  • The purpose of this study was to investigate the extraction yield and quality stability as to the oleoresin process with large amount of onion at one time. The first mixed-product is raw onion juice which was reduced the compression and concentrated by Brix 70% mixed together wit the residue which was extracted and concentrated by ethanol, the second product manufactured by the same method above after the autoclaving with onion, and the other product is made by grinding by 50mesh to freeze-dried onion. Each of yields were 7.3, 9.1 and 0.8% and each of total sugar content was 616.4, 712.3 and 150.3mg/g. Therefore the product extracted by ethanol from freeze-dried onion was very low in yield and total sugar content. By the index of the overall odor intensity, contents of total pyruvate were 1,733.7, 520.6, and 2,716.5$\mu\textrm{g}$/g for each product. As a result, oleoresin onion processing that desired to use raw onion was remarkable for odor recovery. For the homogenous mixture with concentrate of onion juice and ethanol extract were emulsified by the addition of 2% of PGDR(polyglycerol condensed ricinoleate) and agitation(10,000rpm, 30 minutes). At this time, interfacial tension was 1.9 dyne/cm and the formation of emulsion was for 96.2% when left over 24hours in 6$0^{\circ}C$. When it was to be centrifuged(2,000$\times$G, 80 minutes) after emulsification, the volume of emulsion level without seperation was 92.6%, and very high in emulsification stability. The induced heating-oxidize with soy bean oil and sesame oil added to 1% of onion oleoresin, induction-time extension effect appeared with antioxidant activity that was applicable for 80.8~82.2% as to the effect of addition of 0.02% BHA.

  • The study aimed to investigate in vitro lipid deposition of oleic acid, oleic acid methyl ester and cholesterol on a daily disposable (1-day lenses) and 3-days lenses over 3 days and changes of optical characteristics is also investigated. Artificial tear solutions were prepared to simulate actual tear compositions. Two types of contact lenses (1-day lenses (Senofilcon A) and 3-days lenses (silicone tripolymer)) were soaked in the artificial tear solutions within an incubator at 37 ℃ with 150 rpm for 8, 16, 24 hours. Lipid deposition (oleic acid, oleic acid methyl ester and cholesterol) were measured using high performance liquid chromatography (HPLC) instrument. In addition, measurements of oxygen transmissibility, light transmittance and observation of lens surface were conducted. The amount of lipid deposition on the 1-day lenses were 127.55 ㎍/lens for Day 1, 302.96 ㎍/lens, for Day 2, and 353.30 ㎍/lens for Day 3. The 3-days lenses were 46.22 ㎍/lens for Day 1, 66.07 ㎍/lens for Day 2, and 67.45 ㎍/lens for Day 3. Oxygen transmissibility were 81×10-9(cm/sec)(ml O2/ml×mmHg)(Baseline) and 48×10-9(cm/sec)(ml O2/ml×mmHg) (Day 3) for the 1-day lenses, it were 13.23×10-9(cm/sec)(ml O2/ml×mmHg)(Baseline) and 9.6×10-9(cm/sec)(ml O2/ml×mmHg) (Day 3) for the 3-days lenses. Transmittance of each lenses were 97.21% (Baseline) and 94.25% (Day 3) for the 1-day lenses, 97.65% (Baseline) and 95.15% (Day 3) for the 3-days lenses. Observation of surface deposition indicated greatest deposition for the 3-days lenses type on Day 3. Lipid deposition for both lens types increased by day and was greater for the 1-day lenses type. Surface deposition appeared to differ as it was greatest for the 3 days lens type, which may suggest other deposits such as protein may be present.

  • Recently, the studies about rock physics model (RPM) in shale reservoir are widely performed. In shale reservoir, the degree of the maturity can be estimated by kerogen and GOR (Gas-Oil Ratio). The researches on the rock physics model of shale reservoir with the amount of kerogen have been actively carried out but not with GOR. Thus, in this study, we analyzed the changes in seismic velocity and density, and AVO (Amplitude Variation with Offset) response depending on changes in GOR and the amount of kerogen. Since the shale consists of plate-like particles, it has vertical transverse isotropy (VTI). Therefore we estimated the seismic velocity and density by using Backus averaging method and analyzed AVO responses based on these estimated properties. The results of analysis showed that the changes in the velocity with the GOR variation are small but the velocity changes with the variation in kerogen amount are relatively larger. In case, GOR 180 (Litre/Litre) which is boundary between heavy oil and light oil, when volume fraction of kerogen increased from 5% to 35%, the P-wave velocity normal to the layering increased 51%. That is, it helps estimating maturity of kerogen through the velocity. Meanwhile, when rates of oil-gas mixture are large, the effect of GOR variation on the velocity change became larger. In case volume fraction of kerogen is 5%, the P-wave velocity normal to the layering was estimated $1.46km/s^2$ in heavy oil (GOR 40) but $1.36km/s^2$ in light oil (GOR 300). The AVO responses analysis showed class 4 regardless of the GOR and amount of kerogen because variation of poisson's ratio is small. Therefore, shale reservoir has possibility to have class 4.

  • Purpose Estradiol (E2) is a steroid hormone mainly produced in women and is a useful indicator for diagnosis of gynecological diseases, menstrual cycle, menopause, and precocious puberty. E2 measurement is performed by diluting the $^{125}I$ radioactive tracer and tracer buffer in the kit. However, It was not precisely specified when the period of tracer is available after activating. The purpose of this study was to determine the appropriate dilution time based on the measurement value with dilution time. Materials and Methods From December 2016 to February 2017, 60 E2 samples with concentrations ranging from 8 to 4577 pg/mL were divided into low, medium, and high concentrations. Dilution of the $^{125}I$ tracer was performed on a 230 RPM agitator for 30 minutes, 1 hour 30 minutes, and 2 hours 30 minutes, respectively. 24 hour dilution was gently shaken and refrigerated. To verify the difference and significance of the results according to the dilution time, a test of normality was performed using SPSS 18.0 and analyzed by Kruskal-Wallis test. The measured value according to the dilution time was compared with the interquartile range of the absolute error. Results The results of Kruskal-Wallis test were not significant (P>0.05). Measurement results are showed as interquartile range of absolute error. At low concentration, it is 0.052 between 1 hour 30 minutes and 2 hours 30 minutes, and 0.105 between 30 minutes and 1 hour 30 minutes. At medium concentration, 0.062 between 30 minutes and 1 hour 30 minutes, and 0.038 between 1 hour 30 minutes and 2 hours 30 minutes. At high concentration, it is 0.029 between 1 hour 30 minutes and 2 hours 30 minutes, and 0.06 between 2 hours 30 minutes and 24 hours. Conclusion There were no statistically significant differences. However, the change in the measured value is the smallest between 1 hour and 30 minutes to 2 hours and 30 minutes. Therefore, we recommend diluting time between 1 hour 30 minutes and 2 hours 30 minutes.

  • This is an experimental study on friction welding and heat treatment of engine exhaust valve materials whose welding combination is SCr4 as stem to 21-4N as head and SUH3 to 21-4N. In this study, not only the experiments of friction welding under the selected optimum welding condition and the examination of the mechanical properties were carried out, but also the heat treatment of friction welded specimens under the two selected conditions was taken to obtain the better welding character, eliminating the latent stress and the hardness peak which appeared at the welded zones of heat resisting steel(21-4N, SUH3) and low alloyed steel ($SCr_4$) friction weldments. The results obtained by the experiments and consideration in this study are as follows: I) It was experimentally proved quite reasonable that 'speed=3,OOO rpm, heating pressure Pl=8 kg/ mm2, upsetting pressure p, = 20 kg/mm', heating time $t_1$ = 3 see, upsetting time TEX>$t_2$ = 2.5 sec' was selected as the optimum welding condition for friction-welding the engine exhaust valve materials $SCr_4$ to 21-4 Nand SUH 3 to 21-4 N. 2) The results of the previous study and this one on friction welding of such dissimilar materials as SUH 3-SUH 31, SCr 4-SUH 31, SCr 4-SUH 3, SUH 3-CRK 22, SCr4-21-4 Nand SUH3-21-4 N agreed with each other substantially in the friction welding characteristics at welded interface zones. 3) It was also certified quite satisfactory that '600\ulcornerCX30 min. Xroom air cooling' as an optimum heat treatment condition of the friction welded materials SCr 4-21-4 Nand SUH 3-21-4 N was experimentally determined to eliminate the latent stress and the hardness peak at welded zones. 4) About 20% of the tensile strength before heat treatment of friction welded specimens was decreased after heat treatment 600\ulcornerCX30 min. Xair cooling, but the location of fracture was moved from heat affected zone to parent $SCr_4$ & SUH3. 5) Microscopic examination of the weld joints friction-welded and heat-treated under the above mentioned conditions revealed that the weld zone is very narrow and has a fine grained intermixed structure without any welding defects. 6) The above mentioned conditions can be also utilized as friction welding parameters of the other dissimiar materials for engine valve production.

  • Specific carotenoids and astaxanthin biosynthesis power of Phaffia rhodozyma mutant 876, which was obtained after NTG a and UV treatments, was higher than those of the wild type by 40% and 50%, respectively. The mutant strain did not show t the catabolite repression even at 22% (w/v) glucose concentration. The optimum C{N ratio was 2.0, and the optimum t temperature and initial pH were $22^{\circ}C$ and 6.0, respectively. 80th cell growth and astaxanthin formation decreased drastically a as the fermentation temperature was increased over $22^{\circ}C$, whereas they were comparable in the pH range between 5.0 and 7 7.0. Inoculum size did not affect the final cell density nor the carotenoids biosynthesis, and 3%(v/v) was selected as optimal. H Higher dissolved oxygen concentration facilitated astaxanthin biosynthesis, and aeration rate of 1.0 v/0/m and agitation speed of 400 rpm were selected as optimum. The final cell dens때 of 43.3 g/L and the volumetric astaxanthin and carotenoids concentrations of 110.6 mg/L and 149.4 mg/L, respectively, were obtained. The specific carotenoids concentration was 3.45 m mg{g-yeast(dry). Yx/s and Yp/s values of 0.37 and 1.08 were obtained. • In order to remediate heavy metal contaminated Nong island, Maehyang-ri shooting range soils through the batch reactor scale washing were evaluated. The experiment texture soil of N3 in the Nong island at north side incline was (g)mS containing 12.9% gravel, 47.0% sand, 35.1% silt and 5.0% clay. And the N3 soil area was contaminated with Cd($22.5\pm1.9$ ppm), Cu($35.5\pm4.0$ ppm), Pb($1,279.0\pm5.1$ ppm) and Zn($403.4\pm9.8$ ppm). The EDTA(ethylene diamine tetra acetic acid, $C_{10}H_{16}N_2O_8$) in the N3 soil was observed as most effective extractants among the 5 extractants(citric acid, EDTA, phosphoric acid, potassium phosphate and oxalic acid) tested. And chemical partitioning of heavy metals after washing N3 soil with EDTA was evaluated. Removal efficiency of residual fractions was higher than that of non-residual fractions. To choose EDTA extractant which is the most effective in soil washing technology using batch reactor process cleaning Pb and Zn contaminated sits; Pb and Zn removal rates were investigated 92.4%, 94.0% removal(1,000 mM, soil:solution=5, $20^{\circ}C$, 24 hour shaking, pH=2, 200 RPM), respectively. The results of the batch test showed that the removal efficiency curve was logarithmic in soil was removal. Thus, EDTA washing process can be applied to remediate the Pb and Zn contaminated soil used in this study.
