Buckwheat has gained scientists concern due to its nutritional and medicinal values in recent years, and many important bioactive compounds. The present study was performed to investigate the influence of the varietal performance and environmental conditions on the content of rutin and quercetin in Tartary buckwheat germplasm. A total of 44 foreign Tartary buckwheat germplasms were examined and compared their contents based on the collected countries, seed shape, and seed color. The highest number of germplasm (16) was found in ranged from 600 mg/100 g to 800 mg/100 g of rutin content. In case of quercetin content, the highest number of germplasm (19) was observed in ranged from 5 mg/100 g to 10 mg/100 g. However, the rutin content showed the highest value (1326.6 mg/100 g) from CBU408 and quercetin content showed the highest value (22.5 mg/100 g) from CBU456. The number of germplasm showed the highest value (23) in ranged from 3000 mg/100 g to 4000 mg/100 g of rutin content. The number of germplasm is the highest value (30) in ranged from 100 mg/100 g to 200 mg/100 g of quercetin content. When rutin and quercetin content in seed was compared according to collected countries, seed shape, and seed color respectively, no association was appeared with rutin and quercetin content. In sprout of whole Tartary buckwheat germplasm, mean of rutin content and quercetin content was 3362.9 mg/100 g and 14.2 mg/100 g respectively. In conclusion, the results observed from the present study suggest that highly rutin germplasm could be used more efficiently via breeding to develop a highly rutin content variety for using in sprout. No correlation was appeared in the case of quercetin. However, the quercetin content in sprout was 4~90 times more than seed. So, sprout could be used more efficiently than seed in the case of quercetin. Our results suggest that the varietal variation and qualitative characteristic differences of rutin and bioactive compounds may provide important nutrient sources and industrial application of Tartary buckwheat.
From the leaves of Ulmus parviflora isoquercitrin and rutin were isolated and identified by chemical and spectral analysis. The content of rutin was determined by HPLC, and it was about 2.81% in methanol extract.
This study aimed to enhance rutin contents by controlling germination condition for manufacturing buckwheat sprouts. Two kinds of buckwheat, a common buckwheat (Fagopyrum esculentum Moench) and a tartary buckwheat (Fagopyrum tataricum Gaertner) were used. By comparing the rutin content of two buckwheats, tartary buckwheat was 487 ppm, about 36 times higher than common buckwheat. Both common buckwheat and tartary buckwheat which germinated and grew under the light had higher rutin content relatively. In case of tartary buckwheat, rutin content of over 10 cm sprout was 4,579 ppm (without the light), and 5,160 ppm (with the light). Furthermore, tartary buckwheat was germinated and grew under different light strengths from 2,000 to 22,000 Lux. The rutin contents of tartary buckwheat sprout that was grown under the 22,000 Lux light was the highest. The rutin content was increased dramatically at 14,000 Lux of light. From 14,000 to 22,000 Lux, there was a little change on rutin content. Therefore, the condition of 14,000 Lux light was determined optimal for manufacturing tartary buckwheat sprouts. Also, rutin contents of extracts treated with 60, 70, 80 and $90^{\circ}C$ during different time had no significant difference. Therefore, rutin of tartary buckwheat sprout extract had thermostability up to $90^{\circ}C$.

For utilization of buckwheat as vegetable, rutin and minerals of the leaves at growing stages were investigated. The contents of crude protein and crude fat in seed of nine improved kinds of Korean buckwheat were $16.2{\sim}20.4%$ and $2.2{\sim}2.9%$ respectively. Ash content $(2.4{\sim}4.3%)$ in the improved kinds were higher than those in the native kinds. Content of crude fiber in the improved kinds were lower than those in the native kinds. Suwon 11 (0.12%) and Suwon 5 (0.11%) had high contents of rutin among the improved kinds. The composition of K, Ca, Mg and Fe in the improved kinds were better than that in the native kinds. Vegetable of the improved kinds had higher content of crude protein than the native kinds. Suwon 10 and 11 among vegetables of the improved kinds contained high amount of crude protein. Swon 5 had the highest content of rutin (0.40%) in the improved kinds. Suwon 5 was tested for the changes of rutin content in growing. Rutin content was 0.40% in the first growing stage, whereas 1.32% in the final growing stage. Insoluble materials in 10% HCl was also 69.8% in the first growing stage, whereas 4.68% in the final growing stage. Iron and calcium contents were increased with growing of vegetable.

Objectives: Neutrophils represent the front line of human defense against infections. Immediately after stimulation, neutrophilic enzymes are activated and produce toxic mediators such as pro-inflammatory cytokines, nitric oxide (NO) and myeloperoxidase (MPO). These mediators can be toxic not only to infectious agents but also to host tissues. Because flavonoids exhibit antioxidant and anti-inflammatory effects, they are subjects of interest for pharmacological modulation of inflammation. In the present study, the effects of rutin on stimulus-induced NO and tumor necrosis factor $(TNF)-{\alpha}$ productions and MPO activity in human neutrophils were investigated. Methods: Human peripheral blood neutrophils were isolated using Ficoll-Hypaque density gradient centrifugation coupled with dextran T500 sedimentation. The cell preparations containing > 98% granulocytes were determined by morphological examination through Giemsa staining. Neutrophils were cultured in complete Roswell Park Memorial Institute (RPMI) medium, pre-incubated with or without rutin ($25{\mu}M$) for 45 minutes, and stimulated with phorbol 12-myristate 13-acetate (PMA). Then, the $TNF-{\alpha}$, NO and MPO productions were analyzed using enzyme-linked immunosorbent assay (ELISA), Griess Reagent, and MPO assay kits, respectively. Also, the viability of human neutrophils was assessed using tetrazolium salt 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT), and neutrophils were treated with various concentrations of rutin ($1-100{\mu}M$), after which MTT was appended and incubated at $37^{\circ}C$ for 4 hour. Results: Rutin at concentrations up to $100{\mu}M$ did not affect neutrophil viability during the 4-hour incubation period. Rutin significantly decreased the NO and $TNF-{\alpha}$ productions in human peripheral blood neutrophils compared to PMA-control cells (P < 0.001). Also, MPO activity was significantly reduced by rutin (P < 0.001). Conclusion: In this in vitro study, rutin had an anti-inflammatory effect due to its inhibiting NO and $TNF-{\alpha}$ productions, as well as MPO activity, in activated human neutrophils. Treatment with rutin may be considered as a therapeutic strategy for neutrophil-mediated inflammatory/autoimmune diseases.
In this study, we prepared polymer micelles containing quercetin and rutin, known as antioxidants, using poly(${\varepsilon}$-caprolactone)-b-poly(ethylene glycol), and evaluated in vitro skin permeation of the active materials. Quercetin and rutin loaded micelles were characterized by DSC (differential scanning calorimetry), HPLC (high performance liquid chromatography) and DLS (dynamic light scattering) measurements. The particle size of the polymer micelles increased in a concentration dependent manner (0.5~2.0% PCL-b-PEG). The Zeta potential of quercetin and rutin loaded micelles remained constant. To evaluate the skin penetration of PCL-b-PEG micelles, Franz diffusion cell experiment was performed. The aqueous solutions of quercetin and rutin were used as the control groups. Quercetin and rutin loaded PCL-b-PEG micelles showed more efficient skin permeation than the control groups. Safety assessment (patch test) of quercetin and rutin loaded PCL-b-PEG micelles on skin was performed to test application possibility of the polymer micelles to cosmetics. Any adverse symptoms were not observed.
Flavonoid contents of common buckwheat (cv. Kitawase) and tartary buckwheat (cv. Hokkai T 8, Hokkai T 9 and Hokkai T 10) were determined by high-performance liquid chromatography (HPLC). Moreover, they were measured at different plant developments such as 10, 18, 20, 22 and 30 days after sowing (DAS) and with plant parts including leaf, stem and flower harvested at 30 DAS. Total flavonoids including chlorogenic acid, four kinds of C-glycosylflavones (orientin, isoorientin, vitexin, isovitexin) and rutin of tartary buckwheats (range of 44.2-54.7, mean 44.2) were found 35% higher than those of common buckwheat (28.9 mg/g dry wt.). Among them, rutin was measured above 80% of total flavonoid contents. The other flavonoids (chlorogenic acid and four kinds of C-glycosylflavones) presented the highest level at 10 DAS and decreased according to plant developments. On the other hand, rutin content of Kitawase presented the highest level (33.6 mg/g dry wt.) at 22 DAS and decreased up to 30 DAS. Rutin content in tartary buckwheat temporarily decreased from 10 to 18 DAS and then reversely increased up to 30 DAS presented the highest level as 'U' curve. In Hokkai T 10, rutin content was found the highest level (53.8 mg/g dry wt.) at 30 DAS. In different plant parts harvested at 30 DAS, rutin content of leaf (range of 42.8-68.0, mean 57.0) was 5.3-fold higher than that of stem (range of 8.0-15.9, 10.8 mg/g dry wt.), regardless of cultivar. Significantly, rutin content (78.7) in the flower of Kitawase was 1.8 times higher than in the leaf and 9.8 times in the stem. Especially, chlorogenic acid content (14.6 mg/g dry wt.) in the flower of Kitawase was 63-fold higher than in the leaf, and 20-fold in the stem.

Flavonoids including quercetin and rutin are a group of naturally occurring compounds widely distributed in plants, especially in buckwheat. Thus, cereal and the leaf of the plant have increasingly used as a source of nutritional and functional foods such as noodle, cake or soup in Korea, Japan and other countries. This study investigated comparative effects of dietary rutin rich in buckwheat and its aglycone, quercetin, on serum biomarkers and antioxidant parameters in rats treated with chronic ethanol. Rats were fed with the liquid diets prepared by the method of Lieber Decarli. Serum alanine transaminase (ALT) and aspartate transaminase (AST) activities increased significantly by alcohol feeding. Dietary flavonoids including rutin, quercetin and their mixtures (1/1, v/v) decreased significantly the activities of serum ALT whereas the feeding of quercetin decreased only the activity of serum AST. The concentration of serum malondialdehydes elevated by chronic alcohol feeding decreased markedly in all the experimental groups that were fed with the flavonoids; however, the combined administration of quercetin or rutin, but not that of rutin or quercetin alone decreased significantly the concentration of liver malondialdehydes to the normal range in rats fed without ethanol. Our results suggested that dietary combined mixture of rutin and quercetin might be effective in ameliorating adverse responses seen in rats exposed to ethanol chronically.

This study carried out to investigate the variation of flavonoids contents in mungbean (Vigna radiata L.) according to plant parts, harvesting time, growth stage, and sowing time. Vitexin and isovitexin were found only in the seeds but not in the leaves, petioles, stems, and roots. Vitexin and isovitexin in seeds were detected only in the seed coat at concentration of 51.1 and $51.7\;mg{\cdot}g^{-1}$, respectively, but not in the cotyledon. There were no differences in the content of vitexin and isovitexin in mungbean seeds according to early, recommended and late harvesting times. Rutin in leaves was isolated and identified as a functional substance. The content of rutin was the highest in the leaves and higher in the order of petioles and stems. However, there was no rutin in the seeds, roots, and pods. The highest rutin content in the leaves, petioles and stems was observed at the 3rd leaf stage, which was higher in the order of the 5th and 7th leaf stage.

Buckwheat is well-known crop for containing a high contents of flavonoids that are effective in vascular disease. The current study was performed to estimate the influence of morphological characterization of Fagopyrum esculentum (ES) germplasm for seed's two major flavonoids contents: rutin and quercetin. We found that the red stem color, pale green leaf color, arrowhead leaf shape, white flower color, pale brown seed coat color, and egg-shaped seed were significantly associated with 77%, 56.7%, 83.7%, 98.7%, 70.8% and 74.5% germplasm, respectively. Overall, the rutin contents of ES germplasm ranged from 0.30 to 47.86 mg/100g dry weight (DW) and the quercetin contents ranged from 0 to 1.22 mg/100g DW. The rutin contents of germplasm possessing red stem color, pale green leaf color, arrowhead leaves, white flower color, pale brown seed coat color and egg-shaped seed ranged from 7.22 to 47.86 mg/100g DW. However, the quercetin contents of germplasm with red stem color and pale brown seed coat color ranged from 0 to 1.15 mg/100g DW, with pale green leaves ranged from 0 to 0.96 mg/100g, with arrowhead leaves and white flower ranged from 0 to 1.22 mg/100g and with egg-shaped seed ranged from 0.32 to 1.22 mg/100g DW. In PCA analysis, the first three principal components (PCs) showed Eigen value more than 1 and accounted for 51.70% of variation. For both higher contents of rutin and quercetin, the morphological evaluation in ES shows a tendency of red stem color, arrowhead leaves, pale green leaf color, white flower color, pale brown seed coat color and egg-shaped seed. From this information, we can assume the rutin and quercetin contents by the morphological characteristics of the germplasm. And It could be useful in improving the rutin and quercetin contents and selecting proper resources for cultivation in existing buckwheat cultivars.
