Morphological and Cultural Characteristics of Fungi Causing Rice Sclerotial Diseases (수도균핵병(水稻菌核病)을 일으키는 진균(眞菌)의 형태적(形態的) 및 배양적(培養的) 특성(特性))

  • Morphological and cultural characteristics of fungi causing rice sclerotial diseases were examined. Hyphal widths of R. solani and R. oryzae were same and ranged $6.0-12.0\;{\mu}m$ with average $9.0\;{\mu}m$, the widest among those of the sclerotial fungi examined. Hyphal width of R. oryzae sativae ranged $6.0-9.0{\mu}m$ with average $7.4{\mu}m$. Hyphal width of R. cerealis was the narrowest among those of Rhizoctonia species examined, and the same was hyphal width of S. oryzae among those of Sclerotium species. Nuclear staining by HCL-Giemsa method showed that R. solani and R. oryzae had many nuclei within one hyphal cell, S. oryzae one nucleus, and the other sclerotial fungi mostly two nuclei. The nuclear number of R. solani was the largest, which ranged 2-17 with average 6.3. Average size of sclerotia of the sclerotial fungi except S. hydrophilum and S. oryzae produced in lesions ranged 1.0-2.0mm. Average size of sclerotia of S. hydrophilum and S. oryzae was 0.5mm and 0.24mm, respectively. Sclerotia of R. solani and R. oryzae produced in culture were more variable in size and larger than those produced in lesions. However, the sclerotial sizes of the other sclerotial fungi produced in culture were almost the same as those produced in lesions. Sclerotial colors of sclerotial fungi produced in lesions were similar to those produced in culture, but sclerotial shapes of some sclerotial fungi exhibited somewhat difference between the sclerotia produced in lesions and in culture. Optimum temperature for mycelial growth of R. cerealis was $23^{\circ}C$, and that of the other sclerotial fungi ranged from $27\;to\;33^{\circ}C$. Maximum temperature for mycelial growth of some sclerotial fungi was as high as $41^{\circ}C$, while that of R. cerealis was as low as $31^{\circ}C$. Minimum temperature for mycelial growth of R. cerealis was $2^{\circ}C$, and that of the other sclerotial fungi ranged from $6\;to\;10^{\circ}C$.

Various Cultural Factors Associated with Disease Development of Garlic White Rot Caused by Two Species of Sclerotium (마늘 흑색썩음균핵병 발생에 관여하는 여러가지 경종적 요인)

  • This study was conducted to investigate the control possibility of garlic white rot causing severe yield losses of Allium species and cultivars using cultural practices such as optimal sowing date and burial depth, and lime application. Inoculum density in infested field soil was investigated at different soil depth, and that on the diseased plant debris was done. Inoculum density and recovery ratio of white rot pathogen of garlic was highly different between two species of Sclerotium cepivorum forming comparatively small sclerotia and Sclerotium sp. forming comparatively large ones. It was confirmed that S. cepivorum formed more sclerotia on bulbs of garlic than S. sp., and sclerotial recovery of S. cepivorum was higher than that of S. sp. Inoculum density of white rot pathogen in the infested field at garlic seeding period ranged from one to thirteen sclerotia per 30 g soil. Inoculum density of white rot pathogen decreased remarkably with increasing soil depth and above 95% of sclerotia were distributed within 5 cm of soil depth. Disease severity of white rot was higher on slightly planted garlics than deeply-planted ones. Garlic seed bulbs infected by white rot pathogens were confirmed to be one of main inoculum sources of white rot in the field and the disease incidences caused by garlic seed transmission showed big differences among garlic varieties. When nine garlic varieties harvested from infested plots were sown in the field, highly susceptible varieties, ‘Wando’, ‘Daeseo’, ‘Namdo’ and ‘Kodang’ showed high disease incidences, whereas other five varieties were not infected at all. It was confirmed that white rot occurred higher on early-sown garlics, before middle October, than on late-sown ones, after late October. Meanwhile, increasing application rate of lime ranged from 100 to 300 g reduced disease severity of white rot.

Ecological Studies on the Transition of Sheath Blight of Rice in Korea (한국(韓國)에서의 벼 잎집무늬마름병 발생변동(發生變動)에 관(關)한 생태학적(生態學的) 연구(硏究))

  • In an attempt to obtain a basic information to develop an effective integrated system of controlling sheath blight of rice in Korea, the transition of this disease, the variation of cultural characters and pathogenicity of the pathogen, environmental conditions affecting the disease outbreak and varietal resistance have been investigated. 1. Rice sheath blight which has been minor disease in the past was widely spread, especially since 1971. This disease has promptly spread all over the country and infected 65.2% of total rice growing area in 1976. Various factors are considered to be related to such transition of this disease. Above all, increace of application of nitrogenous fertilizer, early season and earlier cultivation of rice, introduction of more susceptible "Tongil" varieties etc. must be important factors influencing the outbreak of this disease. 2. Great variations in cultural characteristics-such as mycelial growth rate, color of the medium, amount of the aerial mycelium, shape and color of the sclerotia- and in the pathogenicity of isolates of the pathogen, Thanatephorus cucumeris Dank were observed. The optimum temperature for mycelial growth also varied with isolates, from $25^{\circ}C$ to $30^{\circ}C$. There were not necessarily any correlation between curtural characteristics and pathogenicity of isolates of Thanatephorus cucumens. 3. Mycelial grow th of isolates of Thanatephorus cucumens on the PDA medium were correlated with the air temperatures of the region where the isolates were collected. The isolates from the regions with high temperature grew well on PDA medium at $35^{\circ}C$ than those from the region with low temperature, on the other hand, the isolates from the regions with the low temperature grew well on the same medium at $12^{\circ}C$ than those from the regions with high temperature. 4. Pectin polygalacturonase (PG) and cellulase (Cx) were most active on the 3rd day after inoculation on the leaves of rice plant with Thanatephorus cucumeris, whereas pectin methylestrase (PE) was most active on the 4th day after inoculation. Relationship between the activities of PE of isolates and the strength of pathogenicity of isolates was obtained, but PG and cellulase activities were not correlated with pathogenicity of isolates. 5. The tolerence of sclerotia from in-vitro culture to low temperature varied with their water content, the dried cultural sclerotia were more tolerent than wet ones, Dried cultural sclerotia maintained almost 100% germinability for 45 days at $-20^{\circ}C$, whereas wet sclerotia lost viability at $-5^{\circ}C$. The germination ratio of the sclerotia after overwintering changed from 18% to 70% according to the water content of the test paddy fields and the ratio was low in wet paddy condition. 6. To investigate the host range of this fungi in and near paddy field, 17 weeds were inoculated with fungi. The lesions of sheath blight disease was obserbed on Sagittaria trifolia L., Echinochloa crusgalli P. Beauv., Monochoria vaginal is Presl, Polygonum Hydropiper L., Eclipta prostrata L., Digitaria sanguinalis Scapoli. 7. When the level of nitrogen applied was doubled over standard level, total nitrogen content in rice sheath increased, ami when silicate was applied, starch content in rice sheath decreased, inducing the rice plants more susceptible to sheath blight disease. Increased dressing of potash ferilizer reduced the incidence of sheat blight disease. 8. The percentage of infected stems in the early period increased more in the narrow hill plot than in the wide hill plot, but in the late period this tendency was inversed; the percentage of infected stems as well as severity in the wide hill plot increased more compared to the narrow hill plot, and the disease severity in the one plant per hill plot was also low. The number of stems in the wide hill plot was more than the number of stems in the narrow hill plot. This indicates that the microclimate, such as the relative humidity, in the narrow hill plot was more favorable for the development of this disease. 9. There was a high negative correlation between the disease severity of varieties to the sheath blight and the maturity of the varieties, that is, the early varieties were more susceptible than the late ones, and much-tillering varieties usually showed more infection than less tillering varieties. 10. No relationship was obtained between the percentage of infected stems in the early period and the severity after heading, whereas a distinct relationship was obtained between former and latter after Aug. 10.

Identification of the Actinomycetes Strain No. 497, Isolated from Soil, Producing Actinomycin Antibiotic MT-497 (Actinomycin계열 항생물질 MT-497 을 생산하는 방선균 분리주 No.497의 동정)

  • Identification of the Actinomycetes isolate strain No. 497 producing an actinomycin antibiotic MT-497 was performed by ISP and chemotaxonomic methods. The strain Nu. 497 formed various shapes of sclerotia and smooth surface spore. Menaquinone MK-9 ($H_6, H_8$) and iso-, anteiso-branched $C_{15}C_{17}$ fatty acids were detected from whole cell extract. The wall chemotype of stram No. 497 was decided as wall chemotype I from the analysis of DAP isomer, peptidoglycan type and sugar pattern. From these morphological, chemotaxonomic characteristics and analysis of various physiological characteristics. the strain No. 497 was identified as Streptomyces nigrifaciens.

First Report of Sclerotium Rot on Cymbidium Orchids Caused by Sclerotium rolfsii in Korea

  • Sclerotium rot was found on Cymbidium orchids at Seosan-si, Chungcheongnam-do, Korea, in July, 2010. Symptoms occurred on low leaves, which turned yellowish, after which the entire plant wilted. Severely infected plants were blighted and eventually died. White mycelial mats and sclerotia appeared on pseudobulbs. Based on the mycological characteristics and pathogenicity, the causal fungus was identified as Sclerotium rolfsii. This is the first report of new Sclerotium rot on Cymbidium spp. caused by S. rolfsii in Korea.

Mulberry popcorn disease occurrence in Korea region and development of integrative control method

  • Mulberry fruits also have tremendous potential for providing various valuable industrial products of very high economic value for human beings. Nevertheless, through global warming, the popcorn disease caused by sclerotia forming fungi reduces the productivity of mulberry fruits in worldwide. So, in this study, we investigated damage ratio of mulberry popcorn disease in mulberry fruit production farm (Buan, Jeongueb, Sangju, Gochang in Korea). In Jeonbuk Buan, popcorn disease rate was the highest about 30%, on the other hand, in case of Gyungbuk Sangju and Jeonbuk Gochang, not damage. Also, we investigated about popcorn disease prevention by various of chemical treatment methods.

Persimmon Gray Mold Caused by Botrytis cinerea (Botrytis cinerea에 의한 감나무 잿빛곰팡이병)

  • Botrytis cinerea was isolated from a gray mo이 leaf lesion on persimmon in fields of Kyeonsangnam-do from 1996 to 1998, and etiological study was conducted including physiological characteristics. It formed gray mold lesion with light green color on leaves of persimmon (Diospyros kaki). The temperature range for mycelial growth was between $5^{\circ}C$ and $30^{\circ}C$ with the optimum temperatures of $20^{\circ}C$ to $25^{\circ}C$. Conidia were oviod, cylindric, and colorless and their dimensions in culture were $8.4~11.5\times7.0~8.9\mu\textrm{m}$. The optimum temperature of conidial germination was $25^{\circ}C$. Sclerotia on potato dextrose agar medium were well formed and brownish condiophores were observed with their size of $18.5~64.9\times4.5~8.0\mu\textrm{m}$. Symptoms on artificially inoculated plants were similar to those of gray mold disease on persimmon caused by Botrytis cinerea in fields.

Conserved Roles of MonA in Fungal Growth and Development in Aspergillus Species

  • MonA is a subunit of a guanine nucleotide exchange factor that is important for vacuole passing and autophagy processes in eukaryotes. In this study, we characterized the function of MonA, an orthologue of Saccharomyces cerevisiae Mon1, in the model fungus Aspergillus nidulans and a toxigenic fungus A. flavus. In A. nidulans, the absence of AnimonA led to decreased fungal growth, reduced asexual reproduction, and defective cleistothecia production. In addition, AnimonA deletion mutants exhibited decreased spore viability, had reduced trehalose contents in conidia, and were sensitive to thermal stress. In A. flavus, deletion of AflmonA caused decreased fungal growth and defective production of asexual spores and sclerotia structures. Moreover, the absence of monA affected vacuole morphology in both species. Taken together, these results indicate that MonA plays conserved roles in controlling fungal growth, development and vacuole morphology in A. nidulans and A. flavus.

  • Thirtythree strains of the Aspergillus spp. isolated from foodstuffs were observed through some physiological characteristics for detection of identification key of Aspergillus spp. 1) Each strain of Aspergillus spp. had their specific characteristics and could be used for identification of species. 2) Excellent amylase-producing fungi were observed among the isolated strains of Aspergillus spp. 3) Amylase activities increased for one week incubation period. 4) In the tests of common characters of aflatoxin-producing fungi among the 33 strains of Aspergillus spp., for example, conidial size, presence of sclerotia, kojic acid, and pigment production, coloration of phenol, reduction of methylene blue, etc.
