case study is to develop a test bench based on MIL-STD-1760E for interoperability testing between aircraft and weapons. We proposed a testing method of the launch sequence based on the defined TIME LINE in the development phase of the missile system from the application of the power of the missile to the targeting, the transfer order, and the missile separation process. Furthermore, it will be a reference model that can maximize the verification scope in the development phase of the air to surface missile system by simulating abnormal situation to the inert missile using the error insertion function.
