Undifferentiated suspension cells had the ability to transfer glucose and fructose actively, but the suspension culture cells were unable to transfer saccharide without previously splitting to monosccarides. The uptake of fructose showed the low- and high-affinity system compared to of glucose, which possessed only one saturable uptake system. In this paper, the low affinity system of the uptake of fructose has been studied intensively. Glucose did not inhibit the low affinity system of fructose competitively. The Km value was 47 mM for fructose, 7.4 mM for glucose and Vmax was 69 $\mu$mol/h.g fresh weight for fuctose, 9.8 $\mu$ mol/h.g fresh weight for glucose. Metabolizer inhibitors, both 50 $\mu$M of CCCP and DNP, inhibited 70% of the uptake of the low affinity system of fructose. The proton ions were accompanied by the uptake of fructose. The stoichiometry showed ratio of proton to fructose was 0.17. The mechanism ofthe uptake was fructose-proton-symport. The molecules of fructose accmululated inside 25 times more than outside. Therefore, the low affinity system of fructose was not mere diffusion, but depended on metabolic energy and thus transported actively. The importance of this system was discussed.
Fourteen compounds were isolated from the n-BuOH fraction of the roots of Aralia cordata (syn. = A. continentalis). Through spectroscopic method, the chemical structures were elucidated as: caffeic acid (1), protocatechuic acid (2), thymidine (3), uridine (4), methyl-$\alpha$-D-fructofuranoside (5), a mixture (3 : 1) of $\beta$-D-fructopyranoside and $\beta$-D-fructofuranoside (6), 1-methyl 1,2,3,4-tetrahydro-$\beta$-carboline-3-carboxylic acid (7), methyl-$\beta$-D-fructofuranoside (8), sucrose (9), 5-caffeoylquinic acid (chlorogenic acid) (10), 3-caffeoylquinic acid (neochlorogenic acid) (11), 4-caffeoylquinic acid (cryptochlorogenic acid) (12), 3,5-di-O-caffeoylquinic acid (13), and 1-kestose [$\beta$-D-fructofuranosyl-($2{\rightarrow}1$)-$\beta$-D-fructofuranosyl-($2{\rightarrow}1$)-$\alpha$-D-glucopyranoside] (14). Among them, compounds 5, 7, 8, and 10 - 14 were isolated from this plant for the first time.
Hyaluronic acid (HA) has long been studied in diverse applications. It is a naturally occurring linear polysaccharide in a family of unbranched glycosaminoglycans, which consists of repeating di-saccharide units of N-acetyl-D-glucosamine and D-glucuronic acid. It is almost ubiquitous in humans and other vertebrates, where it participates in many key processes, including cell signaling, tissue regeneration, wound healing, morphogenesis, matrix organization, and pathobiology. HA is biocompatible, biodegradable, muco-adhesive, hygroscopic, and viscoelastic. These unique physico-chemical properties have been exploited for several medicinal purposes, including recent uses in the adjuvant treatment for chronic inflammatory disease and to reduce pain and accelerate healing after third molar intervention. This review focuses on the post-operative effect of HA after third molar intervention along with its various physio-chemical, biochemical, and pharmaco-therapeutic uses.
This study was conducted to investigate the effects of fermented capsicum and a capsicum product on lipid metabolism. Fermented capsicum was prepared from red pepper puree tov three months. After 9() days of fermentation, capsaicin and dihydrocapsaicin concentrations were reduced from 24.7 and 14.7 g/mL to 15.5 and 6.45 g/mL, respectively. The capsicum product was prepared from the fermented capsicum mixed with prune extract, green tea extract, neroli extract and oligo-saccharide. Thirty-two male Sprague-Dawley rats were as- signed to four dietary groups (control, high-fat control (BE-control), high-fat-fermented capsicum (HF-S-1), high- fat-capsicum product (HF-S-2)). Plasma and hepatic lipid profiles were examined after three weeks of experimental diet. Food intakes were significantly lower in the HF-S-1 and HF-S-2 groups compared to the control group (p<0.05). The weight of perirenal fat pads was lowest in animals on the control diet (low-fat) and highest in high-fat control diet. The addition of fermented capsicum to high fat diets, HF-S-1 and HE-S-2 groups, resulted in significantly lower fat pad weights compared with the HF-control group. Both fermented capsicum (HF-S-1) and the capsicum product (HF-S-2) groups had lower plasma TG levels, atherogenic-index, and liver TG levels than the BE-control group (p <0.05). Liver TC levels were significantly lower in the HF-S-2 group than the HF-control group. The results demonstrate a hypolipidemic effect of fermented capsicum and the fermented cap-sicum product.

A new method to utilize spent mushroom substrates (SMS) for ethanol production was investigated. Analysis of the chemical properties of SMS revealed that they were decomposed by the mushrooms during cultivation. In particular, the free sugar content in SMS was reduced to half of that in mushrooms. Of the tested SMS, the Pleurotus eryngii SMS was determined to be suitable for saccharification. Upon pretreatment with a 1% alkaline solution, Pleurotus eryngii SMS achieved 80.7% of its maximum saccharification ratio. The optimum pretreatment conditions for enzyme saccharification were 1% NaOH solution at $120^{\circ}$ for 60 min. Further studies are required to determine ethanol production using Pleurotus eryngii SMS.

The proteinpolysaccharides (PPS) from Polyporus umbellatus (P. umbellatus) screlotium is composed by 78.2% of saccharide, 16.8% of protein, and 4.0% of ash. PPS from P. umbellatus showed antitumor activities against 180 solid tumor in ICR mice at the concentration of 20-160 mg/kg/day. PPS from P. umbellatus inhibited cell viability to 47.4% and 45.0% in leukemia cell lines, L-1210 and K-562 cells at 50-400 $\mu\textrm{g}$/mL concentration, respectively. But the hall mark of cell apoptosis, DNA fragmentation was not observed at those concentration. 2.5-10.0% of PPS from P. umbellatus inhibited mutagenecity evoked by 2-nitrofluorene and sodium azide in Salmonella typhimurium TA 98 and TA 100. From these results, it is suggested that the PPS of P. umbellatus has antitumor and antimutagenic effect, and its cytotoxic effect may not be ascribed to the apoptosis.
Galactooligosaccharlde (GOS) is a kind of functional oligosaccharides that can be used as a food ingredient and a cosmetic additive. In this paper, characteristics of GOS synthesis by cellulase, using lactose as a substrate, were investigated. Penicillium funiculosum cellulose was found to be the most efficient for GOS production among six cellulose tested. The optimum pH and temperature for GOS production were 5.0 and $50^{\circ}C$, respectively. There was an optimum ratio of lactose concentration to enzyme loading; the value was 10 (w/w). The reaction pattern of P. funiculosum cellulase is consistent with that of microbial ${\beta}$-galactosidase which shows transgalactosylation activity. Amounts of GOS produced from 20% (w/v) lactose after 6 h incubation at $50^{\circ}C$, were 23% (w/w) based on total saccharide in the reaction medium. The GOS % increased with initial lactose concentration in the range of 5 to 20%. The products mainly consisted of a trisaccharide and tetrasaccharide from HPLC and TLC analysis. Among enzymes involved in transgalactosylation reaction, high molecular weight fractions over 50,000 Da, presumably ${\beta}$-glucosldase, were considered to be responsible for GOS production. Using this cellulose, a direct synthesis of galactosyl g1ucoside including GOS could be readily achieved with lactose as a galactosyl donor.
Fully or nearly fully 6- [4- (4'- (nitrophenylazo)phenoxycarbonyl)]pentanoated polysaccharide derivatives were synthesized by reacting cellulose, amylose, chitosan, chitin, alginic acid, pullulan or amylopectin with 6- [4- (4'- (nitrophenylazo)phenoxy) ] pentanoyl chloride (NA6C) and their thermotropic liquid crystalline behaviors were investigated. Like in the case of NA6C, all the polysaccharide derivatives formed monotropic nematic phases, suggesting that the mesophase structure of the polysaccharide derivatives is dertermined by the mesogenic side groups and not by the polysaccharide backbone. This is the first report of polysaccharide derivatives, except cellulose derivative, that form thermotropic nematic phases. The thermal stability and degree of order of the nematic phases observed for poly saccharide derivatives were significantly different from those reported for the polymers in which the azobenzene groups are attached to flexible or rigid backbones through flexible spacers. The results were discussed in terms of the difference in the arrangement of the main and side chains and the flexibility of the main chain.

The purpose of this study was to investigate the effects of Cuscuta chinensis Lamark ethanol extract (CL) on wrinkle improvement. Cuscuta chinensis Lamark is known to contain dried saccharide, alkaloids, flavonoids, lignans and rein glycoside as major components of dried mature seeds of Cuscuta japonica Choisy. In this study, we evaluated the anti-wrinkle effects of CL and investigated bio-markers (e.g ; MMP-1, MMP-3, MMP-9, TIMP-1, type I procollagen) associated with skin wrinkle improvement. We tested the anti-wrinkle effect of CL using human fibroblast called CCD-986Sk cell. We observed an increase in MMPs, TIMP-1, and type 1 pro-collagen CL in CCD-986Sk cells irradiated with UVB at an intensity of $2mJ/cm^2$ for 60 seconds. As a result, CL decreased UVB-induced MMPs levels and mRNA expressions in CCD-986Sk cell. The levels and mRNA expressions of type I procollagen and TIMP-1 were increased by CL. These results suggest that CL has activities on improvement of skin wrinkle, which is induced by UVB radiation. Taken together, this study proposed the possibility of developing herbal medicine and functional herbal cosmetic materials with wrinkle-improving effects of Cuscuta chinensis Lamark.
The purpose of this study was to evaluate the histological toxicity of chitosanoligosaccharide on the rat. A healthy male of Wistar rat that weighted $250{\pm}350g$ was used for experiment The experimental group was divided into five groups. Group 1 was control group which treated with general food Group 2 was $F_1$ generation which was born by mating of 0.1% (1 mg/ml) chitosanoligosaccharide was supplied by feeding ad libitum for 30 days. Group 3 was $F_2$ generation which was born by mating $F_1$ generation. Group 4 was treated with 90 days of 0.1% (1 mg/ml) chitosan oligosaccharide. Group 5 was treated with 365 days of 0.1% (1 mg/ml) chitosanoligo saccharide. All experimental groups were used to 10 rat. The results were as follow: The RER dilation was observed Group 4. However, there were no significantly changes of ultrastructures in the other groups compared to the control. It was concluded that chitosanoligosaccharide can be used for nontoxic natural material.
