Phosphate solubilizing bacteria (PSB) were isolated from the rhizosphere of Chinese cabbage and screened on the basis of their solubilization of inorganic tricalcium phosphate in liquid cultures. Ten strains that had higher solubilization potential were selected, and they also produced indole-3-acetic acid, 1-aminocyclopropane-1-carboxylate (ACC) deaminase, and siderophores. The strains were identified to be members of Pseudomonas, by 16S rDNA sequence analysis. Seed bacterization with PSB strains increased the root elongation and biomass of Chinese cabbage in seedling culture, although they had no effect on phosphorus uptake of plants. The plant growth promotion by PSB in this study could be due to the production of phytohormones or mechanisms other than phosphate solubilization, since they had no effect on P nutrition.
High temperature stresses have caused growth inhibition and delayed heading in highland cultivation Chinese cabbage during summer in Korea. We have studied high temperature stress responses in the terms of changes of inorganic components and proteins by proteomic analyses. Insufficiencies of nitrogen and phosphorus have affected growth rate and calcium deficiency has caused blunted heading. Proteins extracted from Brassica seedling grown at the altitude of 600m and 900m in the Mount Jilun were extracted and analysed by 2-dimentional polyacrylamide gel electrophoresis. Profiles of protein expression was then analyzed by 2-dimentional gel analyses. Protein spots showing different expression level were picked using the spot handling workstation and subjected to MALDI-TOF MS. Total 48 protein spots were analyzed by MALDI-TOF MS and 30 proteins spots out of 48 were identified by peptide mass fingerprinting analyses. Fourteen proteins were up-regulated in extracts from the altitude of 900m and they were identified as oxygen-evolving proteins, rubisco activase and ATPase etc. Sixteen proteins were up-regulated in extracts from the altitude of 600m and they were identified as glutathione S-transferase(1, 28kD cold induced- and 24 kD auxin-binding proteins) and salt-stress induced protein etc. These stress-induced proteins were related to the mediated protective mechanism against oxidative damage during various stresses. The results indicated that physiological phenomenon in response to high temperature stresses might be resulted by complex and multiple array of responses with drought, heat, oxidative, salt, and cold by high temperature.
Plants response to phosphate starvation include the changes of activity of some enzymes, such as phosphatases, fructose-1,6-bisphosphatase, sucrose-phosphate synthase and nitrate reductase. In this study, to determine the effects of phosphate starvation on the change of activities of acid and alkaline phosphatase, fructose-1,6-bisphosphatase, sucrose-phosphate synthase, and nitrate reductase were studied in melon seedlings (Cucumis melo L.). The content of the protein and chlorophyll tended to relatively reduced in melon seedlings subjected to phosphate starvation. Acid phosphatase activity in first and second leaves of melon seedlings was relatively higher than that of third and fourth leaves of seedlings in 14 days after phosphate starvation treatment, respectively. Active native-PAGE band patterns of acid phosphatase in melon leaves showed similar to activities of acid phosphatase, whereas alkaline phosphatase activity was different from the change in the activity of acid phosphatase. Inorganic phosphate content in melon seedlings leaves was constant. The changes of Fructose-1,6-bisphosphatase and sucrose phosphate synthase activities showed similar patterns in melon seedlings leaves, and between these enzymes activities and phosphate nutrition negatively related. Fructose-1,6- bisphosphatase and sucrose phosphate synthase activities showed significant difference in second and fourth leaves, but nitrate reductase showed significant difference in first and second leaves in 14days after phosphate starvation treatment. We concluded that phosphate nutrition could affect the distribution of phosphate, carbon and nitrogen in melon seedlings.

The inhibitory substance on the growth of mung bean seedling was amorphous crystal that had a single spot on silica gel thin layer chromatography under ultra-violet irradiation in dark room. The Rf value was 0.85 in n-butanol-acetic acid-formic acid-water(15 : 12 : 3 : 10) system, and 0.92 in n-butanol-acetic acid-water(4 : 1 : 5) system, respectively. Biochemical reaction of the inhibitor was negative to $FeCl_3$, pyrimidine, ninhydrine, folin, fehling, Salkowski and Ehrlich reaction, but the inhibitor had a blue fluorescence from irradiation of ultra-violet rays and had maximum absorption at 252nm on UV spectrum. On IR spectrum, the Peaks of the inhibitor appeared at $3300{\sim}3500,\;2900{\sim}3000,\;1600{\sim}1700,\;and\;1400cm^{(-1)}$. The strain tested was identified to be similar Streptomyces iuteogriseus by morphorogical and physiological characteristics.
Rice seedlings were used to examine the effect of UV-B radiation on leaf elongation and development. Leaf elongation in both rice seedlings showed differently depending on each leaf age. UV-B radiation strongly reduced leaf elongation, 58-66% compared to without UV-B radiation, of two rice seedlings, therefore, those seedlings could not grow further. Both control and plants grown under UV-B regime showed a diurnial fluctuation in growth rate, showing maximum growth during the light period and minimum during the dark period. Leaf growth at the third leaf stage by UV-B treatment was considerably reduced by 1.7-fold than the control whereas at the fifth leaf stage was not changed. Hydrogen peroxide was considerably increased in the second phase of UV-B-induced response as catalase and peroxidase are deactivated with an increase of UV-B radiation.

Sorghum (Sorghum bicolor (L.) Moench.) is one of the most important crops for human and animal nutrition. Nonetheless, sorghum has a cyanogenic glucoside compound which can be degraded into hydrogen cyanide, toxic to humans and animals even with tiny amount. In consequence, breeding materials with a low cyanide level has been a top priority in sorghum breeding programs. To fulfill our long-term goal, we are screening sorghum accessions with low cyanide level, which would be an important breeding material for food safety. We collected seeds of various sorghum accessions and analyzed relevant metabolites to find useful breeding materials of sorghum accessions containing low cyanide. Fourteen wild relatives were obtained from the University of Georgia in US, a reference accession BTx623, and three local varieties from National Agrobiodiversity Center of Rural Development Administration in Korea, and one wild species from the Wild Plant Resources Seed Bank of Korea University in Korea. Sorghum plants were grown in plastic greenhouse under natural conditions. After growing, leaf samples were harvested at different developmental stages: seedling phase, vegetative phase (right before flowering), and reproductive phase (ripening). Using collected samples, quantification analysis were performed by an HPLC system for three metabolites (dhurrin, 4-hydroxybenzaldehyde, and 4-hydroxyphenylacetic acid) in sorghum plants. Prior to metabolome analysis, specific experimental condition for HPLC system was set to be able to separate three metabolites simultaneously. Under this condition, these metabolites were quantified in each accession by HPLC system. We observed that the metabolite contents were changed differently by developmental stages and accessions. We clustered these results into five groups as patterns of their contents by developmental stages. Most of accessions showed that 4-hydroxybenzaldehyde content was very high at seedling stage and decreased rapidly at vegetative phase. Interestingly, the patterns of dhurrin content were very different among clusters. However, 4-hydroxyphenylacetic acid content was maintained at low levels by developmental stages in most accessions. The results would demonstrate how dhurrin and alternative degradation pathways are differentiated in each accession.
Differences in competitive characteristics for mineral nutrition were compared with rice and weeds according to rice planting types like the traditional hand transplanting, machine transplanting of young(10days old) and mature(30days old) seedling, flood direct seeding of germinated seeds and dry direct seeding of dry seeds. There was a slight competition for such nutrients as N, P, K, Ca, and Mg by E. crus-galli and E. kuroguwai in transplanting cultivations. However, nutrient uptake by rice in direct-seedings was severely inhibited throughout the whole growth period by the two weeds including annual weeds such as S. Juncoides and C. difformis. The competition for nutrients at different cultivation types was mainly governed by the dominant weeds.

This study was carried out to analyze ecophysiological responses for seedling of Quercus serrata, Quercus mongolica and Quercus variabilis that are the typical species of deciduous broadleaved forests in Korea. And executed experiments in the climatic control room to provide necessary information to ecological forest management and methods of natural regeneration through the analysis of seedling responses. The details of experimental analysis were growth processes of 4 months after seeding that vary with the condition of three light intensity[relative light intensity(RLI) 8%, 20%, 52%] and three soil moisture[water suction(WS) Ψ=100 hPa, Ψ=280 hPa, Ψ=330 hPa] gradient, growth factors after harvesting and the nutrition condition of leaves. The results of this study are followings: 1) Early growth was prosperous after germination for the species which have more weight of acorn. 2) The formation of lammas shoot was favourable with Q. variabilis and Q. mongolica. And the rate of the occurrence was the highest in the RLI 20%, and it was remarkably reduced in the RLI 8%. 3) As the height growth of seedling of all 3 species were greater in the RLI 20% and 8% than that of the RLI 52%, they showed strong shade tolerance. 4) The increase of light intensity promoted the diameter at root collar growth, and development of main and lateral roots with all 3 species. 5) It showed that the increase of light intensity in the experimental radiation condition raised special leaf area weight(mg/cm$^2$) and leaf area productivity(mg/cm$^2$). Consequently, these resulted in the increase of leaf thickness and total dry biomass per the unit area of leaf. 6) As the increase of light intensity, the minerals contents of leaves such as N, P and K were lowered, and the increase of soil moisture resulted in the increase of P, K, Ca and Mg.
The mobilization of proteins in the cotyledons of germinating soybean seeds (Glycine mar [L.] Merr.) and seedlings was studied by using light microscopy and transmission electron microscopy. The cotyledon tissues of soybean. were packed with protein bodies(diameter $0.1-15{\mu}m$) where storage protein of soybean is deposited. Degradation of protein bodies started in the epidermis and vascular tissues. After swelling of the protein bodies, autolysis of storage proteins began while the external membrane remained unbroken. Hydrolysis of proteins could be internal or peripheral and fusion might begin before complete protein degradation. Possible instances of vacuolar fusion were encountered in some cells. In all cases, the result of degradation was the same; the central vacuole of the cell. At the late stages of seedling growth, breakdown of tonoplast was observed in some cells.
Seedlings of two rice cultivars i.e. cv. Daesanbyeo and cv. Dongjinbyeo were analyzed for growth and endogenous gibberellins (GAs) in response to nitrogen nutrition applied in the forms of $KNO_3$, $(NH_4)_2SO_4$ and $NH_4NO_3$. All the growth parameters showed an increase in N applied treatments and their magnitudes of increase were different depending on different nitrogen fertilizer forms. The endogenous GAs contents were increased with N application but differentially affected by various N-forms in both rice cultivars. In cv. Daesanbyeo, maximum amount of bioactive $GA_1$ was recorded for $(NH_4)_2SO_4$, while maximum amount of bioactive $GA_1$ in cv. Dongjinbyeo was observed in $NH_4NO_3$ applied treatments. In both rice cultivars, $KNO_3$ applied rice plants contained least $GA_1$ contents. Also, $GA_{19}$ was the most abundant GA found in rice seedlings whereas $GA_4$ was absent at seedling stage. Our study indicated that different rice cultivars showed different responses for the same fertilizer depending upon the response potential of each cultivar and the pertinent physiological responses to changes of endogenous GAs in rice cultivars, which were comparatively lesser in magnitude.
