Screening of Bacillus sp. No. M-71 with High Alkaline Protease Productivity and Some Properties of the Enzyme (Alkaline Protease 고 생산성 세균의 선별 및 효소 특성)

  • A bacterial strain No.71, which produced alkaline protease, was isolated from soil and identified to the genus Bacillus. With the successive mutation, a mutant strain No. M-71, having high alkaline protease productivity, was obtanined from the parental strain No 71. Alkaline protease productivity of mutant strain No. M-71 was about 50 times as much as that of the parental strain No.71. The enzyme preparations showed strong activities toward casein, the optimum pH being 11.0 and the optimum temperature about $55^{\circ}C$.

  • The aim of the present study was to develop strains of actinomycetes producing low molecular weight cathepsin B inhibitor. Among 700 isolates from soil samples, a strain of Streptomyces sp. SMF30 producing cathepsin B inhibitor showing specificity and heat stability was selected by an economical and effective screening method. 50 units characteristics for major cluster analysis and 34 units characteristics for minor cluster were tested and the data were analyzed numerically using the TAXON program. The Isolate SMF30 was identified as a strain of Streptomyces aburabiensis

  • Yeast strains capable to perform malo-alcoholic fermentation in wine were screened. Out of 54 strains isolated from apples, tomatos, grapes, and strawberries, two strains showed strong assimilation of malic acid. After further screening one strain of strawberry origin was selected and identified as Schizosacch-aromyces japonicus var. japonicus based on the morphological and physiological characteristics examined.

  • A different form from Gibberella fujikuroi was isolated from the paddy field of Naju area. The strain, designated as Y107, was identified as Gibberrella sp. based on its morphological, physiological, and biochemical characteristics. The highest production of Gibberellic acid by the strain was achieved in a fermentation medium containing corn starch, glucose, soybean oil, soybean meal, NH$_{4}$NO$_{3}$, K$_{2}$HPO$_{4}$, MgSO$_{4}$, and trace elements.

  • Fifty-eight strains of Aspergillus spp. isolated from various grains were examined for the screening of aflatoxins by the method of the Thin-Layer Chromatography (TLC), using the aflatoxin producing strain of Aspergillus flavus ATCC 15517 as a control. The results as follows: 1. Samples of Aspergillus spp. isolated from various grains were extracted with chloroform and chromatographied by the thin-layer chromatography method. 2. Three strains of Aspergillus spp. among the 58 strains can be found that the spots having the same Rf value as control with culture extract of Aspergillus flavus ATCC 15517. 3. The further prove studies of aflatoxins were proceed by the methods of in vivo and in vitro tests. And this methods considered to capable for the use of mass screening among the samples.

  • Several protease-deficient mutants of Aspergillus niger have been isolated by halo-screening on skim milk plate after UV irradiation of conidiospores. The extracellular proteolytic activities of the mutant strains grown in an optimized medium varied from 3% to 85% of that of the parental strain. Especially, two mutant strains named as ANPD-129 and ANPD-153, which had 3% and 49% of acid protease activity of the parental strain, respectively, were further characterized both physiologically and genetically. The growth rates of the mutants, ANPD-129 and ANPD-153, were similar to that of the parental strain, unlike other protease-deficient mutants. The diploid formed between the two mutants restored protease activity to a similar level of that of the parental strain. This result revealed that ANPD-129 and ANPD-153 had mutations at different loci. Using master strains with marked chromosomes these loci were assigned to linkage groups. The mutation locus (prt129) in ANPD-129 was assigned to linkage group VI and the locus (prt153) in ANPD-153 to linkage group III.

  • Petroleum energy is the major source of the world energy market, and its massive usage, and the corresponding extreme environmental pollution, imposes a serious threat on the ecological cycles. By screening oil-contaminated soil, we isolated, identified, and characterized a novel strain that represents a considerable diesel-degrading potentiality; the Bacillus aryabhattai DA2 strain is registered in the NCBI with the accession number MG571630, and it possesses an efficient tributyrin-degrading capacity. The optimal condition for diesel degradation by DA2 strain was observed at pH between 7-8 and at the temperature of $30^{\circ}C$. The strain is resistant to salt as well as the antibiotics like ampicillin and streptomycin. These results indicate B. aryabhattai is one of the potential candidates for the remediation of the diesel-contaminated sites.

  • In the screening of actinomycetes culture filtrate for inhibitor of adenosine deaminase, a novel inhibitor was found in a cultured broth of strain J-144K. The optimum conditions for the adenosine deaminase inhibitor production from the isolated strain J-144K were evaluated. This strain showed the maximum yield of adenosine deaminase inhibitor when grown at pH 7.0 and 30$\circ$C for 60 hours in the medium of 1.0% dextrose, 0.5% yeast extract, 0.5% peptone and 0.1% KH$_{2}$PO$_{4}$ under the aerobic condition. Through the activated charcoal extraction, methanol fractionation, Dowex 50 H$^{+}$ X-8 ion exchange column chromatography, Dowex CI$^{-}$ X-8 ion exchange column chromatography, and Sephadex G-15 gel filtration procedures, this inhibitor was purified with three materials.

  • Methylglyoxal (MG) is a reactive metabolite known to accumulate in certain physiological conditions. We attempted to isolate genes associated with this metabolite by genome-wide mutagenesis with TnphoA derivative. After screening on methylglyoxal-containing plate, we obtained insertions in three different genes, ydbD, yjjQ, and yqiI, which gave rise to reproducible MG-sensitive phenotypes in glyoxalase-deficient strain. In addition to its MG sensitivity, the insertion in yqiI exhibited an impaired motility resulting from a reduced flagellar expression.

  • Club root is one of the major diseases that occur in crucifers. It is caused by Plasmodiophora brassicae. In order to discover microbial biopesticides against P. brassicae, forty-eight Streptomyces isolated from soil were screened. Among these, three strains showed excellent pesticidal activities. We report results on in vivo screening with fermentation broths of these strains and identification of the strain taxa.
