Effects of Safflower Seeds on the Serum Levels of Insulin-like Growth Factors, Insulin-like Growth Factor Binding Protein-3 and BALP in Osteoporosis Induced-ovariectomized Rats (흰쥐의 난소제거로 유발한 골다공증에 대한 홍화씨의 IGFs, IGF binding protein-3 그리고 BALP에 대한 혈청내 효과)

  • This study was carried out to investigate the effects of the Korean Safflower (Carthamus inctorius L) seed powder on serum level of hormones and trabecula area during the recovery from osteoporosis induced ovariectomized rats. Four month-old rats were ovariectomized (OVX), remained untreated for 8 weeks, and were subsequently administered safflower seed (0.03 g/kg) every other day 30 for days. We examined the effects of treated safflower seed every 10 days on ovariectomy-related changes in Insulin-like Growth Factors, Insulin-like Growth Factor binding protein-3 (IGFBP-3), Estrogen, Bone-specific alkaline phosphotase, Calcium, and Phospotase in the serum, and also histomorphology of the proximal fibula metaphysis and femur/body weight rate. Ten and 20 days after safflower seed treatment in OVX rats, serum levels of IGF-I, -II and IGFBP-3 were not different from the Sham and OVX groups. In 30 days, serum levels of IGF-I,-II and IGFBP-3 were higher after safflower seed treatment in OVX rats as compared to the other two groups (p<0.05). Bone alkaline phosphatase levels were increased through safflower seed treatment in OVX rats compared to the other two groups in 30 days. There were no differences between OVX and safflower seed treated OVX rats in serum levels of estrogen and femur/body weight rate, but estrogen levels for the sham group were higher than for the other two groups. The safflower seed is increased to serum levels of IGFs, IGFBP-3 and BALP of osteoporosis induced by ovariectomized rats. Thus, we conclude that the safflower seed is a possible role for improvement of osteoporosis induced-ovariectomized rats.

Supplementation of Safflower Seed Powder and Extracts Enhances Bone Metabolism in Rib-Fractured Rats

  • The current study investigated the effect of Korean safflower (Carthamus tinctorius L.) seed powder and its water and ethanol extracts on bone metabolism during recovery from rib-fracture induced by surgical operation in rats. 10-week-old male Sprague-Dawley rats weighing about 320 g were divided into 9 groups after arrival: 10d control (AIN 76 semi-purified diet), 10d safflower seed powder (10d SS-powder), 10d safflower seed ethanol extract (10d SS-EtOH), 10d safflower seed water extract (10d SS-$H_2O$), 20d control (AIN-76 semi-purified diet), 20d safflower heed powder (20d SS-powder), 20d safflower seed ethanol extract (20d SS-EtOH), 20d safflower seed water extract (20d SS-$H_2O$), and 20d sham-operation (20d sham), The dietary level for all the supplements was 5% based on the raw material weight. The rats were fed the experimental diets for 10 days before the rib fracture operation and for a further 10 or 20 days after the operation. A number 9 rib was fractured surgically and a sham-operation also performed. The rats were then sacrificed on the l0th or 20th day after the operation. The body weight initially decreased after the operation in all the rib-fractured groups, then gradually recovered. The concentrations of plasma osteocalcin were higher in the control group than in all the safflower-supplemented groups 10 and 20 days after the rib-fracture (p < 0.05). The bone-specific ALP (alkaline phosphatase) activity was significantly higher in the SS-EtOH group than in the other groups 20 days after the rib-fracture (p < 0.05). The level of urinary DPD (deoxypridinoline) was significantly higher in the SS-EtOH and SS-$H_2O$ groups than in the other groups 10 days after the rib-fracture. When comparing the PTH (parathyroid hormone) and calcitonin levels, the SS-$H_2O$ group exhibited the highest PTH level among the groups 10 and 20 days after the rib-fracture. Thus, it was concluded that the bone turnover during the fracture-healing period was more rapid in the rats supplemented with safflower seed powder or its fractions than in the control rats. Furthermore, the SS-$H_2O$ fraction was identified as the most effective in stimulating bone remodeling, as bone resorption and bone formation were both significantly increased during fracture healing when compared to the control group.

Properties of the Chemical Composition of Safflower (Carthamus tinctorius L.) (홍화(Carthamus tinctorius L.)의 부위별 이화학적 특성)

  • In order to develop new materials for the functional feed, the components of safflower (Carthamus tinctorius L.) was studied. Chemical composition, minerals, amino acids, and fatty acids of the seed and the flower of safflower were analyzed. The chemical composition of safflower seed was 3.48% (w/w) moisture, 17.10% crude protein, 30.10% crude fat, 6.11% crude ash and these of safflower flower was 7.47% moisture, 26.30% crude protein, 11.50% crude fat, 5.73% crude ash. Mineral contents of the seed were K 170.70ppm, P 14.82ppm, Ca 13.17ppm, Mg 7.83 ppm whereas these of the flower were K 64.99 ppm, P 49.90 ppm, Ca 10.43ppm. Other mineral contents were less than 7.00ppm in all parts. The composition of the amino acid were approximately as follow, the major amino acid in all parts were aspartic acid, leucine, glycine, and arginine, the contents of these were 12.17mg/g, 11.52mg/g, 8.27mg/g, 6.99mg/g, 4.86mg/g in the seed, 19.35mg/g, 31.67mg/g, 10.30mg/g, 9.06mg/g, 12.51mg/g in the flower, respectively. The mahor fatty acids in the all parts were linoleic acid (C아래첨자). The linoleic acid and the palmitic acid (C아래첨자) in the seed and the flower parts were 77.75% (w/w), 19.32% and 8.37%, 25.62% respectively. On the basis of chemical analysis, the safflower showed to have relatively high contents of crude protein and crude fat, minerals, polyunsaturated fatty acid as linoleic acid. These results suggested that safflower was found to be a useful material of natural health food for the functional food development.

Effect of Pre-treatment with BTCA on Lightfastness of Cotton Fabric dyed with Combination of Safflower red / Amur Cork Tree (BTCA 전처리가 홍화/황벽 혼합 면염색물의 일광견뢰성에 미치는 영향)

  • Safflower red and Amur Cork Tree dyes have used for dyeing on cotton fabrics largely. But they have low degree of lightfastness, So this study investigated the effect of BTCA pre-treatment on lightfastness of cotton fabrics dyed with Safflower red and Amur Cork Tree. The result is as the following; By FT-IR spectra, crosslingking was proved in cotton fabric pre treated with BTCA. K/S value of Safflower red dyed cotton fabric pretreated with BTCA was much more decreased than untreated cotton fabric with BTCA. It showed that the hue of the Safflower red dyed cotton fabric changed from reddish purple to red. However, K/S value of Amur Cork Tree dyed cotton fabric pre treated with BTCA was much more increased than untreated cotton fabric with BTCA. It showed that the hue of the Amur Cork Tree dyed cotton fabric changed from green yellow to yellow. For K/S value of Combination dyed cotton fabric pre treated with BTCA, Safflower red color was much more decreased than untreated cotton fabric. But Amur Cork Tree color was much more increased than untreated cotton fabric. It changed from reddish purple, red, yellowe red, to yellow in hue. ${{\Delta}E^*}_{ab}$ of Combination dyed cotton fabric in 40 hours irradiation was 15.7, however ${{\Delta}E^*}_{ab}$ of Combination dyed cotton fabric treated with 2% BTCA was 8.0, and that treated with 3% BTCA was 9.8. So, pre-treatment with BTCA to combination dyed cotton fabric with Safflower red and Amur Cork Tree showed more effective improvement of Lightfastness than untreated cotton fabric with BTCA.

Molecular Species of Triglycerides in Safflower Oil (홍화유의 triglyceride 분자종에 관한 연구)

  • Triglyceride molecular species of safflower oil was examined from the profiles of triglyceride composition and the fatty acid at ${\beta}-position$ of glycerol. From the results, triglycerides were found to be 26kinds in safflower oil. The major triglyceride molecular species in safflower oil were identified to be PLL; 10,4%, OLL; 22.3%, LLL; 31.95%. Triglyceride molecular species of safflower oil characterized that species occupied by unsaturated fatty acids at ${\beta}-position$ were more than 90%.

Quality Characteristic of Drink and Tea-Bag Processed with Safflower Seed Powder (홍화씨분말을 이용한 음료 및 티백차의 품질특성)

  • Drink and tea-bag were processed with safflower seed powder. Drinks were processed with 90$^{\circ}$C hot water extraction. Yield, soluble solid, pH and centrifugation residue of drinks were ranged in 79.2~89.3%, 0.6~0.99%, 5.98~6.40 and 1.00~1.18, respectively. Sensory score of overall acceptance in drinks were highest at that of enzyme treated drink. Tea-bag was processed with roasted safflower seed powder, alone(1.2g) and the mixtured tea-bags were consisted of persimmon leaves, pine needle and angelica gigas powder in same amount, respectively. Teas were prepared with extraction at 80$^{\circ}$C water for 2 min. Soluble solid and sensory evaluation score of teas were the highest when the tea was processed with safflower seed alone.

Anti-wrinkle Effect of Herbal Medicine Plant and Its Applications in Cosmetics

  • Mt. Jiri located in the southwestern part of Korea is a treasure trove of wild medicinal plants. More than 1200 species currently classified as herbs are grown or cultivated in the area. Recently, safflower has attracted interest because of its ability to control fine wrinkle formation on the neck. The objective of this study therefore was to determine whether the active ingredient of safflower could be used in the form of an extract to reduce wrinkle formation in individuals aged 30 to 59 years. In particular, this study was aimed at determining the extract’s elastase activity and anti-oxidant effect by using DPPH assay in vitro and evaluating the anti-wrinkle efficacy of different types of safflower extracts in improving fine wrinkles on the neck. This study will provide a basis for future studies to develop safflower extracts using advanced composition technology and contribute to the development of the herbal cosmetics industry.

Properties of Chemical Compositions of Safflower Seeds (산지별 홍화씨의 화학적 성분 특성)

  • This study was carried out to investigate the chemical properties of safflower seeds cultivated at some areas in Korea. Safflower was used as one of the functional foods and medicinal plants for many centuries. Porximate compositions of safflower seeds were moisture, 4.8~8.1%, crude protein 16.5~19.5%, crude ash 2.7~3.3%, curde fiber 38.5~43.4%, and crude fat 16.6~24.4%, respectively. Glutamic acid, aspartic acid and arginine were the major amino acids and their contents were 2,587.4~3,143.5 mg%, 1,315.8~1,654.8 mg%, and 1,171.9~1,484.2 mg%. K, P, Ca, and Mg were major minerals and their contents were 611.6~886.3 mg%, 501.5~596.7 mg%, 208.5~641.2 mg%, and 530.6~639.5 mg%, respectively. The free sugars that were identified include raffinose, sucrose, glucose, fructose. ${\alpha}$-tocopherol content was 0.14~3.82 mg%. Contents of vitamin C was 0.43~3.39 mg%. The safflower seeds fatty acids were palmitic acid, stearic acid, oleic acid, linoleic acid and linoleic acid. The major fatty acid was linoleic acid it's content was 74.8~82.9%.

Effects of Safflower Seed Extract on the Osteoblastic Activity and Bone Regeneration (홍화씨 추출물이 조골모유사세포활성 및 골재생에 미치는 영향)

  • The purpose of the present study is to examine the effect of cell proliferation and alkaline phosphatase activity in osteoblastic cells and to compare the bone healing ability of rat calvarial defects between the control group and the safflower seed extract treated group. Osteoblastic cells were obtained from calvariae of a fetal rat. Cells were cultured containing DMEM and safflower seed extract ($10^{-6}g/ml$, $10^{-3}g/ml$) at $37^{\circ}$ with 5% $CO_2$ in 100% humidity for 3 days. MTT was performed to examine the viability of the cells, and alkaline phosphatase activity was analyzed to examine the mineralization in vitro. Rat calvarial defects($5{\times}5mm$) in 250g Sprague-Dawly were made using round bur. Rats were administrated with safflower seed extract(0.35g/kg/day) for experimental periods. Calvarial defects were studied histopathologically and immunohistochemically at 1,4, and 8 weeks. High concentration group($10^{-3}g/ml$) of safflower seed extract significantly increased in the cell proliferation and alkaline phos phatase synthesis in osteoblastic cells. The infiltration of inflammatory cells and osteoclastic activities were decreased in the safflower seed extract treated group as compared with control group. Bone maturation was accelerated in the safflower seed extract treated group as compared to control group. No difference in osteoinductive process was observed between the control and the safflower seed extract treated group. Immunohistochemical observation revealed that protein expression of TGF-$\beta$and osteonectin during early healing phase in the safflower seed extract treated group was slightly increased as compared to control group. These results indicate that safflower seed extract promotes the healing process in bony defect of rat calvariae, and retains a potential applicability as an adjuvant therapeutic modality for regeneration of periodontal bony defect.

Possible Process of Safflower Seed on New Bone formation by 2-Dimensional Electrophoresis (이차원적 전기영동을 이용한 홍화씨의 신생골 형성 기작에 관한 연구)

  • Korean safflower (Carthami Flos) seed has been known to have healing effects on both bone fracture and osteoporosis. On the base of such a notice, this experiment was carried out to explore the effects of safflower seed on bone formation and bone repair. In addition, the healing mechanism was evaluated by analysing serum after feeding the seed to experimental. animals. The effect of Korean safflower seed were evaluated with 40 rats,3-month old. Forty Sprague-Dawley rats composed of 20 male and 20 female were underwent unilateral tibial defect and then fastened with unilateral fixators. The operated rats were divided into two groups depending on the composition of diet, such as positive control group fed normal diet (C-OP group) and safflower seed group fed 30% of safflower seed diet and 70% of normal diet (S-OP group). Postoperative radiographys were taken once in 2 weeks to evaluate callus formation for operated groups. In addition, a possible protein spots involved in bone recovery were examined using 2-Dimensional Gel Electrophoresis (2-DE). The comparison of the radiography between C-OP and S-OP group were showed that the safflower seed diet appeared to stimulate the formation of callus in the rat. On the images of 2-BE, it was able to identify possible five protein spots, having pl from 4 to 5 and molecular weight range from 24 to 26 kDa, involved in bone formation and repair, since no differing protein spots were found the two between groups except the five spots. No differences were observed between two groups before operation, but clear and bigger protein spots were observed from the S-OP group compared with C-OP group on 6 and 9 weeks post operation. These protein spots were, however, showed similar sizes and densities between two groups in 12 weeks later. The transformation of protein spots was suggested that these protein spots were involved in bone formation and recovery, in addition safflower seed might induce the formation of factors and activate these factors. In conclusion, this study suggest that safflower seed influence a variety of factors in the course of bone formation or the periods of remedy.
