Effect of Storage Duration and $GA_3$ of Seed Germination of Celosia argentea (개맨드라미 종자 수확 후 저장 기간 및 $GA_3$처리가 발아율에 미치는 영향)

  • This experiment was conducted to improve germination rate of the seed treated duration of storage and $GA_3$ in Celosia argentea. With the seed directly after harvest, as increasing concentration of $GA_3$, ranged from 50ppm to 200ppm, and dipping hour, ranged from 6 to 24 hour, the rate of germination was also increased. The germination of seed dipping in 200ppm $GA_3$ for 24 hours was the highest with $52\%$ compared to the control with $12\%$. As increasing storage duration in $22{\pm}2^{\circ}C$, ranged from 2 weeks to 12 weeks, the rate of germination was also increased showing $84\%$ at 12 week. However, the rate of germination was not increased with the storage of $5^{\circ}C,\;0^{\circ}C,\;and\;-70^{\circ}C$. With the seed storage 16 weeks in $22{\pm}2^{\circ}C$ after harvest, the rate of germination was $84\%$. The rate of germination was increased $94\%\;to\;99\%$ by the $GA_3$ concentration, ranged from 50ppm to 400ppm, and dipping hour, ranged from 6 to 24 hour. The germination was finished 4 days after sowing.

Effects of Natural Plant Materials on Color and Lipid Oxidation of Ground Pork (식물 소재 첨가가 분쇄 돈육의 육색 및 지방 산화에 미치는 영향)

  • The principal objective of this study was to assess the effects of the addition of natural plant materials on the color and lipid oxidation of ground pork. Ground pork was blended with 0.3% (w/w) of cactus, fennel seed, orange peel, and rosemary, respectively, and stored for 8 days at $2^{\circ}C$. The pH, meat color, thiobarbituric acid reactive substances (TBARS) values, and metmyoglobin (MetMb) contents were measured. A gradual increase in pH was noted with increases in the storage time. The Hunter a (redness) value decreased until 6 days of storage, and evidenced no noticeable changes there after, and the cactus evidenced significantly higher a values than other groups over time (p<0.05). The TBARS formation was effectively inhibited by the addition of rosemary for 6 days of storage, reflective of strong antioxidative activity (p<0.05). The antioxidative activity on the lipid oxidation of cooked pork patties was noted in the following order: rosemary, fennel seed, cactus, and orange peel. The MetMb contents increased according to storage period, and no significant differences were noted among the meat patties.

Physicochemical Properties of Ground Pork with Safflower (Carthamus tinctorius L.) Seed during Refrigerated Storage (홍화씨가 분쇄돈육의 냉장 중 이화학적 품질에 미치는 영향)

  • This study was carried out to investigate the effect of safflower seed powder on the physicochemical characteristics of ground pork during refrigerated storage. Three types of ground pork were evaluated: 20% pork fat added (PF, control), 10% pork fat and 10% added safflower seed powder (PFS), and 20% added safflower seed powder (SS). The pH increased during storage, but decreased after 10 days of the storage (p<0.05). The pH was lower in PFS and SS than that in PF after 10 days of storage (p<0.05). The TBARS (2-thiobarbituric acid reactive substances) values increased with longer storage period (p<0.05), and those of PF, PFS and SS were 1.186, 0.686 and 0.577 mg MA/kg, respectively, after 10 days of storage. The $L^*$ values for external color of PF and PFS decreased (p<0.05), but that of SS was not significantly different after a longer storage period. The $a^*$ values decreased (p<0.05), but the $b^*$ values were not significantly different with longer storage period. The $L^*$ values for internal color of PFS and SS decreased (p<0.05), but that of PF was not significantly different with longer storage period. The $a^*$ value of PF decreased (p<0.05), but that of SS increased with longer storage period. The $b^*$ value decreased (p<0.05), but those of PFS and SS were not significantly different with longer storage period. Water holding capacity decreased with longer storage period, and that of SS was the highest (p<0.05). Cooking loss of PFS and SS was not significantly different with longer storage period, and that of PF was the highest (p<0.05). The reduction in diameter of the samples was not significantly different with longer storage period, and that of PF was the highest (p<0.05). Hardness and chewiness of samples increased, but springiness and cohesiveness decreased with longer storage period (p<0.05). Replacing animal fat with safflower seed powder was effective and may be useful as an innovative meat product.

Seed Structure and Effects of Storage on Germination of Astragalus membranaceus (황기 종자구조 및 저장이 휴면에 미치는 영향)

  • This experiment was conducted to examine the seed structure and to investigate the changes of seed vigor and germinability according to storage condition of Astragalus membranaceus. In case of the hard seed, there observed a gap between a testa and an embryo with abnormal shapes of an hilum and its micropyle. Seeds stored at room temperature maintained vitality for 4 months but continued to lose its vitality, while increase the hand seeds ratio as storing time longer. The content of Ca increased in proportion to the length of storing time.

Effect of Seeds Treatment on Germinablity of Tetragonia Tetragonides Seeds (번행초의 대량번식을 위한 종자처리가 발아력에 미치는 영향)

  • Tetragonia tetragonides is a medicinal plant native to ocean sand soil of southern provinces and has significant effects on the prevention and curing of gastroenteric disorders. Despite of its popularity, supply of the plant has never met the level of demand because of the absence of an adequate culturing method. The present study, thereby, was conducted for classifying the plants with geographically different characteristics, studying growth habits, developing a new culturing method and establishing a large scale propagation system of selected superior individual plants. The study was also aimed for revealing optimum conditions for seed treatment, fertilization, and efficient culturing system and thereby, for utilizing the plant as a new income source for rural communities. The seed was elongated with size of 2.6 mm (width) ${\times}$ 1.8 mm (length). No difference in seed size was observed depending on different inhabitate. Each flower produced about 4.5~4.8 seeds. Germination rate was high for seeds matured for 40 days after fertilization, but deceased to 50% for seeds matured only for 20 or 30 days. Seed dormancy lasted 6 months and seed storage at humid $5^{\circ}C$ facilitated germination. Mechanical obstruct of seed germination was due to seed coat and removal of seed coat enhanced the germination rate. Optimum temp. for seed storage was $5^{\circ}C$, and high germination rate was maintained for 350 days. However, for stratification condition or at room temperature, germination was significantly reduced as storage time increased Optimum treatment of plant growth regulators was soaking in $GA_3$ 250 mg/L for 1 hr. The priming treatment with 50 mM $Ca(NO_3)_2$ at $20^{\circ}C$ for two days improved the seed germination with 10% compared to non-treated control. The treatment of 20% NaOCl for 3 hr. improved the seed germination rate up to 10% and 1 day ahead.

Desiccation Tolerance and Storage Behavior of Sarcandra glabra Seeds for Use in ex-situ Conservation (현지외 보존을 위한 죽절초 종자의 건조 내성과 저장성 평가)

  • This study sought to determine the desiccation tolerance and storage behavior of Sarcandra glabra seeds to assist in the establishment of an ex-situ conservation strategy for the species. The basic germination characteristics of S. glabra seeds were investigated. Subsequently, the seeds were dried to various desiccation levels to allow for analysis of their germination characteristics and seed integrity. In addition, the seeds were subjected to germination tests after being stored for different durations. The initial seed moisture content following the removal of the fleshy fruit was 32.8%, and the fresh seeds maintained their seed viability even after a decrease in the seed moisture content to 4.8%, while the germination percentage was 81.0%. The germination percentage of the seeds decreased to 55.0% and 30.0% when they were desiccated to moisture contents of 2.3% and 2.2%, respectively. By contrast, when the stored seeds were desiccated, they maintained their seed viability and vigor at a moisture content of 18.6%; however, when the moisture content decreased below 5.0%, both the seed viability and vigor significantly declined and the germination percentage fell below 20.0%. Furthermore, analysis of the seed leachate revealed that the decrease in the seed viability and vigor was associated with both increased electrical conductivity and increased concentrations of inorganic compounds such as potassium and calcium. In conclusion, the findings of this study show S. glabra seeds to possess desiccation tolerance that falls somewhere between the recalcitrant and intermediate seed types, with the observed variations depending on the degree of deterioration.

Chemical Components of Seed Oil of Sapium japonicum Pax. et Hoffm. (사람주나무 종실유의 화학적 조성)

  • Physico-chemical characteristics and chemical composition of seed oil of Sapium japonicum were determined by various analysis methods. Physio-chemical characteristics of the extracted oil from fresh seed were specific gravity (0.928), refraction index (1.477), acid value (2.30), saponification number (190.0), and iodine value (126.0). Neutral lipid (93%) in seed oil was identified as major components, followed by glycolipid (4.9%) and phospholipid (1.3%). In GC analysis, nine fatty acids were presented in the seed oil. Among fatty acids, predominant fatty acids were oleic acid (45.8%) and linoleic acid (35.6%). The contents of fatty acids of seed oil somewhat varied with their storage period. Composition of fatty acids from lipid fractions which were isolated on silica open column was investigated. Major fatty acids in three lipid fractions were linoleic acid and linolenic acid. The fatty acid contents of fresh seed oil derived from each lipid fraction were generally high comapred to storage seed oil. From the above results, seed oil of Sapium Japonicum could be useful in cosmetics, detergents and a few pharmaceuticals.

Variations in Seed Storage Protein among Different Colored Soybean Varieties

  • This study was carried out to know the variation of soybean seed proteins, 11S and 7S globulins, and their amino acid compositions among different colored soybean varieties, 'Danbaegkong' (yellow), 'Pureunkong' (green) 'Jinyulkong' (brown), and 'Geoumjeongkong l' (black). Soybean seed proteins showed a wide range in molecular size, but the electrophoresis patterns of total seed protein subunits showed a similarity among different colored soybean varieties. Amino acid compositions of total seed proteins were similar for all soybean varieties tested. However, soybean varieties showed low composition rates in sulfur containing amino acids. The composition rates of cysteine and methionine in the 11S globulins were higher than those of total seed proteins and 7S globulins. Glutamic acid and glycine were higher in the 11S and 7S globulins than those of total seed proteins. However, the levels of methionine and phenylalanine are high in the 11S globulins, but those of valine and lysin are slightly lower than the 7S globulins. By using HPLC, we tried to analyse the soybean seed proteins. The 11S globulin was composed of 10 major peaks whereas the 7S globulin was composed of 4 major peaks. The composition rates of 11S related proteins have a tendency to increasing during the maturing whereas those of 7S related proteins have a tendency to decreasing. Composition rates of each peaks among different colored soybean varieties suggested that soybean seed proteins are varied, although they showed similarity in the electrophoresis patterns, and understanding of this characteristics is important for the utilization of soybeans.

Germination Characteristics of Chinese Milk Vetch(Astragalus sinicus L.) Seeds Produced in China and Korea (수입 및 국내 채종 자운영 종자의 발아 특성)

  • Germination characteristics of Chinese milk vetch(CMV) seeds produced from Hanam Province, China and Milyang, Korea were investigated to give basic information on the stability of seedling establishment in the CMV cultivation. The germination percentage of the imported CMV seed from China varied according to importation year and seed collection site ranging from 79~95%. The germination of black colored seed coat was lower than the light green colored ones and germination by seed weight was not significantly different. Although the seed germination was lower under dark than in the presence of light, it was not significantly different. The germination of the imported CMV seed slightly declined to only less than 6% after one-year of storage under natural environment conditions but it significantly decreased after two years. However, when the seed was stored at the $5^{\circ}C$, the seed germination was the same as after two years of storage. On the other hand, fresh CMV seed produced in Milyang, Korea had only 8% germination due to seed coat dormancy but the germination increased to 73~85% after breaking seed dormancy after a year of storage. The high germination percentage of 72~82% was still maintained even after 27 months of seed storage unlike the CMV seed produced from China. These results indicate that CMV seeds do not require light for germination and the seed from China should be used within one-year after importation while the seeds produced from Korea can be used even after two years from harvest for stable CMV seedling establishment in the CMV-rice cropping system.
