Stringent Factor Regulates Antibiotics Production and Morphological Differentiation of Streptomyces clavuligerus

  • The involvement of the relA and rsh genes in the morphological and physiological differentiation of Streptomyces clavuligerus was evaluated with the relA and rsh genes mutants. The morphological differentiation of S. clavuligerus was greatly affected by the disruption of the relA gene, but not very much by the disruption of the rsh gene. The altered morphological characteristics were completely restored by the complementation of the corresponding disrupted genes. Thus, it was apparent that the mycelial morphology and clavulanic acid production were severely affected by the disruption of the relA gene. Production of clavulanic acid in the submerged batch culture and glycerol-limited chemostat showed that production was inversely related to the specific growth rate in the wild-type strain. However, the production of clavulanic acid in the ${\Delta}relA$ and ${\Delta}rsh$ null mutants was completely abolished. Therefore, it seems plausible that the stringent response of S. clavuligerus to starvation for amino acids is governed mainly by ReIA, rather than Rsh, and that the (p)ppGpp synthesized immediately after the depletion of amino acids triggers the initiation of pathways for both morphological and physiological differentiation in this species.

  • RSH (relA/spoT homolog) has been known to determine the level of guanosine tetraphosphate (ppGpp) and guanosine pentaphosphate (pppGpp), which are the effector nucleotide of the prokaryotic stringent response and also play a role in antibiotic production and differentiation in Streptomyces species but not a little in eukaryotic organism, especially in plant. Salicylic acid (SA), a critical signal molecule of establishing systemic acquired resistance (SAR), could induce SAR in Pepper (Capcicum annuum) against Phytophthora capsici. And the extent of SAR induction was in proportion to the dosage of SA (or BTH). Suppression subtractive hybridization (SSH), a PCR-based method for cDNA subtraction, was carried out between SA-treated and non-SA-treated pepper leaves to isolate genes which may be responsible for defense signaling against pathogens. Early upregulated gene was selected from reverse northern and kinetics of SSH-genes transcripts in SA-treated pepper leaves upon SA treatment. Full-length cDNA of the gene (PepRSH; Pepper RelA / SpoT homolog) had an open reading frame (ORF) of 2166 bp encoding a protein of 722 amino acids and a significant homology with (p)ppGpp phosphohydrolase or synthetase. Genomic DNA gel blot analysis showed that pepper genome has at least single copy of PepRSH. PepRSH transcripts was very low in untreated pepper leaves but strongly induced by SA and methyljasmonic acid (MeJA), indicating that PepRSH may share common SA and MeJA-mediated signal transduction pathway Functional analysis in E. coli showed PepRSH confers phenotypes associated with (p)ppGpp synthesis through a complementation using active site mutagenesis.

An Approach for Identifying the Temperature of Inductance Motors by Estimating the Rotor Slot Harmonic Based on Model Predictive Control

  • In order to satisfy the urgent requirements for the overheating protection of induction motors, an approach that can be used to identify motor temperature has been proposed based on the rotor slots harmonic (RSH) in this paper. One method to accomplish this is to improve the calculation efficiency of the RSH by predicting the stator winding distribution harmonic order by analyzing the harmonics spectrum. Another approach is to increase the identification accuracy of the RSH by suppressing the influence of voltage flashes or current surges during temperature estimation based on model predictive control (MPC). First, an analytical expression of the stator inductance is extracted from a steady-state positive sequence motor equivalent circuit model developed from the rotor flux field orientation. Then a procedure that applies MPC for reducing the identification error of the rotor temperature caused by voltage sag or swell of the power system is given. Due to this work, the efficiency and accuracy of the RSH have been significantly improved and validated our experiments. This work can serves as a reference for the on-line temperature monitoring and overheating protection of an induction motor.

Chemical Quality Evaluation of Commercial Infant Formulas (시판 조제 분유의 화학적 품질 평가)

  • In this study, three domestic and one foreign formulas for the infants up to 5 month old were examined to detect chemical changes such as pH, reactive sulfhydryl groups(RSH) content, 5-hydroxymethylfurfral(HMF) content, available lysine content, electrophoresis, and surface color caused by heat treatment for long term storage. In the SDS-polyacrylamide gel electrophoresis, A and B products showed similar pattern, while C product had a clearly distinguishable $\beta$-lacto-globulin band, but in casein, only D product showed a few strong casein band. RSH content, which indicate the extent of whey protein denaturation, ranged from 4.40 to 5.93 mmole/g protein. HMF content. which indicate the extent of Maillard reaction, ranged from 192 to 432 $\mu$mole/100g in formulas. B product showed the highest RSH and HMF content. Available lysine content ranged from 31 to 46 mg/g protein. Among them D product contain the highest available lysine content and others showed no significant difference. In conclusion, the domestic infant formulas showed higher RSH and HMF content than the foreign product and the available lysine content of the domestic products were lower than of the foreign product.

  • Rectus sheath hematoma (RSH) is an uncommon but well-documented clinical condition. It is usually caused by direct trauma or anticoagulation, although there are many other causes. However, RSH after noncontact strenuous exercise is very rare. We present a rare case of RSH after playing volleyball without direct trauma that was successfully treated by angiographic embolization.

Speed Estimation of Induction Motor in Steady State Using the RSH (RSH를 이용한 정상상태 운전 유도전동기의 회전속도 추정)

  • The slip frequency is included in feature frequency for fault diagnosis of rotor bar, so rotating rotor speed is needed. In this study, rotor slot harmonic(RSH) method is suggested for speed estimation of induction motor. When the rotor is rotating, motor current signal include the harmonic signal of back-emf voltage related with number of rotor slot. So from the power spectrum of current signal, the rotor speed can be founded. This method of rotor speed estimation gives the slip frequency, and the feature frequency of rotor bar fault can be calculated. Comparing with stroboscope speed meter, the error rate of suggested method is less than 0.1[%].

Differential Stringent Responses of Streptomyces coelicolor M600 to Starvation of Specific Nutrients

  • This study focused on the involvement of the unusual nucleotide (p)ppGpp, a stringent factor, during the morphological and physiological differentiation of Streptomyces coelicolor. Two genes, relA and rshA, were disrupted to demonstrate the roles of the stringent factor in the differentiation. The intracellular concentration of (p)ppGpp in the wild-type (M600) and disrupted mutants was measured in relation to the intentional starvation of a specific nutrient, such as carbon, nitrogen, and phosphate or the in situ depletion of nutrients in a batch culture. As a result, it was found that the morphological characteristic of the ${\Delta}relA$ mutant was a bld phenotype forming condensed mycelia, whereas the ${\Delta}rshA$ mutant grew fast-forming spores and straightforward mycelia. In both mutants, the production of actinorhodin (Act) was completely abolished, yet the undecylprodigiosin (Red) production was increased. Intracellular (p)ppGpp was detected in the ${\Delta}relA$ mutant in the case of limited phosphate, yet not with limited carbon or nitrogen sources. In contrast, (p)ppGpp was produced in the ${\Delta}rshA$ mutant under limited carbon and nitrogen conditions. Therefore, (p)ppGpp in S. coelicolor was found to be selectively regulated by either the RelA or RshA protein, which was differentially expressed in response to the specific nutrient limitation. These results were also supported by the in situ ppGpp production during a batch culture. Furthermore, it is suggested that RelA and RshA are bifunctional proteins that possess the ability to both synthesize and hydrolyze (p)ppGpp.

Bending Stress에 따른 Ag 및 Al 금속전극의 저항 변화에 관한 연구

  • OLED 소자가 소형화됨에 따라 Flexible display를 넘어 Foldable display를 연구 중이며 동시에 신뢰성 및 수명이 중요시 되고 있다. 따라서 본 연구에서는 신뢰성 및 수명 평가에 대한 한 가지 방법으로 Bending test를 이용하여 소자의 Resistivity 변화를 측정하여 소자의 신뢰성을 확인 하여 보았다. Flexible substrate위에 Ag와 Al을 Cross bridge structure로 각각 증착한 후 bending 시간에 따른 Sheet resistance (Rsh)와 Resistivity (비저항)을 분석 하였다. 100시간 동안의 bending test결과 Ag전극의 Rsh는 $0.104{\Omega}$에서 $0.098{\Omega}$으로 5.67% 감소하였고 비저항은 5.70% 감소하였다. Al전극의 Rsh는 $0.091{\Omega}$에서 $0.063{\Omega}$으로 30.4% 감소하였고 비저항은 30.3% 감소하였다. Foldable에서는 저항 변화가 크게 되면 접히는 부분의 흐르는 전류가 많아지게 되어 소자의 저하를 발생시킨다. 저항변화가 거의 없다는 것은 물질의 안정성이 좋다고 할 수 있다. 실험 결과 Ag의 저항 변화가 Al보다 작으므로 Ag가 Flexible 관련 물질로 더 유용하다는 것을 확인 할 수 있다.

Thermal Degradation of BZO Layer on the CIGS Solar Cells

  • We investigated a study on the thermal degradation of boron doped zinc-oxide (BZO) layer which used as a transparent conducting layer on the Cu (In1-xGax) Se2 (CIGS) based thin film solar cells. Devices were annealed under the temperature of $100^{\circ}C$ or 100 hours and then Hall measurement was carried out to characterize the parameters of mobility (${\mu}Hall$), resistivity (${\rho}$), conductivity (${\sigma}$) and sheet resistance (Rsh). The initial values of ${\mu}Hall$, ${\rho}$, ${\sigma}$ and Rsh were $29.3cm^2$/$V{\cdot}s$, $2.1{\times}10^{-3}{\Omega}{\cdot}cm$, $476.4{\Omega}^{-1}{\cdot}cm^{-1}$ and $19.1{\Omega}$/${\Box}$ respectively. After the annealing process, the values were $4.5cm^2$/$V{\cdot}s$, $12.8{\times}10^{-3}{\Omega}{\cdot}cm$, $77.9{\Omega}^{-1}{\cdot}cm^{-1}$ and $116.6{\Omega}$/${\Box}$ respectively. We observed that ${\mu}Hall$ and ${\sigma}$ were decreased, and ${\rho}$ and Rsh were increased. In this study, BZO layer plays an important role of conducting path for electrons generated by incident light onthe CIGS absorption layer. Therefore, the degradation of BZO layer characterized by the parameters of ${\mu}Hall$, ${\rho}$, ${\sigma}$ and Rsh, affect to the cell efficiency.

  • Rectus sheath hematoma (RSH) is a rare condition caused by hemorrhage into the rectus sheath. It is usually associated with severe cough, abdominal surgery, coagulopathy, and anticoagulation treatment. RSH can be difficult to diagnose and can be misdiagnosed as acute appendicitis, as diverticulitis, or as an ovarian mass. Although RSH usually presents as a benign condition, it can be life threatening, especially in the critically-ill patient. Here, we report a case of fatal RSH due to hypovolemic shock in a critically-ill 73-year-old woman, who had received heparin treatment due to acute myocardial infarction in the intensive care unit and who had been successfully treated by conservative management.
