Changes of Moisture Uptake. Respiration and Chemical Contents in Germinating Tobacco(Nicotiana tabacum L.) Seeds (담배종자의 발아조건에서 수분흡수, 호흡 및 저장물질의 변화)

  • The physiological aspects of tobacco seeds from seeding to germination under light and dark condition, and different temperature were studied to obtain basic information on the production of good seedlings. There were distinct three phases in moisture uptake and respiration process during germination that classifiable into 'Imbibition', 'Lag', and 'Growth' phase under light condition but such growth phase were not observed in the dark germination. Great changes of sugar and fatty acid content of tobacco seeds were observed during germination in light condition but such changes were slight in dark germinated seeds and fatty acid content of seeds during germination were decreased as corresponding to increasing in respiration.

Effect of chlorella culture solution using animal liquid manure on improving seed germination in perennial ryegrass

  • This experiments were conducted to evaluate the influence of Chlorella culture solution on seed germination of perennial ryegrass seeds. Chlorella are known to contain different bioactive compounds. In present research work, Chlorella culture solution using liquid manure as medium have been used to study their effects on germination and root length. The study conducted a germination experiment in petri-dishes. Four treatments were compared: non-treated control treated with distilled water, Chlorella culture solution and Chlorella culture filtrate, and liquid manure. The germination percentage of perennial ryegrass seeds was highest in the Chlorella culture solution treatment. Days required for 50, 70% seed germination was the fast in Chlorella culture solution and the Chlorella culture filtrate treatment. Root length of perennial ryegrass seeds was long by 1~2cm in the Chlorella culture solution compared with no treated control. The germination index of perennial ryegrass seeds was high by 180~202% in the Chlorella culture solution treatment compared to no treatment. Chlorella culture solution and the Chlorella culture filtrate have shown stimulatory effects in germination and development of root. Use of Chlorella culture solution and the Chlorella culture filtrate can be recommended to farmers as a ecofriendly practice for better germination and growth. Present research work reveals that Chlorella contain certain growth promoting substances which enhances seed germination.

Somatic embryogenesis and in vitro plant regeneration from various explants of the halophyte Leymus chinensis (Trin.)

  • The halophyte Leymus chinensis (Trin.) is a perennial rhizome grass (tribe Gramineae) that is widely distributed throughout China, Mongolia and Siberia. This study was conducted to investigate an optimal condition for plant regeneration from mature seeds, leaf base segments, and root segments in L. chinensis. Plant growth regulators affecting embryogenic callus induction and plant regeneration were investigated by four-factor-three-level [L9 (34)] orthogonal test in this study. The effects of explants types (mature seeds, leaf base segments and root segments), callus types, medium types were examined in this study. Wild type (WT) and Jisheng No. 1 plants (JS) were used for primary callus induction. A clear explants difference was seen during callus induction; mature seeds were considered as the preferred explants; and the highest frequency of callus induction was obtained in Medium 6 using mature seeds as explants in WT. Plant regeneration ability was evaluated by frequencies of green callus forming, shooting, rooting, and shooting with roots. Effect of α-naphthalene acetic acid (NAA) on shoot regeneration was remarkable with the highest frequency of 70.8% in WT after 2-month culture. The medium with 0.2- 0.5 mg/L NAA was found to have the highest shoot induction. All regenerated shoots were successfully rooted when transferred on half-strength Murashige and Skoog (MS) basal medium. The acclimatized plantlets were grown to mature with flowering and seeds setting in green house conditions.

Antifreeze proteins promote the germination of low temperature-treated petunia seeds via regulation of antioxidant- and proline-related genes

  • The involvement of antifreeze proteins (AFPs; type I and III) in the germination of low temperature-treated petunia seeds (cv. 'Mirage Rose') was investigated. The addition of AFPs (300 or 500 ㎍/l) in low-temperature treatment significantly promoted the germination of seeds compared with that in which AFPs were not added. Among all treatments, treatment with AFP I added at 300 ㎍/l showed the highest germination percentage and improved plant growth. The expression levels of antioxidant-related genes such as superoxide dismutase, peroxidase, and proline synthesis were associated with the germination of low temperature-treated seeds. Overall, this study demonstrated that AFP I may potentially function as a cold-protective agent for the germination of low temperature-treated seeds.

Effects of local conspecific abundance on seed set and seed predation, and control of Carpinus laxiflora (Betulaceae) population density

  • In this study, the dependence of local conspecific abundance on seed set and predation was examined, and determinates of population density of the temperate tree species Carpinus laxiflora (Betulaceae) in Inagi city, Tokyo, Japan were identified. During a good seed year (2013), seeds were sampled from 27 individuals and categorized as Sound, Empty, Predated, Immature, Decayed, or Broken. Empty seeds were identified as those that failed to fertilize and predation rates were defined as the proportion of seeds that were affected by predators. The proportion of the seed set that was fertilized was significantly positively correlated with local abundance of conspecific trees, while the proportion of seeds that escaped predation was significantly negatively correlated with local abundance of conspecific trees. Thereby, the production of Sound seeds was highest at sites with moderate local conspecific abundance. Although this phenomenon was only observed in a few cases, it clearly showed optimal reproductive success under conditions of moderate density of conspecific reproductive adults, suggesting a mechanism that controls the population density of this species.

Effect of Priming on Germination of Aged Soybean Seeds

  • The object of this study is to determine the effect of priming on the germination ability and seedling emergence of aged soybean seeds in lab and field conditions. Artificial or natural procedure for seed aging was applied in this study. One seed lot was artificially aged for 3 to 5 days at 42$^{\circ}C$ with high relative humidity (nearly RH 100%), and the other one was stored at room temperature for 17 months. Aged seeds were osmoconditioned in -1.1 MPa polyethylene glycol 8000 (PEG) solution for 3 days at $25^{\circ}C$ and air-dried. When Danyeobkong was aged for 4 days average germination was 61.5%, however, this improved to 98.5% after the priming treatment. Improvement of seed germination by priming the aged seed was consistent with large seed sized Jangyeobkong cultivar, indicating that the priming was effective in enhancing seed germinability regardless of seed size. Priming aged seeds also resulted in good stand establishment in the field trials. Germination of aged seeds of Danyeobkong without priming was 17.0%, whereas that of primed ones was 66.4%.

Antifungal Activity of Root Colonizing Pseudomonas fluorescens MC07 is Responsible for Its Disease Suppression Ability (근권 정창 세균 Pseudomonas fluorescens MC07의 항진균 활성과 병 억제 능력)

  • An antagonistic bacterium, Pseudomonas flurorescens MC07 inhibited the mycelial growth of Rhizoctonia solani, Pythium ultimum, Fusarium oxysporum, and Phytophthora capsici in on potato dextrose agan (PDA) and other media. The strain MC07 conlonizes various plant roots and possesses antifungal activity. To determine the role of antifungal activity of the bacterium in disease suppression, a mutant Okm3-4 which lost its activity was isolated after screening 2,500 colonies generated by Omegon-Km insertions. The mutant Okm3-4 showed diminished growth inhibition of R. solani, P. ultimum, F. oxysporum, and Ph. capsici in vitro and had reduced suppressive effects on sesame damping.-off compared to the parental strain. In soils, accumulation of the pathogens by continuous cropping, 90% of sesame plants were killed by natural infection of damping-off whereas, only 29% of plants grown from seeds treated with MC07 were killed. On the other hand, 85% of plants died when sesame seeds were treated with the Okm3-4 cells. This indicated that antifungal activity of MC07 in vitro is directly responsible for the suppression of damping-off disease. Emergence rates of sesame seeds in pots containing diseased soil were 33%. However, MC07 treatments on seeds significantly improved emergence rates, which has similar effects of Benomyl treatment. The mutant Okm3-4 exhibited 53% of emergence rate. This indicated that antifungal activity of MC07 also affects the emergence rate of sesame seeds.

Effect of Viscosity of Binder and[ Storage Times of Pelleted Seed on Physical and Germination Characteristics of Tobacco Seeds. (종자피복용 binder 점도와 피복후 저장기간이 종자의 발아에 미치는 영향)

  • In order to improve the sowing practice, pelleted seeds of tobacco NC 82(Nicotiana tobacco L.) were manufactured in use of binders at the different levels of viscosity, and their physical properties according to pellet size and biological activity in seed germination were investigated while storage time elapsed. Proper range of the binder viscosity for the pellet formation was 20.3-37.2 m.pas. At the high level of viscosity(45.7 m.pas) took longer time to form the pellet and pelleting was not easy. The high binder viscosity and large pellet size revealed higher level in hardness of the pelleted tobacco seeds, which caused the longer splitting time of pellets in water. High level of binder viscosity(37.2 m.pas) in pelleted seeds led to decrease the biological activity of tobacco seed germination by the storage at 4 t over 30 days. But at the level of 20.3m.pas in binder viscosity, the germination of pelleted seeds was little affected by the long storage time to 120 days.

Determination of Germination Quality of Cucumber (Cucumis Sativus) Seed by LED-Induced Hyperspectral Reflectance Imaging

  • Purpose: We developed a viability evaluation method for cucumber (Cucumis sativus) seed using hyperspectral reflectance imaging. Methods: Reflectance spectra of cucumber seeds in the 400 to 1000 nm range were collected from hyperspectral reflectance images obtained using blue, green, and red LED illumination. A partial least squares-discriminant analysis (PLS-DA) was developed to predict viable and non-viable seeds. Various ranges of spectra induced by four types of LEDs (Blue, Green, Red, and RGB) were investigated to develop the classification models. Results: PLS-DA models for spectra in the 600 to 700 nm range showed 98.5% discrimination accuracy for both viable and non-viable seeds. Using images based on the PLS-DA model, the discrimination accuracy for viable and non-viable seeds was 100% and 99%, respectively Conclusions: Hyperspectral reflectance images made using LED light can be used to select high quality cucumber seeds.

Seed Deterioration Response of Different Genes of Sweet Corn during Long-tenn Storage

  • Sweet com seeds deteriorate faster due to low starch content than field com seeds when stored for a long tenn. This study had been conducted to observe the seed deterioration of four different sweet corns in a long tenn storage conditions in room temperature. Four kinds of sweet com genes (sh2, bt, su, and se) were harvested from 15 days to 50 days after silking with 5-day intervals. These seeds were stored in the room temperature and tested for germination percentages from 3 months to 18 months period with 3-month interval. su seeds germinated better than other types of gene. Hybrid Mecca which is sh2 gene germinated better when stored for 3 months to 18 months. For all genes, mean regression equations in relation to storage periods showed linear responses. For regression equation, the slope of sh2 gene was lower than that of su gene. The highest slope value was observed in bt gene showing faster deterioration rate. The rate at which seed deteriorates seems to be affected by the date at which it was harvested. The seeds that were harvested at the optimum time deteriorated more slowly than those which were not.
