$NH_2-terminal$ Amino acid Sequence Analysis of Monoclonal Antibody by Electroblotting Method (Electroblotting을 이용한 단일클론항체의 $NH_2$-말단 아미노산 배열분석)

  • $NH_2-terminal$ amino acid sequence analysis of monoclonal antibody is very important to identify gene family and diversities of antigen-antibody recognition. When we used the PVDF (Polyvinylidene difluoride) membrane blotting method, we could easily analyze $NH_2-terminal$ sequence of monoclonal-antibody which specifically binds to phosphatidylinositol 4,5-biphosphate. PVDF membrane is an ideal solid-phase support for sequence analysis, especially when used with electroblotting method. This method is superior to continual method and will be applied to the sequence analysis of picomole quantities of proteins by gel electrophoresis.

  • Traditional Korean morphological analysis and POS tagging methods usually consist of two steps: 1 Generat hypotheses of all possible combinations of morphemes for given input, 2 Perform POS tagging search optimal result. require additional resource dictionaries and step could error to the step. In this paper, we tried to solve this problem end-to-end fashion using sequence-to-sequence model convolutional features. Experiment results Sejong corpus sour approach achieved 97.15% F1-score on morpheme level, 95.33% and 60.62% precision on word and sentence level, respectively; s96.91% F1-score on morpheme level, 95.40% and 60.62% precision on word and sentence level, respectively.

  • The present study was performed to analyze the molecular mechanism which dictates the transcription regulation of the $O^6$-methylguanine-DNA methyltransferase (MGMT) gene in Saccharomyces cerevisiae. Previously we identified one possible upstream repressing sequence (URS) in MGMT promoter by promoter deletion and competition analysis. In this paper we report another regulatory element (UAS: upstream activating sequence. -213 to -136) which affects the transcription activity of MGMT promoter. Gel mobility shift assay and Southwestern blot analysis using UAS probe showed several specific proteins which were able to bind to this sequence.

  • The purpose of this thesis is to suggest objective basic data for the environmental design through the quantitative analysis of the visual quality included in the physical environment of Basemsagol valley sequence landscape. For this, visual volumes of physical elements have been evaluated by using the mesh analysis, spatial images structure of physical elements have been analyzed by factor analysis algorithm, and degree of visual quality have been measured mainly by questionnaires. Also, this study aims to understand semiotics and to grope the possibility of application to the sequence landscape assessment. A semiological approach suggests a new dimension in sequence landscape assessment, which is a contrast to the existing scientific evaluation methods. Result of this thesis can be summarized as follows. Visual volumes of the immediate vegetation, rock, bridge, road and distant vegetation are found to be the main factor determining the visual quality. Factors covering the spatial image of natural park sequence landscape have been found to be the overall synthetic evaluation, potentiality, natural quality, spatial, appeal and dignity. By using the control method for the number of factors, T.V. has been obtained as 40.22%. The characteristics of the semiological approach is qualitative, open, holistic, and experiential, whereas that of the scientific approach is quantitative, closed, reductive, and experimental.

Displacement prediction in geotechnical engineering based on evolutionary neural network

  • It is very important to study displacement prediction in geotechnical engineering. Nowadays, the grey system method, time series analysis method and artificial neural network method are three main methods. Based on the brief introduction, the three methods are analyzed comprehensively. Their merits and demerits, applied ranges are revealed. To solve the shortcomings of the artificial neural network method, a new prediction method based on new evolutionary neural network is proposed. Finally, through two real engineering applications, the analysis of three main methods and the new evolutionary neural network method all have been verified. The results show that, the grey system method is a kind of exponential approximation to displacement sequence, and time series analysis is linear autoregression approximation, while artificial neural network is nonlinear autoregression approximation. Thus, the grey system method can suitably analyze the sequence, which has the exponential law, the time series method can suitably analyze the random sequence and the neural network method almostly can be applied in any sequences. Moreover, the prediction results of new evolutionary neural network method is the best, and its approximation sequence and the generalization prediction sequence are all coincided with the real displacement sequence well. Thus, the new evolutionary neural network method is an acceptable method to predict the measurement displacements of geotechnical engineering.

  • To clarify some confusions concerning identification of the Korean Peridinium species, genotypic analysis was performed with their SSU rDNA sequences. PCR was used to amplify the partial SSU rDNA of Peridinium isolates collected from three different Korean waters (Juam, Sang-sa and Togyo Reservoirs). The PCR products were allowed directly to sequence, which revealed each 942 bp of rDNA sequence. Analyses of the rDNA sequences showed that all the Korean isolates had the same genotype (100% sequence homology), and they were nearly identical to a Japanese strain of P. bipes f. occultatum (NIES 364; 99.8% sequence similarity). The sequence-based comparisons could clearly resolve P. bipes f. occultatum isolated from three different Korean waters.

  • Complementary DNA of the movement protein (MP) gene of tobacco mosaic virus Korean pepper strain (TMV-KP) was synthesized from purified TMV-KP RNA by using the reverse transcription and polymerase chain reaction (PCR) system. The synthesized double stranded cDNA was cloned into the plasmid pUC9 and transformed into Escherichia coli JM110. The movement protein gene of TMV-KP of the selected clones was subjected to sequence analysis by Sanger's dideoxy chain termination method. The complete sequence of viral MP gene from TMV-KP strain was 807 nucleotides long. The nucleotide of MP gene from TMV-KP has thirteen and two nucleotide differences from TMV vulgarae (TMV-OM) and Korean (TMV-K) strains, respectively. Thus, the nucleotide sequence of TMV-KP MP gene showed higher homology of 99% with that of TMV-K MP gene.

  • Several methodologies analyzing welding distortions of large shell structures like ship blocks have been developed and utilized in shipyards for a long times ago. In general, one of objects of thermal distortion analysis is to find welding sequence making least-deformation without any suppliance, and it can be solved easily and rapidly by EP strain-boundary method. But after assembly construction, there are usually more than 10 weldments in each process, and the number of sequence will follow the Factorial calculation. In this research, a method has been suggested to decide the best welding sequence by minimum analysis. Using this method, welding deformation could be reduced just analyzing some cases as many as the number of weldments. Experiments and Analysis of all cases were also done, and their best results are good agreements with predictation by suggested methodology.

  • Expert system for automating HAZOP analysis of batch processes In contrast with continuous processes must consider the discrete variables such as time and sequence. So in this study, we have developed the expert system for automating HAZOP analysis of batch processes to analyse time and sequence on the basis of the relation between discrete variables and continuous ones. Because these variables can not be explained by the method used in the HAZOP analysis of continuous processes. The proposed expert system have been discussed on a Latex batch process to evaluate its effectiveness.

  • With the establishment of rapid sequence analysis of 16S rRNA and the recognition of its potential to determine the phylogenetic position of any prokaryotic organism, the role of 16S rRNA similarities in the present species definition in bacteriology need to be clarified. Comparative studies clearly reveal the limitations of the sequence analysis of this conserved gene and gene product in the determination of relationship at the pathogenic strain level for which DNA-DNA reassociation experiments still constitute the superior method. Since today the primary structure of 16S rRNA is easier to determine than hybridization between DNA strands, the strength of the sequence analysis is to recognize the level at which DNA pairing studies need to be performed, which certainly applies to similarities of 97% and higher.
