Detection of Anticancer Activity from the Root of Angelica gigas In Vitro

  • Anticancer activity of a fraction of the ethanol extract from the root of Korean angelica (Angelica gigas Nakai) was recognized in human cancer cell lines HeLa $S_3$, K-562, and Hep $G_2$. The extract blocked the phorbol ester-inducing megakaryocytic differentiation of K-562 cells, which indicated the modification of protein kinase C (PKC) activity. In vitro assay showed the activation of PKC by the extract. An effective fraction of the Angelica gigas extract, of which $R_f$ value was 0.64 in a thin layer chromatography, was a different component from those of European angelicas. The $ED_50$ value of the fraction was 8, 9, and $16\;\mu\textrm{m}/ml$ against HeLa $S_3\;Hep\;G_2$, and K-562 cells, respectively, while the fraction showed higher $ED_50$ values against normal cell lines.

상백피 추출물로부터 향균성 물질의 분리정제 (Purification of Antimicrobial Substance for the Extract from the Root Bark of Morus alba)

  • The ethanol extract from the root bark of Morus alba showed the strongest antimicrobial activity on the growth of almost all the tested microorganisms which were food-borne pathogens and food-related microorganisms. 1) In order to isolate and purify of antimicrobial substance extracted from the root bark of Morus alba, the antimicrobial substance from the ethanol extract which exhibited a strong antimicrobial activity was purified by solvent fractionation, silica gel column chromatography, TLC and HPLC. Among the fractions fractionated by 4 kind of solvents from the ethanol extract, the antimicrobial activity of ethyl acetate fraction had the strongest antimicrobial activity against B. subtilis. Unknown compound were isolated from the ethyl acetate fraction by silica gel column chromatography, TLC and HPLC and the compounds showed strong absorbance at 207, 217 and 285 nm, therefore, it was supposed to be a kinds of aromatic compound.

Pinus rigida Allelochemicals가 차풀종자의 발아과정에서 동위효소의 활성에 미치는 영향 (Effects of Pinus rigida Allelochemicals on Isozyme Activities during Seed Germination of Cassia mimosoides var.nomame)

  • Eleven phenolic compounds including caffeic acid were identified through analyzing the aqueous extracts of Pinus rigida by HPLC. Among them, protocatechuic acid was the maximum amount of 6.84 ppm. Seed germination of Cassia mimosoides var. nomame was significantly stimulated by the extract of P. rigida leaves in the proportion ot concentration. However, root growth was elevalted at a threshold concentration below 25%, but it was inhibited at high concentrations. In 50% extract of P. rigida, upward root tip of C. mimosoides var. nomame showed negageotropism which the root end showed necrosis. New isozyme bands were induced indicating concentration activity of peroxidase from the extract of C. mimosoides var. nomame, especially in the cathodic region. Although it reduced the mumber of isozyme bands of esterase, esterase activities were stimulated in the anodic region of C. mimosoides var. nomame. The activity of amylase was not remarkably different between control and treatment.

발효 균주에 따른 삼채뿌리 열수추출물의 항산화능 비교연구 (Effect of Starter Cultures on the Antioxidant Activities of Allium hookeri Root-Hot Water Extract)

  • Allium hookeri, a member of the onion family, has long been mainly cultivated for food and medicinal use in Southeast Asian countries, owing to its various biological properties. However, no studies of the anti-oxidative effects of fermented A. hookeri root extracts have been conducted to date. Therefore, this study investigated the effect of different starter cultures on the antioxidant activities of hot water extract of A. hookeri root by using the following five strains: Lactobacillus acidophilus, Bifidobacterum longum, Enterococcus faecium, Streptococcus thermophilus, and Saccharomyces cerevisiae. DPPH and ABTS radical scavenging activity, total phenolic acid contents, and total antioxidant capacity were higher in the hot water extract of A. hookeri root fermented with starter cultures than those of A. hookeri root. Among hot water extract of A. hookeri root fermented with starter cultures, fementing with S. cerevisiae showed the highest antioxidant activities. The results of this study provide new evidence of the anti-oxidative properties of A. hookeri root with starter cultures, indicating that it may be highly valuable as a natural product owing to its high-quality functional components.

해당화 줄기 및 뿌리의 주성분 분리 및 항산화활성 (Isolation of main component and antioxidant activities on the stem and root of Rosa rugosa)

  • 1. 해당화의 총페놀화합물 함량은 줄기추출물이 평균 7.8(g/100g, D.W.)이고 뿌리추출물이 평균 10.5(g/100g, D.W.)이며, 모든 시료에서 뿌리추출물의 함량이 높게 나타났다. 2. K-1의 줄기와 뿌리 추출물(0.5g)에서 주성분의 하나로 추정되는 S-4(84.1mg)과 R-2(70.0mg)의 화합물을 단리했고, 그 구조를 catechin으로 결정했다. 3. UPLC 분석에 의해 줄기추출물의 탄닌함량은 평균 752.5(mg/100g, D. W.)로, gallic acid 6.3, epicatechin 61.5, catechin 684.8이고, 뿌리추출물의 탄닌함량은 줄기의 약 2배 이상 높은 1676.0(mg/100g, D.W.)으로 gallic acid 6.8, epicatechin 160.3, catechin 1508.5로 나타났다. 4. 해당화의 free radical 소거활성에서 추출물의 IC50는 뿌리가 $13.7{\mu}g/ml$, 줄기가 $14.5{\mu}g/ml$으로 높은 소거활성을 나타냈고, SOD 유사활성은 $50{\mu}g/ml$의 농도에서 뿌리추출물이 평균 75.1%, 줄기추출물이 71.9%로써 높은 활성을 나타냈다.

C57BL/6 mouse에서 뽕나무 추출물의 모발성장효과 (Effect of Morus alba extract for hair growth promotion in C57BL/6 mouse)

  • Morus alba has been well known for its tonic effect or preventing of hypertension, aging, or diabetes mellitus in Asian countries. To evaluate the its effect on hair growth promotion, we performed the hair regrowth experiment with leaf, branch, root and fruit extract of Morus alba on animal model of C57BL/6 mouse. The hair regrowth effect was remarkable in the mouse treated branch and root extract (with what %), however, little effect on hair regrowth appeared on the mouse treated with leaf extract. Morus alba extract displayed antibacterial effects on Bacillus subtilis, Staphylococcus aureus, Pseudomonas and Candida albicans. These results indicated that branch and root extract of Morus alba could be applicable for hair regrowth or prevention of hair loss in human.

Bioactivity of Cajuput Seedling n-Hexane Extract as an Attractant for Subterranean Termite Coptotermes curvignathus Holmgren (Isoptera: Rhinotermitidae)

  • Subterranean termite attacks on cajuput (Melaleuca cajuputi) seedling roots were widespread in several of Java's Perum Perhutani Forest Management Units. This attack was suspected to be related to the chemical components of the cajunput seedling roots. This study was conducted to determine the bioactivity of cajuput seedling root extract as an attractant for the subterranean termite Coptotermes curvignathus Holmgren (Isoptera: Rhinotermitidae). The extraction process was performed according to ASTM D1108-96, and the extract was characterized using gas chromatography-mass spectrometry (GC-MS). Bioactivity testing of the extracts was carried out using attractiveness and no-choice feeding bioassays. The results showed that the average root extraction yield from cajunput seedlings was 4.94%. The attractiveness of the extract solutions at concentrations of 0.50%, 0.75%, and 1.00% were 45.33%, 62.00%, and 74.67%, respectively. The mortality rate of C. curvignathus termites ranged from 9.63% to 24.44%. Cajuput seedling root extract's lethal concentration 50 was 2.45% (non-toxic). GC-MS analysis showed that the extract contained linoleic acid, which has the potential to attract insects.

갓무를 첨가한 김치의 발효 및 관능 특성 (Fermentative and Sensory Characteristics of Kimchi added Mustard Root (Brassica juncea))

  • This study was conducted to examine the effect of mustard root on the shelf life of kimchi. Ethanol extract was prepared from the mustard root (Brassica juncea) and antimicrobial activities were examined against Lactobacillus plantarum and Leuconostoc mesenteroides which are the representative lactic bacteria in kimchi. Growth of lactic bacteria were inhibited by ethanol extract from mustard root and the growth time of Leuconostoc mesenteroides was delayed more than 24 hours compared with that of Lactobacillus plantarum. Also, the fermentative and sensory characteristics of kimchi with mustard root were evaluated during fermentation at $10^{\circ}C$. The pH and acidity was lower than that of the control kimchi and the fermentation period was retarded in proportion to the increase in mustard content. The total numbers of microorganism and lactic bacteria in the control kimchi were higher than those in kimchi with mustard root during fermentation. Leuconostoc sp. in the initial stage of growth was more inhibited than Lactobacillus sp. While the pH of kimchi were in the initial stage, the sensory evaluation score for savory flavors of the kimchi with mustard root was lower than that of control due to bitterness and pungency. But when the pH of kimchi reached the optimum stage, the scores for texture and savory flavor of kimchi with mustard root were higher than that of the control. Then overall quality score for kimchi with mustard root increased as bitterness decreased.

우엉발효추출물을 이용한 생활악취물질 제거효율 조사 (Removal Efficiency of odor substance Using Arctium Lappa Root extract)

  • Increasing public concerns over odors and air regulations necessitates the remediation of a wide range of odor substances for industrial and indoor purpose. Currently, the wet scrubbing technique by neutralization using oils is being used to treat odor substances such as Ammonia($NH_3$), Methyl Mercaptan($CH_3SH$), Hydrogen Sulfide($H_2S$), Methylamine($CH_3NH_2$), Acetaldehyde($CH_3CHO$). The chemical analysis is performed to analyze the composition of an Arctium Lappa Root extract by VOC analyzer(Phocheck 5000Ex, ION SCIENCE co.) The objectives of the this study are to clarify the possibility of the neutralization of odors sprayed in the fixed bed and determine the removal efficiencies for different input odor concentrations. It is found that Ammonia($NH_3$), Methyl Mercaptan($CH_3SH$), Hydrogen Sulfide($H_2S$), Methylamine($CH_3NH_2$), Acetaldehyde($CH_3CHO$) are significantly removed and their removal efficiencies are higher than 98%. The kitchen detergent with Arctium Lappa Root extract showed excellent removal efficiencies of odor substances and high possibility for the development of kitchen detergent with odor removal.

까마중-에탄올 추출물의 기능적 특성과 항산화 효과 (Functional Properties and Antioxidant Effects of Solanum nigrum-Ethanol Extract)

  • Functional properties of Solanium nigrum-ethanol extract were investigated, nitrite scavenging ability(NSA) and antioxidant activities based on thiobarbituric acid(TBA) value were measured. The contents of potassium in fruit and root, calcium in leaf were the highest mineral components in each parts of Solanium nigrum. Total aromatics contents(TAC) was order of fruit>root>leaf, total flavonoid contents(TFC) in leaf was 6 and 10 times, total phenolic compounds contents(TPC) in leaf was 15 and 30 times of those in fruit and root, respectively, and TFC was high with TPC increase. NSA of leaf-ethanol extract was 61.9% on pH 1.2, 53.5% on pH 3.0, 54.8% on pH 4.0 and 14.3% on pH 6.0, where as NSA of root and fruit-ethanol were considerably low to 3.5~7.9% over pH 1.2~6.0. TBA value of soybean oil with the dosage of leaf-extract showed few differences from control within 3 days, but it fairly decreased with considerable antioxidant effect to 47.6% of the control in 8 days.
