Scopolia parviflora Nakai, a rare and endangered species, is the sole plant producing tropane alkaloids (TA) among the Korean native species. In order to enhance TA productivity the SP72 root line was selected by screening 100 of root line, and the optimal culture media for root growth and TA production were investigated with the SP72 roots. Based on the several media, SH and 2B5 medium were determined as growth medium and White and NN medium as production medium. Among the four combinations of two-stage culture, 2BN (2B5 as growth medium plus NN as production medium) showed more enhanced root growth and TA production as compared with production media of White and NN medium and growth media of SH and 2B5 medium, respectively. However, bubble column bioreactor (BCB) cultures applying two-stage culture did not reveal the effective results despite of the each successful operation of two-stage culture in conical flasks and BCB cultures.
The important component for medical effect in ginseng is ginsenoside. Korea Ginseng & Tobacco Research Institute contains approximately 200 lines produced by inbred selection. It is assumed that ginseng lines containing high level of ginsenoside should be included in those lines. Besides, new breeding methods such as cell line selection in vitro and hairy root were recently developed. Therefore, this study was carried out to detect genes related to ginsenoside, and to use it for selection marker to select and distribute lines containing high level of ginsenoside. DNA was extracted from both ginseng roots and hairy roots, and the difference between the line containing high ginsenoside(KG101) and normal ginsenoside(KG103) were analysed. As a result, 28 out of 36 primers showed bands, and many primers showed band difference between ginseng lines. It is considered that the bands should be analysed using DNA sequence comparison to check if those are related to ginsenoside. In case of hairy roots of ginseng, almost no differences were found between two lines.

Activation tagging that uses T-DNA vectors containing multimerized transcriptional enhancers from the cauliflower mosaic virus (CaMV) 35S gene is a powerful tool to determine gene function in plants. This approach has been successfully applied in screening various types of mutations and cloning the corresponding genes. We generated an activation tagged hairy root pool of ginseng (Panax ginseng C.A. Meyer) in an attempt to isolate genes involved in the biosynthetic pathway of ginsenoside (triterpene saponin), which is known as the major active ingredient of the root. Quantitative and qualitative variation of ginsenoside in activation tagged hairy root lines were profiled using LC/MS. Metabolic profiling data enabled selection of a specific hairy root line which accumulated ginsenoside at a higher level than other lines. The relative expression level of several genes of triterpene biosynthetic pathway in the selected hairy root line was determined by real time RT-PCR. Overall results suggest that the activation tagged ginseng hairy root system described in this study would be useful in isolating genes involved in a complex metabolic pathway from genetically intractable plant species by metabolic profiling.
Hyoscyamus species is sources of the hypnotic and sedative drugs hyoscyamine and scopolamine. Single cells of Hyoscyamus niger were dissociated from suspension cultures and adventitious roots obtained from single-cell clones which were cultured on B5 medium containing 3% (w/v) sucrose, 0.1 mg/L IBA and 0.4% (w/v) gelrite. H. niger adventitious root lines showed wide variation in tropane alkaloids production and growth. An effective selection of 200 root lines was made possible by the application of the 'Dragendorff's reagent' for qualitative detection of the alkaloids from root. A high correlation coefficient (r=0.9390) was observed between the values obtained with the two methods based on HPLC and Dragendorff's reagent analysis. Among the selected roots, the highest scopolamine content was 16.64 mg/g DW (Hn-59), which was 8.82-fold more productive than the lowest alkaloid producing line (Hn-25). Here, we established an efficient selection method on tropane alkaloids production and suggest that the Dragendorff's reagent is of great practical value in selection of invisible compounds.

The combination of a radiation technique with an in vitro culture system was appiled to develop submergence tolerant rice. The 3,000 $M_3$ lines with an average 80 percent of fertile grain were utilized for the selection of submergence tolerance. Salt tolerant lines were selected based on high plant height, root length and root number after submergence in plastic pots. Of the lines tested, the tolerant line (403-6) showed a dramatic difference in morphological traits under submergence compared to its original variety (Dongjinbyeo). It was suggested that genetic variations between the original variety and $M_3$-403-6 did exist. The levels of $\alpha$-amylase and alcohol dehydrogenase activities were significantly increased in the mutant line compared to its original variety. The mutant with greater tolerance showed less electrolyte leakage indicating a greater membrane integrity and better survival. Also, this line was much more resistant to a salt stress of $1.25\%$ than the original variety. The proline level of the line was significantly (p<0.01> higher than that of the original variety. The relationships between the inhibition of growth caused by stress and the physiological changes in the plant cell were discussed.
This study was conducted to obtain the basic information on setting up selection procedure using seedling characteristics in Angelica gigas Nakai lines. Morphological characteristics of the seedlings including height, leaf and petiole length among lines tested showed difference significantly, meaning that earlier selection was possible. Bolting rate at maturity ranged from 0.6 to 74.6% and root yield did from 350 to 3,893kg/ha. Root yield was increased in the lines with below 5% bolting rate. Heritabilities of petiole length, seedling height and leaf length in seedling stage bolting rate and root yield were high, while those of leaf length, root diameter and root length were relatively low. Bolting rate at maturity was positively correlated with seedling height, leaf length, leaf width, petiole length and number of leaves. A highly significant negative correlation was showed between root yield and seedling height, leaf length and width, petiole length, number of leaves, root length or bolting rate. From the above results the selection markers were found to be height, leaf length and petiole length in seedling stage.

This paper describes an approach for synthesizing a modularized gain scheduled PD type fuzzy logic controller(FLC) of nonlinear magnetic bearing system where the gains of FLC are on-line adapted according to the operating point. Specifically the systematic procedure via root locus technique is carried out for the selection of the gains of FLC. Simulation results demonstrate that the proposed gain scheduled fuzzy logic controller yields not only maximization of stability boundary but also better control performance than a single operating point (without gain scheduling)fuzzy controller.
Ginseng(Panax ginseng C. A. Meyer) is important medicinal plant but requires 4-year cultivation for root harvest because of slow growth. In contrast, ginseng hairy roots induced by introducing Ri-plasmid of Agrobacterium rhizogenes into genomic DNA of plant cells show vigorous growth, and the hairy roots produce the same or more saponins than natural ginseng roots. Therefore, hairy roots can be used for commercial purposes. The present study was carried out to induce hairy roots with both active growth and high saponin contents. Numerous hairy roots of Panax ginseng were obtained after root disks of three-year old roots were infected with Agrobacterium rhizogenes R1000 A4T in dark condition after one month of culture. About 3 hundred lines of hairy roots were selected according as morphological characters on medium with carbenicillin. After pre-selection of fifteen lines of hairy roots with active growth, KGHR-l and KGHR-8 lines were finally selected which had characters of high content of ginsenoside-Rd and ginsenoside-Re, respectively. The optimum growth of hairy roots was achieved in the culture of 1/2 MS liquid medium in dark (22 $^{\circ}C$) under 60 rpm gyratory shaking. Hairy roots grew well in 5L Erlenmeyer flasks, lL roller drums, 10L jar-fermenters, and especially in 20L air-lift culture vessels.
A new Sikbangpoong(Peucedanum japonicum Thunberg) variety, 'Sikbangpoog 1', was developed through a pure line selection at the Kyougpuk Provincial RDA during the period of 1990 to 1995. The variety was characterized to have higher stem height, leaf number and seed production, and larger root diameter compared with the check variety of Bonghwa Sikbangpoong but plant height of 'Siknagpoong 1' was similar to that of the check variety and it was also more tolerant to water and heat stress. Root color of 'Sikbangpoong 1' was yellowish brown and root shape of the variety was straight and conical. The root yields of Sikbangpoong 1 in yield trial, regional adaptation trial and farmer's field trial were always 10-28% higher than those of the check variety.
This study was to determine optimal sizeof root top using Suwon 2 selected from the Jinbu local population though pure line selection and five root head diameters of $0.11{\sim}0.30,\;0.31{\sim}0.50,\;0.51{\sim}0.70,\;0.71{\sim}0.90\;and\;0.91{\sim}1.10cm$. Emergence rate, characteristics of top, and root were increased with increasing diameterof seedling root. In the group of root head diameter from 0.51cm to 0.70cm, bolting rate was the lowest being less than 3.3% and root yield was the highest being more than 300kg/l0a. Bolting rate was positively correlated with plant height while showed negatively correlated with root yield. The study showed that the optimum of root head diameter raised seedlings in the open field appeared to be from 0.31 to 0.70cm.
