Determination of Salt Type, Salt Concentration, and Salt Application Method and Timing for Suppression of Stem Elongation in Grafted Cucumber Seedlings (오이 접목묘의 도장억제를 위한 염 스트레스 처리 효과)

  • This study aimed to examine a suppression effect of salinity on extension rate of stem elongation of cucumber seedlings (Cucumis sativus L.) grafted with roots of figleaf gourd plant. The effects of application methods, timings, and concentrations of two salt types, sodium chloride (NaCl) and calcium chloride ($CaCl_2$), were compared to untreated control plants. In result, an obvious suppression effect on the excessive elongation of stem was obtained by both sub-irrigated and medium-mixed NaCl salt. An improvement in quality of transplants was also obtained by the sub-irrigated NaCl salt. Foliar-applied NaCl caused visible leaf injury when the concentration was higher than 40 mM; but, with no effect on suppressing the stem elongation. When the NaCl was applied at 7 days after grafting, a higher concentration of NaCl was demanded for suppressing the stem elongation compared to an application at the day of grafting. No effect of the NaCl salt on the fresh weights of 36-day grown plants was observed; but, there was a negative effect on the number of female flowers at a high temperature season. Overall, the NaCl salt was more effective on slowing down the stem elongation and had the lower incidence of leaf injury than the $CaCl_2$ salt.

A Study on the Effect Factor to the Foam Generating Characteristics of High Expansion Foam (고팽창포소화약제 발포특성에 영향을 미치는 요인 연구)

  • Using the high expansion foam generator of ISO 7203-2 which spray 6 liter at 5 bar, foam generation characteristics was studied. Wind flow rate, foam screen, concentration of foam agent solution and concentration of salt of water were varied to find the effect of the parameters on foam generation. Research result showed that expansion ratio of foam was increased with wind flow rate. The expansion ratio of foam in the perforated type standard screen was higher than the wire mesh screen. Expansion ratio and drainage time were increased with increase of foam solution concentration. But a increase of salt concentration in solution showed the decrease of expansion ratio and drainage time.

  • This paper will begin by describing osmosis and how reverse osmosis works. It will show how osmotic pressure affects reverse osmosis operations. It uill explain salt rejection, membrane flux, and recovery rates and the affect that salt built up has on membrane performance. It wil 1 explain the limitations of RO performance and why pretreatment is important. It will describe the two basic types of membrane, asymmetric and thin-film composite and explain the difference between these types plus compare cellulose acetate types to aromatic polyamide type membranes. It will discuss operating efficiences as it compares to feedwater pressure, concentration, temperature and pH. Finally, it will discuss the differences between tubular, plate and frame, hollow fiber and spiral wound element design. It will be a paper that talks about the basics of RO systems and should give a person who is unfamiliar with RO a basic introduction to this type of separation technology.

Quality characteristics of Doenjang depending on various salt concentration during long-term fermentation period (다양한 염도에서 제조한 된장의 장기 숙성 시 품질변화)

  • This study presented the change of quality characteristics of Doenjang with various salt concentrations (4, 8, 15, and 20%) during 12 months. The pHs of Doenjang during fermentation were gradually increased for 12 months, whereas the pHs of Doenjang with 4 and 8% salt increased rapidly. Titratable acidity of Doenjang with 4 and 8% salt was high at initial time, while Doenjang with 15 and 20% salt was high at the 9 and 12 months. The amino-type nitrogen content of Doenjang with 4 and 8% salt was increased and ${\alpha}-amylase$ activity of all Doenjang was decreased during whole fermentation period. The protease activity of Doenjang with 4 and 8% salt was higher than Doenjang with 15 and 20% salt until 7 months. Total aerobic bacteria number 8.0~9.1 log CFU/g for 12 months, which was steady during fermentation period. Doenjang with 8% salt showed the highest score in umami taste whereas the lowest score in bitterness, astringency, and sourness tastes between 5 and 12 months. In conclusion, Doenjang with 8% salt was a suitable concentration for low-salt fermented foods.

Isolation and Identification of Staphylococcus sp. from Korean Fermented Fish Products

  • In order to find out if staphylococci occur in significant numbers in Korean fermented fish products, a total of 40 different fermented fish products were collected from different markets in Korea and analyzed for their physico-chemical and microbiological states. The pH, salt concentration and water activity of the products were measured and the total viable cell count and the number of Staphylococcus grown on mannitol salt agar were determined. The identification of the strains of Staphylococcus were made by API Staph Strip and MIS identification kits, and the physiological properties of the identified strains were further characterized by different conventional methods. The pH, salt content and water activity of fermented fish samples varied widely from 4.8 to 7.1, 7.4-28.7$%$ and 0.77-0.84, repectively, depending on the type of product. The total viable cell count varied from $10^4-10^9$ cfu/ml, and most of the samples had $10^5-10^6$ cfu/ml No correlation was found between the viable cell count and the pH, NaCl concentration and water activity of the samples. Among the 35 colonies identified as Staphylococcus strains by the identification kits, S. xylosus was the most frequently occurring strain marking 17, and S. warneri was 8, S. epidermidis 4 and S. cohnii 2. S. hominis, S. saprophyticus, S. haemolyticus and S. aureus were also identified once each. In some samples (K-3, P-6, K-8, G-5 and G-10), 2-3 different species of Staphylococcus were found. Considering the region of sampling, among the 10 samples from Kunsan 5 were identified as S. warneri, while in the other regions S. xylosus was predominant. Although the physiological characteristics of the identified strains were generally consistent with those in Bergey's Manual, some discrepances were also observed. All the strains were highly salt tolerant, growing in the media containing over 18$%$ NaCl. All the strains except S. aureus (G-11) showed negative in hemolysis activity, plasma coagulation and DNase tests. All the strains including S. aureus (G-11) showed negative in enterotoxin test.

Assessment of Bile Salt Effects on S-Layer Production, slp Gene Expression and, Some Physicochemical Properties of Lactobacillus acidophilus ATCC 4356

  • In many conditions, bacterial surface properties are changed as a result of variation in the growth medium and conditions. This study examined the influence of bile salt concentrations (0-0.1%) on colony morphotype, hydrophobicity, $H_2O_2$ concentration, S-layer protein production, and slpA gene expression in Lactobacillus acidophilus ATCC 4356. It was observed that two types of colonies (R and S) were in the control group and the stress condition. When the bile level increased in the medium, the amount of S type was more than the R type. A stepwise increment in the bile concentration resulted in a stepwise decline in the maximum growth rate. The results showed that hydrophobicity was increased in 0.01%-0.02% bile, but it was decreased in 0.1% bile. Treatment by bile (0.01%-0.1%) profoundly decreased $H_2O_2$ formation. S-Layer protein and slpA gene expression were also altered by the stress condition. S-Protein expression was increased in the stress condition. The slpA gene expression increased in 0.01%-0.05% bile and it decreased in 0.1% bile. However, we found that different bile salt concentrations influenced the morphology and some surface properties of L. acidophilus ATCC 4356. These changes were very different in the 0.1% bile. It appears that the bacteria respond abruptly to 0.1% bile.

Application of Capacitive Deionization Packed Ion Exchange Resins in Two Flow Channels (두 가지 유로 형태에 따라 이온교환수지를 채운 축전식 탈염기술)

  • To desalinate the aqueous solutions with high salt concentration using the capacitive deionization technology, two resin/membrane capacitive deionization(RMCDI) cells were fabricated by filling mixed ion exchange resins in two different flow channels (spacer and spiral type). The salt removal efficiency of the spacer- and spiral-RMCDI was 77.21 and 99.94%, respectively. Many ions were significantly removed in a spiral RMCDI cell because the feed solution could be more evenly contacted with the ion exchange resins filled on the spiral type flow channel. As the result of the changes of pH and accumulative charges, it was observed that Faradaic reaction was diminished for a spiral RMCDI cell filled by the mixture of cation and anion exchange resins. Therefore, the desalination of the aqueous solutions with high salt concentration by the capacitive deionization technology was proven. In addition, further studies on the optimization of the mixing ratio with ion exchange resins and the introduction of the regeneration process generally occurred in the continuous electrodeionization (CEDI) technology are required to improve the RMCDI technology.

Influence of Notch Change on Corrosion Fatigue Fracture in F.E.M. Dual phase Steel of SS41 Steel (SS41강의 F.E.M.복합조직강에서 노치변화가 부식피로파괴에 미치는 영향)

  • The rotated bending fatigue test was conducted in air md in 3.5% NaCl salt solution to investigate the fatigue fracture behaviour of raw material and F.E.M dual phase steel made from raw material(SS41) by a suitable heat treatment. This study has compared the initial microcrack creation of material by tensile test with that by fatigue test. And the rotated bending test of cantilever type under the condition of 3.5% NaCl salt solution and air has investigated the corrosion fatigue fracture behaviour with the variation of stress concentration factor determined by each of notch shapes. The initial microcrack have been developed in fragile grainboundary with general corrosion occurring in raw material : in the pits built up by corrosion in F.E.M. dual phase steel because pits bring out stress concentration. It is small that the degree of decrease in corrosion fatigue life for F.E.M. dual phase steel compared with raw material because the notch sensitivity of F.E.M. dual phase steel is lower than raw material in reason of characteristics with two-phase construction.

Antihypertensive effect of Ganjang (traditional Korean soy sauce) on Sprague-Dawley Rats

  • BACKGROUND/OBJECTIVES: Although Korean fermented foods contain large amounts of salt, which is known to exacerbate health problems, these foods still have beneficial effects such as anti-hypertension, anti-cancer, and anti-colitis properties. We hypothesized that ganjang may have different effects on blood pressure compared to same concentrations of salt. MATERIALS/METHODS: Sprague-Dawley rats were divided into control (CT), NaCl (NC), and ganjang (GJ) groups and orally administered with 8% NaCl concentration for 9 weeks. The systolic blood pressure (SBP), serum chemistry, $Na^+$ and $K^+$ concentrations and renal gene expressions were measured. RESULTS: The SBP was significantly increased in the NC group compared to the GJ and CT groups. In addition, the $Na^+$ concentration in urine was higher in the GJ and NC groups than the CT group, but the urine volume was increased in the GJ group compared to the other groups. The serum renin levels were decreased in the GJ group compared to the CT group, while the serum aldosterone level was decreased in the GJ group relative to the NC group. The mRNA expression of the renin, angiotensin II type I receptor, and mineralocorticoid receptor were significantly lower in the GJ group compared to other groups. Furthermore, GJ group showed the lowest levels of genes for $Na^+$ transporter in kidney cortex such as $Na^+/K^+$ $ATPase{\alpha}1$ ($NKA{\alpha}1$), $Na^+/H^+$ exchanger 3 (NHE3), $Na^+/HCO_3{^-}$ co-exchanger (NBC), and carbonic anhydrases II (CAII). CONCLUSIONS: The decreased SBP in the GJ could be due to decreased renin and aldosterone levels in serum and increased urinary volume and excretion of $Na^+$ with its transporter gene alteration. Therefore, ganjang may have antihypertensive effect despite its high contents of salt.

The Effect of Sodium Chloride on the Serine-type Fibrinolytic Enzymes and the Thermostability of Extracellular Protease from Bacillus amyloliquefaciens DJ-4

  • By adding sodium chloride (2.5%) into a Bacillus amyloliquefaciens DJ-4 culture broth, two serine-type fibrinolytic proteases with a molecular weight of 29 (subtilisin DJ-4) and 38-kDa were stimulated on the SDS-fibrin zymogram or inhibitor gels. B. amyloliquefaciens DJ-4 showed the highest proteolytic activity (5.52 plasmin NIH unit/ml) on the fibrin plate based on the molar ratio when cells were subjected to the 2.5% NaCl. Using a fibrin plate, the secreted protease from this strain in the presence of 5% NaCl showed that about 49% of the enzyme's activity remained after incubation at $60^{\circ}C$ for 30 min, but as the salt concentration was increased (10% NaCl) the activity nearly disappeared (0.14 plasmin NIH unit/ml). However, through a fibrin zymography assay, three fibrinolytic enzymes (38, 53 and 80-kDa) from the cells in the presence of 10% NaCl were detected. Also, two salt-activated serine-type fibrinolytic professes (29 and 38kDa) showed thermostability from 65 to $70^{\circ}C$ for 30 min. Furthermore, these professes also showed stability, pH 6-11. In particular, 29-kDa (subtilisin DJ-4) was very stable in the pH range of 4-11 at $4^{\circ}C$ for 48 h.
