Role of Osmotic and Salt Stress in the Expression of Erythrose Reductase in Candida magnoliae

  • The osmotolerant yeast, Candida magnoliae, which was isolated from honeycomb, produces erythritol from sugars such as fructose, glucose, and sucrose. Erythrose reductase in C. magnoliae (CmER) reduces erythrose to erythritol with concomitant oxidation of NAD(P)H. Sequence analysis of the 5'-flanking region of the CmER gene indicated that one putative stress response element (STRE, 5'-AGGGG-3'), found in Saccharomyces cerevisiae, exists 72 nucleotides upstream of the translation initiation codon. An enzyme activity assay and semiquantitative reverse transcription polymerase chain reaction revealed that the expression of CmER is upregulated under osmotic and salt stress conditions caused by a high concentration of sugar, KCl, and NaCl. However, CmER was not affected by osmotic and oxidative stress induced by sorbitol and $H_2O_2$, respectively. The basal transcript level of CmER in the presence of sucrose was higher than that in cells treated with fructose and glucose, indicating that the response of CmER to sugar stress is different from that of GRE3 in S. cerevisiae, which expresses aldose reductase in a sugarindependent manner. It was concluded that regulation of CmER differs from that of other aldose reductases in S. cerevisiae.

  • Parkinson's disease (PD) is a neurodegenerative disease caused by selective degeneration of dopaminergic neurons in the substantia nigra. Mutations in ${\alpha}$-synuclein have been causally linked to the pathogenesis of hereditary PD. In addition, it is a major component of Lewy body found in the brains of sporadic cases as well. In the present study, we examined whether overexpression of wild type or PD-related mutant ${\alpha}$-synuclein induces unfolded protein response (UPR) and triggers the known signaling pathway of the resulting endoplasmic reticulum (ER) stress in SH-SY5Y cells. Overexpression of wild type, A30P, and A53T ${\alpha}$-synuclein all induced XBP-1 mRNA splicing, one of the late stage UPR events. However, activation of ER membrane kinases and upregulation of ER or cytoplsmic chaperones were not detected when ${\alpha}$-synuclein was overexpressed. However, basal level of cytoplsmic calcium was elevated in ${\alpha}$-synuclein-expressing cells. Our observation suggests that overexpression of ${\alpha}$-synuclein induces UPR independent of the known ER membrane kinase-mediated signaling pathway and induces ER stress by disturbing calcium homeostasis.

Temperature Dependence of Optical Energy Gaps of $CdGaInS_4:Er^{3+}$ Single Crystals for Optoelectronic device (광전 소자용 $CdGaInS_4:Er^{3+}$ 단결정의 광학적 에너지 갭의 온도의존성)

  • $CdGaInS_4$ and $CdGaInS_4:Er^{3+}$ single crystals crystallized in the rhombohedral(hexagonal) structure. with lattice constants $a=3.913{\AA},\;c=37.245{\AA}$ for $CdGaInS_4$, and $a=3.899{\AA}$ and $c=36.970{\AA}$ for $CdGaInS_4:Er^{3+}$. The optical absorption measured near the fundamental band edge showed that the optical energy band structure of these compounds had a direct and indirect band gap. the direct and indirect energy gaps are found to be 2.771 and 2.503 eV for $CdGaInS_4$, and 2.665 and 2.479 eV for $CdGaInS_4:Er^{3+}$ at 10 K. The temperature dependence of the optical energy gap was well represented by the Varshni equation. In $CdGaInS_4$, the values of ${\alpha},\;{\beta}$ of the direct and the indirect energy gap were found to be $7.57{\times}10^{-4}eV/K$. $6.53{\times}10^{-4}eV/K$ and 240K. 197K. and the values of ${\alpha}$ and ${\beta}$ of the direct and the indirect energy gap in the $CdGaInS_4:Er^{3+}$ were given by $8.28{\times}10^{-4}eV/K,\;2.08{\times}10^{-4}eV/K$ and 425 K, 283 K, respectively.

  • This study demonstrated that upregulation of gene expression of endoplasmic reticulum (ER) stress chaperones (Bip, ERp29, calnexin, and PDI), ER stress sensors (PERK, ATF6, and Ire1), and cAMP phosphodiesterase 7A1 (cAMP PDE7A1) was induced by ER stresses in FRTL5 cells. While removing A23187 from the culture medium restored upregulation of cAMP PDE7A1 gene expression, removal of thapsigargin did not recover its expression. In addition, cAMP PDE7A1 gene expression was strongly inhibited by treatment with A23187 combined with thyroid stimulating hormone (TSH). The results are the first to show that ER stress induces cAMP PDE7A1 gene expression.

Analysis of Factors Related to Length of Stay Time in Patients with Back Pain at Emergency Department

  • Purpose: Most patients with acute low back pain visit emergency room (ER). They mostly need beds, and if their length of stay is longer, it can become difficult to accommodate new patients at the ER. We analyzed the treatment process of patients with back pain and tried to find method for shortening of the length of stay at the ER. Methods: We retrospectively analyzed the medical records of patients with back pain who visited at our ER for one year. Patients were divided into two groups according to their length of stay at ER and were compared the charateristcs of between two groups. Results: A total of 274 patients were included in the study. Eigthy-nine patients (32.5%) were in the group with less than 3 hours and 185 patients (67.5%) were in the other group. In the comparison of the two groups according to the medical departments, the number of patients who were in group with more than 3 hours were 25 (14.0%) in the emergency department, 94 (50.5%) in neurosurgery, 66 (35.5%) in orthopedic surgery. Length of stay was significantly increased in orthopedic surgery and neurosurgery (p=0.014). In addition, the length of stay was longer when computed tomography and magnetic resonance imaging examinations were performed (p=0.000). Regardless of the type of analgesic agent, the median time to the analgesic treatment was shorter in the group with less than 3 hours (p=0.034). Conclusions: In patients with back pain who visit the ER, the emergency medicine doctor will early control the pain and do not unnecessary image examination to reduce a length of stay at the ER.

Activation of Estrogen Receptor by Bavachin from Psoralea corylifolia

  • In this study, we examined the estrogenic activity of bavachin, a component of Psoralea corylifolia that has been used as a traditional medicine in Asia. Bavachin was purified from ethanolic extract of Psoralea corylifolia and characterized its estrogenic activity by ligand binding, reporter gene activation, and endogenous estrogen receptor (ER) target gene regulation. Bavachin showed ER ligand binding activity in competitive displacement of [$^3H$] $E_2$ from recombinant ER. The estrogenic activity of bavachin was characterized in a transient transfection system using $ER{\alpha}$ or $ER{\beta}$ and estrogen-responsive luciferase plasmids in CV-1 cells with an $EC_{50}$ of 320 nM and 680 nM, respectively. Bavachin increased the mRNA levels of estrogen-responsive genes such as pS2 and PR, and decreased the protein level of $ER{\alpha}$ by proteasomal pathway. However, bavachin failed to activate the androgen receptor in CV-1 cells transiently transfected with the corresponding receptor and hormone responsive reporter plasmid. These data indicate that bavachin acts as a weak phytoestrogen by binding and activating the ER.

A Continuously Variable ER Damper with Energy Generation (재생 에너지를 이용하는 연속가변 ER댐퍼에 관한 연구)

  • This study presents a so-called regenerative ER damper which does not require external power sources with aid of energy conversion from kinetic energy into electrical energy. The ER damper is manufactured by employing a rack and pinion mechanism which converts a linear piston motion to a rotary motion. This rotary motion is amplified by gears and subsequently activates a generator to produce an electrical energy. In order to demonstrate the efficiency of the mechanism, with respect to the frequency of the ER damper is investigated a generated electric voltage, which is amplified via a transformer prior to being applied to the ER damper. In addition, the damping force of the ER damper obtained by the regenerated voltage is evaluated.

  • Electro-Rheological(ER) fluids consist of suspensions of fine polarizable particles In a dielectric oil, which upon application of an external electric field control take on the characteristics of the Bingham solid. In this study, the temperature dependence of the viscosity was Investigated for an ER fluid consisting of 35 weight % of zeolite particles in hydraulic oil 46cSt. Thermal activation analysis was performed by changing the ER fluid's temperature from $-10^{\circ}C$ to $50^{\circ}C$. According to the analysis, the activation energy for flow of the ER fluid was 79.6 kJ/mole without applying electric field. On the other hand, with the electric field of 2kV/mm, the linearity between viscosity and temperature was not existed By changing the temperatures the viscosity (or shear stress) versus shear rates were measured. In this experiment shear rates were increased from 0 to $200s^{-1}$ in 2 minutes. Generally, the hydraulic oil 46cSt will be operated at the temperature of about $40^{\circ}C$, thus, the ER fluid's electric field dependence of viscosity was examined at this temperature. Also, an influence of adding the dispersant(Carbopl 940) on ER effect was discussed.

  • $Yb^{3+}$, $Er^{3+}$ co-doped $SrMoO_4$ ($SrMoO_4$ : $Yb^{3+}/Er^{3+}$) specimens have been successfully synthesized via the complex citrate-gel method and their structural and optical properties were investigated in detail. Under 980 nm excitation, $SrMoO_4$ : $Yb^{3+}/Er^{3+}$ UC phosphors have been emitted strong green luminescence at 530 and 550 nm with weak red emission around 670 nm corresponding to the intra 4f transitions of $Er^{3+}$ ($^4F_{9/2}$, $^2H_{11/2}$, $^4S_{3/2}$) ${\rightarrow}$ $Er^{3+}$ ($^4I_{15/2}$). The optimal doping concentrations of $Er^{3+}$ and $Yb^{3+}$ ions were verified to 2/16 mol% and a possible upconversion mechanism depending on pump power dependence is studied in detail.

  • Thermal stabilizing effect of $Yb^{3+},\;Er^{3+}$ codoping into $TiO_2$ powder prepared by sol-gel method and its upconversion characteristics were analyzed. The effect of $TiO_2:Yb^{3+},\;Er^{3+}$ ions on crystallinity and phase transition was studied by X-ray diffraction (XRD). The change of band-gap energy induced from Yb and Er codoping was analyzed by UV-Vis. The band-gap energy of $TiO_2$ have been slightly narrowed by $Yb^{3+},\;Er^{3+}$ codoping, which indicated that the $Yb^{3+},\;Er^{3+}$ ions can enhance the photo-catalytic property of $TiO_2$. green and red up-conversions of $Yb^{3+}$ and $Er^{3+}$ co-doped $Y_2O_3:Yb^{3+},\;Er^{3+}$ phosphor were analyzed by PL equipped with 980 nm laser.
