  • To efficiently separate a protein stimulating sperm swim-up migration and movement from follicular proteins, the effect of paper chromatography and liquid chromatography with reverse phase column and superose column on protein separation was examined. And the results obtained were as follows; 1. The band component that was separated with paper chromatography stimulated sperm migration and movement depending on its additional levels. Especially, band I component significantly increased sperm migration. But, all components of bands 1, 2 and 3 showed lower sperm migration and movement, compared to follicular fluid at the same additional level. 2. Among the components separated from follicular protein of 2~5mm follicles with reverse phase column ($\mu$RPC), components at retention time (RT) of 3.33, 7.00, 13.87, and 16.6A minutes stimulated sperm migration within a limited range. 3. All components separated from follicular protein of 10mm follicles with $\mu$RPC column didn't stimulate sperm migration and movement. 4. Among the components separated from follicular protein of 2~5m follicles with superose column, components at retention volume (RV) of 1.35 and 0.82 ml significantly stimulated sperm migration and movement. In conclusion, protein components stimulating sperm migration and movement were efficiently separated with superose column in Smart system. Especially, components of RV 1.35 and RV0.82 stimulated sperm swim-up separation.

  • In order to improve the operability in a fully thermally coupled distillation column the main column is separated into an upper and a lower column, and its design method is explained. With a distillation experiment using a two-inch column the operability of the proposed column is examined. Readily available methanol, ethanol and propanol are used as the feed, and the experimental result is compared with that of the HYSYS simulation. It is found from the experiment that the recovery ranges between 58.8% and 68% and the distillation efficiency is lower than that of the simulation. consumes less energy than an original column, it is not widely implemented in practice due to its operational difficulty. However, it is proved that the proposed distillation column can be operated without compressors and it can be practically utilized.

  • Among proteins separated from methanol extract of follicular fluid with superose column, the components inducing sperm swim-up separation through sucrose layer were analysed with superose column in Smart system and SDS-PAGE. And the results obtained were as follows; The fractions of retention volume (RV) 0.83ml and RV 1.36ml separated with superose column should stimulate sperm migration and movement. However, RV 0.83 fraction was consisted of complex materials containing RV 1.36 component. RV 1.36 fraction contained a BSA analogue of 67 kilodaltons (Kd) and showed identical peak pattern with BSA fraction V. In conclusion, the protein of 67 Kd in follicular fluid should stimulate sperm migration and movement.

  • Free phenolic acids (FPA), soluble phenolic acid esters (SPA) and insoluble-bound phenolic acids (IPA) were extracted from defatted Perillae semen flour and the antioxidative components in FPA extract was separated by column chromatography and HPLC. Total phenolic content of defatted Perillae semen flour was 0.38% as chlorogenic acid and each percent ratio of the content of FPA, SPA and IPA to total phenolic content was 71.1%, 15.8% and 13.1%, respectively. The antioxidative activity was compared by measuring of electron donating ability (EDA) and thiobarbituric acid value (linoleic acid substrates). The FPA extract was showed the highest antioxidative activity among the three kinds of phenolic extracts. The FPA extract showing the highest antioxidative activity was separated by silica gel column chromatography and then the separated fractions were compared in terms of antioxidative activity. The fractions of acetone : methanol (8 : 2) showing the highest antioxidative activity was further separated by HPLC. Five fractions (F-I, F-II, F-III, F-IV and F-V) were observed on the HPLC chromatogram and F-I fraction showed the highest antioxidative activity.

  • A new method for liquid chromatography with post column reaction is suggested for the separation and quantification of tertiary amines. A mixture of triethanolamine and N-ethyl diethanolamine was separated by a strong cation exchange column, followed by spectrophtometric detection of the blue colors generated from the reaction of each amine with the Folin-Ciocalteau reagent. The tertiary amines were properly separated when an eluent of pH 9.5 containing 0.5M sodium nitrate was used. Under this condition, calibration curve of triethanolamine in 2-10mg/100ml concentration range was attained. Good results were obtained when cream and shampoo preparations containing known amount of triethanolamine were analysed according to this method. In case the sample did not contain any other interfering reducing substances, the amine was quantitatively determined by the simple reaction of the samples with Folin-Ciocalteau reagent, and the subsequent spectrophotometric measurement.

  • A method is developed for simultaneously determining ethanol and acetic acid in vinegars by quantitative packed-column gas chromatography. Vinegars were filtrated and directly chromatographed on a $2\;m{\times}2\;mm$ stainless steel column packed with Tenax-GC, 80/100. Ethanol, isopropy alcohol as an internal standard, and acetic acid were completely separated within 20 min of running time without any interfering peaks. The accuracy of packed column gas solid chromatography (PCGSC) was discussed compared to the analytical data by titration, high performance liquid chromatography and capillary column gas liquid chromatography (CCGLC).

  • Mill scale, an iron waste, was used to separate magnetite particles for the adsorption of phosphate from aqueous solution. Mill scale has a layered structure composed of wustite (FeO), magnetite ($Fe_3O_4$), and hematite ($Fe_2O_3$). Because magnetite shows the highest magnetic property among these iron oxides, it can be easily separated from the crushed mill scale particles. Several techniques were employed to characterize the separated particles. Mill scale-derived magnetite particles exhibited a strong uptake affinity to phosphate in a wide pH range of 3-7, with the maximum adsorptive removal of 100%, at the dosage of 1 g/L, pH 3-5. Langmuir isotherm model well described the equilibrium data, exhibiting maximum adsorption capacities for phosphate up to 4.95 and 8.79 mg/g at 298 and 308 K, respectively. From continuous operation of the packed-bed column reactor operated with different EBCT (empty bed contact time) and adsorbent particle size, the breakthrough of phosphate started after 8-22 days of operation. After regeneration of the column reactor with 0.1N NaOH solution, 95-98% of adsorbed phosphate could be detached from the column reactor.

  • Achiral-chiral column switching HPLC assay was developed to allow the separation and quantification of the enantiomers of terbutaline in human plasma by means of fluorescence detection. Plasma samples were prepared by solid-phase extraction with sep-pak silica, followed by HPLC assay. The enantiomers of terbutaline and the internal standard were separated from the biological matrix on a silica column, and the two enantiomers were resolved and quantified on a Sumichiral OA-4900 column. The two columns were connected by a switching valve equipped with silica trap column, The trap column was used to concentrate the terbutaline in the eluent from the achiral column before back flushing onto the chiral phase. For each enantiomers, the assay was linear between 2.5-125 ng/$m\ell$ (r=0.9999) and detection limit was 1.0 ng/$m\ell$ .

  • Though a fully thermally coupled distillation column consumes less energy than an original column, it is not widely implemented in practice due to its operational difficulty. A new fully thermally coupled distillation column is proposed for the operability improvement, and its performance is investigated. The main improvement is the separation of a main column to give an upper and lower columns and the installation of an intermediate heat exchanger between them to regulate the fluctuation of product compositions. A proper manipulation of column pressure in the separated main columns made easy vapor flow without a compressor. The operability improvement is examined in a hexane process from the dynamic simulation using a commercial design software HYSYS. The simulation results indicate that the coupling among inputs and outputs is loosened to make easy manipulation of product compositions in the proposed distillation system.
