Induction of Heme Oxygenase-1 by Traditional Herb Mix Extract Improves MKN-74 Cell Survival and Reduces Stomach Bleeding in Rats by Ethanol and Aspirin in vivo

  • Chinese herb medicines have traditionally been used to treat or alleviate the symptom of various diseases. The rationale for use of certain herbs to certain disorder is now getting unveiled by modern technology. In the present study, we investigated whether herb mix extract(HMX), which is alleged to be useful for gastric ulcer, protects stomach from oxidative stress. Rats were allowed to normal diet with and without HMX (1, 5, 10 mg/kg) for 30 days. To induce gastric ulcer, ethanol (75%, 1.5 ml) or acidified aspirin (100 mg/kg in 0.2 N HCl) was administered by oral route in 24 h-fasted rats and examined the gastric ulceration(bleeding) by measuring the size 1 h after the treatment. Results indicated the area of gastric bleeding was significantly less in HMX fed rats than in normal diet fed ones, and it was dependent on the duration and amount of HMX. To investigate the underlying mechanism by which HMX protects stomach from oxidative stress, expression of enzymes like heme oxygenase (HO), cyclooxygenase (COX), and inducible nitric oxide (iNOS) were investigated in MKN-74 cells, where aspirin or H. pylori was introduced. The results were compared with RAW 264.7 cells to check if there's cell specificities exist. The expression of HO-1 but not COX-2, iNOS was significantly increased by HMX. Furthermore, HO-1 inhibitor, SnPP IX reduced the HO-1 activity and reversed the survival rate in HMX-treated MKN-74 cells. There's no difference between RAW 264.7 cells and MKN-74 cells. We, thus, concluded that HMX is beneficial for protection from oxidative injury, and induction of HO-1 by HMX in gastric cells is, at least, responsible for protection from oxidative stress such as ethanol, aspirin and possibly H. pylori infection.

The Relationship among Marital Dissatisfaction, Loneliness and SNS Addiction - The Moderated Mediating Effect of Social Capital - (결혼불만족과 외로움, SNS중독의 관계 - 사회자본의 조절된 매개효과를 중심으로 -)

  • This study was carried out to examine whether the moderated mediating effect of SNS addiction resulting from marital dissatisfaction and loneliness is influenced by gender or size and composition of social capital. 714 married men and women in their 20's to 40's were participated in this study. The data were collected by online research center with quota sampling based on demographical proportion. To analyze this research model, the moderated mediating effect utilizing structural equation was used. The results of this study were summarized as follows. First, it is verified that the loneliness moderated between marital dissatisfaction and SNS addiction. Since the relationship between independent and dependent variable is insignificant, comparison of competition model was able to determine if mediating effect was fully or partially shown. As a result, the complete mediated model was ultimately chosen because its various goodness-of-fit index including $x^2$ performed fairly well. Second, the mediating effect which moderates social capital size was presented in the mediated model between the marital dissatisfaction, loneliness and SNS addiction. Specifically, It implies that group who recognizes their social capital big recognizes the route and effect size more than that who considers their social capital small. And the moderated mediating effect depending on different type of social capital among the marital dissatisfaction and loneliness and between SNS addiction was substantiated. It implies that disequilibration of SNS social capital in face-to-face sphere and in online acted as a danger fact with respect to the marital dissatisfaction, loneliness and SNS addiction. The implications of these findings were discussed, and directions for future studies were also proposed.

Tissue Distribution and Toxicokinetics of 4-Tert-Octylphenol in Rats (4-Tert-Octylphenol의 랫드에서의 조직분포 및 독성동태에 관한 연구)

  • 4-Tert-Octylphenol (OP) is a surfactant additive widely used in the manufacture of a variety of detergents and plastic products. OP can disrupt endocrine function in humans and animals. This study was carried out to obtain toxicokinetic parameters of OP in male Sprague-Dawley (SD) rats. Male rats were administered with OP by single oral application of 200 mg/kg body weight. Blood, urine and tissues samples were taken at several time intervals after administration. Analysis of samples for OP was performed by column-switching high performance liquid chromatography (HPLC). In addition, we exam-ined tissue distribution and accumulation of OP after single oral application of 50, 100, and 200 mg/kg, single intravenous injection of 1, 5 and 10 mg/kg or daily application of 50 mg/kg for 14 consecutive days. After single oral administration of 200 mg/kg, Cmax of 213 $\pm$ 123 ng/ml was reached within the first 1.3 hr (Tmax) in the plasma. AUC was calculated for 1,333$\pm$484 ngㆍhr/ml. The final elimination half-life of plasma was longer than that of urine, but urinary clearance was lower than oral. A very small fraction of OP (Fe < 0.0017%) was excreted in urine within 24 hr. These results indicated that the major excretion route of OP was not urine. The mean maximal tissue distribution of OP was obserbed at 6 hr after treatment and slowly decreased time-dependently. High OP concentrations were detected in fat at 24 hr. The OP in fat was slowly released with longer elimination half-life and lower clearance than that of other tissues. OP was not accumulated in the liver following single oral application but 14-day oral treatments resulted in two-fold accumulation. It was probably due to the saturation of detoxification pathways. On the other hand, the mRNA expression of cytochrome P450 isoforms except CYP2C11 was not affected by OP at any dose. The expression of CYP2C11 mRNA decreased in a dose-dependent manner. This result suggests that OP changes expression of the male-specific cytochrome P450 isoforms in rat liver, and these changes are closely related to the toxic and estrogenic effect of OP.

Placenta Transfer and Toxicokinetics of Valproic Acid in Pregnant Cynomolgus Monkeys

  • Placenta transfer study in non-human primate (NHP) is one of the crucial components in the assessment of developmental toxicity because of the similarity between NHP and humans. To establish the method to determine placenta transfer in non-human primate, toxicokinetics of valproic acid (VPA), a drug used to treat epilepsy in pregnant women, were determined in pregnant cynomolgus monkeys. After mating, pregnancy-proven females were daily administered with VPA at dose levels of 0, 20, 60 and 180 mg/kg by oral route during the organogenesis period from gestation day (GD) 20 to 50. Concentrations of VPA and its metabolite, 4-ene-VPA, in maternal plasma on GDs 20 and 50, and concentrations of VPA and 4-ene-VPA in placenta, amniotic fluid and fetus on GD 50 were analyzed using LC/MS/MS. Following single oral administration of VPA to pregnant monkeys, concentrations of VPA and 4-ene-VPA were generally quantifiable in the plasma from all treatment groups up to 4-24 hours post-dose, demonstrating that VPA was absorbed and the monkeys were systemically exposed to VPA and 4-ene-VPA. After repeated administration of VPA to the monkeys, VPA was detected in amniotic fluid, placenta and fetus from all treatment groups, demonstrating that VPA was transferred via placenta and the fetus was exposed to VPA, and the exposures were increased with increasing dose. Concentrations of 4-ene-VPA in amniotic fluid and fetus were below the limit of quantification, but small amount of 4-ene-VPA was detected in placenta. In conclusion, pregnant monkeys were exposed to VPA and 4-ene-VPA after oral administration of VPA at dose levels of 20, 60 and 180 mg/kg during the organogenesis period. VPA was transferred via placenta and the fetus was exposed to VPA with dose-dependent exposure. The metabolite, 4-ene VPA, was not detected in both amniotic fluid and fetus, but small amount of 4-ene-VPA was detected in placenta. These results demonstrated that proper procedures to investigate placenta transfer in NHP, such as mating and diagnosis of pregnancy via examining gestational sac with ultrasonography, collection of amniotic fluid, placenta and fetus after Caesarean section followed by adequate bioanalysis and toxicokinetic analysis, were established in this study using cynomolugus monkeys.

Gastrointestinal and Hepatic First-pass Effects of Triflusal in Rats (흰쥐에서 트리플루살의 위장관 및 간 초회통과효과)

  • In order to elucidate the influence of intestinal and hepatic first-pass effect on the pharmacokinetics of triflusal, the biotransformation of triflusal in the gastrointestinal tract and liver was designed. Moreover, we tried to establish an HPLC method applicable for bioassay and available to pharmacokinetics, not only with the simultaneous determination of triflusal and its active metabolite, 2-hydroxy-4-trifluoromethyl benzoic acid (HTB), but also with improving sensitivity. After the administration of triflusal (10 mg/kg) and HTB (10 mg/kg) into femoral vein, portal vein (only triflusal) and oral route (only triflusal), pharmacokinetic parameters were investigated from the plasma concentration-time profiles of triflusal and HTB in rats. An HPLC method was developed for the simultaneous determination of triflusal and HTB in rat plasma, urine and bile. The HPLC analysis was carried out using a C18 column and acetonitrile-methanol-water (25:10:65, v/v/v) as the mobile phase and UV detection at 234 nm. Furosemide was used as the internal standard. The calibration curves were linear over the concentration range $0.05-5.0\;{\mu}g/ml$ for triflusal and $0.2-200.0\;{\mu}g/ml$ for HTB with correlation coefficients greater than 0.999 and with intra-day or inter-day coefficients of variation not exceeding 10.0%. This assay procedure was applied to the study of metabolite pharmacokinetics of triflusal and HTB in rats. It was supposed that triflusal was almost metabolized in vivo because urinary and biliary excreted amounts of triflusal could be ignored as it was lower than 1.2% of the administered dose. According to the gastrointestinal and hepatic biotransformation pathways of triflusal, it was found that triflusal was hydrolyzed by about 5% in intestine and metabolized by about 53% in liver, and that the bioavailability of triflusal after oral administration of triflusal was 0.44, and also that the fraction of total elimination rate of triflusal which formed HTB in liver $(F_{mi},\;%)$ was about 98%. These results showed that triflusal was almost metabolized in liver, and the total elimination of triflusal in the body was dependent to the formation rate of HTB from triflusal in liver.

Human Health Risk Assessment of Benzene from Industrial Complexes of Chungcheong and Jeonla Province (충청·전라지역 산업단지 주변지역에서의 벤젠 인체 위해성 평가)

  • This research studied human health risk assessment of benzene from industrial complexes of Chungcheong Province (Seosan industrial complex) and Jeonla Province (Iksan industrial complex and Yeosoo industrial complex). The residents near the industrial complexes areas can be often exposed to volatile organic compounds (e.g., benzene, toluene, xylenes) through a number of exposure pathways, including inhalation of the organic pollutant via various environmental matrices (air, water and soil), contaminated water, and soil intake. Benzene is well known to be a common carcinogenic and toxic compound that is produced from industrial and oil refinery complexes. In this study, a number of samples from water, air, and soil were taken from the residential settings and public school zones located near the industrial complex sites. Based on the carcinogenic risk assessment, the risk estimates were slightly above $10{\times}10^{-6}$ at all three industrial sites. According to deterministic risk assessment, inhalation was the most important route. The distribution of benzene in the environment would be dependent on vapor pressure, and the physical property influencing the extent of the potential risks. Non-carcinogenic risk assessment of benzene shows that the values of Hazard Index(HI) were much lower than 1.0 at all industrial complexes. Therefore, benzene was not a cause of concern in terms of non-carcinogenic risk posed to the residents near the sites. When compared to probabilistic risk assessment, the CTE(central tendency exposure) cancer risk values of deterministic risk assessment were close to the mean values predicted by the probabilistic risk assessment. The RME(reasonable maximum exposure) values fell within the range of 95% to 99.9% estimated by the probabilistic risk assessment. Since the values of carcinogenic risk assessment were higher than $10{\times}10^{-6}$, further detailed monitoring and refined risk assessment for benzene may be warranted to estimate more reliable and potential inhalation risks to receptors near the industrial complexes.

The Influence of Worker's Stress and Anxiety on Work Commitment: Focused on Mediating Effect of Smartphone Addiction (직장인의 스트레스와 불안이 업무몰입에 미치는 영향 : 스마트폰 중독의 매개효과를 중심으로)

  • The purposes of this study were to find smart-phone addiction how to mediate between worker's stress, anxiety and work commitment. The subject of survey was 329 workers in a public cooperation, and the data were analyzed by AMOS 18.0. The major findings were as follows; First, the higher the stress and anxiety, the more the direct effect on smartphone addiction. Second, smartphone addiction showed a significant negative relationship with work commitment. Third, smartphone addiction played a mediating role in the relationship between stress, anxiety, and work commitment. Workers with high levels of stress and anxiety were over-dependent on smartphones, which resulted in a decrease in work commitment. In order to prevent the problem of smartphone addiction of workers, it is necessary to prepare guidelines for correct use of smartphone in the workplace and service support should be provided according to the level of addiction. In addition, in order to improve work commitment, active support from the aspect of corporate welfare is needed to manage stress well. Finally, this study has a policy implication in that it reveals the route leading to smartphone addiction.

Flux Melting Route to 2-and 3-dimensional Fibrous Potassium Titanates, K$_2Ti_{2n}O_{4n+1}$ (n = 2 and 3) (Flux 용융법에 의한 2차원 및 3 차원 구조의 티탄산칼륨 섬유의 합성)

  • Two-dimensional potassium tetratitanate ($K_2Ti_4O_9$) and three-dimensional potassium hexatitanate ($K_2Ti_6O_{13}$) fibers have been prepared by the combined method consisting of the flux melting (1150$^{\circ}C$)-slow cooling (cooling rate = 5$^{\circ}C$/h) process from the starting raw materials of $K_2CO_3$, and $TiO_2$ with the flux of $K_2MoO_4$. It was found that the fiber growth reaction is strongly dependent upon the mole ratio of flux (F) to raw material (R), which is 7 : 3 (F : R) as for the optimum growth condition. Relatively long fibers (average length ${\thickapprox}$ 4 mm) with a mixture of $K_2Ti_4O_9$ (major) and $K_2Ti_6O_{13}$ (minor) could be obtained when the reaction was carried out for the $K_2MoO_4-$K_2O{\cdot}4TiO_2$ (F : R = 7 : 3) system, but for the $K_2$MoO_4$-$K_2O{\cdot}6TiO_2$ (F : R = 7: 3) one, only the short fibers with ${\thickapprox}$ 2 mm long could be grown as the mixed phase of $K_2Ti_6O_{13}$ and $K_2Ti_4O_9$.

Behavior of the herbicide metolachlor in carps (제초제 metolachlor의 잉어체내 행적)

  • In order to investigate the behavior of the herbicide metolachlor [2-chloro-6'-ethyl-N-(2-methoxy-1-methylethyl)aceto-O-toluidide] in fish, carps (Cyprinus carpio L.) were exposed to the herbicide at $LC_{10}$(1.93 mg/L) for 4 days. Metolachlor dissolved in water was absorbed rapidly into carps to mark the maximum concentration 6 hours after exposure. The amounts of the $^{14}C$-metolachlor and its metabolites absorbed in gall were much higher than those in the other parts, strongly suggesting that biliary excretion involving enterohepatic recirculation could be an important route for the elimination of metolachlor. The $^{14}C$-radioactivity distributed into aqueous phase fraction in test water and in carp extract was increased in time-dependent manner. Extraction rate of $^{14}C$ absorbed in carp tissues was decreased remarkably up to 6 hours after exposure, suggesting that the possible polar metabolites of metolachlor were transformed into the conjugates to form non-extractable bound residues.

Making a Civil War Surrounding History in Cyber Space Focused on 5·18 Discourses in Ilbe Storehouse (사이버 공간에서의 역사의 내전(內戰)화 '일간베스트저장소'의 5·18 언설을 중심으로)

  • Officially given a historical signifier as Gwangju Democratic Movement since 1987, far-right and conservative group have restated that $5{\cdot}18$ is a rebellion and a riot that was committed by rebellious elements who obeyed North Korea's command or who were connected with North Korea. As those who had been responsible for the rebellion, revolt and riot were rewarded, far-right and conservatives' collective narrative that a country was born where the pro-North Korea left became dominated aroused extreme hostility towards $5{\cdot}18$. Far-right and conservatives involved in many different fields such as political party, university, press and media and civil group carry out incendiary discourse politics with intention to reestablish history and memory of $5{\cdot}18$ in their own story. Many people at online sites such as Ilbe Storehouse who are considered 'young right wing' is a main route to spread the far-right groups' remarks on $5{\cdot}18$. Ilbe is a main channel to reconstitute and reproduce the far-right conservatives' remarks and information on $5{\cdot}18$. Ilbe is one of main area where remarks of disparagement and ridicule, hostility and hatred on $5{\cdot}18$ unfurl. This study collects $5{\cdot}18$-related remarks and stories unfolded at Ilbe and examines how these remarks and stories make significance as to $5{\cdot}18$ and how information resources which remarks are dependent upon are connected each other. In this process, this study intends to find implications of incendiary politics that echoed of remarks on $5{\cdot}18$ have which at the online site Ilbe and by the far-right conservatives.
