Bacillus sp. BS061 Suppresses Powdery Mildew and Gray Mold

  • The use of a microorganism, or its secretions, to prevent plant disease offers an attractive alternative or supplement to synthetic fungicides for the management of plant disease without the negative effects of chemical control mechanisms. During a screening for microorganisms with the potential to be used as microbial fungicides, Bacillus sp. BS061 was isolated from a plant leaf. The strain BS061 potently inhibited the mycelial growth of Botrytis cinerea, and significantly reduced disease incidence of powdery mildew in cucumber and strawberry. We also found that the culture filtrate of BS061 inhibited the mycelial growth of various plant pathogens.

  • This study is to predict the possible roles of the aryisulfate sulfotransferase (ASST) in the microorganism. At first we studied the spectrum of a distribution of the ASST enzyme through about 1,300 bacteria and the several selected strains were compared with Klebsiella K-36 previously reported in the level of DNA homology using the Southern blot method. From this study, we could predict that this enzyme would not exist in specific bacteria and it might not be a critical enzyme for the life of bacteria.

  • This study was carried out to select the useful bacteria which secret extracellula enzymes for enzymatic deinking agent of old newspaper. CMCase, FPase and xylanase activities of the bacteria liquid culture were measured at optimal growth conditions. Clear zone test was checked on the solid culture. The results of this study were as follow: Eight strains of 28 bacteria isolated from a paper mill soil ground were shown strong CMCase and xylanase activity with the clear zone test. The optimal pH and temperature for culture growth were 6~8 and 26~$34^{\circ}C$, respectively and optimal culture period were less than 60 hours. Based on CMCase, FPase and xylanase activity, strain No. 18, 21, 22 and 28 which were relatively higher than the other strains, were selected for further enzymatic deinking research.

Heptelidic Acid, a Sesquiterpene Lactone, Inhibits Etoposide-Induced Apoptosis in Human Leukemia U937 Cells

  • In the course of screening for substances that inhibit etoposide (10 ${\mu}g$/ml)-induced apoptosis in human leukemia U937 cells, fungal strain F000120, which exhibits potent inhibitory activity, was selected. The active compound was purified from an ethyl acetate extract of the microorganism by Sep-pak $C_{18}$ column chromatography and HPLC, and was identified as heptelidic acid (koningic acid) by spectroscopic methods. This compound inhibited caspase-3 induction in U937 cells with an $IC_{50}$ value of 40 ${\mu}M$ after 8 h of etoposide treatment. Fluorescent dye staining with acridine orange and ethidium bromide showed that heptelidic acid inhibited apoptosis. Furthermore, it was found that DNA fragmentation and caspase-3 activation, the biological hallmarks of apoptosis, were inhibited by the compound in a dose-dependent manner, suggesting that heptelidic acid inhibits etoposide-induced apoptosis via downregulation of caspases.

Isolation and Identification of Antagonistic Microorganisms for Biological Control to Apple Tree Diseases, Canker(Valsa ceratosperma) (사과 부란변 방제를 위한 길항미생물 분리 및 동정)

  • For the purpose of acquiring microbial agents that can be utilized to billogically control the major airborne disease to apple trees, such as canker(Valsa ceratosperma), the effective microorgaisms were isolated, tested for antagonistic activity to the pathogen causing major disease to apple trees and identifed. Screening of more than 3, 000 species of microorganisms collected in nature for them antagonistic action to the pathogen, Valsa ceratosperma causing disease to apple tree resulted in selection of effective species. Out of the 11 species, one species designated as CAP141 demonstrated outstanding activity. The bacterial strain, CAP141 exerted antagonistic efficiency of 65% on Valsa ceratosperma. The CAP141 was identified as a bacterial strain to Bacillus subtilis based on morphology, culture conditions, and physio-biochemical characteristics.

  • To utilize hemicellulosic biomass efficiently, the microorganism producing xylanase was isolated from fermented sawdust. It was a Gram positive, aerobic and rod shape bacterium. It had endospore and secreted strong hydrolases, such as amylase and protease. Through morphological, cultural and physiological tests, it was identified as Bacillus sp. GS. To increase the productivity of xylanase from Bacillus sp. GS, the enzyme production medium was optimized. The composition of the medium and incubation conditions were like follows xylan 1.25%, yeast extract 0.1%, NaN030.2%, K2HP04 0.1%, MgSO4 0.02%, mineral salt 0.005%, pH 6.5, incubation temperature 37$^{\circ}C$.

  • Erythritol is a four-carbon sugar alcohol with a low calorific value and non-cariogenicity. Erythritol is a new functional sweetener which can be used as sugar alternative. Erytheitol dose not cause discomfort such as diarrhoea and flatulence upon ingestion. The purpose of this study is to develope a novel process of erythritol economically in a large scale. To obtain a high erythritol producer, we have screened 3500 colonies from molasses, honey and honey combs. We have selected 40 erythritol-producing microorganisms, one of which yields 140g/L erythritol in 40% glucose medium. We have tested this strain in 5L fermentor to examine the fermentation characteristics. Results of fermentation show that the erythritol production was about 1.4g/L$.$hr in 400g/L glucose media with a 42% conversion. Further improvements require mutation for a higher producer, process optimization to reduce glycerol, and suppression of excessive foaming.

Atromentin-Induced Apoptosis in Human Leukemia U937 Cells

  • In the course of screening for apoptotic substances that induce apoptosis in human leukemia U937 cells, a fungal strain, F000487, which exhibits potent inducible activity, was selected. The active compound was purified from an ethyl acetate extract of the microorganism by Sep-pak $C_{18}$ column chromatography and HPLC, and was identified as atromentin by spectroscopic methods. This compound induced caspase-3 processing in human leukemia U937 cells. The caspase-3 and poly (ADP-ribose) polymerase (PARP) were induced by atromentin in a dose-dependent manner. Furthermore, DNA fragmentation was also induced by this compound in a dose-dependent manner. These results show that atromentin potently induces apoptosis in U937 cells and that atromentin-induced apoptosis is related to the selective activation of caspases.

  • The study was performed to investigated the excellent microbial anticaries substance which is more effective than the chlorohexidine in the dental caries treatment. For the screening of alkaliphilic microorganism, more than 1200 bacterial strains were isolated from sea soil sample. A typ-ical strain which produced the most excellent antimicrobial substance was selected. The strain was identified novel alkalophilic Bacillus sp. through the results of morphological, biochemical and chemotaxonomical characteristics and 16S rDNA sequencing and designated as Bacillus alkalophilshaggy JY-827.
