Nutrient Uptake and Growth Kinetics of Chattonella antiqua (Hada) Ono (Raphidophyceae) Isolated from Korea

  • The red tide-causing flagellate Chattonella anticfua can cause mass fish kills by their clogging in fish gills. Thisstudy examined the nutrient requirements of C. antiqua isolated from Korea. C. anticfua displayed maximum growthat the day five, followed by a decrease in cell density. Nitrate and nitrite were the preferred nitrogen sources, alonewith adenosine diphosphate for phosphorus compounds. In medium that contained ammonium, a significantdecrease in cell density was observed. Half-saturation constants, Ks, calculated from the maximum growth ratewere 4.94 U|M for NC>3 and 0.79 flM for P04. The growth of C. antiqua was not within the function of the N:P ratio (RU= 0.29). With an N:P ratio as low as 10, the increase in cell density was apparent, with a higher division rate. At lev-els above 50 fiM of NaNOg or 8 ;uM of NaHUPCU, the growth rates were somewhat decreased. Phosphate was thelimiting factor for C. antiqua growth since the starvation of phosphate had brought about a rapid decrease in celldensity in semi-continuous culture. Studies about the temporal modification of the efficiency of nitrate or phosphateuptake may be necessary to explain the bloom dynamics of C. antiaua.

  • Recently MR CDC damper has been applied to semi-active suspension control system gradually. Compared to former hydraulic CDC damper, it has rapid time response performance as well as simple internal structure and wide range of damping force. In order to develop control logic algorithm which enables to take maximum advantage of unique characteristics of MR CDC damper, it is inevitable to perform a thorough investigation into its nonlinear performance. In many previous researches, MR fluid model was either simply assumed as Bingham Plastic, or a phenomenological model based on experiment was established instead to predict damping performance of MR CDC damper. These experimental flow model which is not based on flow analysis but intentionally built to fit damping characteristics, may lead to totally different results in case of different configuration or structure of MR CDC damper. In this study, a generalized flow formula from mathematical flow model of MR fluid for annular orifice is derived to analyze and predict damping characteristics when current is excited at piston valve.

  • In recent years, ultrasonic has been widely applied in measurement and industrial fields and its application range has been expanded as a result of continuous research and development. Wire Bonding Machine, an instrument fabricating semi-conductor, makes use of ultrasonic bonding method. Specially, the method utilizes the longitudinal vibration of ultrasonic transducer composed of piezoelectric vibrator and horn. This work investigates the design conditions affecting the dynamic characteristics through the theretical and experimental analysis. It conducts separately the system identification of piezoelectric vibrator in time domain and the modal analysis of horn in frequency domain. The integrated modeling is conducted via a combbination of dynamic identification of piezoelectric vibrator and theroretical analysis of horn. Then comparison is made for theroretical and experimental results of the dynamic characteristics of the ultrasonic transducer comprised of piezoelectric vibrator and horn. Form the results of the comparison we develop the design technique of ultrasonic transducer using dynamic characteristics analysis and propose the possibility of ultrasonic bonding considering the optimal conditions for the longitudinal vibration of ultrasonic transducer and other conditions.

Extraction of Volatile Essential Oil from Citrus junos Peel by Supercritical Carton Dioxid (초임계 이산화탄소를 이용한 유자과피로부터 휘발성 정유성분의 추출)

  • The extraction of volatile essential oil using supercritical carbon dioxide with 2%(v/v) ethanol and non-ethanol was performed in a semi-continuous flow extractor at the range of pressures and temperatures 6.9 to 17.2 MPa and 30 to 45$^{\circ}C$, respectively. When ethanol was added to the co-solvent, the solubility of volatile essential oils was increased up to 90% over the neat CO$_2$ value. The most efficiency of extraction of the voilatile essential oils was achieved at 13.8㎫ and 40$^{\circ}C$ by supercritical carbon dioxide with entrainer from Citrus junos peel.

Preventive Strategies of Ventilator Associated Pneumonia

  • Purpose: Despite numerous evidence based preventive strategies of ventilator associated pneumonia (VAP) have been introduced, the incidence rate of VAP continues in an unacceptable range. The purposes of this review were to identify risk factors and diagnosis of VAP and to introduce current evidence based preventive strategies of VAP. Methods: A comprehensive literature search using keywords, including ventilator associated pneumonia were entered into a search engine. A number of highly pertinent papers relevant to the purpose of the review were identified. The papers that discussed specific preventive strategies of VAP were selected for analysis and inclusion in this review. Results: A number of evidence based preventive strategies that nurses can implement in their clinical practice to prevent VAP were identified. Such strategies include hand washing, use of protective gloves and gowns, oral care, stress ulcer prophylaxis, avoidance of unnecessary intubation, weaning protocol, sedation vacation, use of non-invasive ventilation, semi-recumbent position, continuous aspiration of subglottic secretions, and maintenance of proper endotracheal tube cuff pressure. Staff education is essential in preventing VAP. Conclusion: Preventive strategies of VAP should be applied to daily nursing care and each critical nurse should play a functional role in preventing VAP.

Immobilization of Trigonopsis variabilis and Conversion of Cephalosporin C to 7$\beta$-(4-Caboxybutanamido)Cephalosporanic Acid (Trigonopsis variabilis의 고정화 및 Cephalosporin C로부터 7$\beta$-(4-Carbohybutanamido)Cephalosporanic Acid의 전환)

  • An immobilized Trigonopsis variabilis cells having an high activity of D-amino acid oxidase(DAO) was used to convert CPC into GL-7-ACA. The optimal pH of the reaction system was 8.0-8.5, and the optimal temperature was 40$\circ$C. When immobilized cell was used repeatedly in semi-batchwise reaction, the system retained 80% of the initial activity after used of 12 times for over 12 hours. The storage stability of the immobilized cell was maintained for 30 days at 4$\circ$C. The CPC concentration for the maximal reaction rate was about 30 mM and 40 mM for free and immobilized cells, respectively. Substrate inhibition of CPC concentration more than 50 mM was overcomed by 20~25% by immobilization. Pure oxygen supply into reaction system was most efficient in D-amino acid oxidase reaction. Continuous conversion to GL-7-ACA from CPC has been developed with an bioreactor system containing immobilized T variabilis cells. By opera- tion of the reactor for 5 hours, the average conversion yield of >80% and GL-7-ACA production of 40~45 mM per hour could be obtained.

Degradation of Chlorophenols and Phenol Mixtures by Cooperative Activities of Chlorophenol-degrading Strains

  • Three strains capable of degrading a chlorophenol were isolated by selective enrichment from soils contaminated with industrial wastewater. A Pseudomonas solanacearum TCP114 could use 2,4,6-trichlorophenol (TCP) as sole carbon and energy source, while two strains of Pseudomonas testosteroni CPW301 and Arthrobacter ureafaciens CPR706 could use 4-CP. All isolates also grew well on phenol. The degradation of one component by a pure strain was strongly affected by the presence of other compounds in the medium, CPW301 and CPR706 entirely lost the ability to degrade 4-CP and phenol in the presence of TCP. TCP114 also lost the ability to degrade phenol when 4-CP was added to the culture medium. These restrictions on the degradability could be overcome by employing defined mixed cultures (TCP114 and one strain of 4-CP degrading strains). All three components were successfully degraded by defined mixed cultures through their cooperative activities. It was also demonstrated that defined mixed cultures could be immobilized by using calcium alginate for the semi-continuous degradation of the three component mixture. Immobilization could not only accelerate the degradation rate, but also allowed the reuse of the cell mass several times without loss of the cells' degrading capabilities.

Relationship Between Geometrical Stiffness of Diaphragm and Resonance Frequency for Micro-speaker (마이크로스피커 진동판의 등가탄성과 공명진동수의 연관성)

  • Information technology devices, such as cellular phones, MP3s and so on, due to restrictions of space, require thin and small micro-speakers to generate sound. The reduction of the size of micro-speakers has resulted in the decrease of sound quality, due to such factors as frequency range and sound pressure level. In this study, the acoustical properties of oval microspeakers has been studied as a function of pattern shape on the diaphragm. The other conditions of micro-speakers, except for the pattern, was not changed. When the pattern is present on the diaphragm and the shape of pattern was a whirlwind, the resonance frequency was reduced due to the decrease of tensile strength of diaphragm. The patterns presented in the semi-minor axis of diaphragm did not effect a change of resonance frequency. However, increasing the number of patterns in the semimajor axis of diaphragm became a reason for the decrease of resonance frequency on edge side. When the depth of pattern on edge side was increased, the resonance frequency was decreased due to reduction of geometrical stiffness. If the height of edge and dome were increased, the resonance frequency and geometrical stiffness rapidly increased. After reaching the maximum values, they began to decrease with the continuous increase of height.

TREATMENT OF ZINC DEFICIENCY IN SHEEP BY ZINC CONTAINING BOLUSES

  • A study was conducted to investigate the release pattern of zinc form the zinc containing boluses and to see whether the released zinc can cure a zinc deficiency in sheep. Three sheep were used in this experiment and were fed a low zinc semi-synthetic diet throughout the experimental period. Each sheep was given a single pre-weighed zinc containing bolus when blood variables showed continuous zinc deficiency. The zinc containing boluses when placed within the reticulo-rumen of zinc deficient sheep, release zinc at the rate of 106.6 mg zinc/day for 39 days. At the end of depletion period there was a reduced feed consumption, plasma zinc concentration, plasma alkaline phosphatase activity and increased plasma zinc binding capacity which were 409 g, 0.18 mg/l, 87 U/l and 88.7% respectively and 521 g, 0.18 mg/l, 142 U/l, and 89.5% respectively before first and second blousing. After the administration of the first and second boluses, the feed consumption, plasma zinc levels and plasma alkaline phosphatase activities rose rapidly and far exceeded the starting values. The zinc binding capacity was reduced to 21.9% due to the administration of the first and second boluses. It is concluded that zinc boluses can be used for curing a zinc deficiency in sheep.
