Two-Phase Genetic Algorithm for Solving the Paired Single Row Facility Layout Problem

  • This paper proposes a two-phase genetic algorithm (GA) to solve the problem of obtaining an optimum configuration of a paired single row assembly line. We pair two single-row assembly lines due to the shared usage of several workstations, thus obtaining an optimum configuration by considering the material flow of the two rows simultaneously. The problem deals with assigning workstations to a sequence and selecting the best arrangement by looking at the length and width for each workstation. This can be considered as an enhancement of the single row facility layout problem (SRFLP), or the so-called paired SRFLP (PSRFLP). The objective of this PSRFLP is to find an optimal configuration that seeks to minimize the distance traveled by the material handler and even the use of the material handler itself if this is possible. Real-world applications of such a problem can be found for apparel, shoe, and other manual assembly lines. This research produces the schematic representation solution using the heuristic approach. The crossover and mutation will be utilized using the schematic representation solution to obtain the neighborhood solutions. The first phase of the GA result is recorded to get the best pair. Based on these best matched pairs, the second-phase GA can commence.

  • A cDNA that was rapidly induced upon abscisic acid, cold, drought, mechanical wounding and to a lesser extent, by high salinity treatment, was isolated from Arabidopsis seedlings. It was classified as DREB subfamily member based on multiple sequence alignment and phylogenetic characterization. Since it encoded a protein with a typical ERF/AP2 DNA-binding domain and was closely related to the TINY gene, we named it TINY2. Gel retardation assay revealed that TINY2 was able to form a specific complex with the previously characterized DRE element while showed only residual affinity to the GCC box. When fused to the GAL4 DNA-binding domain, either full-length or its C-terminus functioned effectively as a trans-activator in the yeast one-hybrid assay while its N-terminus was completely inactive. Our data indicate that TINY2 could be a new member of the AP2/EREBP transcription factor family involved in activation of down-stream genes in response to environmental stress.

Rapid and Direct Detection of Vibrio vulnificus in Small Octopus (Octopus variabilis) Using Polymerase Chain Reaction

  • The cells of Vibrio vulnificus can be induced to the viable but nonculturable (VBNC) state by natural environmental parameters. The V. vulnificus cells in the VBNC state can not be recovered by ordinary laboratory techniques. This nonculturability could often hamper development of effective processing strategies to minimize the number of V. vulnificus in seafoods. Even with V. vulnificus cells in a culturable state, the length of time required to identify the bacteria in contaminated food by phenotyphic characterization may prevent appropriate in-time responses by public health agencies to infections of the bacteria. In the present study, we used polymerase chain reaction (PCR) to develop a rapid and direct detection method for V. vulnificus in small octopus (Octopus variabilis) which is consumed as a raw food in Korea. The region targeted was a 704-base pair (bp) portion of the hemolysin gene, vvhA, of V. vulnificus. The primers designed for PCR amplification were specific for all V. vulnificus sp. tested. Several methods were examined to extract total DNA directly from V. vulnificus seeded into the octopus homogenate and the guanidine isothiocyanate (CITC) method appeared to be most effective. From the octopus homogenate seeded by V. vulnificus at an initial level of $10^2$ CFU/ml of the homogenate and then incubated for 12 h, the targeted sequence was successfully amplified by PCR and the 704-bp DNA fragment was observed by gel electrophoresis. The total completion of this assay requires less than one day.

Proxy Design for Improving the Efficiency of Stored MPEG-4 FGS Video Delivery over Wireless Networks

  • The widespread use of the Internet and the maturing of digital video technology have led to an increase in various streaming media application. However, new classes of hosts such as mobile devices are gaining popularity, while the transmission became more heterogeneous. Due to the characteristics of mobile networks such as low speed, high error bit rate, etc., the applications over the wireless channel have different needs and limitations from desktop computers. An intermediary between two communicating endpoints to hide the heterogeneous network links is thought as one of the best approaches. In this paper, we adopted the concept of inter-packet gap and the sequence number between continuously received packets as the error discriminator, and designed an adaptive packet sizing mechanism to improve the network efficiency under varying channel conditions. Based on the proposed mechanism, the packetization scheme with error protection is proposed to scalable encoded video delivery. Finally, simulation results reveal that our proposed mechanism can react to the varying BER conditions with better network efficiency and gain the obvious improvement to video quality for stored MPEG-4 FGS video delivery.

Molecular Cloning and Characterization of Maltooligosyltrehalose Synthase Gene from Nostoc flagelliforme

  • A genomic DNA fragment encoding a putative maltooligosyltrehalose synthase (NfMTS) for trehalose biosynthesis was cloned by the degenerate primer-PCR from cyanobacterium Nostoc flagelliforme. The ORF of NfMTS was 2,799 bp in length and encoded 933 amino acid residues constituting a 106.6 kDa protein. The deduced amino acid sequence of NfMTS contained 4 regions highly conserved for MTSs. By expression of NfMTS in E. coli, it was demonstrated that the recombinant protein catalyzed the conversion of maltohexaose to maltooligosyl trehalose. The $K_m$ of the recombinant enzyme for maltohexaose was 1.87 mM and the optimal temperature and pH of the recombinant enzyme was at $50^{\circ}C$ and 7.0, respectively. The expression of MTS of N. flagelliforme was upregulated, and both trehalose and sucrose contents increased significantly in N. flagelliforme during drought stress. However, trehalose accumulated in small quantities (about 0.36 mg/g DW), whereas sucrose accumulated in high quantities (about 0.90 mg/g DW), indicating both trehalose and sucrose were involved in dehydration stress response in N. flagelliforme and sucrose might act as a chemical chaperone rather than trehalose did during dehydration stress.

New record of Dumontia contorta and D. alaskana (Dumontiaceae, Gigartinales) in Korea

  • During a survey of marine algal flora, two gigartinalean species were collected from Pohang and Youngdeok located on the eastern coast of Korea. They share the generic morphological features of Dumontia. One is characterized by cylindrical to complanate thallus with multi- and uniaxial structure, somewhat inflated and contorted branches, and hollow medulla and cortex consisting of progressively smaller cells outwards. The other shows basically the same features as the former species but was smaller in size, as having 4-7 cm in thallus length and 1-2 mm in branch width rather than 15 and 2-5 mm. Both species are distinguished from each other only by these morphometric features. However, it is supported by molecular analysis that both species are genetically distinct. In a phylogenetic tree based on internal transcribed spacer sequence, the two species nest in the same clade as Dumontia contorta and D. alaskana, respectively. The genetic distance between both sequences within the clade was calculated as 0.0 0.2%, considered to be intra-specific for Dumontia. Based on the morphological and molecular analyses, the two Korean species are identified as D. contorta and D. alaskana described originally from Netherlands and Alaska, respectively. This is the first record of the two Dumontia species in Korea.

  • Experimental studies on determination of the supply leading time of propellants to combustion chamber have been made to stably and efficiently guarantee the ignition process with liquid rocket engine. The propellant used is a Jet A-1 as fuel and a liquid oxygen as oxidizer. Unlike impinging FOOF type of injectors are arranged radially and the designed O/F ratio is 2.34. The present experiment program also includes the stability on the quadlet type of ignitor using the triethylalumimum as an ignition source and injector life tests. Experimental results clarifies that the propellant supply through LOx leading to combustion chamber is proper for stable ignition and combustion processes based on the fuel and oxidizer manifold pressures, combustion chamber pressure, and the variation of flame length from the nozzle exit with lapse time, and shows that the leading supply time of propellants affects the engine performance little. The effect of positioning cooling holes is remarkable to protect the injector face.

  • In this paper we introduce a time series model using the triangle fuzzy numbers in order to construct a statistical relation for the data which is a sequence of observations which are ordered in time. To estimate the proposed fuzzy model we split of a universal set includes all observation into closed intervals and determine a number and length of the closed interval by the frequency of events belong to the interval. Also we forecast the data by using a difference between observations when the fuzzified numbers equal at successive times. To investigate the efficiency of the proposed model we compare the ordinal and the fuzzy time series model using examples.

  • recently out door stadiums have been built as multi purpose stadium for athletic sports It hold but also various events and huge concerts. in the past, out-door stadium usually was built, but recently the out-door stadium which of the 50 % of roof covered by doom have been built increasingly. as the result, the sound obstacle is becoming very important. but a design of sound has become unplaned except some of the stadium, also rarely been built by sound-absorption material. as the result, the purpose of this study is investigating, analyzing the theory for plan of electric acoustic sound, a drawing of acoustic design and measurement a result value, comparison, evaluating a main sound factor with criteria of the Seoul City design. and reservation time, clearness, sound pressure level suitable for using purpose, so the necessary quantities will be added to financial budget of building acoustic design. and For verification, this contains acoustic analyser measurement and computer simulation and this study will find the solution of helping method.

  • This paper deals with the prediction of behavior of composite girder bridges according to the placing sequences of concrete deck. Based on a degenerate kernel of compliance function in the form of Dirichlet series, the time-dependent behaviors of bridges are simulated, and the layer approach is adopted to determine the equilibrium condition in a section. The variation of bending moments along the bridge length caused by the slab casting sequence is reviewed and correlation studies between section types and placing sequences are conducted with the objective to establish the validity of the continuous placing of concrete deck on the closed steel box-girder which is broadly used in practice.
