Human sebocyte-based assay system for the screening of compounds to lower the lipid synthesis in sebaceous gland

  • SZ95 cell is an immortalized human sebaceous gland cell line that shows the morphologic, phenotypic and functional characteristics of normal human sebocytes. Sebocytes may play crucial parts in the pathophysiologic processes and disorders of the pilosebaceous unit. The secretory activity of the sebaceous gland is remarkably species-specific and acne is an exclusively human disease. Thus, this SZ95 cells offer possibilities for investigations on the physiology of the sebaceous gland and its role in sebum-associated skin disease such as acne. In this study, we investigated the effects of 13-cis-retinoic acid (13-cis-RA) and spironolactone, frequently used as therapeutic agents of acne, on the lipid synthesis and proliferation of human sebocytes. Cell proliferation was determined by MTT assay and cytoplasmic lipid droplets was shown by Oil-red a staining. Total lipid levels were biochemically estimated by the sulfo-phospho-vanilline reagent. 13-cis-RA and spironolactone significantly inhibited proliferation and lipid levels in a dose-dependent manner. Combined treatment with testosterone and 13-cis-RA or spironolactone resulted in a lower total lipid levels than that with androgen alone. These observations indicate that 13-cis-RA and spironolactone are potent inhibitors of both cell proliferation and lipid synthesis in human sebocytes. We will provide experimental evidence that this human sebocyte cell line serves as an adequate tool for evaluating the anti-lipogenic activity of various compounds potentially useful for the bioactive cosmeceutical ingredients on acne skin, and studying the intracellular biochemical markers depending on the types of compounds from various sources.

Correlation Between Sasang Constitution and Korean Adult Scalp and Hair Condition (사상체질과 두피상태의 상관관계에 대한 연구보고)

  • Objectives ; Sasang Constitional Medicine is a part of Korean traditional Medicine, and classify human constitution into four types. Scalp and hair is a part of skin. Skin is one of constitutional charasteristic element. So we researched a correlation between Sasang constitutions and Korean adult scalp and hair condition. This is a basic study to find constitution can be a important factor to care scalp and hair. Methods ; We examined Sasang constitutions and scalp and hair condition of 80 human who aged from 20-50. Scalp disease, dandruff was observed by video micro scope and sebum tape diagnosis. Thickness of hair was measured by Laser Scan Micrometer(LSM-3100). Itching sense and life habit was also investigated. Constitution diagnosis and scalp, hair diagnosis was investigated seperately as a blind test and the data was statistically analyzed with chi-square test, correlation analysis, one-way analysis of varience. Results ; 1. In Sasang Constitution, Taeumin group had significant tendency of oilily scalp. 70% of Taeumin group had oilily scalp. 2. The group of dry or oilily scalp had more itching sense and more sensativity of scalp compaired to neuter scalp group. 3. There was no correlation between height, weight, thickness of hair, itching sense, scalp disease and Sasang constitution. 4. There was correlation between loss of hair and drinking alcohol, obesity, curly hair, soft scalp. Conclusions ; Korean adult scalp and hair condition had correlation with Sasang constitution.

Analysis the skin physical properties of periorbital region and cheek based on female's aging pattern by 7 years (한국 성인 여성의 눈가 피부 특성 분석)

  • Objectives : We investigated the skin properties on the periorbital region and cheek of Korean female according to age pattern by 7year based on Sang Go Cheun Jin Ron (上古天眞論) in So Moon (素問,) Methods : 49 Korean females, aged 21-56 years ($39.54{\pm}10.04$ years) were participated in this study. We measured the skin properties of periorbital region and cheek with SkinTouch (Amorepacific, Korea). Skin moisture, sebum, elasticity, lightness, roughness, area of wrinkle, depth of wrinkle were assessed. Results : Differences were noticed depending on the site and age. The level of moisture was higher on the eyelid than crow's feet. The level of lightness and elasticity were lower on the periorbital region than cheek. Skin lightness was highest on group1(under 28y) and lowest on group5(over 49y). Skin roughness was highest on group1(under 28y) and lowest on group4(over 42y under 49y). The level of wrinkle are was lowest on group1(under 28y) and highest on group 5(over49y). The level of wrinkle depth was lowest on group1(under 28y) and highest on group 4(over 42y under 49y). Conclusion : Unique aging pattern by 7 years of female is reflected to diverse skin properties of eyes. It can be possible to apply the theory on Sang Go Cheun Jin Ron (上古天眞論) to practical use in dermatology and cosmetic.

Sustainability of Textile Products based on Washing Conditions: Focusing on the washing temperature and washing time (의류제품의 세탁조건과 지속가능성: 세탁온도와 세탁시간을 중심으로)

  • The use stage of a textile product impacts sustainability more significantly than other stages of the product's life cycle due to repeated washing and drying. This study determines efficient washing conditions, with high detergency, to reduce energy consumption from excessive washing and improve the washing process sustainability. Detergency was measured at various washing temperatures ($20^{\circ}C$, $40^{\circ}C$, and $60^{\circ}C$) and time (10 min, 20 min, and 30 min) using standardized soiled fabrics, i.e., 100% cotton, polyester/cotton (65%/35%), and 100% polyester woven fabric soiled with pigment/sebum, carbon black/mineral oil, soot/mineral oil, cocoa, blood, and red wine. Detergency at the washing condition of $20^{\circ}C$ and 30 min was higher than that at $40^{\circ}C$ and 10 min. In addition, detergency at the condition of $40^{\circ}C$ and 30 min was also higher than that at $60^{\circ}C$ and 10 minutes. This may be because a reduced washing effect at low washing temperatures was complemented by increased mechanical action over a long time. Further, washing temperature and time, with the same detergency, differed based on the type of fiber and soil. Also, the influence of a detergent on the detergency depends on the type of soil. The results suggest that energy and detergent have been consumed more than necessary in actual laundry. According to each type of fiber and soil, washing conditions designed to reduce the energy consumption of the washing process while maintaining the same detergency, were determined.

Orobol, A Derivative of Genistein, Inhibits Heat-Killed Propionibacterium acnes-Induced Inflammation in HaCaT Keratinocytes

  • Acne is a chronic skin disease that typically occurs in the teens and twenties, and its symptoms vary according to age, sex, diet, and lifestyle. The condition is characterized by hyperproliferation of keratinocytes in the epidermis, sebum overproduction, excessive growth of Propionibacterium acnes, and P. acnes-induced skin inflammation. Interleukin (IL)-1α and IL-6 are predominant in the inflammatory lesions of acne vulgaris. These cytokines induce an inflammatory reaction in the skin in the presence of pathogens or stresses. Moreover, IL-1α accelerates the production of keratin 16, which is typically expressed in wounded or aberrant skin, leading to abnormalities in architecture and hyperkeratinization. Orobol (3',4',5,7-tetrahydroxyisoflavone) is a metabolite of genistein that inhibited the P. acnes-induced increases in IL-6 and IL-1α levels in human keratinocytes (HaCaTs) more effectively compared with salicylic acid. In addition, orobol decreased the IL-1α and IL-6 mRNA levels and inhibited the phosphorylation of inhibitor of kappa-B kinase, nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor, alpha, and mitogen-activated protein kinase induced by P. acnes. Finally, the expression of Ki67 was decreased by orobol. Thus, orobol ameliorated the inflammation and hyperkeratinization induced by heat-killed P. acnes and thus has potential for use in functional foods and cosmetics.

A Meta-Analysis of Influencing Collagen Intake on Skin Utilizing Big Data (빅데이터 분석을 활용한 콜라겐 섭취가 피부에 미치는 영향에 관한 메타분석)

  • Big data analysis, in the large amount of data stored as the data warehouse which it refers the process of discovering meaningful new correlations, patterns, trends and creating new values. The important issue of a meta-analysis is not the significance test, the effect size of the predictor variable on the criterion variable. We reviewed a total of 236 samples among 6 studies published on the topic related Collagen intake on skin between 2000 and 2016 in Korea. The results of the study are summarized as follows. First, we concluded that the path between before and after of Sebum (SB) had the largest effect size of (r = .416) Therefore, the effect of the Collagen intake intervention showed an explanatory power of 17 (%) about. Next, the path between before and after of Moisture (MS) had the higher the effect size of (r = .318). Thus, we present the theoretical and practical implications of these results.

  • Tests of stability and toxicity, and clinical evaluation of anti-acne activity suggest that cryptotanshinone, a constituent of the roots of Salvia miltiorrhiza Bunge, is an effective active ingredient for acne vulgaris treatments. Acne vulgaris, called acne or pimples, is the most common disease of the pilosebaceous follicle unit of the skin. It affects nearly 80% of people between the ages of 11 and 30. Approximately 30% of teenagers have acne of sufficient severity to require medical treatment. Acne is a follicular disorder of the skin. It occurs in specialized pilosebaceous units on the face and body. Acne develops when these specialized follicles undergo pathologic alterations that result in the formation of non-inflammatory lesions (comedones) and inflammatory lesions (papules, pustules and nodules). An abnormality of keratinizing epithelium of these follicles, thought to be due to the action of sebum synthesized and secreted by the androgen-sensitive sebaceous glands, leads to inflammation induced by the follicular bacterium Propionibacterium acnes. Therapy involves treatments that modify these pathogenic factors and includes drugs with antikeratinizing, antibacterial and antiseborrheic actions. Acne vulgaris is a very frequent disease, seen primarily in adolescents, involving the sebaceous follicles. Acne vulgaris is characterized by a great variety of clinical inflammatory and non-inflammatory lesions: comedones, papules, pustules, nodules, cysts and scars. Acne vulgaris is a multi-factorial disease. Although its pathogenicity is unclear, extensive studies have shown that hyperseborrhea, superinfection by P. acnes and endocrinologic androgenic changes play a role in the development of acne vulgaris.

A comparison of detergency and dimensional stability between wet cleaning and dry cleaning (물세탁과 드라이클리닝의 세탁성능과 형태안정성 비교)

  • The washability, redeposition, fill power, and fabric damage of wet cleaning and dry cleaning solvents were measured to identify the optimal type of washing that would increase washability while maintaining dimensional stability. The soiled fabric is a polyester cotton blend and the types of soil were wine, blood, make-up and sebum with carbon black. Petroleum and silicone solvents were used in dry cleaning. Results from this study are as follows. First, detergency is significantly influenced by the type of washing and type of soil. Wet cleaning is superior to dry cleaning. Wet cleaning shows a strong washing performance against hydrophilic soils, whereas, dry cleaning is stronger against hydrophobic soils. Second, redeposition is significantly affected by the type of washing, fabrics, and soils. Redeposition occurred little on cotton during wet cleaning, but showed a high rate for nylon. However, when the two types of fabric were dry cleaned, redeposition occurred on both types. Third, the fill power of duck-down is very affected by the type of washing. Resilience is the best in wet cleaning; and in dry cleaning, petroleum solvents showed a higher resilience when as compared to silicone solvents. Last, the level of fabric damage to cotton fabrics is highly influenced by the type of washing. Wet cleaning damages cotton fabrics significantly more than dry cleaning. For dry cleaning, petroleum solvents damage these fabrics slightly more than silicone solvents. In conclusion, the type of soil must initially be identified to determine the optimal type of washing. Special caution is required when textiles with particulate soil and nylon are washed. When considering the resilience of duck-down clothing, wet cleaning is more appropriate than dry cleaning. Dry cleaning, especially when using silicone-based solvents, is more suitable than wet cleaning for maintaining the shape of clothing.

Effect of Paraffin Wax Mask Pack on Skin Improvement (파라핀왁스 마스크 팩의 피부 개선 효과)

  • This study examined the effect of paraffin wax mask pack on skin improvement. The subjects were 13 males and females in 20s. Subjects applied the paraffin wax mask pack eight times for four weeks. To determine the effects of paraffin wax mask pack on the skin, we measured oil, moisture, skin surface temperature, softness of skin and satisfaction in subjective skin condition. After using the paraffin wax mask pack, the moisture, oil content of the facial skin, skin surface temperature and softness of skin increased. In addition, the subjects were highly satisfied with the improvement in subjective skin conditons such as dryness, oil and skin tone. Therefore, these results suggest that paraffin wax mask pack has beneficial effects on skin improvement and it will have high utility and value as development of skin care devices for skin care.

In vitro Study and Clinical Trial of Natural Essential Oils and Extract Against Malassezia Species

  • Background: Malassezia, a lipophilic yeast, is a causative agent for dandruff and seborrheic dermatitis. Many biological agents have been studied for anti-Malassezia effect but further studies are needed for their clinical application. Objective: The study was conducted to evaluate the inhibitory effect of different natural essential oils and a fruit extract on Malassezia species in an in vitro study and a clinical trial. Methods: The antifungal effects of natural essential oils and a fruit extract on Malassezia species (M. furfur and M. sympodialis) were evaluated by measuring the minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) and using the disc diffusion method. Natural essential oils of citron seed, lavender, and rosemary and citrus junos fruit extract were used for the in vitro study. The clinical trial was conducted with a shampoo containing four ingredients. A total of 22 subjects used the shampoo every day for 4 weeks and were evaluated using clinical photography, trichoscopy, and sebumeter at baseline, 2 weeks, and 4 weeks after treatment. Results: Antifungal activity of agents was relatively lower in lavender and rosemary essential oils at MIC and MFC. Disc diffusion method revealed same results. In the clinical trial, the amount of sebum decreased statistically significantly and erythema, dandruff, and lesion extent also improved. Conclusion: The natural essential oils and fruit extract are effective for suppressing Malassezia activity, therefore these might be used as an alternative for treatment of dandruff and seborrheic dermatitis.
