Studies on Screening of Seaweed Extracts for Peroxynitrite and DPPH Radical Scavenging Activities (과산화아질산염과 DPPH 라디칼에 대한 해조추출물의 소거 활성 효과)

  • As a part of our search for novel antioxidants from the seaweeds, we have investigated radical scavenging effect for their crude extracts using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, authentic peroxynitrite, and 3-morpholinsydnonimine (SIN-1), a peroxynitrite-generating species in vitro. Thirty-four seaweeds were screened for $ONOO^-$ and DPPH radical scavenging activities. A potent inhibitory effect against peroxynitrite generated by SIN-1 at $5{\mu}g/ml$ of methanol extracts was observed in order of Ishige okamurae(95.3%), Sargassum hemiphyllum(90.2%), Symphyocladia latiuscula(89.6%), Porphyra suborbiculata(86.7%), and Gelidium amamsii(85.9%), Also, a significant scavenging effect against direct authentic peroxynitrite was revekaled for methanol extracts of Ishige okamurae(66.2%) and Sargassum hemiphyllum(55.2%) and the acetone/methylene chloride(1:1) extract of Gigatina tenella (61.0%). In our measurement for evaluating the capacity to scavenge the stable free radical of DPPH, acetone/methylene chloride(1:1) extracts of Symphyocladia latiuscula, Gloiopeltis furcata, and Sargassum thunbergii and the methanol extract of Sargassum sp. showed an inhibitory potency of 85.8%, 82.8%, 74.1%, and 64.0%, respectively.

The Scavenging Effect on Nitric Oxide in Carthami Flos Herbal-Acupunture Solution (홍화 약침액의 Nitric Oxide에 대한 소거효과)

  • Objectives : Free radical metabolism seems to occupy a remarkably common position in the mechanisms of aging and aging related disease. Oxidative damage to DNA, lipids. proteins and other molecules may contribute to the development of cancer, cardiovascular disease and possibly neurodegenerative disease. This study was designed to find out whether Carthami Flos Herbal-Acupuncture Solution can scavenge Nitric Oxide(NO) or not. SNAP is NO generator. NO concentration was estimated after 2,6, 12 and 24 hrs in no treatment group, after treatment with Vit. C or 1, 10, 100${\mu}$g/m1 of Carthami Flos Herbal-Acupuncture Solution. There was no significant scavenging effect of Carthami Flos Herbal-Acupuncture Solutionon NO after 2 hrs. But there was a significant scavenging effect of Cafhami Flos Herbal-Acupuncture Solution on NO in 10${\mu}$/m1 group after 6hrs. And there was a significant scavenging effect of carthami Flos Herbal-Acupuncture Solution on NO in 1, 10${\mu}$g/ml group after 12, 24 hrs. These results suggest that Carhami Flos Herbal-Acupuncture Solution has scavenging effect on NO. This study shows that Carthami Flos Herbal-Acupuncture Solution can be used for aging related disease and further studies are required to investigate the antioxidative effects of it.

The Scavenging Effect on NO, DPPH and Inhibitory Effect on IL-4 in Radix Cynanchi Wilfordii Herbal-Acupunture Solution (백하수오약침액(白何首烏藥鍼液)의 NO, DPPH 소거(消去) 및 IL-4 억제효과(抑制效果))

  • Objective: For designing an optimal countermeasure against ageing and ageing related disease, it is necessary to understand the ageing mechanism and other reactive species. Free radicals are associated with the mechanisam of biological ageing and age-related diseases. This study was performed to identifiy the scavenging effect of Radix Cynanchi Wilfordii Herbal-Acupunture Solution(RCWHA) on Nitric Oxide(NO), 1, 1-diphenyl-2-picryl-hydrazyl(DPPH) and Interleukin-4(IL -4). Methods: The activity to scavenge DPPH radicals and NO was observed in vitro, We evaluated the antioxidant activity of RCWHA by using DPPH or NO free radical method. Also the inhibitory effect on IL-4 in RCWHA was observed in vitro, Results: The followings are the summary of the results; (1) There is a significant scavenging effect of RCWHA on NO in 0.1mg/ml and 10mg/ml group after 24hrs. (2) There is no significant scavenging effect of RCWHA on DPPH. (3) There is a significant scavenging effect of RCWHA on IL-4 in $10{\mu}g/m{\ell}$ and $100{\mu}g/m{\ell}$ group. Conclusions: These findings indicate that RCWHA can be used as antioxidant or antiimflamatory drug. But further study is needed about the effect of RCWHA.

  • The ethanol extracts of mixed vegetables (Bioactive Vegetables, BV), mixed fruits (Bioactive Fruits, BF) and their liquid formulation (Chungpae Plus , CP) were evaluated for their antioxidative and antigenotoxic activities. They were shown to possess the significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and were revealed to show the inhibitory effect of lipid peroxidation as measured by malondialdehyde (MDA) formation although the potencies were not higher than those of vitamin C. They did not possess any pro-oxidant effect on bleomycin-Fe(III) dependent DNA degradation, whereas vitamin C showed strong pro-oxidant effect. Furthermore, oral administration of BV and BF inhibited micronucleated reticulocyte (MNRET) formation of mouse peripheral blood induced by KBrO3 treatment in vivo. CP also showed significant inhibition under same experimental condition. Therefore, the liquid formulation (CP) containing BV and BF may be a useful natural antioxidative and antigenotoxic agent by scavenging free radicals, inhibition of lipid peroxidation and protecting chromosomal damage.

Effect of Corydalis tuber Acua-acupuncture Solution on Antiacetylcholinesterase and Antioxidants (현호소약침액(玄胡索藥鍼液)의 acetylcholinesterase 억제효과와 항산화에 미치는 영향(影響))

  • It has been investigated about aging theory. However, aging mechanism still remains to be unknown. Aging and aging related diseases might be due to oxidative damage and these were modifiable by genetic and environmental factors. For designing an optimal medical treatment and countermeasure against aging and aging related disease, it is necessary to understand the aging mechanism. Acetylcholine(Ach) plays an important role in memory. If someone doesn't have enough Ach, he has a tendency to catch a Alzheimer's disease. Corydalis tuber has been clinically used to treat heart disease, gastrointestinal disease and other diseases including endocrine disease in Oriental medicine. The purpose of this article is to investigate the inhibitory effect on Acetylcholinesterase and scavenging effects on NO, DPPH of Corydalis tuber Acua-acupuncture solution(CTAS). The results are summerised as follows; 1. There is a significant inhibitory effect of $0.01mg/m{\ell}$ CTAS group at 20, 30, 60 minutes and $0.1mg/m{\ell}$ CTAS group at 10, 20, 30, 60 minutes on AchE. 2. There is no significant scavenging effect of CTAS on NO. 3. There is a significant scavenging effect of $0.1mg/m{\ell}$ and $0.01mg/m{\ell}$ CTAS group at 10 minutes but there is no significant scavenging effect at 20, 30, 60 minutes on DPPH. There is a significant scavenging effect of $1mg/m{\ell}$ CTAS group at 10, 20, 30, 60 minutes on DPPH.

  • The scavenging characteristics have a great influence on the performance of a diesel engine, especially slow-speed two-stroke diesel engines which are usually used as a marine propulsion power plant, and they are greatly affected by the conditions in the cylinder, intake and exhaust manifolds, and the opening and closing timing of scavenging ports or exhaust valves during the gas exchange process. Besides, there are many other factors to affect the scavenging characteristics and these factors interact each other very complicatedly. Therefore the simulation program of the gas exchange process is very useful to improve and predict the scavenging characteristics, due to the high costs associated with redesign and testing. In this paper it was attenpted to investigate the effect of the variation of the pressure ratio of intake to exhaust manifolds, and the variation of the opening and closing timing of a exhaust valve by using a computational program for a three-zone scavenging model which was developed by authors. The computed results showed that the scavenging efficiency and delivery ratio increased considerably, but the trapping efficiency decreased with increasing of the pressure ratio of intake to exhaust manifolds. The scavenging efficiency, trapping efficiency, and th conditions of the cylinder gases were affected by the opening timing of the exhaust valve, but the delivery ratio by the closing timing.

Experimental Study on Antioxidative Effect of Luffae Fructus Retinervus and Their Effects on Cytokines to 3T3-L1 Cell Lines (사과락(絲瓜絡)의 항산화효과 및 3T3-L1분화 시 Cytokine류에 미치는 영향)

  • In recent year, We are concerned in anti-aging, disease-protection, long-life, many method are used in solving this problem. Recently, We heard that Luffae Fructus Retinervus(LFR) has effect of anti-aging, disease-protection, long-life. So I let made a experiment for this result. The purpose of this study is to; 1) the anti-oxidant effect of Luffae Fructus Retinervus(LFR) used for 3 methods, those are DPPH radical scavenging activity, Nitric oxide(NO) radical scavenging activity, Superoxide anion radical scavenging activity, 2) cultivation 3T3-L 1 Preadipocytes and Protein chip used for ProteoPlexTM 16-Well Murine Cytokine Array Kit. We measured level of DPPH radical scavenging activity. And we experienced that the ability of DPPH radical's elimination was increased by rising concentration of LFR. When the concentration of LFR was 5 mg/ml, the ability of DPPH radical's elimination was Maximum. We measured level of Nitric oxide(NO) radical scavenging activity. And we founded that the ability of NO radical's elimination was significant when concentration of LFR was from 1.25 mg/ml to 2.5 mg/ml. We measured level of Superoxide anion radical scavenging activity. And we founded that the ability of Superoxide anion radical's elimination was maximum when concentration of LFR was 0.3125 mg. When we inspected Antioxidative Effects with BSA, we experienced that ability of defense was increased by rising concentration of LFR. We known the immunity of LFR about 3T3-L1 Preadipocytes and gained the increase of Cytokines(IL-2, IL-4, GM-CSF) without IL-12p70, $INF-{\gamma}$, $TNF-{\alpha}$ So I guess that Luffae Fructus Retinervus(LFR) has effects of anti-aging, disease-protection, long-life, etc.

This study was designed to find out whether Ursi Fel ${\cdot}$ Bovis Calculus Herbal-Acupuncture Solution(UBHAS) can scavenge nitric oxide(NO) or not. NO concentration was estimated after treatment with Vit. C or 1, 10, $100{\mu}g/m{\ell}$ of UBHAS. There was no significant scavenging effect of UBHAS on NO in 1, $100{\mu}g/m{\ell}$ group. while significant scavenging in $l0{\mu}g/m{\ell}$ group after 2hrs. UBHAS showed significant scavenging effects on NO in 1, 10, $100{\mu}g/m{\ell}$ group after 6hrs. There was a significant scavenging effect in 10, $100{\mu}g/m{\ell}$ group. even though no significant in $1{\mu}g/m{\ell}$ group after 12, 24 hrs. This study showed that UBHAS might be used for aging related disease. These results suggest that UBHAS has scavenging effect on NO. however, further studies are required to investigate the antioxidative effects of it.

Radical Scavenging and Antioxidant Effects of Juglandis Semen Extract(JSE) (호도약침액(胡桃藥鍼液)의 유리기(遊離基) 소거(消去)와 항산화(抗酸化) 효과(效果)에 대한 실험적(實驗的) 연구(硏究))

  • This study was performed to determine if Juglandis semen extract(JSE) has free radical scavenging and antioxidant activities. Superoxide anion generation by xanthine oxidase/xanthine and in neutrophils activated by phorbol-12, 13-dibutyrate was inhibited by JSE and its effect was dose-dependent. JSE also inhibited generation of $H_2O_2$ induced by glucose oxidase/glucose and in opossum kidney cells treated with antimycin A. JSE exerted a direct $H_2O_2$ scavenging effect. Exposure of opossum kidney cells to 1mM tBHP caused a significant increase in lipid peroxidation, which was prevented by JSE. JSE also prevented tBHP-induced LDH release. These data suggest that JSE has free radical scavenging and antioxidant activities. However, further studies should be carried out to find the active ingredient(s) of JSE that exerts radical scavenging action.

Effect of Heating on DPPH Radical Scavenging Activity of Meat Substitute

  • This study was conducted to evaluate the increase of DPPH radical scavenging activity of meat substitute by heating. The meat substitute showed higher DPPH radical scavenging activity than those of other foods rich in protein such as beef, pork, chicken, and soybean curd. The DPPH radical scavenging activity of meat substitute was dependent upon concentration, heating temperature and heating time of meat substitute. The DPPH radical scavenging activity of meat substitute was enhanced with increasing heating temperature and time. The increase of DPPH radical scavenging activity was only applied to meat substitute without showing any activation in other foods rich in protein such as beef, pork, chicken, and soybean curd.
