Identification of 26 Germplasms of Safflower (Carthamus tinctorius L.) with ISSR and SCAR Markers

  • Safflower (Carthamus tinctorius L.) is a herb primarily distributed throughout in the world. We have used the inter-simple sequence repeats (ISSR) technique to investigate the phylogenetic relationships and genetic diversity of C. tinctorius. Of all germplasms, 88.7% were polymorphic among all germplasms. Mean genetic diversity within germplasms was very low (0.048). The Turkey germplasm had the highest expected diversity (0.082) and Australia germplasm was the lowest (0.020). These values indicate that most of the genetic diversity of safflower is found among germplasms and there is a high among-germplasm differentiation. We found eight phenetic bands for determining the specific marker of germplasm with SCAR markers. The regions of the Mediterranean Sea and India may be the most probable candidates for the origin of safflower. The tree showed four major clades: (1) European germplasms, (2) Azerbaijan, Egypt, and Ethiopia, (3) Australia, and (4) America.

한국산과 중국산 홍화꽃의 화학적 성분조성비교 (Comparison of the Chemical Compositions of Korean and Chinese Safflower Flower(Carthamus tinctorius L.))

  • 홍화꽃이 식품재료적 가치를 높이는 기초적 연구의 일환으로 한국산과 중국산의 화학적성분을 비교 분석한 결과는 다음과 같다. 홍화꽃의 일반성분은 한국산 홍화꽃의 조단백질, 조지 방, 조회분, 조섬유, 무질소물의 함량이 14.70%, 3.10%, 6.90%, 18.20%, 57.10% 이었으며 중국산 홍화꽃은 12.60%, 2.70%, 5.80%, 16.40%, 62.50%로 나타나 무기질소물을 제외하고 한국산 홍화꽃이 중국산 홍화꽃 보다 높게 나타났다. 유리당은 한국산과 중국산 홍화꽃에서 fructose, glucose, sucrose가 대부분이었다. 폴레페놀 함량은 수용성 추출물이 한국산 13.85%, 중국산이 9.39%로 한국산 홍화꽃이 많이 함유되었으며 또한 메탄올 추출물에서도 한국산 홍화꽃이 9.70%, 중국산 홍화꽃이 7.04%로 한국산 홍화꽃이 중국산 홍화꽃보다 비교적 많이 함유되어 있는 것으로 나타났다. 지방산 함량은 한국산 홍화꽃은 포화지방산과 불포화지방산이 6.80%, 93.20% 중국산 홍화꽃은 16.0%, 84.0%로 한국산이 월등히 높은 불포화지방산 함량을 나타냈다. 한국산과 중국산 홍화꽃 모두에서 linoleic acid 와 oleic acid 가 함량이 높게 나타났다. 아미노산중 필수아미노산은 한국산 홍화꽃이 46.67% 중국산 홍화꽃이 36.79%로 한국산 홍화꽃이 높게 나타났으며 모든 필수아미노산 조성이 한국산 홍화꽃에서 높게 나타났다. 그리고 비필수아미노산이 차지하는 비율은 한국산 홍화꽃이 65.17% 중국산 홍화꽃이 54.49%로 한국산 홍화꽃이 차지하는 비율이 필수아미노산과 더불어 높게 나타났다. 무기질은 한국산 홍화꽃이 중국산 홍화꽃에 비해 Ca, Cu, Fe, Mn이 높게 나타났으며 반면에 A1, Ba, Mg, K, Na, Zn, Sr, P은 중국산 홍화꽃에서 더 많은 함량을 나타냈다.

  • For the purposed of improving the utilization of natural chemical pigment, carthamin, of Carthamus tinctorius, the effective extraction methods on this compound were pursued in the present study. The best solvent for the extraction was found to be the 1 %(v/v) NaOH solution, at 25 hours. In addition, more carthamin was extracted flowers from main stem than fromb-ranches . The carthamin content of Korean local safflower was shown to be higher than that of Japanese variety used for medicinal uses.

Anthracnose of Safflower Caused by Colletotrichum acutatum

  • Anthracnose occurred severely on safflower plants grown in Euiseong and Jecheon areas of Korea in 1997 and 1998. The disease incidence was up to 100% in some fields, and symptoms developed on seedlings, leaves, stems, roots and hulls of the plants. Coletotrichum sp. was consistently present on the diseased plant parts, and all the isolates from the lesions were identified as Colletotrichum acutatum based on the morphological and cultural characteristics. Similar symptoms were produced on the host plants by artificial inoculation with isolates of the fungus. The fungus was reisolated from lesions on the plants inoculated. This is the first report that c. acutatum causes anthracnose of safflower.

황색계 천연염색 견직물의 색채 이미지 연구 (A Study of Color Image on Silk Fabrics Dyed with Yellow Natural Materials)

  • Silk fabrics were dyed with yellow natural materials. Gardenia, turmeric, amur cork tree, safflower, Japanese pagoda tree, and onion were used as natural materials. Color image of natural dyed silk fabrics was classified by 4 factors(Cheerfulness, Comfortness, Pastrol, Revealation). Cheerfulness factor affected significantly with color image. Amur cork tree or turmeric dyed fabrics were shown as light and cute, safflower or gardenia dyed fabric shown as comfort, and onion dyed fabrics shown as mature and simple. Color image with specialty was significant difference in Preference and Revelation factor. Preference was appeared as amur cork tree>turmeric>gardenia>safflower>Japanese pagoda tree>onion.

天然染料에 관한 硏究(10) -홍화 황색소의 견섬유에 대한 염색성- (Studies on the Natural Dyes(10) -Dyeing properties of safflower yellow for silk fibers-)

  • In order to study the properties of safflower yellow colors, thermodynamic parameters and dyeing properties on the silk in several dyeing conditions were investigated. The uv-visible spectra of safflower yellow colors in several solvents show hypsochromic shift with the polarity of solvent but bathochromic shift with increasing acidity of solution. The apparent diffusion coefficients and standard affinities of dyeing increased with the increase of dyeing temperature. The standard heat of dyeing(${\Delta}H^0$), entropy change(${\Delta}S^0$) and activation energy($E_{act}$) were calculated to be - 1.144kcal/mol, -7.498(5$0^{\circ}C$)~-3.804(9$0^{\circ}C$)cal/molㆍdeg and 0.123kcal/mol, respectively. The concentration of safflower yellow colors in the silk fiber increased with dyeing temperature, time, concentration of colors and acidity of initial dyebath. Silk fabrics were dyed bright yellow by pre-mordanting with tin chloride. Lightfastness of silk fabrics pre-mordanted by tin chloride was not excellent.

Metabolic Discrimination of Safflower Petals of Various Origins Using 1H NMR Spectroscopy and Multivariate Statistical Analysis

  • The metabolic discrimination of safflowers from various geographical origins was performed using 1H nuclear magnetic resonance (NMR) spectroscopy followed by principal components analysis. With a combination of these techniques, safflower samples from different origins could be discriminated using the first two principal components (PC) of the 1H NMR spectra of the 50% methanol fractions. PC1 and PC2 accounted cumulatively for 91.3% of the variation in all variables. The major peaks in the 1H NMR spectra that contributed to the discrimination were assigned to fatty acid (terminal CH3), lactic acid, acetic acid, choline derivatives, glycine, and safflower yellow derivatives. In this study, we suggest that various types of safflower can be discriminated using PCA and 1H NMR spectra.

Occurrence of Powdery Mildew on Safflower Caused by Sphaerotheca fuliginea in Korea

  • The powdery mildew of safflower (Carthamus tinctorius L.) extensively occurred at 1999 at the experimental farm of Kyongsangnam-do Agricultural Research and Extension Services. Both sides of the leaves and the older stems were covered with the fungus, and then the leaves and stems turned yellow. The conidia, conidiophores and perithecia were observed on the leaf lesion. Perithecia were ellipsoidal, $80-117\;{\mu}m$ in diameter. Asci were subglobose and $84{\sim}99{\times}59{\sim}73\;{\mu}m$ in size. Ascospore were ellipsoidal to ovoid, and $15{\sim}34{\times}11{\sim}23\;{\mu}m$ in size. Conidia were ellipsoid to barrel-shaped, $25{\sim}37{\times}11{\sim}22\;{\mu}m$ in size and formed in long chains. The causal organism was identified as Sphaerotheca fuliginea. This is the first report on powdery mildew of safflower caused by Sphaerotheca fuliginea in Korea.

Antioxidative Activity of Phenolic Compounds in Roasted Safflower (Carthamus tinctorius L.) Seeds

  • Antioxidative compounds contained in roasted safflower seeds were investigated. Six phenolic compounds, N-feruloylserotonin, N-(p-coumaroyl) serotonin, matairesinol, 8'-hydroxyarctigenin, acacetin 7-Ο-β-D-glucoside(tilianine) and acacetin were isolated and identified from the extract of seeds. The inhibitory effects of six phenolic compounds on 1,1-dipheny1-2-picrylhydrazyl(DPPH) radical and lipid peroxidation induced by H₂O₂/FeSO₄in rat liver microsomes were determined. Two serotonins showed more potent DPPH radical scavenging activity, and a stronger inhibitory effect on the lipid peroxidation than that of α-tocopherol. In addition, acacetin and matairesinol also considerably inhibited lipid peroxidation, while 2-hydroxy-arctigenin and tilianine were inactive. These results suggest that phenolic compounds, including serotonins, lignans and flavonoids in the roasted safflower seeds can be used as potential dietary natural antioxidants.
