Renal Manifestations and Imaging Studies of Kawasaki Disease (가와사키 질환에서의 신증상과 영상 검사 소견)

  • Purpose: The aim of this study was to verify renal inflammation following Kawasaki disease (KD) using single photon emission computed tomography along with Technetium-99m dimercaptosuccinic acid scintigraphy (DMSA renal SPECT). Methods: From March 2011 to October 2011, 15 patients diagnosed with KD at the National Health Insurance System Ilsan Hospital were enrolled in the study. All patients underwent DMSA renal SPECT to evaluate renal involvement during the acute phase of KD. Urine ${\beta}2$-microglobulin (${\beta}2$-MG), a marker of renal proximal tubular dysfunction, was also measured to assess renal damage. Results: All 15 patients had normal renal function test results. However, microscopic hematuria and pyuria were observed in 13% and 33% of the patients, respectively. Moreover, urine ${\beta}2$-MG was elevated in 46% of the patients. In addition, patients were divided into two groups based on ${\beta}2$-MG level: those with an increased ${\beta}2$-MG level, and those with a normal ${\beta}2$-MG level. No significant differences were found between these two groups in clinical characteristics, laboratory, sonography, and echocardiography findings. All patients' DMSA renal SPECT scans were normal. Conclusion: Our study showed that mild abnormalities in the urinalysis and elevated urine ${\beta}2$-MG were the only findings of renal involvement in KD. However, no aggressive renal manifestations were detected on DMSA renal SPECT.

  • Bombyx mori nuclear polyhedrosis virus (BmNPV) was successfully multiplied in the nuclear of BmN4 cells cultured with insect Grace's medium. By electron microscopic observation, the virons had a single nucleocapsid in an envelope. Polyhedral protein synthesis of BmNPV in BmN4 cells was detected at 18 hr p.i. and polyhedral protein was a singlepolypeptide with a M.W of 30 kd. At 48 hr p.i. polyhedra formation was observed by inverted mociroscope and electron microscope. Genome analysis of BmNPV by restriction endonucleases was not revealed the difference between virus produced in vivo and that in vitro.

Purification and Characterization of Phytoferritin

  • Ferritins from germinated pumpkin seeds were isolated by ammonium sulfate precipitation (0.55 saturation), ion-exchange chromatography on DEAE-cellulose, and gel filtration chromatographies on Sephacryl S-300 and Sephadex G-100. Pumpkin ferritin contains less iron than soybean ferritin. Pumpkin ferritin cross-reacted with anti-soybean ferritin antiserum made in rabbit, and showed two distinct antibody reactive bands, both of equal intensity. The pumpkin ferritins corresponding to the two bands were separable by centrifugation in a sucrose gradient (20~50%). The molecular weights of the native pumpkin ferritins based on the estimation of sucrose gradient centrifugation, gel filtration on Sephacryl S-300 and non-denaturing polyacrylamide gel electrophoresis appeared to be: 530~580 KD (the large molecular weight pumpkin ferritin) and 330-360 KD (the small molecular weight pumpkin ferritin) The large molecular weight pumpkin ferritin contains less iron. Both pumpkin ferritins cross-reacted with anti-soybean ferritin antibody with a spur formation suggesting partial antigenic recognition.

Association between Kawasaki disease and acute respiratory viral infections (가와사끼병과 급성 호흡기 바이러스 감염증의 연관성에 관한 연구)

  • Purpose:The etiology of Kawasaki disease (KD) is still unknown. Recently, an association between human coronavirus NL63 (HCoV-NL63) and KD was implicated. Hence, we attempted to determine the association between KD and acute respiratory viral infections. Methods:Nasopharyngeal aspirate samples were obtained from 54 patients diagnosed with KD at the Seoul National University (SNU) Children's Hospital and SNU-Bundang Hospital between October 2003 and September 2006. Viral diagnoses of 11 respiratory viruses were made using multiplex reverse transcriptase-polymerase chain reaction (RT-PCR): respiratory syncytial virus (RSV), adenovirus, rhinovirus (RV), parainfluenza viruses (PIVs) 1 and 3, influenza viruses (IFVs) A and B, human metapneumovirus (HMPV), human bocavirus (HBoV), HCoV OC43/229E, and HCoV-NL63. Clinical data were reviewed retrospectively. Results:The median age was 32 months (6 months-10.4 years). Respiratory symptoms were observed in 37 patients (69%). The following respiratory viruses were identified in 12 patients (22%): RV (n=4), PIV-3 (n=2), HBoV (n=2), and adenovirus, RSV, PIV-1, IFV-A, and HCoV-NL63 (n=1). Co-infection with PIV-3 and RV was observed in one patient. Respiratory symptoms were observed in 7 (58.3%) and 30 (71.4%) patients of the virus-positive and virus-negative groups (P>0.05). Response rate to intravenous immunoglobulin administration was 67% (n=8) and 86% (n=36) in the virus- positive and virus-negative groups (P>0.05). Conclusion:Respiratory symptoms were commonly observed in KD patients but the association between respiratory viruses and KD were not found. Large multicenter-based investigations are required to confirm the association between acute respiratory viral infections and KD.

The Binding Study of $^{125}I-LSD$ to Platelet Membrane in Alcoholism (주정중독환자의 혈소판막과 $^{125}I-LSD$ 결합에 관한 연구)

  • This research was performed to find out the changes of serotonin receptors in the platelet of alcoholics. Using $_{125}I-LSD$ as a radioligand. We obtained the following results. 1) In the comparison of patients vs controls, $B_{max}$ of the former was significantly higher than that of the latter(P=0.0017). But there was no significant difference in the Kd between the two groups. 2) In the comparison of type 1 vs type 2, alcoholics by cloninger's classification $B_{max}$ of the latter was significantly higher than that of the farmer(P=0.0396). But there was no significant difference in the Kd between the two groups. 3) In the comparison of alcohol abuse vs alcohol dependence, there were no significant differences in the value of $B_{max}$ and Kd between the two groups. These results support the hypothesis of serotonin-deficit in alcoholism. Furthermore, it can be inferred that the increase of platelet serotonin receptors in alcoholics might be used as a trait marker.

Studies on the Effect of Polyamine on Chlorophyll Contents and Chloroplast Peroxidase Activities in Rice Leaf Segments (벼잎 절편에서 Polyamine이 엽록소 함량 및 Chloroplast Peroxidase활성에 미치는 영향에 관한 연구)

  • The effect of polyamine on chlorophyll amount, chloroplast peroxidase and chloroplast thylakoid protein in rice leaf segments which were grown for 10 days(16 hrs, light : 8 hrs, dark) in a hormone-free MS medium containing polyamine was studied. Polyamine treatment increased the chlorophyll contents compared with the control in rice leaf segments. Especially spermine was most effective. Also, in rice leaf segments treated with polyamine chloroplast peroxidase activity was higher than in the control. The treatment with lmM of spermidine increased the enzyme activity by 100%. In polyamine treatment and control two major polypeptide bands corresponding to 56 and 25Kd molecular weight were clearly resolved with other minor bands by SDS-PAGE in the insoluble protein fraction. However, in these bands (56, 25Kd), the total area of protein in treating with polyamine were higher than that of the control. These results suggest that polyamine was an important factor in the chloroplast development of rice seedlings.

Antimicrobial drug susceptibility and plasmid profiles of Salmonella species isolated from poultry (가금에서 분리한 Salmonella속 균의 항균물질에 대한 감수성 및 plasmid profile)

  • In this study, we aim to find the presence of virulence-related plasmid in Salmonella isolates from poultry, and the difference between S pullorum and S gallinarum on the plasmid profile and antibiotics resistance. We used seventeen isolates of Salmonella spp that were isolated from poultry. Thirteen isolates, S typhimurium(ST), S pullorum(SP) and S gallinarum(SG), contained virulence-related plasmids. These are 95Kd plasmid in ST and 85Kd plasmid in SP and SG. Three(1/4 of ST, 1/1 of SE, and 1/9 of SP) isolates have no detectable plasmids. The isolates of ST have relatively variable plasmid profile but the isolates of S pullorum except No 12(additional 3.0Kb plasmid) have common 85K6, 8.1Kb, 4.0Kb and 2.3Kb plasmid and two of three isolates of S gallinarum have common 85Kb, 4.0Kb and 2.3Kb plasmid but the rest has only 85Kb plasmid. Interestingly, all of the isolates of SP have 8.1Kb plasmid, and same size of plasmid is also found in one of ST isolates. All of the isolates have the resistance to penicillin, chloramphenicol, rifampicin, streptomycin, sulfamethazine and some isolates show the resistance to ampicillin and tetracycline. There is no relatedness between plasmid profile and antibiotics resistance and no differences between SP and SG in antibiotics resistance. Therefore further differentiation of each isolates by restriction enzyme assay and, if possible, charaterization of each plasmid, especially, 8.1Kb plasmid in SP and ST, may be necessary.

  • In order to investigate the changes and characteristics of biochemical metabolic substances of soybean tissue culture during the cultural period, immature cotyledons were detached form the plant on 15th days after flowering and cultured in vitro for 3 weeks. The cultures were classified into embryogenic(EC) and non-embryogenic callus(NEC). A part of the EC lines were subcultured for another 3 weeks and classified into root forming(RFC), and shoot forming cultures(SFC). Another part of the EC lines were used for isolation of protoplasts, which were subsequently cultured in vitro for 4 weeks. The cultures were classified into embryogenic(PEC) and non-embryogenic callus(PNEC) derived from the protoplasts. The cultures of EC and PEC lines showed higher phenylalanine content and lower methionine content than those of NEC and PNEC. At organ differentiation stage, both cultures showed the content of aspartic acid decreased, while the other amino acids increased as a whole. The protein pattern analysis of the cultures revealed that EC and NEC lines contained distinctive polypeptides, with mass of ca. 18KD for EC and ca. 22KD for NEC respectively. The EC and PEC lines also showed high activity of peroxidase isozyme A(piA), while the RFC and SFC lines showed that of peroxidase isozyme B(piB).

Extraction of Water Depth in Coastal Area Using EO-1 Hyperion Imagery (EO-1 Hyperion 영상을 이용한 연안해역의 수심 추출)

  • With rapid development of science and technology and recent widening of mankind's range of activities, development of coastal waters and the environment have emerged as global issues. In relation to this, to allow more extensive analyses, the use of satellite images has been on the increase. This study aims at utilizing hyperspectral satellite images in determining the depth of coastal waters more efficiently. For this purpose, a partial image of the research subject was first extracted from an EO-1 Hyperion satellite image, and atmospheric and geometric corrections were made. Minimum noise fraction (MNF) transformation was then performed to compress the bands, and the band most suitable for analyzing the characteristics of the water body was selected. Within the chosen band, the diffuse attenuation coefficient Kd was determined. By deciding the end-member of pixels with pure spectral properties and conducting mapping based on the linear spectral unmixing method, the depth of water at the coastal area in question was ultimately determined. The research findings showed the calculated depth of water differed by an average of 1.2 m from that given on the digital sea map; the errors grew larger when the water to be measured was deeper. If accuracy in atmospheric correction, end-member determination, and Kd calculation is enhanced in the future, it will likely be possible to determine water depths more economically and efficiently.
