Large-Scale Screening of the Plant Extracts for Antifungal Activity against the Plant Pathogenic Fungi

  • Plants produce chemicals of immense diversity that provide great opportunities for development of new antifungal compounds. In search for environment-friendly alternatives to the fungicide of current use, we screened plant extracts obtained from more than eight hundred plant materials collected in Korea for their antifungal activity against the model plant pathogenic fungus, Magnaporthe oryzae. This initial screening identified antifungal activities from the eleven plant extract samples, among which nine showed reproducibility in the follow-up screening. These nine samples were able to suppress not only M. oryzae but also other fungal pathogens. Interestingly, the plant extracts obtained from Actinostemma lobatum comprised five out of eight samples, and were the most effective in their antifungal activity. We found that butanol fraction of the A. lobatum extract is the most potent. Identification and characterization of antifungal substances in the A. lobatum extracts would provide the promising lead compounds for new fungicide.

Screening for In Vitro Antifungal Activity of Soil Bacteria Against Plant Pathogens

  • Antifungal bacteria for biological control of plant diseases or production of novel antibiotics to plant pathogens were isolated in 1997 from various soils of Ansung, Chunan, Koyang, and Paju in Korea. Sixty-four bacterial strains pre-screened from approximately 1,400 strains were tested on V-8 juice agar against eight plant pathogenic fungi using in vitro bioassay technique for inhibition of mycelial growth. Test pathogens were Alternaria mali, Colletotrichum gloeosporioides, C. orbiculare, Fusarium oxysporum f. sp. cucumerinum, F. oxysporum f. sp. lycopersici, Magnaporthe grisea, Phytophthora capsici, and Rhizoctonia solani. A wide range of antifungal activity of bacterial strains was found against the pathogenic fungi, and strain RC-B77 showed the best antifungal activity. Correlation analysis between inhibition of each fungus and mean inhibition of all eight fungi by 64 bacterial strains revealed that C. gloeosporioides would be best appropriate for detecting bacterial strains producing antibiotics with potential as biocontrol agents for plant pathogens.

Candida와 Penicillium 속 진균에 대한 천연물의 항진균 효과 검색 (Screening of the Antifungal Activity from Natural Products against Candida albicans and Penicillium avellaneum)

  • For the research of antifungal active constituents from natural products, 226 plants were extracted with ether and methanol, separately, and screened antifungal activity against Candida albicans and Penicillium avellaneum cells. The results demonstrated that 30 samlpes showed antifungal activity in ether or methanol extracts and 17 samples in ether extracts and 20 samples in methanol extracts against C. albicans. Against P. avellaneum, 19 samlpes showed antifungal activity in ether or methanol extracts and 17 samples in ether extracts and 11 samples in methanol extracts, respectively. The antifungal activity of natural products against C. albicans were showed more sensitive than P. avellaneum, and the polarity of the solvent was not specific in antifungal activity.

  • To search and develop the compounds exhibiting antifungal activities against the mycelial phase of Candida albicans, approximately 2,900 microorganisms isolated from soil were examined for antifungal activity. Among them, a strain with preferential activity against the mycelial phase of Candida albicans was isolated and identified as Streptomyces sp. A393. Isolation and purification of compounds A393 showing antifungal activity against the mycelial phase of C. albicans were performed using XAD-7 column chromatography, silica gel chromatography, preparative thin- layer silica gel chromatography, and HPLC. The molecular weights of compounds isolated from Streptomyces sp. A393 were determined as 774, 790, 804 and 820. These compounds appeared to have a structure of macrolide antibiotics, oligomycin A, B, C and E. Especially, oligomycin E, which is formerly reported to have no antifungal activity, showed antifungal activity against the mycelial phase of Candida albicans.

Streptomyces with Antifungal Activity Against Rice Blast Causing Fungus, Magnaporthe grisea

  • Screening tests against fungus causing rice blast, Magnaporthe grisea, were performed in order to develop biopesticides. More than 400 actinomycetes collected at several sites near Hanla Mountain on Jeju Island, Korea were tested, and strain BG2-53 showed potent antifungal activity. The in vivo screening was performed with fermentation broth, and the strain taxon was identified.

Antifungal Activity of Streptomyces sp. Against Puccinia recondita Causing Wheat Leaf Rust

  • To discover a potent strain against wheat leaf rust, soil samples collected from Ilgamho, Seoul, Korea were tested in vivo and a strain belonging to Streptomyces sp. was found to show good antifungal activity when fermented in a broth. The identification of the strain was carried out based on 16S rDNA analysis, and the active compound was separated from the fermented broth. Even though its structure was not determined completely, the authors report the results obtained so far indicate that the fermented broth of the strain showed activity against wheat leaf rust. Therefore, we propose that this may be a potential novel strain showing antifangal activity against Puccinia recondita.

Identification of a Gene Encoding Adenylate Kinase Involved in Antifungal Activity Expression of the Biocontrol Strain Burkholderia pyrrocinia CH-67

  • Burkholderia pyrrocinia CH-67 is a biocontrol bacterium with strong antifungal activity against several plant pathogenic fungi. Transposon mutagenesis was performed to identify the genes responsible for the antifungal activity of B. pyrrocinia CH-67. Of the 2,500 mutants tested using the Fulvia fulva spore screening method, a mutant deficient in antifungal activity, M208, was selected. DNA sequence analysis of the transposon-inserted region revealed that a gene encoding an adenylate kinase-related kinase was disrupted in M208. Antifungal activity was restored in M208 when a full-length adenylate kinase gene with its promoter was introduced in trans. The deduced amino acid sequence of adenylate kinase from CH-67 was 80% identical to that of B. cenocepacia MCO-3. Adenosine diphosphate supplementation or high levels of adenosine triphosphate and adenosine monophosphate together restored antifungal activity in M208, suggesting that adenylate kinase of B. pyrrocinia CH-67 is involved in antifungal activity expression.

버섯중 항균물질의 검색 및 개발에 관한 연구 -버섯중 항진균활성 물질의 검색 (I)- (Studies on the Screening and Development of Antibiotics in the Mushroom -The Screening of Antifungal Components in Basidiomycetes (I)-)

  • 32종의 한국산 버섯을 석유에테르, 80% 에탄올 및 증류수로 각각 추출하여 항진균 활성을 검색한 결과, 수원그물버섯, 거친껄껄이그물버섯, 나팔버섯, 해면버섯, 주걱간버섯 및 석이버섯의 에탄올 추출물 그리고 큰낙엽버섯의 석유에테르 추출물이 효모에 대하여 활성을 보였다. 특히 석이버섯의 에탄올 추출물은 C. neoformans균에 큰 활성을 보였으며, 그 MIC 값은 $600\;{\mu}g/ml$ 이었다. 애광대버섯, 거친껄껄이그물버섯, 나팔버섯, 해면버섯, 주걱간버섯, 큰낙엽버섯 및 석이버섯의 에탄올 추출물과 석유에테르 추출물은 곰팡이에 대하여 각각 항균활성을 보였다. 또한 나팔버섯의 에탄올 추출물은 M. gypseum균에 대하여 $1,000\;{\mu}g/ml$ 이었고, 석이버섯의 에탄올 추출물은 T. mentagrophytes, M. gypseum 및 P. oryzae균에 대하여 각각 $300\;{\mu}g/ml,\;600\;{\mu}g/ml$$600\;{\mu}g/ml$ 였으며, 이 버섯의 석유에테르 추출물은 M. gypseum균에 대하여 $300\;{\mu}g/ml$의 MIC 값을 보였다.

당귀와 백지로부터 분리한 Coumarin계 물질들의 식물병원균에 대한 항균활성 (Antifungal activities of coumarins isolated from Angelica gigas and Angelica dahurica against Plant pathogenic fungi)

  • 식물자원으로부터 식물병원균에 대한 항균물질을 탐색하고자 국내에서 재배한 40종의 약용식물 추출물을 대상으로 항균활성을 검색하였다. 우선, 각 식물을 methanol (MeOH)로 추출, 농축한 MeOH추출물을 시료로 하여 $2000{\mu}g/mL$ 농도수준에서 6가지 식물병에 대한 in vivo 방제효과를 조사하였다. 40종의 시료 중 14종의 시료가 적어도 한가지이상의 식물병에 대하여 90% 이상의 방제효과를 나타내었다. 벼 도열병, 토마토 역병 및 밀 붉은녹병에 대해서는 각각 8개, 7개 및 3개의 시료가 90% 이상의 효과를 나타내었으나, 벼 잎집무늬마름병, 토마토 잿빛곰팡이병 및 보리 흰가루병에 대해서는 90% 이상의 방제효과를 보이는 시료는 없었다. 벼 도열병에 대하여 높은 활성을 보이는 당귀(Angelica gigas, 당귀(當歸))와 백지 (Angelica dahurica, 백지(白芷))의 MeOH추출물로부터 solvent partitioning과 column chromatography를 통하여 각각 1개 및 2개의 항균물질을 분리하였으며 이들 물질은 질량분석 및 핵자기공명분석에 의하여 모두 coumarin계 물질인 decursin, imperatorin 및 isoimperatorin으로 동정되었다. 구조와 항균활성간의 관계를 알아보기 위하여 분리한 3개의 물질을 7번 위치에 free hydroxyl기를 가지고 있는 umbelliferone (7-hydroxycoumarin)과 scopoletin (6-methoxy-7-hydroxycoumarin)과 비교하여 in vitro 및 in vivo 항균활성을 조사하였다. In vitro에서 균사생육억제활성을 조사한 결과, 이들 물질들은 대상식물병원 곰팡이에 대하여 50% 균사생육억제농도 ($IC_{50}$)가 대부분 $200{\mu}g/mL$ 이상으로 나타나 항균활성이 비교적 약한 것으로 나타났다. 또한 7번 위치에 hydroxyl기가 노출되어 있는 umbelliferone과 scopoletin보다는 7번 위치의 free hydroxyl기 대신에 cyclic alkoxy기로 존재하는 decursin과 imperatorin이 곰팡이에 대한 항균활성이 강한 것으로 나타났다. 특히, decursin과 imperatorin은 각각 Pythium ultimum과 Magnaporthe grisea에 대하여 $25{\mu}g/mL$ 이하의 $IC_{50}$를 보여 강한 항균활성을 나타냈다. In vivo 실험에서는 decursin과 imperatorin은 여섯 가지 식물병 중 벼 도열병에 대하여 항균활성이 높은 것으로 나타났으며, 다른 세 개의 물질은 식물병 방제활성이 거의 없는 것으로 나타났다. 이상의 실험결과 7-hydroxycoumarin계 물질들의 항진균활성과 관련하여 7번 위치에 free hydroxyl기가 있는 것보다는 이 hydroxyl기가 보다 안정한 cyclic alkoxy기로 치환되는 것이 높은 항균활성을 나타내는데 중요하다는 것을 알 수 있었다.

근권 정창 세균 Pseudomonas fluorescens MC07의 항진균 활성과 병 억제 능력 (Antifungal Activity of Root Colonizing Pseudomonas fluorescens MC07 is Responsible for Its Disease Suppression Ability)

  • An antagonistic bacterium, Pseudomonas flurorescens MC07 inhibited the mycelial growth of Rhizoctonia solani, Pythium ultimum, Fusarium oxysporum, and Phytophthora capsici in on potato dextrose agan (PDA) and other media. The strain MC07 conlonizes various plant roots and possesses antifungal activity. To determine the role of antifungal activity of the bacterium in disease suppression, a mutant Okm3-4 which lost its activity was isolated after screening 2,500 colonies generated by Omegon-Km insertions. The mutant Okm3-4 showed diminished growth inhibition of R. solani, P. ultimum, F. oxysporum, and Ph. capsici in vitro and had reduced suppressive effects on sesame damping.-off compared to the parental strain. In soils, accumulation of the pathogens by continuous cropping, 90% of sesame plants were killed by natural infection of damping-off whereas, only 29% of plants grown from seeds treated with MC07 were killed. On the other hand, 85% of plants died when sesame seeds were treated with the Okm3-4 cells. This indicated that antifungal activity of MC07 in vitro is directly responsible for the suppression of damping-off disease. Emergence rates of sesame seeds in pots containing diseased soil were 33%. However, MC07 treatments on seeds significantly improved emergence rates, which has similar effects of Benomyl treatment. The mutant Okm3-4 exhibited 53% of emergence rate. This indicated that antifungal activity of MC07 also affects the emergence rate of sesame seeds.
