Separation of Trichlorinated Hydrocarbons by ZSM-5 Zeolite Membrane (ZSM-5 제올라이트 분리막에 의한 3가 염소화합물의 투과증발 분리)

  • MFI(Mobil Five) structured hydrophobic ZSM-5 zeolite membrane was used for selective pervaporation of trichlorinated organic compounds(trichloromethane, trichloroethane, trichloroethylene) from their aqueous solutions. ZSM-5 zeolite membrane was hydrothermally synthesized on the inside of a porous stainless steel tube by secondary growth employing ZSM-5 seed powders. Separation factors for each binary mixtures were observed $16{\sim}66$ for trichloromethane/water, $3.3{\sim}4.6$ for trichloroethane/water and $1.4{\sim}8$ for trichloroethylene/water at the experimental conditions of the feed mole fraction from 0.0001 to 0.001 with temperature ranged $25{\sim}35^{\circ}C$.

Capillary Electrophoretic Method for the Determination of (+)-Catechin, (-)-Epicatechin in Grape Seed Ethanol Extract (포도종실 에탄올 추출물에 함유된 (+)-카테킨, (-)-에피카테킨의 모세관 전기영동법에 의한 분석)

  • Capillary electrophoresis (CE) method was developed to determine (+)-catechin and (-)-epicatechin contents in grape seed ethanol extract. CE separation was achieved using 100 mM phosphate and borate buffer at pH 6.0 as background electrolyte and fused silica capillary with 50 microns x 375 microns O.D. (effective length 20.0cm) maintained at $25^{\circ}C$. The applied voltage was 10kV, and detection was performed by DAD at 210 nm, Two catechins were well separated within 6 min with repeatability of <0.8% RSD for migration time and <2.0% RSD for peak area, and correlation coefficients higher than 0.994 were obtained from 58.0 to 174.0 mg/L with detection limit of 0.035 mg/L. Separated compounds were successfully determined. CE method was easy to handle and showed good reproducibility. CE method was compared with conventional coloring and HPLC methods, and main advantages of CE method were low amount of sample required, simple pre-sample treatment, good recovery rate, and short analysis time.

  • This study has main purpose to understand and interpret the house Nok-U-dang, an upper class built in Chosun dynasty, not by outsider researcher's view but insider dweller's view. To interpret correctly, dweller's everyday life in the space and form is examined on a microscale beyond the physical space and form of the house, main object of architectural history To understand the present form exactly, the study restore traditional life in past era, 1940s. Main method of restoration is the ethnographic interview, based on cultural anthropology. Like any other upper-class house, the house has been influenced under ruling Confucian ideology in Chosun dynasty: separation of man's and woman's quarters and hierachical arrangement by generation, and worshipping ceremony for ancestor. However, it is by practical management for agricultural production that every court and building of the Noku-Dang can be explained correctly; preparing seed for sowing, tool storing, preparing and serving meal for laborers, making manure, harvesting, threshing grain, storing grain and so on. Precedent studies interpreted the house by the Confucian principle too much and made conclusion of dignity and austerity of ritual: woman's quarter, is closed and serene space. However this study shows that the space is semi-opened and composite space by agricultural works. And the Sarangchae, master's quarter, is located properly at visual center to control every agricultural activity.

Mixing Effect by Tone-Excitation In Round Jet Diffusion Flame (원형분류확산화염에서의 음파가진에 의한 혼합효과)

  • An experimental investigation has been conducted with the objective of studying the mixing mechanism near the nozzle exit in a tone-excited jet diffusion flame. The fuel jet was pulsed by means of a loudspeaker-driven cavity. The excitation frequencies were chosen for the two cases of the non-resonant and resonant frequency identified as a fuel tube resonance due to acoustic excitation. The effect of tone-excitations on mixing pattern near the nozzle exit and flame was visualized using various techniques, including schlieren photograph and laser light scattering photograph from $TiO_2$ seed particles. In order to clarify the details of the flame feature observed by visualization methods, hotwire measurements have been made. Excitation at the resonant frequency makes strong mixing near the nozzle. In this case, the fuel jet flow in the vicinity of nozzle exit breaks up into disturbed fuel parcels. This phenomena affects greatly the combustion characteristics of the tone excited jet and presumably occurs by flow separation from the wall inside the fuel nozzle. As a result, in the resonant frequency the flame length reduces greatly.

Spatial Autocorrelation within Three Populations of Sasa borealis in Korea (한국 조릿대집단의 공간적 상관관계)

  • Spatial autocorrelation was applied to microgeographic variations of Sasa borealis populations in Korea. Separate counts of each type of join (combination of genotypes at a single locus) for each allele, and for each distance class of separation, were tested for significant deviation from random expectations by calculating the Standard Normal Deviation. Moran's I was significantly different from the expected value in 25 of 150 cases $(16.7\%)$. Seven of these values $(4.7\%)$ were negative, indicating genetic dissimilarity among pairs of individuals in the ten distance classes. Populations of S. borealis are small in Korea, and are distributed with occasional cutting of seed-bearing stems used for sieves. Thus, artificial disturbance may contribute to the fact that the S. borealis population of Jirisan is unusual in lacking spatial genetic structure.

Identification and Changes of Physiologically Active Substances During Chilling Storage of Dehisced Ginseng Seeds (저온저장중 개갑인삼종자내의 생리활성물질 동정 및 변화)

  • Identification and changes of physiologically active substances during chilling storage of dehisced ginseng (Panax ginseng C. A. Meyer) seeds were analyzed using various preparatory separation methods and purification columns; Dowex 50W and silica gel columns. Seven components with Rf values of 0.20, 0.40, 0.58, 0.66, and 0.70 In solvent system, $CHCl_3$:MeOH=3:1 (v/v), Rf values of 0. 63 and 0.74 in solvent system, $CHCl_3$:MeOH:$H_2O$:=7:3:1 (v/v) were obtained through Dowex 50W and silica gel column chromatographies. Two components with Rf values of 0.20 and 0.63 in the all chilling treatments were detected in the extract obtained through both chromatographies, and only the former component was gradually increased till 4 weeks of chilling storage and then rapidly decreased from 8 to 16 weeks. UV spectra of Rf values of 0.66 and 0.56 were similar to that of cytokinin, but their physiological activities were not found. Rf values of 0.20 showed activity by radish cotyledon expansion bioassay. The component with Rf value of 0.20 was revealed to have a naphthalene in the proposed chemical structure by various NMR techniques.

An Analysis of Morphological Variation in Abies koreana Wilson and A. nephrolepis (Traut.) Maxim. of Korea (Pinaceae) and Their Phylogenetic Problems (한국산(韓國産) 분비나무와 구상나무의 형질분석(形質分析)과 종간유연관계(種間類緣關係))

  • Ten total populations of Korean fir (Abies koreana Wilson) and Manshurian fir [A. nephrolepis (Traut.) Maxim.] were sampled from south Korea to investigate patterns of intraspecific variation in these species and to evaluate a recognition of the two species. Principal components analysis and cluster analysis were performed both on seed-cone data and on needle morphology data. The characters that contributed most to the separation between A. koreana and A. nephrolepis along three principal components axis were leaf width, length of seed, width of seed wing, length of seed wing, cone width, width of scale, and length of bract tip, but these characters were not diagnostic because of overlap in reality. Therefore, all these characters were not reliable in distinguishing these two taxa including bract position (exerted and recurved vs. exerted and straight). The individuals of A. koreana from Mt. Chi-ri appeared quite unique probably on account of its larger cone size and longer scale tip, while those from Mt. Hal-la of A. koreana were generally distinct from others in terms of their larger seed and seed wing and longer scale width. The Mt. Duk-yu specimens of A. korecana appeared somewhat smaller but more data were needed due to the small sampling size. Generally, the gradual clinal geographic trends made evident by the position of resin ducts in leaves of A. koreana can be detected. The southern populations, Mt. Hal-la (an insular population) were generally distinct from the northern populations (Mt. Chi-ri, Mt. Ga-ya and Mt. Duk-yu) in terms of their position of resin duct (medial, within mesophyll vs marginal, close to epidermis : 100% vs 75 or 50%). Although no sharp boundary separating these two species could be detected based on cone and needle morphology, the observed clinal pattern was distinct in northern populations of A. koreana and southern population of A. nephrnlepis. In a preceding study of the flavonoids variation of 20 species in eastern Asia, flavanone (5-deoxyflavanone) was found to be characteristic of A. faxoniana Rehder et Wilson, A. georgei Orr of China and A. koreana of Korea. A. faxoniana, which is assumed to be primitive species, has position of resin duct relative to both the medial and the marginal, while A. georgei and A. koreana are identified by marginal position of resin duct. With respect of foliar flavonoids chemistry, A. koreana was distinct from A. nephrolepis : the southmost samples (Mt. Hal-la and Mt. Chi-ri) contained additional flavonoids derivatives (mainly flavanone) that were not found in the northmost samples of A. nephrolepis except a few individuals from Mts. Seo-rak and Tae-bak populations of Kwang-won province. The presence of A. koreana type flavonoids in two Chinese species suggested that position of resin duct may be a phyletic character. Abies koreana including two Chinese taxa, exhibited the most elaborate and specialized flavonoids profile within the Abies in eastern Asia. Contrary to our initial expectations, the apparent intermediates between A. nephrolepis and A. koreana in Duk-yu and Ga-ya mountains were found. The pattern of variation on position of resin duct and flavonoids chemistry in these populations of A. kareana suggested that genetic interchange or natural hybridization had occurred between these two species. The evidence needed to resolve the status of this taxon is still inconclusive in our opinion until intermediate individuals from Mts. Duk-yu and Ga-ya show indication of hybridization between the two species.

Seedling Emergence and Growth Affected by Priming and $GA_3$ Treatments to Three Campanulan Plant Seeds (Priming과 $GA_3$ 처리에 따른 도라지, 더덕 및 만삼의 출아(出芽)와 생장(生長))

  • Seed germination test done in laboratory does not coincide with field emergence in general. The experiments were carried out to examine the effect of priming and $GA_3$, treatment to seeds of Platycodon grandiflorum; Codonopsis lanceolata and C. pilosula on lapsed time to first seedling emergence, seedling emergence, morphological characters and growth and the cause of poor emergence of C. pilosula. No-treatment as Control (water), priming or $GA_3$ treatment was done with only distilled water for 2 days, $CA(NO_3)_2$ 150 mM for 2 days or $GA_3$ 0.1 mM for 3 days, respectively. Seedling emergence rate was counted every 2 days but morphological characters and dry weight of shoot and root were measured on 38 days after sowing. Their internal seed structures were examined with Scanning Electron Microscope. C. pilosula had poorer seedling emergence rate than P grandiflorum and C. lanceolata showing nearly same rate: Compared to the other treatment (s) P. grandiflorum displayed higher rate in priming and $GA_3$, treatments but C. lanceolata or C. pilosula did the greatest rate in only $GA_3$ or priming treatment, respectively. $GA_3$ treatment to seeds of P. grandiflorum and C. lanceolata shortened the lapsed time to seedling emergence in comparison with Control, 2-days water imbibition before sowing. In all the species plant height and number of leaves per seedling became shorter and less in priming treatment than the other treatments except plant height of C. Pilosula while their hypocotyl length was nearly same in all treatments. Although priming treatment had nearly similar effect to morphological characters, $GA_3$ treatment forced greater shoot, root and aftermath total dry weight per seedling. Poor seedling emergence of C. pilosula was caused by its seed defect like cleavage or lack of embryo, poor development of embryo and endosperm or their separation.

Industrial Utilization and Function of Omega Fatty Acid and Their Content Variation in Perilla (들깨 오메가 지방산의 기능과 함량변이 및 이용)

  • The n-3 family fatty acids containing ${\alpha}$-linolenic acid(18:3, ALA) have been known as physiological activation materials such as inhibitory effects on the incidence of hyper-tension, coronary heart disease and cancers as well as the control of senilc dementia. Although a lot of ALA(about $63\%$) are contained in perilla oil, it has not been commercialized yet because the purification technique of the ALA has not been well established. The procedure of purification of ALA from perilla oil was saponified with 1 N-KOH /ethanol and then saturated and low level unsaturated fatty acids were removed by low-temperature crystallization method. The concentrated unsaturated fatty acids (containing about $75\%$ ALA) went down through the silver nitrate-impregnated silica column chromatography for separation of high purity of ALA. The results obtained we Fraction B, C and D contained ALA more than $85.5\%$(recovery, >$88.9\%,\;95.4\%$(recovery, >$54.4\%$) and $99.9\%$(recovery, >$31.5\%$) in purity, respectively. Seed oil content of the tested varieties were ranged from 34.8 to $54.1\%$ with $45.3\%$ of varietal means. The major omega fatty acids contained in the oil were oleic acid(n-9) $15.2\%$, linoleic acid(n-6) $13.9\%$ and linolenic acid(n-3) $63.1\%$ in the mean value. Varietal variation of n-9, 6 and 3 fatty acids ranged of $9.5\~21.4\%,\;9.1\~20.4\%$ and $50.6\~70.5\%$ respectively. Unsaturated fatty acid were averaged $92.2\%$ of seed oil in fatty acid composition. The ratios of n-6 to n-3 ranged of $0.13\~0.34\%$($0.22\%$ in mean value). The highest n-3 fatty acid variety was Yecheonjong being $70.5\%$. The lowest variety in ratios of n-6 to n-3 was Goseongjong being $0.13\%$. Oil content showed positive correlation with stearic acid and linolenic acid, while the negative correlation with oil content and linoleic acid. On the other hand, A significant negative correlation were showed between linolnic acid and the ratios n-6/n-3 fatty acid, saturated fatty acid. Saturated fatty acid was highly correlated with unsaturated fatty acid negatively being $r= -0.723^{**}$.

Metabolic Pharmacokinetics in Rats: Differences between Pure Amygdalin and Amygdalin in a Decoction of Peach Seeds

  • The pharmacokinetics of prunasin after oral administration of amygdalin or a decoction of peach seeds was determined and compared in rats. A $C_{18}$ column was used for separation at a column temperature of $25^{\circ}C$. The mobile phase consisted of 20% aqueous acetonitrile, and the flow rate was 0.5 mL/min. After oral administration of a decoction of peach seeds, prunasin was absorbed rapidly, reaching a maximum plasma concentration ($C_{max}$) of 62.1 mg/L within 45 min. After oral administration of amygdalin, the absorption of prunasin was delayed. The $C_{max}$ of prunasin was 42.9 mg/L and was reached at 60 min. Values for the pharmacokinetic parameters of prunasin, including $T_{max}$, $C_{max}$, AUC, $T_{1/2}$, CL/F, and V1/F, were significantly different for the oral administration of amygdalin compared with that of a decoction of peach seeds.
