  • 활성있는 새로운 농약을 창출하기 위해, saccharin 모체에 imidazolidine-2,4,5-trione과 2-thio-imidazolidine-4,5-dione 기를 도입시켰다. 각 saccharin 유도체들은 1-치환된 urea (혹은 1-치환된 thiourea)와 oxalyl chloride의 반응을 시작으로 4단계를 거쳐 합성하였다. 1-치환된 urea를 사용해서 1-치환된-3-(1,1,3-trioxo-1,3-dihydro-1$^{6}$-benzo[d]isothiazol-2- ylmethyl)-imidazolidine-2,4,5-trione 5a, 5b, 5c를 합성하였고, 1-치환된 thiourea를 사용하여 1-치환된-2-thioxo-3-(1,1,3-trioxo-1,3-dihydro-1$^{6}$-benzo[d]isothiazol-2-ylmethyl)-imidazolidine-4,5-dione 12a,12b, 12c를 합성하였다.

Effects of Saccharin Intake on Hippocampal and Cortical Plasticity in Juvenile and Adolescent Rats

  • The sensory system is developed and optimized by experiences given in the early phase of life in association with other regions of the nervous system. To date, many studies have revealed that deprivation of specific sensory experiences can modify the structure and function of the central nervous system; however, the effects of sensory overload remains unclear. Here we studied the effect of overloading the taste sense in the early period of life on the synaptic plasticity of rat hippocampus and somatosensory cortex. We prepared male and female Sprague Dawley rats with ad libitum access to a 0.1% saccharin solution for 2 hrs per day for three weeks after weaning on postnatal day 22. Saccharin consumption was slightly increased in males compared with females; however, saccharin intake did not affect chow intake or weight gain either in male or in female rats. We examined the effect of saccharin-intake on long term potentiation (LTP) formation in hippocampal Schaffer collateral pathway and somatosensory cortex layer IV - II/III pathways in the 6-week old saccharin-fed rats. There was no significant difference in LTP formation in the hippocampus between the control group and saccharin-treated group in both male and female rats. Also in the somatosensory cortex, we did not see a significant difference in LTP among the groups. Therefore, we conclude that saccharin-intake during 3~6 weeks may not affect the development of physiological function of the cortical and hippocampal synapses in rats.

  • Introduction: This study examined the regulatory mechanism underlying the meal-induced changes in the hypothalamic-pituitary-adrenal gland (HPA) axis activity. Materials and Methods: Male Sprague-Dawley rats (250-300 g) were hired for two different experiments as follows; 1) rats received either 8% sucrose or 0.2% saccharin ad libitum after 48 h of food deprivation with the gastric fistula closed (real feeding) or opened (sham feeding). 2). rats received 5 ml of intra-oral infusion with 0.2% saccharin or distilled water after 48 h of food deprivation. One hour after food access, all rats were sacrificed by a transcardiac perfusion with 4% paraformaldehyde. The brains were processed for c-Fos immunohistochemistry and the cardiac blood was collected for the plasma corticosterone assay. Results: Real feedings with sucrose or saccharin and sham feeding saccharin but not sucrose, following food deprivation decreased the plasma corticosterone level. c-Fos expression in the nucleus tractus of solitarius (NTS) of the fasted rats was increased by the consumption of sucrose but not saccharin, regardless of the feeding method. On the other hand, the consumption of sucrose or saccharin with real feeding but not the sham, induced c-Fos expression in the paraventricular nucleus (PVN) of the fasted rats. The intra-oral infusion with saccharin or water decreased the plasma corticosterone level of the fasted rats. Intra-oral water infusion increased c-Fos expression in both the PVN and NTS, but saccharin only in the NTS in the fasted rats. Conclusion: Neither restoration of the fasting-induced elevation of plasma corticosterone nor the activation of neurons in the PVN and NTS after refeeding requires the palatability of food or the post-ingestive satiety and caloric load. In addition, neuronal activation in the hypothalamic PVN may not be an implication in the restoration of the fasting-induced elevation of the plasma corticosterone by oropharyngeal stimuli of palatable food.

PVM 및 FBRM 기반 인라인 모니터링을 통한 indomethacin-saccharin 공결정의 생성 메커니즘이해 (Understanding the Mechanism of Indomethacin-Saccharin Co-crystal Formation Using In-line Monitoring System based on PVM and FBRM)

  • 난용성 약물의 용해도를 증가시키고 안정성을 향상하기 위한 제약 공결정은, 미국 FDA가 지난 2016년 8월에 공결정을 solvate의 특별한 경우라고 정의를 수정하면서, 개량신약의 개발에 더욱 박차를 가할 것으로 추측된다. 본 연구에서는 모델 조합으로 잘 알려진 indomethacin-saccharin 공결정을 반용매 방법으로 제조할 때, 인라인 모니터링 기법을 적용하여, 반용매의 주입속도에 따른 indomethacin 준 안정상의 일시적인 생성 및 indomethacin-saccharin 공결정의 생성을 관찰하고 메커니즘을 제안하고자 하였다. 그간 인라인 모니터링을 위해서 매우 다양한 분석도구가 연구되어 왔는데, 본 연구에서는 PVM (particle vision measurement)와 FBRM (focused beam reflectance measurement)를 조합하여 공결정화 공정에서의 변화를 실시간으로 측정하였다. 공결정화 결과물의 오프라인 분석은 PXRD (powder x-ray diffraction)와 DSC (Differential scanning calorimeter)를 이용하여 수행하였다. 반용매의 주입 속도에 따라서 공결정이 생성되는 경로에 분명한 차이가 있음을 관찰하였으며, 이러한 상관 관계의 이해를 통해서 제약학적 특성이 더욱 일정하고 품질이 보증된 indomethacin-saccharin Co-crystal을 얻는 조건을 확립하였다. 본 연구결과, PVM과 FBRM을 조합한 인라인 모니터링은 제약 공결정 제조공정에서 활용성이 매우 높은 기술이라고 할 수 있다.

Studies of Silyl-Transfer Photochemical Reactions of N-[(Trimethylsilyl)alkyl]saccharins

  • Photochemical studies of N-[(trimethylsilyl)alkyl]saccharins were carried out to investigate their photochemical behavior. Depending on the nature of the substrate and the solvent system employed, reactions of these substances can take place by either SET-promoted silyl migration from carbon to either the amide carbonyl or sulfonyl oxygen or by a N-S homolysis route. The results of the current studies show that an azomethine ylide, arising from a SET-promoted silyl migration pathway, is generated in photoreactions of N-[(trimethylsilyl)methyl]saccharin and this intermediate reacts to give various photoproducts depending on the conditions employed. In addition, irradiation of N-[(trimethylsily)ethyl]saccharin produces an excited state that reacts through two pathways, the relative importance is governed by solvent polarity and protic nature. Finally, photoirradiation of N-[(trimethylsilyl)propyl]saccharin in a highly polar solvent system comprised of 35% aqueous MeOH gives rise to formation of a tricyclic pyrrolizidine and saccharin that generated via competitive SET-promoted silyl transfer and $\gamma$-hydrogen abstraction pathways.

식품 중 함성첨가물 사용실테 조사연구 - 사카린나트륨, 안식향산나트륨, 소르빈산칼륨 중심으로 - (A Study on the Sodium Saccharin, Sodium Benzoate and Potassium Sorbate Used in Foods)

  • 본 연구에서는 식품 중에 사카린나트륨, 안식향산나트륨, 소르빈산칼륨의 사용실태를 알아보고자 시중에서 유통되고 있는 단무지, 곡물튀김과자, 혼합음료 등 총7종 230품목을 대상으로 HPLC에 의해 분석하여 얻은 결과 사카린나트륨은 탄산음료, 과실·채소류 음료, 조미건포류에서 검출되지 않았고 단무지, 장아찌, 곡물튀김과자, 혼합음료에서 N.D.-1234.8mg/kg 범위로 검출되어 곡물튀김과장의 경우 허용하지 않는 사카린나트륨을 사용하고 있는 것으로 나타났다. 안식향산나트륨은 단무지, 장아찌, 곡물튀김과자, 과실·채소류음료, 조미건포류에서는 불검출, 혼합음료, 탄산음료에서는 N.D.-663.2 mg/kg이 검출되어있으며 소르빈산 칼륨은 곡물튀김과자, 혼합음료, 탄산음료, 과실 ·채소류 음료등에서 검출되지 않았고 단무지, 장아찌 , 조미건포류에서 N.D.-2725.1 mg/kg이 검출되어 국내허용기준을 초과한 제품이 장아찌 1 품목에서 나타났다. 조사 대상 식품 중 사카린 나트륨이 검출된 곡물 튀김 과자 42품목과 소르빈산칼륨 허용량을 초과한 장아찌 1품목을 제외한 6종 187품목은 사카린나트륨, 안식향산나트륨, 소르빈산칼륨의 사용이 국내 사용기준이내였으며 단무지, 장아찌 등 절임 식품은 대부분이 감미료와 보존료를 병행해서 사용하는 것으로 나타났다.

Ni Sulfamate-chloride 전기도금 용액에서 전류밀도와 첨가제의 농도 변화가 Ni 박막에 미치는 영향 (Effects of the Changes of Current Density and Additive Concentration on Ni Thin Films in Ni Sulfamate-chloride Electrodeposition Baths)

  • Sulfamate plating solution containing a small amount of chloride bath was fabricated to study the properties of the electrodeposited Ni thin films. Effects of the changes of current density and additive concentration on current efficiency, residual stress, surface morphology and microstructure of Ni thin films electrodeposited from Ni sulfamate-chloride baths were investigated. The current efficiency was measured to be more than about 95%, independent of the changes of current density and saccharin concentration in the baths. Residual stress of Ni thin film was appeared to be the compressive stress modes in the range of $5{\sim}30mA/cm^2$ current density. Maximum compressive stress was observed at the current density of $10mA/cm^2$. Compressive stress values of Ni thin/thick films were increased to be about -85~-100 MPa with increasing saccharin concentration from 0 to 0.0195 M(4 g/L). Surface morphology was changed from smooth to nodule surface appearance with increasing the current density. Smooth surface morphology of Ni thin films electrodeposited from the baths containing saccharin was observed, independent of the saccharin concentration. Ni thin/thick films consist of FCC(111), FCC(200), FCC(220), FCC(311) and FCC(222) peaks. It was revealed that the FCC(200) peak of Ni thin films is the preferred orientation in the range of $5{\sim}30mA/cm^2$ current density. The intensity of FCC(200) peak was gradually decreased and the intensity of FCC(111) peak was increased with increasing saccharin concentration in the baths.

전해액 내 사카린의 농도 변화에 의한 전기도금 니켈-철 퍼멀로이 박막의 미세구조와 자기적 특성 변화 (Microstructure and Magnetic Properties of Electroplated Ni-Fe Permalloy Thin Films by Saccharin Concentration in Electrolytes)

  • 전기도금 니켈-철 퍼멀로이 박막에 대한 사카린의 영향에 대해 조사하였다. 1 ${\mu}mol/L$ 이하 농도의 사카린이 도금용 전해액에 첨가되면, 도금 박막의 결정립 크기를 감소시키고, 표면 거칠기를 낮추는 효과가 있다. 이러한 물성의 변화는 자기적 특성 중 보자력의 감소와 투자율과 자기임피던스의 증가로 나타난다. 결정립 크기의 감소는 증분투자율과 자기임피던스의 증가와 매우 밀접한 상관관계를 나타내고 있다. 사카린은 전기도금 연자성 퍼멀로이 박막에 유용한 첨가제이며 사카린의 농도변화로 연자성의 조절 가능성을 보였다.
