Effects of Anti-ecotoxicological Curcumin Nanospheres on Feed Efficiency and Fecal Odor in Mice (환경 독성 억제효과를 가진 커큐민 나노스피어가 마우스의 사료 효율 및 악취저감에 미치는 영향)

  • Curcumin ($C_{21}H_{20}O_6$) is a hydrophobic polyphenol found in turmeric. Although curcumin has been used as a natural medicine, its major limitation is related to poor absorption from the gut. Therefore, we developed a method for preparation of Curcumin Nanospheres (CN) to improve the aqueous-phase solubility of curcumin and investigate the functional role of CN in promoting feed efficiency and odor reduction in mice. CN showed inhibitory effects on actate dehydrogenase (LDH) cytotoxicity induced by ecotoxic substance toluene in gut epithelial HCT116 cells. In addition, the weights of internal organs (liver, heart, kidneys, and spleen) and the levels of serum Glutamate Oxaloacetate Transaminase (GOT), Glutamate Pyruvate Transaminase (GPT), and LDH did not show significant differences between mice administered oral CN for two weeks and compared to the control group. Interestingly, CN not only reduced hydrogen sulfide ($H_2S$) and ammonia ($NH_3$) levels and fecal odor, but also improved feed efficiency in mice. These results demonstrate that oral nano-delivery of anti-ecotoxicological CN is a functional system to deliver curcumin to the gut to improve feed efficiency and reduce fecal odor in mice.

  • Role of low degradable protein in milk production of early lactating Murrah buffaloes has been studied in relation to energy status of test animals. Replacement of conventional concentrate mixture with low degradable cotton seed cake resulted in appreciable changes in circulatory transaminases and phosphatase levels. The enzymes viz. glutamate oxaloacetate and glutamate pyruvate transaminase and alkaline phosphatases increased with feeding of said cake indicating stress on hepatic tissue. Animals seemed to overcome stress by feeding enhanced levels of same protein along with improved feed intake, body weight and milk production.

  • Methanol extracts of the roots of Atractylodes japonica (alba) (Compositae) and Gentiana scabra (Gentianaceae) were further fractionated with n-butanol, chloroform, n-hexane and water. Then each fraction was p.o. administered to an animal model of hepatitis caused by $CCl_4$ intoxication. Measurements of sleeping time of mice induced by hexobarbital administration indicated that n-butanol and chloroform fractions of Atractylodes japonica (alba), and chloroform and n-hexane fractions of Gentiana scabra showed significant liver protective activities. Measurements of the activities of serum transaminase, S-GOT and S-GPT appeared to support these results.

  • Doxorubicin induces oxidative stress leading to cardiotoxicity causing electrocardiogram abnormalities and increases in biomarkers associated with toxicity. Green tea extract (GTE) is reported to possess antioxidant activity mainly via its polyphenolic constituent, catechins. This study was intended to determine the effect of various doses of GTE (25, 50 and 100 mg/kg/day p.o. for 30 days) on doxorubicin-induced electrocardiographic and biochemical changes in rat heart. The latter included lactate dehydrogenase, creatine kinase, and glutamic oxaloacetate transaminase in serum and superoxide dismutase, catalase, and reduced glutathione, as well as membrane bound enzymes like $Na^+K^+ATPase,\;Ca^{2+}ATPase,\;Mg^{2+}ATPase$ and decreased lipid peroxidation in heart tissue Results demonstrated that rats which received GTE were less susceptible to such changes indicating protection afforded by GTE.

  • The anti-oxidant activities of fucosterol isolated from the marine algae Pelvetia siliquosa were investigated. Fucosterol exhibited a significant decrease in serum transaminase activities elevated by hepatic damage induced by CCl$_4$-intoxication in rats. Fucosterol inhibited the sGOT and sGPT activities by 25.57 and 63.16%, respectively. Fucosterol showed the increase in the anti-oxidant enzymes such as hepatic cytosolic superoxide dismutase, catalase and glutathione peroxidase activities by 33.89, 21.56 and 39.24%, respectively, in CCl$_4$-intoxicated rats. Effects of Vitamin E and Selenium on the Antioxidative Defense System in Streptozotocin-induced Diabetic Rats (Vitamin E와 Selenium이 Streptozotocin 유발 당뇨쥐의 항산화계에 미치는 영향)

  • The purpose of this study was to investigate the effect of vitamin E and selenium on the antioxidative defense mechanism in the liver of streptozotocin(STZ)-induced diabetic rats. Sprague-Dawley male rats(120$\pm$10gm) were randomly assigned to one control and five STZ-diabetic groups. Diabetic groups were classified to STZ-0E (vitamin E free diet), STZ-40E(40mg vitamin E/kg of diet), STZ-400E(400mg vitamin E/kg of diet), STZ-S(0.5ppm Se/kg of diet) and STZ-400ES(400mg vitamin E and 0.5ppm Se/kg of diet) according to the level of vitamin E and selenium supplementation. Diabetes was experimentally induced by intravenous adminstration of 55mg/kg of STZ in citrate buffer(pH 4.3) after 4-weeks feedng of six experimental diets. Animals were sacrificed at the 4th day of diabetic states. Activities of the serum glutamic oxaloacetate transaminase(GOT) and the glutaminc pyruvate transaminase(GPT) in STZ-0E, STZ-40E and STZ-S rats were higher than those of control. Liver xanthine oxidase activities were similar to serum GOT and GPT. Liver superoxide dismutase(SOD) activities were higher in STZ-0E and STZ-40E groups by 33%, 22%, respectively than that of control. Glutathione S-transferase(GST) activities of liver were similar to GSH-Px activities. The contents of vitamin E in liver tissue were significantly lower STZ-0E, STZ-40E and STZ-S groups by 50%, 36%, 45% than that of control. Reduced glutathione(GSH) contents of liver were lower STZ-0E, STZ-40E, STZ-400E, STZ-S and STZ-400ES groups by 57%, 51%, 19%, 18%, 12% than that of control. Lipid peroxide values (LPO) in liver were higher 5.6, 2.3 and 2.3 times in STZ-0E, STZ-40E and STZ-S group than that of control. The present results indicate that STZ-induced diabetic rats are more sensitive to oxidative stress, leading to the acceleration of lipid peroxidation process, which can be more accelerated by feeding the low level of dietary vitamin E. In the coincident supplementation of high dietary vitamin E and selenium antioxidative enzymes activities and physiolosical antioxidants were increased more than those of the separate supplementation of vitamin E or selenium. Therefore, dietary vitamin E and selenium reduced peroxidative damage of tissue, promoting antioxidative defense mechanism against lipid peroxidation by diabetes.

Association Factors for CT Angiography Spot Sign and Hematoma Growth in Korean Patients with Acute Spontaneous Intracerebral Hemorrhage : A Single-Center Cohort Study

  • Objective : This study was conducted to clarify the association factors and clinical significance of the CT angiography (CTA) spot sign and hematoma growth in Korean patients with acute intracerebral hemorrhage (ICH). Methods : We retrospectively collected the data of 287 consecutive patients presenting with acute ICH who arrived within 12 hours of ictus. Baseline clinical and radiological characteristics as well as the mortality rate within one month were assessed. A binary logistic regression was conducted to obtain association factors for the CTA spot sign and hematoma growth. Results : We identified a CTA spot sign in 40 patients (13.9%) and hematoma growth in 78 patients (27.2%). An elapsed time to CT scan of less than 3 hours (OR, 5.14; 95% CI, 1.76-15.02; p=0.003) was associated with the spot sign. A CTA spot sign (OR, 5.70; 95% CI, 2.70-12.01; p<0.001), elevated alanine transaminase (GPT) level >40 IU (OR, 2.01; 95% CI, 1.01-4.01; p=0.047), and an international normalized ratio ${\geq}1.8$ or warfarin medication (OR, 5.64; 95% CI, 1.29-24.57; p=0.021) were independent predictors for hematoma growth. Antiplatelet agent medication (OR, 4.92; 95% CI, 1.31-18.50; p= 0.019) was significantly associated with hematoma growth within 6 hours of ictus. Conclusion : As previous other populations, CTA spot sign was a strong predictor for hematoma growth especially in hyper-acute stage of ICH in Korea. Antithrombotics medication might also be associated with hyper-acute hematoma growth. In our population, elevated GPT was newly identified as a predictor for hematoma growth and its effect for hematoma growth is necessary to be confirmed through a further research.

The Effects of Solanum nigum Linne extract on the Hepatotoxicity of Rats Induced by Lipopolysaccharide (Lipopolysaccharide로 유발된 간독성에 대한 까마중의 효과)

  • This study aimed to investigate the protective effect of Solanum nigum Linne total extract (SNT), Solanum nigum Linne leaf extract (SNL), Solanum nigum Linne root extract (SNR) on liver injury induced by Lipopolysaccharide(LPS) in Sprague-Dawley rats. SNT, SNL, SNR of 100 mg/kg concentration was intraperitoneally administered into rats at dose of 1.5 ml/kg for 20 days. on the day 1.5 ml/kg of LPS was injected. Four hours later, they were anesthetization with ether and dissected. Glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT) were measured in serum and superoxide dismutase (SOD), catalase and glutathione peroxidase (GPX) were measured in liver homogenate. SNT, SNL, SNR extract inhibited GOT and GPT activities in LPS-induced rats, whereas increased SOD, Catalase and GPX activity in liver tissue of LPS-induced rats. These suggested that SNT, SNL, SNR could be used for functional beverage.

  • The effects of alcohol. extract of acacia on total cholesterol, ${\beta}-lipoprotein$ content and S-GOT activity in serum of rabbit were examined. Rabbits were divided into four groups. A : Control group B : Cholesterol group C : Cholesterol+aca. alc. ex. (A) group D : Cholesterol+aca. alc. ex. (B) group Each group had four rabbits. A Group was fed with the stock diet only for the experimental period of 28 days. B Group was fed with the stock diet containing 500mg of cholesterol. C Group was fed with the stock diet containing 500mg of cholesterol and 300mg of acacia alcohol extract. D Group was fed with the stock diet containing 500mg of cholesterol and 500mg of acacia aocohol extract. Total cholesterol ${\beta}-lipoprotein$ content and S-GOT activity in serum was determined with spectrophotometer, spectronic 20. It was confirmed that the extract of acacia alcohol inhibits the increase of total cholesterol, ${\beta}-lipoprotein$ content and S-GOT activity in serum but the statistical probability was not significant.

Effects of Dietary Fiber Isolated from Synurus deltoides on Constipation in Loperamide-Induced Rats (수리취 식이섬유가 Loperamide로 유발된 변비에 미치는 영향)

  • The effects of dietary fiber isolated from Synurus deltoides on constipation induced by loperamide (4 mg/kg/day) were investigated. Food intake and body weight both decreased in the 5% S. deltoides dietary fiber and loperamide-treated group (SD5) and 10% S. deltoides dietary fiber and loperamide-treated group (SD10), whereas fecal water contents increased by 2.4 and 3.4-fold in the SD5 and SD10 groups, respectively. The concentrations of total-cholesterol, HDL-cholesterol and triglyceride in the sera of the SD5 and SD10 groups were lower than those in the control (C) group. However, the biochemical parameters, GOT (glutamic oxaloacetic transaminase), GPT (glutamic pyruvic transaminase), and glucose levels, were not affected by the level of S. deltoides. In addition, the concentrations of total-cholesterol and triglyceride in the livers of the SD5 and SD10 groups were also significantly lower than those in the control group. These results suggest that dietary fiber isolated from S. deltoides might ameliorate constipation symptoms, and lower lipid concentrations in the blood and liver.
