Estimation of Water Production Cost from Seawater Reverse Osmosis (SWRO) Plant in Korea (국내 해수담수화 플랜트 생산수 단가 추정)

  • This study was conducted to supply information that can be utilized as data for desalination plant construction in the future by estimating unit cost of water production in the potential site of Incheon, Daesan, Yeosu, Busan, Ulsan and Sokcho in Korea. The production costs in Sokcho and Ulsan were similar to those of Busan and Yeosu. Those four sites showed better economic range due to low construction cost for intake facility compared to Incheon and Daesan. Although the salinity measured in the above 6 sites did not show perceptible effect on the production cost, the difference of seasonal seawater temperature needs to be considered due to the change of flux in reverse osmosis (RO) membrane. It turned out that the most critical parameters are the amortization in a year by the analysis of life cycle and the capacity of plant. Incheon and Daesan showed the difference of production cost up to 29% at the condition of amortization in 25 year, and up to 22% depending on plant capacity. However, the production cost in this study did not take into account of other indirect costs, therefore, this should be considered as the minimum cost.

Effects of butaphosphan and cyanocobalamin mixture on immunity and stress in olive flounder, Paralichthys olivaceus

  • Background: The study evaluated the effects of a butaphosphan and cyanocobalamin mixture on the immune system and stress in olive flounders, Paralichthys olivaceus. Methods: The mixture was intramuscularly injected into olive flounders at the current recommended dose. Furthermore, to determine the toxicity of overdose, a histological examination was performed after injection of 1-, 2-, and 4-fold higher than the recommended dose. Results: Immunity parameters were altered during the first 2 weeks after a single intramuscular injection of the mixture in olive flounders (average weight 20.5 ± 1.1 g). The levels of all tested items, except glutathione and antiprotease, were higher in the treated group than in the control group in the first week; the levels of all tested items were even higher in the second week in the treated group than in the control group. The level of nitro-blue tetrazolium, myeloperoxidase, and superoxide dismutase between the two groups differed significantly. Changes in the stress response to different seawater temperatures (increase or decrease in seawater temperature by 3-5 ℃ using 50 L heated or cooled seawater tanks) were studied by determining the changes in cortisol and glucose levels on days 1 and 7. Both cortisol and glucose levels were significantly lower in the treated group than in the control group. Histological analysis did not reveal any abnormalities after intramuscular injection of the mixture at doses that were 1-, 2-, and 4-fold higher than the recommended dose. Conclusions: Intramuscular injection of a butaphosphan and cyanocobalamin mixture is safe and effective in reducing stress and improving immunity in olive flounders.

Tidal variations of Nutrient Concentration in Hampyeong Bay, West coast of Korea (서해 함평만에서 조위변화에 따른 영양염 변동)

  • In order to understand the circulation of nutrient between muddy tidal flat and the surrounding coastal area, tidal time-scale variations in nutrient concentrations were seasonally investigated at the entrance of Hampyeong Bay. The results show that the temperature was higher in ebb tide and lower in flood tide during the summer, but it was lower in ebb tide and higher in flood tide during the autumn/winter. The salinity was higher in flood tide and lower in ebb tide during the summer/winter because of the inflow of external sea water resulting from the increase in the tide level. By contrast, the salinity was lower in flood tide and higher in ebb tide during the autumn. Salinity difference was lower than 0.3 psu between flood tide and ebb tide during survey period. Meanwhile, all nutrient concentrations observed in Hampyeong Bay was lower in flood tide and higher in ebb tide during the summer, and by contrast, it was higher in flood tide and lower in ebb tide during the winter. Characteristically, no clear variation of concentrations was found depending on the tide level during the autumn. This tidal variations imply that the muddy tidal flat of Hampyeong Bay supplies nutrients to the seawater in summer and removes nutrient from the seawater in winter. During tidal cycle, seasonal variation of nutrient concentration in seawater is considered as the result of complex interactions between the mud flat and external sea water.

Sanitary assessment of the oyster rack culture waters in Wando, Korea (완도군 군외면 연안 수평망식 참굴 (Crassostrea gigas) 양식해역의 세균학적 수질 평가)

  • To introduce an oyster rack culture in tidal flat of Wando-gun, Korea, we investigated sanitary conditions from 2012 April to 2013 March in oyster growing waters and adjacent area in Wando. Average seawater temperature and salinity ranged $8.6-28.0^{\circ}C$ and 26.2-33.6 psu, respectively. The coliform group and fecal coliform of seawater ranged < 1.8-13.0 MPN/100 mL each. Only one of 60 samples was exceeded in the European criteria of fecal coliform. Influence of adjunct basin's pollution load on cultural waters was negligible, which half radius of dilution was not above 300 m. According to our observation, however, landing of oyster should be banned for at least 5 days after the heavy rainfall. Our findings meet the sanitary seawater criteria for domestic use and export of shellfish and classified as 'Class A' in European Community regulations for bivalve culture.

Production and Characterization of Alkaline Protease of Micrococcus sp. PS-1 Isolated from Seawater (해수에서 분리한 Micrococcus sp. PS-1이 생산하는 단백질 분해효소의 생산과 효소학적 특성)

  • The purpose of this research was to investigate the production and characterization of alkaline protease from Micrococcus sp. PS-1 newly isolated from seawater. Micrococcus sp. PS-1 was grown in Luria-Bertani (LB) medium. Its optimal temperature and pH for growth were $30^{\circ}C$ and 7.0, respectively. The effect of nitrogen sources was investigated on optimal enzyme production. A high level of alkaline protease production occurred in LB broth containing 2% skimmed milk. The protease was purified in a 3-step procedure involving ultrafiltration, acetone precipitation, and dialysis. The procedure yielded a 16.43-purification fold, with a yield of 54.25%. SDS-PAGE showed that the enzyme had molecular weights of 35.0 and 37.5 kDa. Its maximum protease activity was exhibited at pH 9.0 and $37^{\circ}C$, and its activity was stable at pH 8.0-11.0 and $25-37^{\circ}C$. The protease activity was strongly inhibited by PMSF, EDTA, and EGTA. Taken together, the results demonstrate that the protease enzyme from Micrococcus sp. PS-1 probably belongs to a subclass of alkaline metallo-serine proteases.

Generation of Hydrogen from Hydrolysis Reaction of NaBH4 Using Sea Water (바닷물을 이용한 NaBH4 가수분해에 의한 수소발생)

  • Sodium borohydride,$NaBH_4$, has many advantages as hydrogen source for portable proton exchange membrane fuel cells (PEMFC). When PEMFC is used for marine use, $NaBH_4$ hydrolysis using seawater is economical. Therefore, in this study, hydrogen was generated by using seawater instead of distilled water in the process of hydrolysis of $NaBH_4$. Properties of $NaBH_4$ hydrolysis reaction using activated carbon supported Co-B/C catalyst were studied. The yield of hydrogen decreased as $NaBH_4$ concentration and NaOH concentration were increased during $NaBH_4$ hydrolysis using sea water. At higher concentrations of $NaBH_4$ and NaOH, byproducts adhered to the surface of the catalyst after hydrolysis reaction using sea water, reduced hydrogen yield compared to distilled water. The activation energy of $NaBH_4$ hydrolysis is 59.3, 74.4 kJ/mol for distilled water and sea water, respectively. In order to increase the hydrogen generation rate in seawater as high as distilled water, the reaction temperature has to be increased by $80^{\circ}C$ or more.

Fluctuation Characteristics of Seawater Temperature Identified by Time Series Analysis Off the Southern Coast of Korea (한국 남해안에서 시계열 분석을 통한 수온 변동 특성)

  • In order to understand the fluctuation characteristics of water temperature in August, water temperature, tide level, air temperature and wind data were analyzed using a power spectrum and coherence analysis for a region off the southern coast of Korea. Spectrum analysis showed that the water temperature and tide level in Wando, Goheung, Yeosu, Tongyeong and Masan have peaks over a semi-diurnal period. Coherence showed that water temperature was affected by tide in Wando, Goheung, Yeosu and Tongyeong where tidal range is relatively high. In Masan and Busan, where tidal range is relatively low, however, the tide did not affect water temperature significantly. In particular, wind was the most important factor in relation to water temperature in Masan. Time lags were calculated from phase. In Wando, water temperature decreased 1.5 hrs after a flood tide began. Water temperature decreased 0.3 hrs after a flood tide began in Goheung and Tongyeong. In Masan, water temperature increased 3 hrs after a south wind began. Water temperature in Yeosu was affected by tide, but air temperature, tide and wind in Busan affected water temperature very little.

  • the horizontal corrosion fatigue tester has been developed for investigating environmental strength. Using this tester, we investigated about corrosion fatigue caracteristic for A106 GrB steel weldments in 3.5% synthetic seawater and room temperature. Considered parameter is only frequency of 1,3 and 5Hz. and Corrosion fatigue crack length was measured by DC potential difference method. From the results, we could find that the horizontal corrosion fatiued tester could be well applied to estimation of fatigue strength. and In case of 5Hz, corrosion fatigue crack growth path of A106 GrB steel weldment was transgranular, and of 1 and 3Hz showed that transgranular and intergranular was mixed. Also, Corrosion fatigue crack growth caracteristic values estimated in each frequency were C=9.33 x 10-9 and m=2.93 in 1Hz, C=9.77x10-10 and m=3.47 in 3Hz, C=1.02x10-10 and m=4.05 in 1Hz

Comparative Morphology and Seasonality of Campylaephora borealis and C. crassa (Ceramiaceae, Rhodophyta)

  • Populations of Campylaephora borealis (Nakamura) Seo, Cho et Boo and C. crassa (Okamura) Nakamura show a year-around occurrence of all life-history stages. Such a concurrency of life-history stages produces problems in recognizing species in the field. Here, we invesitgated the morphological variation and life-history stages of both species using a statistical character analysis. Life-history stage was correlated with the seawater temperature in C. borealis, whereas it was dependant on biomass in C. crassa. Thalli had dichotomous branches with adaxial branchlets. The statistics showed that the seasonal change in morphology of C. borealis was significantly different from that of C. crassa in seven qualitative characters and five quantitative characters (p < 0.001), although six quantitative features including tetrasporangial size were similar in both species. The morphological difference between the two species may be due to the annual variation of branchlet number and the variance of branch subangle.

Characterization of Crude Oil Degradation by Klebsiella sp. KCL-1 Isolated from Sea Water (해양에서 분리된 Klebsiella sp. KCL-1에 의한 원유분해 특성의 규명)

  • Several bacterial strains utilizing crude oil as their sole carbon and energy sources were isolated from marine. One of the strains named KCL-1 showed the highest degradative activity for crude oil and the best growth rate. This strain was identified as a Klebsiella sp. based on the morphological, biochemical, and physiological characteristics. The optimum cultural conditions were as follows; $32^{\circ}C$~$37^{\circ}C$ for temperature and 7.0 for initial pH. Additionally, the optimal concentration of sodium chloride was 3.0%, indicating that this strain was derived from seawater. KCL-1 could use several kinds of n-alkane hydrocarbons from octadecane to hexacosane as a sole carbon source. The degradation of crude oil by KCL-1 was stimulated by addition of octadecane in the culture. The emulsifying activity by KCL-1 was highest after 3 days of cultivation under the condition of 3.0% sodium chloride, pH 7.0 and $37^{\circ}C$.
