The nicotine converter genotypes of burley tobacco (Nicotiana tabacum L.), which convert nicotine to nornicotine, contain a high amount of nornicotine that degrades tobacco quality and smoking taste. Elimination of nicotine converter plants before seed harvesting is required for breeding nicotine low-converter lines and for increasing their seed production. This study aims to develop a rapid and convenient method of identifying nicotine converter plants of burley breeding lines of KB9118(KB108) using thin-layer chromatography (TLC) and isatin coloration method. Out of 223 plants in 10 lines harvested at maturity in 2002, 102 plants ($45\%$) were identified as nicotine converters by TLC of tobacco leaves air-cured. For 16 lines selected as low-converters in 2002, 148 plants grown in the field in 2003 were tested by the isatin coloration method using two detached leaves at the flowering stage thoroughly sprayed with $1\%\;NaHCO_3$ solution and cured in conditioned chambers for the early identification of nicotine to nornicotine conversion. From these samples, 46 plants ($31\%$) in 4 lines were identified as nicotine converters, indicating that the ratio of converters significantly decreased by one time selection. Mean percent conversion of non-screened lines was $14\%$ higher than that of following generation. Therefore in the burley tobacco, a rapid and convenient means of identifying and removing nornicotine converter plants by the isatin coloration method during growth in the greenhouse or field were effective in reducing the converter plants in the following generation.
The present study was conducted to compare the relative efficiency of four different breeding methods in tobacco varietal development. A single Fl hybrid plant from cross of two flue-cured cultivars of Nicotiana tabacum L Bright Yellow 4(BY4) and NC95, was selfed. F2population above cross was screened for resistance against bacterial wilt caused by Pseudommonas solanacearum E.F.Smith under the naturally infested field conditions, and the 30 lines were developed from F2 individual plant by anther culture (ADH), maternal method utilizing Nicotiana afpicana (MDH), single- seed descent(SSD) and Bulk breeding method, respectively. All characters except content of total alkaloids of ADH and MDH which wore bred by haploid methods reduced more than that of lines bred by conventional methods(SSD & Bulk) : however, the yields were 8% lower than other lines. The total alkaloid content of ADH was higher than that of MDH, and yield was reduced about 4n even though the number of leaf was identical with the MDH. All other characters of ADH were also reduced significantly. In the lines bred by conventional methods, population developed by SSD showed significantly wider leaf width, shorter plant height, later days to flower, and lower in percent reducing sugar than those by Bulk. The populations derived from haploid method showed greater phenotypic variance and wider range of variation than conventionally developed ones. The results obtained indicate that selection will provide a significantly greater genetic gain for leaf number and leaf length in the ADU and MDH populations, and for plant height and days to flower in the SSD and Bulk populations.
In order to control seed-borne diseases, we obtained extracts from commercial fermented food products of Kimchi, Gochujang, Doenjang, Ganjang and Makgeolli and their suppressive effects against seed-borne diseases were studied. In addition, the suppressive effects of bacterial strains isolated from the fermented foods were screened in vitro and in vivo. Among fifty food extracts, twenty food-extracts suppressed more than 92% incidence of seedling rots in vitro and seven food extracts increased 58.3-66.8% of healthy seedling in the greenhouse. Among 218 isolates from the fermented foods, 29 isolates showing high antifungal activity against seven seed-borne fungal pathogens were selected. Among 29 isolates, 13 isolates significantly reduced seedling rot and increased healthy seedlings. Sixteen isolates with high antifungal activity and suppressive effect against sesame seedling rots were identified by 16S rRNA sequencing. Fourteen of sixteen isolates were identified as Bacillus spp. and the other two isolates from Makgeolli were identified as Saccharomyces cerevisiae. It was confirmed that B. amyloliquifaciens was majority in the effective bacterial population of Korean fermented foods. In addition, when the bioformulations of the two selected effective microorganisms, B. amyloliquifacien Gcj2-1 and B. amyloliquifacien Gcj3-1, were prepared in powder forms using bentonite, kaolin, talc and zeolite, talc- and kaolin-bioformulation showed high control efficacy against sesame seed-borne disease, followed by zeolite-bioformulation. Meanwhile control efficacy of each bentonite-bioformulation of B. amyloliquifacien Gcj2-1 and B. amyloliquifacien Gcj3-1 was lower than that of bacterial suspension of them. It was found that the selected effective microorganisms from Korean fermented foods were effective for controlling seed-borne diseases of sesame in vitro and in the greenhouse. We think that Korean fermented food extracts and useful microorganisms isolated from the extract can be used as bio-control agents for suppressing sesame seed-borne diseases based on above described results.
The objective of this study was to obtain the basic informations on variability of germplasm and cultivation practice for the production of biodiesel in sunflower. A total of 275 accessions obtained from the National Agrobiodiversity Center (34 landraces from Korea, 219 and 22 introductions from America and other countries, respectively) were screened to evaluate variations of seed characteristics, oil content and fatty acid composition. Seed types were classified into 4 types (broad ovoid, narrow ovoid, rounded and elongated). The broad ovoid type was the highest proportion of 40.4%, while the elongated type was the lowest proportion of 5.0% among accessions. Also, the seed colors were classified into 4 colors (gray, brown, black and white). Gray and brown seed color showed the highest distribution and white seed color showed the lowest among accessions, respectively. Hundred and one liter seed weight ranged 2.9~15.5g and 178~439g with averages of 6.3g and 322.0g, respectively. Oil content ranged from 11.7% to 45.6% with an average of 25.5%. The frequency distribution of oil content showed the highest in range of 22~28% and the lowest in range of over 40%. Three promising accessions with higher oil content than 40.0% were IT031967, IT031970 and IT031965 introduced from America. Palmitic and stearic acid contents, saturated fatty acid, ranged 3.1~7.6% and 1.3~4.1% with averages of 4.7% and 2.2%, respectively, and a total content of saturated fatty acid ranged 5.4~9.4% with an average of 6.9%. Oleic and linoleic acid contents, unsaturated fatty acid, ranged 18.1~75.7% and 18.1~74.1% with averages of 55.2% and 38.0%, respectively. Five accessions, IT031831, IT031669, IT031895, IT031938 and IT031694, showed higher oleic acid content than 70%. A total content of unsaturated fatty acid ranged 89.6~94.8% with an average of 93.0%.
As part of a study on the ability of fungicides to control disappearing root rot of ginseng (Panax quinquvdius) caused by Cylindruarpn destmtans, 15 fungicides were screened for toxicity to the fungus in vitro. Highly toxic fungicides were Benlate (benomyl), Thiram (thiram), and Orbit (propiconazole). EC5O values (mg a.i./L) were less than 1 and EC95 values were less than 10. Crown (carbathiin and thiabendazole), ASC-66835 (fluazinam), and UBI-2584 (tebuconazole) were moderately toxic, with EC5O values in the range 1-10 and EC95 values in the range 32-45. Weakly toxic fungicides (EC5O in the range 20-80, EC95 in the range 35-140) included UBI-2643 (thiabendazole), UBI-2565 (cyproconazole), and Vitaflo-280 (carbathiin and thiram). Anvil (hexaconazole), Vitaflo-250 (carbathiin), UBI-2383 (triadimenol), Daconil (chlorothalonil), CGA-173506 (fludioxonil), and CGA-169374 (difeno- conazole) were considered nontoxic to C. destmtan (EC5O 1.29->600, EC95>500). Relations between proportional inhibition of growth and concentration of fungicide were linear on arithmetic plots (Benlate, UBI-2643, UBI-2565, Vitaflo-280) or logarithmic plots (all other fungicides). Based on toxicity in vitro and formulation, it is recommended that Benlate, Orbit, and ASC-66835 be tested as soil drenches, and Benlate, Thiram, UBI-2584, and Crown be tested as seed treatments for controlling disappearing root rot.
Non-toxic stimulation of dendritic cells (DCs), which are central immunomodulators, may aid the prevention of cancer. Furthermore, induction of apoptosis in cancer cells by anticancer agents contributes to the induction of DC maturation. We previously reported that extracts from $Pinus$$parviflora$ Sieb. et Zucc pine cone and $Mucuna$ seed induce differentiation of mouse bone marrow cells into mature dendritic cells and also induce apoptosis in various human cancer cell lines. In the present study, we screened 31 kinds of edible beans with biological activity similar to that of extracts from pine cone and $Mucuna$ and found that the heat-stable extract from azuki bean ($Vigna$$angula$) stimulated differentiation of bone marrow cells into immature DCs with the greatest efficacy. The level of IL-6 produced by sequential treatment of DCs with azuki extract and lipopolysaccharide was the highest among the examined beans. Azuki extract also inhibited the growth of human leukemia U937 cells, leading to induction of apoptosis. These results suggest that azuki bean and its extract are immunopotentiating foods that can be used as a dietary supplement for cancer prevention and immunotherapy.
This article aims at demonstrating location specific approach for agricultural technology promotion and adoption in improving the livelihood of the small farmers in the haor basin and coastal belt of Bangladesh. Innovative technologies that have potentials are initially screened by ex-ante investigation and instrumented by the business model canvas, which is used as a bottom-up approach for sustainability of the adoption of proposed technology innovations. Village-level extension farmers, sub-district extension officers and farmers' cooperative are the unique and central features to the business models and forward linkages. Extension service, power tiller, low-lift pump, sunflower, shallow tube well, quality seed, forward linkage for farmed duck eggs, live ducks and open catch fish etc. are the suggested potential technology innovations for the small farmers. The technology adoption business model can be reinvented for different locations within or beyond the country considering the local agricultural problems and prospects for greater sustainability.
Apurinic/apyrimidinic endonuclease 1 (APE1) is an enzyme responsible for the initial step in the base excision repair pathway and is known to be a potential drug target for treating cancers, because its expression is associated with resistance to DNA-damaging anticancer agents. Although several inhibitors already have been identified, the identification of novel kinds of potential inhibitors of APE1 could provide a seed for the development of improved anticancer drugs. For this purpose, we first classified known inhibitors of APE1. According to the classification, we constructed two distinct pharmacophore models. We screened more than 3 million lead-like compounds using the pharmacophores. Hits that fulfilled the features of the pharmacophore models were identified. In addition to the pharmacophore screen, we carried out molecular docking to prioritize hits. Based on these processes, we ultimately identified 1,338 potential inhibitors of APE1 with predicted binding affinities to the enzyme.
We established an efficient plant regeneration system for Catharanthus roseus L. (G.) Don through somatic embryogenesis. Embryogenic callus was induced from hypocotyl of seed germinated in vitro. Somatic embryogenesis in Catharanthus has been categorized into three distinct stages: (1) initiation and proliferation of embryo; (2) maturation, and; (3) germination or plantlet conversion. Beside plant growth regulators, various stages of embryogenesis were screened for their response to a wide variety of factors (pH, gelrite, light, sugar alcohols, polyethyleneglycol and amino acids), which affect embryogenesis. All of the tested factors had a small to marked influence on embryogeny and eventual conversion to plantlets. The plantlets were acclimatized successfully in a greenhouse. To our knowledge, this is the first report describing a detailed study of various cultural factors which regulate embryogenesis in C. roseus. The results discussed in this paper may be used in mass propagation to produce medicinal raw material, and the embryo precursor cells could be used in genetic modification programmes that aim to improve the alkaloid yield as well.
Among 24 isolates of fluorescent Pseudomonads, 5 isolates named as LPT1, LPT2, LPT3, LPT4 and LPT5 were screened in vitro for their nematicidal activity against cyst forming nematode, Heterodera cajani causing patchiness, poor and stunting growth besides discoloration in Sesamum indicum. Second stage juveniles of H. cajani hatched from egg masses were collected from roots of host plant and subjected to fresh and heat-treated culture filtrate of isolates for 24 h. Mortality of H. cajani was recorded on the basis of parameters used for test organism bioassay. Among these isolates, Pseudomonas aeruginosa LPT5 caused maximum mortality towards second stage juvenile of H. cajani in vitro. Five isolates were used as seed coating for the management of cyst forming nematode H. cajani on sesame in green house condition. The strains LPT5 was better than the other strains in reducing the population of H. cajani both in vitro and in vivo. The reduction in cyst and juveniles population was found to be 49 and 60%, respectively when seeds were coated with strain LPT5. Among other strains, LPT4 was also found to inhibit the cyst and juveniles population 12 and 36% respectively. Increases in early vegetative plant growth parameters recorded in both in vitro and in vivo further revealed the significance of indigenous bacteria in comparison to introduced strain.
