Seeds are an ideal repository for recombinant proteins in molecular farming applications. However, in order to use plant seeds efficiently for the production of such proteins, it is necessary to understand a number of fundamental biological properties of seeds. This includes a full understanding of promoters which function in a seed-specific manner, the subcellular targeting of the desired polypeptide and the final form in which a protein is stored. Once a biologically active protein has been deposited in a seed, it is also critical that the protein can be extracted and purified efficiently. In this review, these issues are examined critically to provide a number of approaches which may be adopted for production of recombinant proteins in plants. Particular attention is paid to the relationship between subcellular localization and protein extraction and purification. The robustness and flexibility of seed-based production is illustrated by examples close to or already in commercial production.
This study was conducted to determine feasibility of production system of Perilla frutescens leaf-stem by fertilizing of Sta-Green in pots. Germination rate of Perilla frutescens seeds collected in 2002 was $7\%$, also germination rate of seeds collected in 2003 was $62\%$, while germination rate of seeds collected in 2004 was above $93\%$. Seed germination rate of Perilla frutescens collected in 2004 were higher than seed gathering in 2002. Especially, plant growth and yield of Perilla frutescens grown in pot(The pots was filled with soil mixtures of Sta-Green and Peat Moss mixed with 40:60 ratio.) was the highest. These results indicate that leaf and stem production of Perilla frutescens can be improved by fertilizing of Sta-Green in pots.
The production of abalone seed has grown and been specialized since the 2000s with the growth of the abalone farming industry. Despite the increase in the production of abalone seeds, the sales volume of abalone seeds remained flat and competition among producers increased. This paper will analyze the management efficiency of abalone seed production fishery to diagnose the management status and improve the abalone seed production efficiency. In addition, this study is the result of the basic research on the abalone seed industry and it is meaningful to prepare a platform for further research since the management status survey and the management efficiency survey of abalone seed production fishery have not been conducted until now. The data on the farmed fish prices of abalone seeds were collected from surveys of sample fish as part of the fish seed observation project conducted by the Fisheries Outlook Center (FOC) of Korea Maritime and Fisheries Development Institute (KMI). Management efficiency analysis utilizes DEA (Data Envelopment Analysis) model. The DEA model was analyzed by classifying into CCR (Super-CCR), BCC, and SBM (Super-SBM) models according to the assumptions taking into account the characteristics of the industry. The slack considered in the SBM model was judged as possible decreases in input variables and increase in output variables. The average efficiency from the CCR model was analyzed to be 69%. The BCC model was classified into input and output orientations, and the average efficiency was 79% and 75%, respectively. There were seven production fisheries with an SE value of 1 or more, which remained unchanged in terms of size and could be benchmarked. The average efficiency of the SBM model was 59% for CRS and 66% for VRS. Under the VRS assumptions, the variable increase/decrease efficiency analysis shows that labor costs can be reduced by 37.3%, facility capacity by 18.8%, and operating costs by 8.5%. In order to improve management efficiency, Wando needs to reduce labor and management costs. In Jindo region, sales increase as well as labor cost reduction is urgent. In other regions, reduced facilities and increased sales are recommended.
For better understanding of population dynamics of Symplocarpus renifolius, some aspects of seed production were studied in natural populations for 3 years. The rate of reproducing plants (RP) was 8.06% among the whole studied. The RPs were 0.0% in leaf size class under 500cm2 per individual, and 3.6% in 500~1,000cm2, and 44.3% in over 3,000cm2. The resource allocated to sexual organ was 11.6% of total biomass at the end of growing season, and that to belowground was about 80% regradless of presence or absence of sexual organ. In the previous and the next years of seed production, the energy allocated to sexual organ didn't affect the changes of leaf size, year by year. After flowing season. Especially, a large number of sex organ were degraded in April, a period of pollination and fertilization. But the weight per seed increased along leaf size per individual. Therefore, in S.renifolius population, large individual produced large seeds rather than many seeds.
Seeds of medicinal soybean [Glycine max(L.) Merill] are characterized by a black seed coat, white stripe at hilum border, yellow cotyledon, and very small seed weight. Production of this medicinal soybean has recently increased as a consumption increased. The objective of this study was to evaluate the growth characteristics and seed yield of collected medicinal soybeans and to obtain basic information on production practices and breeding materials. The collected medicinal soybean lines were cultivated at three locations for two years. Twenty-seven lines were planted at the Research Farm, Korea University, Namyangju city, on May 23, and at the Research Farm, Kyungpook National University, Taegu, on May 20, 1995. In 1996 field experiments, forty-four lines were planted on May 25, at Research Farm, Korea University, and twenty-seven lines among those were planted on June 7 at the Research Farm of National Yeongnam Agricultural Experiment Station, Milyang city. The investigated lines had purple or white flower. Flowering and maturing dates were similar or later than those of the control cultivars. Branch number was greater for the investigated lines. One hundred-seed weight of the lines ranged from 8.5 to 15.0 g. Mean seed yields ranged from 1.54 to 2.89 MT/ha. Nine lines of the investigated medicinal soybeans showed higher yield capacity than the control cultivars. Further research should be done on the improvement of the production system and breeding program of medicinal soybeans.
Effects of soybean oil or whole cotton seed addition on conjugated linoleic acid (CLA) and performance of fattening Brahman$\times$Thai-Native cattle were studied. Eighteen fattening cattle averaging $241{\pm}24kg$ body weight and approximately 1 year old were stratified by live weight into three groups and randomly assigned by group to one of three dietary treatments. The treatments were control (concentrated 14% crude protein), control and supplemented with 170 g/d soybean oil, control plus 170 g/d of oil from whole cotton seed. All animals were weighed before and after the experimental period and 4 cattle per treatment were randomly slaughtered then carcass measurements were obtained. There were no statistically significantly differences in the final body weight, average daily gain and dry matter intake among treatments. However, the crude protein intake was significantly decreased (p<0.01) when whole cotton seed was fed compared with control and soybean oil treatments. The carcass composition and carcass characteristics were not significantly different in Longissimus and Semimembranosus muscle by feeding soybean oil and whole cotton seed compared with the control treatment. Supplementation of soybean oil increased (p<0.01) cis-9, trans-11 CLA by 116% in Longissimus muscle and by 240% in Semimembranosus muscle. However, whole cotton seed did not increase cis-9, trans-11 CLA in both muscles. The present study successfully increased cis-9, trans-11 CLA content of muscle lipids by soybean oil but not by whole cotton seed.
Seventeen females of the mud crab Scylla serrata, from the State of Kosrae, Micronesia, were transported to the Fisheries Resources Research Institute, Gyeongsangnam-do, in oxygen-filled plastic bags. After acclimatization to a $30^{\circ}C$ holding temperature, nine females were selected for seed production trials. Spawning was hastened using eyestalk ablations; however, this may not be required in commercialscale mud crab seed production. Primary spawning produced an average of 2.4 million hatched larvae, whereas secondary spawning produced 0.4 million. About 10 days elapsed between spawning and hatching and 30 days between hatching and crablet. Mass mortalities up to 90% were observed between stages zoea 1 and zoea 2 in every trial. The highest survival rate from zoea 1 to crablet was estimated at 0.25%. Most commercial shrimp hatcheries in Korea are equipped with almost all necessary facilities and could be converted easily to mud crab hatcheries, able to run three to four times per year using hatchery technologies developed for blue crabs and Chinese mitten crabs.
Current data on reproductive characters of endemic and native species are essential to provide a strategy for the conservation of these species. Red pine (Pinus densiflora Sieb. & Zucc.) is one of the dominant, native tree species in Korea, but its reproductive ecology is not well-known. In 1997, the pattern of variation in cone and seed yields contributing to the conservation of declining populations of red pines was examined. Plant height and dbh were measured, and several new cones were collected from each tagged tree after counting the number of cones on each tree. For a subset of cones sampled, the number of fertile scales, the number of seeds at three development stages (early/late aborted, and filled seed), seed wing size, wing color, and individual filled seed mass were measured. The three sites which differed significantly in mean plant size also differed in mean cone and seed production per plant. However further analyses showed that most variation in characters examined occurred among plants within sites, but not among sites. An average of 90% of the potential seeds on the cones aborted at an early developmental stage, demonstrating that early abortion is a major factor affecting the number of filled seeds per cone. Individual seed mass was the only character which exhibited significant variations among sites as well as among trees within sites. Individual seed mass was overall negatively correlated with both the percentage of late abortion and the number of old cones per plant, suggesting that both the past and current years' reproductive activities have caused variations in seed mass. The potential dispersal distance of red pine seeds is quite large. However, wing loading was correlated with seed mass and number in a complex pattern across the sites. Distribution of seeds with varied colored wings differed among sites and among trees within sites. These results suggest that red pines at different sites might possess different strategies to cope with selection pressures acting during the final phase of reproduction, from seed dispersal to establishment. Then the ‘fitted’ red pine trees at each site should be identified and managed to conserve or restore populations.
Seed production of italian ryegrass has a problem of lodging during ripening and a decrease in quality due to difficulty in drying seeds during harvest. Therefore, in order to produce high-quality Italian ryegrass in paddy fields, it was carried out to reduce the density and solve the lodging problem through transplanting. In this experiment, Lolium multiflorum cv. Kowinearly was transplanted in autumn from a paddy field in Sindong-ri, Gwansan-eup, Jangheung-gun, Jeollanam-do. var. Kowinearly was made into a bed at 90 g/box and stacked in boxes. It was transplanted on October 27th after 2 days of germination at 30℃ and 15 days of seedling and greening. When transplanting, they were transplanted at intervals of 30×14cm. The existing cultivation method, drill seedling, was sowed at a level of 50 kg/ha, and both transplanting and drilling were carried out at a nitrogen fertilization rate of 45 kg/ha. The number of ears during transplant cultivation was 1,016/m2 and the drilling tended to be higher at 2,278/m2, but this was probably due to the difference in seeding amount. The seed number of an ear tended to be 56% higher in transplantation, which had a significant impact on yield. The seed yield was 2,096 ka/ha in transplantation, which was 21% higher than that of drilling. When looking at the relationship with weed occurrence, there were areas where all the weeds, such as amul foxtail, occurred due to the low density. Even in the same transplanting area, the seed yield was about 1,000kg/ha less in the area where the weeds were abundant. It seems that weed management is important in paddy cultivation. Therefore, it seems necessary to develop an exclusive herbicide for Italian ryegrass cultivation.
Codium fragile (Suringar) Hariot, an edible green alga is farmed in Korea by natural blooming zygotes attachment. Experiments were conducted to reveal the conditions for artificial seed production of C. fragile by sexual and asexual reproduction. Growth was compared between zygotes attachment (sexual reproduction) and isolated utricles with medullary filaments (asexual reproduction). Zygotes and isolated utricles with medullary filaments were cultured under different light conditions (10, 20, 40, 60 and $100\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and temperatures (5, 10, 15, 20 and $25^{\circ}C$) under 16:8LD. Maximum growth of zygote was $261.3{\pm}21.0\;{\mu}m$ under $15^{\circ}C$ and $20\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ after 13 days culture. Maximum regeneration of isolated medullary filament was $8.1{\pm}1.7\;mm$ per one isolated utricle under $20^{\circ}C$ and $100\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ after 15 days culture. After intermediate culture during two months in the field, morphogenesis occurred in both sexual and asexual reproduction, and growth of young thalli was not significantly different (p>0.05) between the both reproduction methods. Even though seed production of C. fragile is possible in both sexual and asexual reproduction, the mass artificial seed production of asexual reproduction is much more effective than that of sexual reproduction that is too much affected by maturity.
