Effects of Tween 80 and Fibrolytic Enzymes on Ruminal Fermentation and Digestibility of Feeds in Holstein Cows

  • The effects of the nonionic surfactant Tween 80 and a mixture of fibrolytic enzymes on total tract digestion, in situ disappearance (ISD) and ruminal fermentation characteristics of orchardgrass hay and barley grain were investigated in a 4${\times}$4 Latin square experiment with 4 non-lactating Holstein cows and 4 diets in 4 periods. Cows were offered a total mixed ration of 50% rolled barley grain and 50% orchardgrass hay treated with either 1) water (control), 2) 0.2% (vol/wt) Tween 80, 3) 0.2% (vol/wt) hydrolytic enzyme, or 4) 0.2% hydrolytic enzyme plus 0.2% Tween 80. Total tract digestibility coefficients of DM, nitrogen, NDF and ADF were not affected (p>0.05) by dietary treatment. Compared to the control, the rate of ISD of DM from orchardgrass hay was faster (p<0.05) in cows receiving diets treated with the enzyme alone or with enzyme plus Tween 80 (0.06/h vs. 0.076 and 0.069/h). The rate of digestion was lower (p<0.05) as compared to control when barley grain was treated with these additives. Ruminal fluid pH and concentrations of total VFA, acetate, isobutyrate and butyrate were not affected (p>0.05) by treatments. Cows that consumed diets treated with enzyme plus Tween 80 had higher (p<0.05) ruminal concentrations of propionate and isovalerate, and lower (p<0.05) acetate:propionate ratios. Compared to the control, microbial protein synthesis tended (p = 0.13) to increase with the addition of enzyme to the diet while nonammonia nitrogen flow to the duodenum increased (p<0.05) with both enzyme and Tween 80 treatments. The study indicated that fibrolytic enzymes alone or in combination with Tween 80 could enhance ISD of orchardgrass hay and ruminal concentrations of propionate, valerate and iso-valerate, but do not affect total tract digestibility.

식용유 정제 폐백토(Spent Bleaching Clay)의 급여가 반추위 발효특성과 사료 이용률에 미치는 영향 (Effects of Spent Bleaching Clay Supplementation on Ruminal Fermentation and Digestibility in Holstein Dairy Cows)

  • 본 시험은 Spent bleaching clay(SBC) 첨가에 따라 반추위내의 발효특성과 이용율을 조사하기 위하여 Fistula가 장착된 홀스타인 3두를 공시하여 반추위내 발효성상을 조사하였다. 반추위내 pH 변화는 대조구와 SBC 2% 첨가구간에는 차이가 없었으나, SBC 4% 첨가에 의하여 낮아지는 경향이 인정되었다. 반추위내 $NH_3-N$의 함량은 SBC 4% 첨가구가 대조구와 SBC 2% 첨가구에 비하여 사료 급여 후 3시간째까지 높은 경향을 나타냈으나 이후 3처리구는 유사한 경향을 나타내었다. 한편 SBC의 첨가수준에 관계없이 반추위내 VFA의 함량은 변화가 없는 것으로 나타났다. 반추위내 건물 및 유기물 손실율은 SBC 4% 첨가구가 대조구 및 SBC 2% 첨가구에 비해서 배양초기에는 낮아지는 경향을 보였으나 이후 4처리간 차이는 인정되지 않았다.

Predicted functional analysis of rumen microbiota suggested the underlying mechanisms of the postpartum subacute ruminal acidosis in Holstein cows

  • Background: The relationships between the postpartum subacute ruminal acidosis (SARA) occurrence and predicted bacterial functions during the periparturient period are still not clear in Holstein cows. Objectives: The present study was performed to investigate the alterations of rumen fermentation, bacterial community structure, and predicted bacterial functional pathways in Holstein cows. Methods: Holstein cows were divided into the SARA (n = 6) or non-SARA (n = 4) groups, depending on whether they developed SARA during the first 2 weeks after parturition. Reticulo-ruminal pH was measured continuously during the study period. Reticulo-ruminal fluid samples were collected 3 weeks prepartum, and 2 and 6 weeks postpartum, and blood samples were collected 3 weeks before, 0, 2, 4 and 6 weeks postpartum. Results: The postpartum decline in 7-day mean reticulo-ruminal pH was more severe and longer-lasting in the SARA group compared with the non-SARA group. Changes in predicted functional pathways were identified in the SARA group. A significant upregulation of pathway "PWY-6383" associated with Mycobacteriaceae species was identified at 3 weeks after parturition in the SARA group. Significantly identified pathways involved in denitrification (DENITRIFICATION-PWY and PWY-7084), detoxification of reactive oxygen and nitrogen species (PWY1G-0), and starch degradation (PWY-622) in the SARA group were downregulated. Conclusions: The postpartum SARA occurrence is likely related to the predicted functions of rumen bacterial community rather than the alterations of rumen fermentation or fluid bacterial community structure. Therefore, our result suggests the underlying mechanisms, namely functional adaptation of bacterial community, causing postpartum SARA in Holstein cows during the periparturient period.

  • Anaerobic rumen microorganisms mainly bacteria, protozoa and fungi degrade ligno-cellulosic feeds consumed by the ruminants. The ruminants in developing countries are predominantly maintained on low grade roughage and grazing on degraded range land resulting in their poor nutrient utilization and productivity. Hence, manipulation of rumen fermentation was tried during last two decades to optimize ruminal fermentation for improving nutrient utilization and productivity of the animals. Modification of rumen microbial composition and their activity was attempted by using chemical additives those selectively effect rumen microbes, introduction of naturally occurring or genetically modified foreign microbes into the rumen and genetically manipulation of existing microbes in the rumen ecosystem. Accordingly, rumen protozoa were eliminated by defaunation for reducing ruminal methane production and increasing protein outflow in the intestine, resulting in improve growth and feed conversion efficiency of the animals. Further, Interspecies trans-inoculation of rumen microbes was also successfully used for annulment of dietary toxic factor. Additionally, probiotics of bacterial and yeast origin have been used in animal feeding to stabilize rumen fermentation, reduced incidence of diarrhoea and thus improving growth and feed conversion efficiency of young stalk. It is envisaged that genetic manipulation of rumen microorganisms has enormous research potential in developing countries. In view of feed resource availability more emphasis has to be given for manipulating rumen fermentation to increase cellulolytic activity for efficient utilization of low grade roughage.

Effects of Candida norvegensis Live Cells on In vitro Oat Straw Rumen Fermentation

  • This study evaluated the effect of Candida norvegensis (C. norvegensis) viable yeast culture on in vitro ruminal fermentation of oat straw. Ruminal fluid was mixed with buffer solution (1:2) and anaerobically incubated with or without yeast at $39^{\circ}C$ for 0, 4, 8, 16, and 24 h. A fully randomized design was used. There was a decrease in lactic acid (quadratic, p = 0.01), pH, (quadratic, p = 0.02), and yeasts counts (linear, p<0.01) across fermentation times. However, in vitro dry matter disappearance (IVDMD) and ammonia-N increased across fermentation times (quadratic; p<0.01 and p<0.02, respectively). Addition of yeast cells caused a decrease in pH values compared over all fermentation times (p<0.01), and lactic acid decreased at 12 h (p = 0.05). Meanwhile, yeast counts increased (p = 0.01) at 12 h. C. norvegensis increased ammonia-N at 4, 8, 12, and 24 h (p<0.01), and IVDMD of oat straw increased at 8, 12, and 24 h (p<0.01) of fermentation. Yeast cells increased acetate (p<0.01), propionate (p<0.03), and butyrate (p<0.03) at 8 h, while valeriate and isovaleriate increased at 8, 12, and 24 h (p<0.01). The yeast did not affect cellulolytic bacteria (p = 0.05), but cellulolytic fungi increased at 4 and 8 h (p<0.01), whereas production of methane decreased (p<0.01) at 8 h. It is concluded that addition of C. norvegensis to in vitro oat straw fermentation increased ruminal fermentation parameters as well as microbial growth with reduction of methane production. Additionally, yeast inoculum also improved IVDMD.

Substitution of Wheat for Corn in Beef Cattle Diets: Digestibility, Digestive Enzyme Activities, Serum Metabolite Contents and Ruminal Fermentation

  • The objective of this study was to evaluate the effect of diets containing different amounts of wheat, as a partial or whole substitute for corn, on digestibility, digestive enzyme activities, serum metabolite contents and ruminal fermentation in beef cattle. Four Limousin${\times}$LuXi crossbred cattle with a body weight ($400{\pm}10kg$), fitted with permanent ruminal, proximal duodenal and terminal ileal cannulas, were used in a $4{\times}4$ Latin square design with four treatments: Control (100% corn), 33% wheat (33% substitution for corn), 67% wheat (67% substitution for corn), and 100% wheat (100% substitution for corn) on a dry matter basis. The results showed that replacing corn with increasing amounts of wheat increased the apparent digestibility values of dry matter, organic matter, and crude protein (p<0.05). While the apparent digestibility of acid detergent fiber and neutral detergent fiber were lower with increasing amounts of wheat. Digestive enzyme activities of lipase, protease and amylase in the duodenum were higher with increasing wheat amounts (p<0.05), and showed similar results to those for the enzymes in the ileum except for amylase. Increased substitution of wheat for corn increased the serum alanine aminotransferase concentration (p<0.05). Ruminal pH was not different between those given only corn and those given 33% wheat. Increasing the substitution of wheat for corn increased the molar proportion of acetate and tended to increase the acetate-to-propionate ratio. Cattle fed 100% wheat tended to have the lowest ruminal $NH_3-N$ concentration compared with control (p<0.05), whereas no differences were observed among the cattle fed 33% and 67% wheat. These findings indicate that wheat can be effectively used to replace corn in moderate amounts to meet the energy and fiber requirements of beef cattle.

Evaluation of Coarsely Ground Wheat as a Replacement for Ground Corn in the Diets of Lactating Dairy Cows

  • Eight multiparous Holstein cows ($569{\pm}47$ kg of BW; $84{\pm}17$ DIM) were used to evaluate the effects of different levels of coarsely ground wheat (CGW) as replacements for ground corn (GC) in diets on feed intake and digestion, ruminal fermentation, lactation performance, and plasma metabolites profiles in dairy cows. The cows were settled in a replicated $4{\times}4$ Latin square design with 3-wk treatment periods; four cows in one of the replicates were fitted with rumen cannulas. The four diets contained 0, 9.6, 19.2, and 28.8% CGW and 27.9, 19.2, 9.6, and 0% GC on dry matter (DM) basis, respectively. Increasing dietary levels of CGW, daily DM intake tended to increase quadratically (p = 0.07); however, apparent digestibility of neutral detergent fiber (NDF) and acid detergent fiber (ADF) were significantly decreased (p<0.01) in cows fed the 28.8% CGW diets. Ruminal pH remained in the normal physiological range for all dietary treatments at all times, except for the 28.8% CGW diets at 6 h after feeding; moreover, increasing dietary levels of CGW, the daily mean ruminal pH decreased linearly (p = 0.01). Increasing the dietary levels of CGW resulted in a linear increase in ruminal propionate (p<0.01) and ammonia nitrogen ($NH_3$-N) (p = 0.06) concentration, while ruminal acetate: propionate decreased linearly (p = 0.03) in cows fed the 28.8% CGW diets. Milk production was not affected by diets; however, percentage and yield of milk fat decreased linearly (p = 0.02) when the level of CGW was increased. With increasing levels of dietary CGW, concentrations of plasma beta-hydroxybutyric acid (BHBA) (p = 0.07) and cholesterol (p<0.01) decreased linearly, whereas plasma glucose (p = 0.08), insulin (p = 0.02) and urea nitrogen (p = 0.02) increased linearly at 6 h after the morning feeding. Our results indicate that CGW is a suitable substitute for GC in the diets of dairy cows and that it may be included up to a level of 19.2% of DM without adverse effects on feed intake and digestion, ruminal fermentation, lactation performance, and plasma metabolites if the cows are fed fiber-sufficient diets.

활성탄 및 목탄의 첨가가 산양의 영양소 이용율 및 반추위내 발효성상에 미치는 영향 (Effects of Activated Carbon and Charcoal on the Nutrients Utilization and Ruminal Fermentation Characteristics in Goat)

  • 본 시험은 활성탄 및 목탄의 첨가가 사료의 소화율, 반추위내 pH 발효성상에 미치는 영향을 조사하기 위하여 실시하였다. 한국재래산양에 대한 급여 실험을 실시하였는데, 활성탄과 목탄의 첨가수준은 1.0% 수준이었으며, 조사료/농후사료 비율은 오차드그래스 건초와 농후사료를 2:8로 하였다. 시험에 공시된 동물은 대사케이지에 수용하여 매일 09시에 사료를 급여하였다. 시험 결과를 요약하면 건물 소화율은 활성탄 및 목탄 첨가구가 무첨가구에 비하여 유의한 결과는 아니었지만 높은 경향을 보였다. 그리고 단백질의 소화율은 활성탄 및 목탄 첨가구가 무첨가구에 비하여 유의하게 높은 결과(p<0.05)를 나타내었다. 그러나 조지방, NDF, ADF 및 hemicellulose의 소화율은 유의한 증가를 나타내지 않았다. 활성탄구와 목탄구 사이에도 유의한 차이를 나타내지 않았다. pH와 반추위내 암모니아의 농도는 활성탄구와 목탄구가 무첨가구보다 다소 높은 경향을 보였으며, 총VFA 농도는 무첨가구에서 유의하게 높게 나타났다. 그리고 $C_2/C_3$ 비율을 보면 활성탄구는 무처리구에 비하여 유의하게 낮은 결과(p<0.05)를 나타내었고, 목탄구도 무처리구보다는 낮은 결과였으나 유의성은 인정되지 않았다. 위 결과에서 보듯이 활성탄은 가축의 생산성 향상에 유리한 조건을 제공하는 경향이 있다고 보여진다. 그러나 이미 보고된 바와 같이 시험 조건에 따라 상반되는 결과를 보이기도 한다. 이것이 외부 환경요인이 사료처리 영향을 압도하는 것인지, 또는 어느 제한된 조건하에서만 활성탄의 영향이 발현되는가에 대하여 추가연구가 요구된다고 하겠다. 그리고 여기에 대한 경제성 여부도 검토되어져야 할 것으로 사료된다.

Influence of an Anaerobic Fungal Culture (Orpinomyces sp.) Administration on Growth Rate, Ruminal Fermentation and Nutrient Digestion in Calves

  • The study was to see the effect of administration of ruminal fungi on feed intake, growth rate, rumen fermentation and nutrient digestion of calves (Tharparkar$\times$Holstein-Friesian, average age: 10 months, average body weight: 130 kg). The 6 calves in first group were fed a mixture consisted of 50% wheat straw and 50% concentrate (Maize 62%, Groundnut cake 35%, Mineral mix. 2% and Common salt 1%) along with 1 kg green oats $animal^{-1}$ $day^{-1}$ while second group calves were fed the above-mentioned diet in addition to a dose of 160 ml ($10^{6}$ CFU/ml) fungal culture $calf^{-1}$ $week^{-1}$. The average dry matter intake per day was slightly lowered in fungal fed calves yet feed conversion ratio was higher. The average daily weight gain was significantly higher (15.37%) in fungal administered group as compared to control. The nutrient digestibility was increased for crude fibre, NDF and ADF with fungal administration. Digestible energy value of straw-based diet in terms of percent TDN also increased. The pH and $NH_{3}$-N were lower whereas TVFA, total-N, TCA-N and number of zoospores were higher in rumen liquor in fungal administered group.

Effect of Unsaturated Fatty Acids on Cellulose Degradation and Fermentation Characteristics by Mixed Ruminal Microbes

  • This experiment was conducted to evaluate the effects of supplemental unsaturated fatty acids (UFA) on fermentation characteristics, especially on gas production, cellulose degradation and volatile fatty acid (VFA) concentration by mixed ruminal microorganisms. In order to attain this objective, unsaturated fatty acids including oleic acid (C 18:1), linoleic acid (C18:2) and arachidonic acid (C22:4) were added at varying level. Mixed ruminal microbes used in this experiment were obtained from the rumen of a cannulated Holstein cow. Medium pH values after 7 d incubation were significantly affected by type and level of unsaturated fatty acids (p<0.01). All of UFA inhibited total gas production, and especially treatment of arachidonic acid at the levels of 0.01% gave the lowest gas. production after 7 d incubation (p<0.01). Comparison of the population of protozoa revealed that UFA did not have any significant effect on the total protozoa number. The addition of UFA did not effect dry matter degradation. Volatile fatty acid (VFA) composition of the culture was influenced little by UFA, although the considerable amount of iso-type VFA were detected in UFA supplemented incubations. The ratio of acetic acids to propionic acids, however, was lower than control in all the treatments after 7 d incubation (p<0.01).
