A Study on the Extraction and Purification of Glutathione from Yeast

  • During the course of studies on the production of glutathione from yeast, process development and optimization was carried out. The optimal pH for the extraction of glutathione was found to be 2.5 to 4.0 and the maximum yield for glutathione was obtained when the extraction temperature was 25 to $45^{\circ}C$. The cuprous salt of glutathione was recovered maximally at the range of 2 to 4g of cuprous oxide per 10 Kg of compressed yeast. Further purification was needed for the removal of impurities from glutathione. Cation exchange resin, anion exchange resin and Sephadex G-25 were employed for this purpose. 13 to 15 g of glutathione was obtained from 10 Kg of compressed yeast and the purity was above 99.3%.

SEPARATION OF CsCl FROM LiCl-CsCl MOLTEN SALT BY COLD FINGER MELT CRYSTALLIZATION

  • This study provides a fundamental understanding of a cold finger melt crystallization technique by exploring the heat and mass transfer processes of cold finger separation. A series of experiments were performed using a simplified LiCl-CsCl system by varying initial CsCl concentrations (1, 3, 5, and 7.5 wt%), cold finger cooling rates (7.4, 9.8, 12.3, and 14.9 L/min), and separation times (5, 10, 15, and 30 min). Results showed a potential recycling rate of 0.36 g/min with a purity of 0.33 wt% CsCl in LiCl. A CsCl concentrated drip formation was found to decrease crystal purity especially for smaller crystal formations. Dimensionless heat and mass transfer correlations showed that separation production is primarily influenced by convective transfer controlled by cooling gas flow rate, where correlations are more accurate for slower cooling gas flow rates.

Property Changes of Salt-seasoned Salmon Roe during Fermentation

  • In order to utilize chum salmon returned in the east coast of Korea, the changes in the properties of Yeonual-jeot (salted salmon roe) fermented at different temperatures were measured for obtaining the basic data for its commercial production. Moisture content of salted salmon roe was significantly lower than raw salmon roe, while other proximate components were in the reverse. pH was constant for 56 days fermentation at $5^{\circ}C$, but decreased with fermentation Reriod at 10 and $15^{\circ}C$. The contents of amino-nitrogen and VBN were almost in constant at $5^{\circ}C$ up to 28 days of fermentation, but increased very slowly at $10^{\circ}C$ and moderately at $15^{\circ}C$. TBA value, and the numbers of total viable cells and lactic acid bacteria increased over all fermentation periods, more at higher fermentation temperatures.

  • Carbaryl was degradaded to monomethylamine(MA) by bacterium. which isolated from lake sediment. Its carbaryl degradation was maximized when grown on mineral salt medium conditioning 20 $\mu$M of carbaryl as a sole carbon source at 26${\circ}$C and initial pH 7.0-9.0 Its degradation ability was minimized at initial pH 3.0 and 5.0 The percent conversions { (moles of MA in excess of control / mole of carbaryl added) $\times$ 100} for 10. 20. 40 and 80 $\mu$M of carbaryl were 18.6. 16.1. 18.1 and 11.6 respectively. It suggests that increasing amount of carbaryl added above 80 $\mu$M. the percent conversion should be decreased. The MA production by the bacterium was lineary related to the cultural time. After 7days culture. its percent conversion was 46.2. and this result suggests that a half amount of carbaryl would be hydrolyzed to MA by the bacterium within 8 days.

  • 국내 및 해외에서의 멸치액젓 관련 제품의 가공공정과 포장에 대하여 살펴 본 후 멸치액젓의 표준화된 가공과 품질관리를 위해 새로운 가공공정 및 포장방법을 제시하였다. 제시된 방법에서는 원료 멸치를 소금과 혼합한 다음(염도 20%), 온도조절된 상태에서 발효숙성시키고, 여과 혹은 원심분리, 여과잔사를 염수첨가 후 2차 발효시켜서 재여과하여 1차 여과액과 혼합시키고, 용기에 포장하고 저온에서 살균하는 과정에 의해 구성되도록 설계되었다. 여과잔사 폐기물의 처리 문제도 함께 고려하였다.

  • In Korea, fermented fish has been playing an important role as a preserved and flavor rich food. It is said that the digestion of fish protein is due to both action of intrinsic (autolytic enzymes) and bacterial enzymes in fish. The mass production of fermented fish has been impeded since traditional method of fermentation requires a long duration for a complete digestion. A high concentration of salt and unsanitary condition are also considered disadvantages of the old method. To improve the quality of the product and to develop mechanized process of fermentation, fermentors which have such control device as temperature, pH and agitation control system have been urgently needed. In this study, a model design of a fermentor is studied. The calculation was based on the optimum conditions for enzymatic hydrolysis of fish protein which involve temperature, pH, viscosity and other factors.

기수호의 습성천이 현상과 생태적 복원정책 (On the Realities of Hydrach Succession in Brackish Water Lakes and Ecological Restoration Policy)

  • The purpose of this study is to present the model of ecological restoration through analysis on the condition of hydrach succession process in brackish water lake. To this study, we investigate and research the present condition of brackish water lake in South Korea. Most brackish water lake in South Korea have characteristics that is come into being malignant hydrach succession. Many lakes were reclaimed to increase foodstuff production. And river conservation work seperate lakes from the ocean. In a embryological view, most lakes based on the brackish water lakes, But present lakes show many different types such as a fresh water lake, a salt lake, a brackish water lake, a swamp, and a marshland. As a result, brackish water lakes have greatly damaged by diverse artificial interventions to promote people's activities. Therefore, in order to manage brackish water lakes systematically, criteria of lake preservation must be coincide with the present conditions of hydrach succession in each lake.

Immunomodulatory activity of Salicornia herbacea L. Components

  • Salicornia herbacea is an annual herb growing in salt marshes and on muddy seashores. Salicornia herbacea has been used as a fork medicine as well as a seasoned vegetable. In fork medicine, Salicornia herbacea has been used to treat a variety of diseases such as constipation, obesity, diabetes, asthma, arthritis and cancer. However, the biological mechanisms for these activities have not been characterized, nor the active components. The immunomodulatory activity of Salicornia herbacea components were studied in the present study. The components of Salicornia herbacea were prepared from the whole plant by passage through a fine screen, and then dialyzed against PBS overnight. Immunomodulatory activities of the Salicornia herbacea components were examined on a mouse macrophage cell line, RAW 264.7 cells. The Salicornia herbacea components were shown to stimulate cytokine production, nitric oxide release, and expression of surface molecules in a dose dependent manner. The Salicornia herbacea components also induced further differentiation of slightly adherent RAW 264.7 cell into strongly adherent macrophages. These results indicate that Salicornia herbacea contains immunomodulator(s) that induces activation of macrophages.

Isolation and Identification of Acid- and Bile-Tolerant Lactobacillus salivarius subsp. salivarius from Human Faeces

  • This study was conducted to isolate lactobacilli having characteristics to be used as health adjuncts with fermented milk products. Acid tolerant strains were selected in Lactobacilli MRS broth adjusted to pH 4.0 from human faeces. Bile tolerant strains were examined in Lactobacilli MRS broth in which 1.0% bile salt was added. Microhemagglutination tests using swine erythrocytes were performed to select lactobacilli having adherence properties to survive in the intestinal tract. By examination of these characteristics the strain Nam 27, which was isolated from adult faeces, was selected and identified as Lactobacillus salivarius subsp. salivarius based on carbohydrate fermentation and 16S rDNA sequencing.

  • Atorvastatin calcium salt (1) was obtained through the preparation of lactone compound (8) from 2-((4R,6R)-6-(2-(2-(4-fluorophenyl)-5-isopropyl-3-phenyl-4-(phenylcarbamoyl)-1H-pyrrol-1-yl)-ethyl)-2-phenyl-1,3,2-dioxaborinan-4-yl)acetic acid tert-butyl ester (9) by hydrolysis in basic condition. Efficient hydrolysis of boronate compound 9 aimed at the viable synthesis for commercial production and purification of Atorvastatin calcium is reported. Detail studies of evaluation procedure are also reported.
