Bacillus spp. as Biocontrol Agents of Root Rot and Phytophthora Blight on Ginseng

  • Ginseng (Panax ginseng) is one of the most widely cultivated medicinal herbs in Korea. However, yield losses reached up to 30-60% due to various diseases during 3 or 5 years of ginseng cultivation in the country. Therefore, successful production of ginseng roots depends primarily on the control of diseases. The objective of this study was to select potential biocontrol agents from rhizobacteria isolated from various plant internal root tissues for the control of multiple ginseng diseases as an alternative to fungicides. Among 106 Bacillus strains, two promising biocontrol agents, Bacillus pumilus strain B1141 and Paenibacillus lentimobus strain B1146, were selected by screening against root rot of ginseng caused by Cylindrocarpon destructans in a greenhouse. Pre-inoculation of selected isolates to seed or l-year-old root of ginseng resulted in stimulation of shoot and/or root growth of seedlings, and successfully controlled root rot caused by C. destructans (P<0.05). Furthermore, drenching of cell suspension of the selected isolates on seedling-growing pots reduced the incidence of Phytophthora blight after the seedlings were challenged with zoospores of Phytophthora cactorum (P<0.05). P. lentimorbus strain B1146 showed antifungal activity against various soil-borne pathogens in vitro, while B. pumilus strain B1141 did not show any. Results of this study suggest that some rhizobacteria can induce resistance against various plant diseases on ginseng.

과실쥬우스용 효소제의 개발연구 (A Study on the Development of A Juice-clarifying Enzyme Preparation)

  • 과실쥬우스용 효소제를 개발할 목적으로 펙틴분해력이 강한 미생물균주를 선택하고 이에서 얻은 효소제에 의하여 포도쥬우스 및 포도주 제조과정에 있어서의 효과를 시험하여 다음과 같은 결과를 얻었다. 1) Aspergillus 및 Rhizopus속 균주에서 펙틴분해효소 생산력이 가장 강한 Aspergillis niger를 선정 하였다. 2) 이 균주에 의한 효소의 생성과정과 정제과정을 실험하고 조효소제품의 활성과 안정도에 미치는 pH 및 온도의 영향을 조사하였다. 3) 포도쥬우스 제조시에 효소제를 사용하면 free run juice의 수율이 증가하고 투명한 쥬우스를 얻을 수 있었다. 4) 포도주 양조시에 효소제를 사용하면 적포도주에는 아무 효과를 가지지 못하나 백포도주의 경우에는 과즙의 여과촉진 및 청정효과가 있을것으로 기대된다.

Transposon Tn5를 이용한 Slow growing Rhizobium japonicum의 돌연변이 유도 (Mutagenesis of Slow Growing Rhizobium japonicum by Transposon Tn5)

  • Slow growing R. japonicum R-l68 균주로부터 spectinomycin 내성 균주를 선발하고 이 Rhizobium내에 Tn-5를 도입시키기 위하여 Tn5가 함유된 E. coli WA 803/pGS9과의 conjugation을 통한 transposon mutagenesis를 실시하였다. 이때 C conjugation을 통한 Tn5 전이 빈도는 $1.0\times 10^{-5}-5.0\times 10^{-7}$ 범위 이였으며, 얻어진 transconjugant들은 spectinomycin (($100{\mu}$g/ml)과 kanamycin ($50{\mu}$g/ml)을 함유한 yeast extract-mannitol 배지에서 8-10일 배양후 colony를 형성하였다. 또한 transconjugant들은 genome상에 Tn-5를 함유하고 있음을 hybridization-을 통하여 확인하였다. 한편 nodule은 형성 하나 질소고정 활성이 없는 돌연변이주 R. japonicum RMa 75 $nod^{+}fix^{-}$ 균주를 선발하였는데 이 균주는 nodule내에 leghemoglobin이 결핍되어 있음이 확인되었다.

Effects of heat stress on growth performance, selected physiological and immunological parameters, caecal microflora, and meat quality in two broiler strains

  • Objective: This study was conducted to investigate the effects of normal and heat stress environments on growth performance and, selected physiological and immunological parameters, caecal microflora and meat quality in Cobb 500 and Ross 308 broilers. Methods: One-hundred-and-twenty male broiler chicks from each strain (one-day-old) were randomly assigned in groups of 10 to 24 battery cages. Ambient temperature on day (d) 1 was set at 32℃ and gradually reduced to 23℃ on d 21. From d 22 to 35, equal numbers of birds from each strain were exposed to a temperature of either 23℃ throughout (normal) or 34℃ for 6 h (heat stress). Results: From d 1 to 21, strain had no effect (p>0.05) on feed intake (FI), body weight gain (BWG), or the feed conversion ratio (FCR). Except for creatine kinase, no strain×temperature interactions were observed for all the parameters measured. Regardless of strain, heat exposure significantly (p<0.05) reduced FI and BWG (d 22 to 35 and 1 to 35), immunoglobulin Y (IgY) and IgM, while increased FCR (d 22 to 35 and 1 to 35) and serum levels of glucose and acute phase proteins (APPs). Regardless of temperature, the Ross 308 birds had significantly (p<0.05) lower IgA and higher finisher and overall BWG compared to Cobb 500. Conclusion: The present study suggests that the detrimental effects of heat stress are consistent across commercial broiler strains because there were no significant strain×temperature interactions for growth performance, serum APPs and immunoglobulin responses, meat quality, and ceacal microflora population.

섬유소 물질의 동시당화발효에 적합한 Glucose/Cellbiose 혼합당 발효균주의 개발 (Development of Strain Fermenting the Glucose/Cellbiose Mixed Sugar for Simultaneous Saccharification of Fermentation of Cellulosic Materials)

  • Brettanomyces custersii CBS 5512 which has reported as a thermotolerant glucose-cellobiose co-fermentable yeast strain was mutated with UV and NTG to improve ethanol yield at higher than 4$0^{\circ}C$ B. custersii H1-23, H1-39, H1-55 and H1062 were finally selected for hyper-fermentable strains at higher than 4$0^{\circ}C$ from thermotolerant 7510 colonies through 5th selection. Among the selected strains, H1-39 mutant had better fermentability at 4$0^{\circ}C$ and 43$^{\circ}C$ from different concentrations of glucose. H1-39 and H1-23 mutants yielded more than 70% of the theoretical ethanol yield in 4 and 8% mixed sugars at above 4$0^{\circ}C$, which was 5-11% higher than those by original strain. Especially, H1-39 mutant had better fermentability in 4% mixed sugar. It showed 78.5% of the theoretical yield at 4$0^{\circ}C$ and 72.2% of the theoretical yield at 43$^{\circ}C$. On the other hand, theoretical yield of ethanol by H1-39 mutant in 8% mixed sugar at 4$0^{\circ}C$ and 43$^{\circ}C$ were 75.2% and 70.2%, respectively. Theses values increased up to 7-11% as compared to those by orginal strain. By the simultaneous saccharification and fermentation, ethanol production by H1-39 mutant increased up to more than 23% as compared to that by original strain.

효모(酵母)에 의(依)한 과실주(果實酒)의 감산(減酸) (Decomposition of Acid in Wine by Yeast)

  • 산미(酸味)가 과다(過多)한 과실주(果實酒)의 감산(減酸)에 이용(利用)하기 위한 시험(試驗)으로는 공시효모(供試酵母)의 균학적(菌學的) 성질(性質)을 검토(檢討)하고 사과주(酒) 양조(釀造)에 있어서의 공시효모(供試酵母)의 특성(特性)을 시험(試驗)하였다. 그 결과(結果)는 아래와 같다. 1. 공시효모(供試酵母)는 Schizosaccharomyces pombe R-73로 동정(同定)되었다. 2. 공시효모(供試酵母)는 사과즙(汁) 중(中)에서의 아황산내성(亞黃酸耐性)이 포도주효모(酒酵母) 보다 강(强)하였다. 3. 사과즙(汁) 중(中)에서 배양(培養)하는 경우 $25^{\circ}C$에서는 공시효모(供試酵母)의 증식속도(增植速度)는 포도주효모(酒酵母)나 청주효모(淸酒酵母) 보다 빨랐다. 그러나 $15^{\circ}C$에서는 공시효모(供試酵母)의 증식속도(增植速度)가 대조균주(對照菌株) 보다 많이 느렸다. 4. 사과주양조(酒釀造)에 있어서 $25^{\circ}C$(고온(高溫))에서는 공시효모(供試酵母)의 Ethanol 생성력(生成力)이 강(强)하였고 감산효과(減酸效果)가 매우 컸다. 그러나 $15^{\circ}C$(저온(低溫))에서는 공시효모(供試酵母)의 Ethanol 생성력(生成力)이 떨어졌으며 감산효과(減酸效果)도 적었다.

Actinoplanes teichomyceticus의 변이주에 의한 Teicoplanin 발효생산

  • Actinoplanes teichomyceticus ATCC 31121 was mutated with UV to obtain a superior mutant strain with increasing level of teicoplanin production. In this investigation lethal curve was obtained and the optimal condition to induce mutagenesis was determined and to isolate the desirable mutant strain. It was also confirmed that teicoplanin activities by agar diffusion method to be compared to the mother strain. One mutant strain, T991014-1 that had the highest teicoplanin productivity was finally selected for further investigation including fermentation pattern. The mutant was characterized by the various tests such as amylase activity, protease activity, antibiotic resistance, autotoxicity, and productivity.

  • In this research, on-line model for prediction of effective strain distribution hi strip on finishing mill process is prescribed. It has been developed using several selected non-dimensional parameters and previously made average effective strain model via series of finite element process simulations, $\Delta$$\varepsilon$ was introduced to describe the effective strain distribution in strip. To confirm adequate non-dimensional variables uniqueness test was done. And to decide the order of polynomial in on-line model equation tendency test for each variables was done. The prediction accuracy of the proposed model is examined through comparison with finite element calculation results.

  • The physical, electrical and piezoresitive characteristics of CrN(chromiun nitride) thin-films on silicon substrates have been investigated for use as strain gauges. The thin-film depositions have been carried out by DC reactive magnetron sputtering in an argon-nitrogen atmosphere(Ar-(5~25 %)$N_2$). The deposited CrN thin-films with thickness of $3500{\AA}$nd annealing conditions($300^{\circ}C$, 48 hr) in Ar-10 % $N_2$ deposition atmosphere have been selected as the ideal piezoresistive material for the strain gauges. Under optimum conditions, the CrN thin-films for the strain gauges is obtained a high electrical resistivity, $\rho=1147.65\;{\mu}{\Omega}cm$, a low temperature coefficient of resistance, TCR=-186 ppm/$^{\circ}C$ and a high temporal stability with a good longitudinal gauge factor, GF=11.17.

中性 Protease 高 生産性 Bacillus sp. KN103N의 分離 및 酵素의 特性 (Isolation of Neutral Protease Hyperproducing Bacillus sp. KN103N and Some Properties of the Enzyme)

  • A bacterial strain KN, which highly produced a protease, was isolated from several soil samples and identified to to belong to the genus Bacillus. We selected mutant strain Bacillus sp. KN103N, which was hyperproducer of protease and was resistant to D-cyclowerine, from the strain KN by several steps of mutagenesis. Neutral protease productivity of mutant strain KN103N was about 55 times as much as that of the original strain KN. The optimum pH and temperature for the enzyme activity were 7.0 and 50$^{\circ}C$, respectively and the enzyme was relatively stable at pH6.0~8.0 and below 40$^{\circ}C$. The enzyme was inactivated by EDTA, but not by DFP. These results indicate that the enzyme from Bacillus sp. KN103N was a neutral (metallo-) protease.
