포도 삽목에서 내생 균근균 접종효과 (Effectiveness on the Inoculation of Arbuscular Mycorrhizal Fungi in Cutting of Grapevine)

  • 본 연구는 균근균 접종이 포도삽목묘의 생장에 미치는 영향과 균근 형성율 그리고 형성 양상을 구명하고자 수행하였다. 포도 삽목시, 균근균 접종원을 처리한 거봉과 탐나라 품종은 무 접종처리 보다 엽수, 엽면적, 총 뿌리길이 그리고 뿌리 표면적이 크게 증가하였다. 그러나 켐벨어리 품종의 총 뿌리길이와 뿌리 표면적은 무처리구 (AMF-)와 비교하여 유의적인 차이가 없었다. 접종 8주후 균근균 접종 처리구 (AMF+)의 형성율은 22.5-32.5%이었으며, 12주 후 켐벨어리, 탐나라 그리고 거봉의 균근 형성율은 각각 29.6%, 28.8% 그리고 48.8%이었다. 반면 균근균 무처리구 (AMF-)는 균근형성 수준이 매우 낮았다.

Involvement of Pyridoxine/Pyridoxamine 5′- Phosphate Oxidase (PDX3) in Ethylene-Induced Auxin Biosynthesis in the Arabidopsis Root

  • As sessile organisms, plants have evolved to adjust their growth and development to environmental changes. It has been well documented that the crosstalk between different plant hormones plays important roles in the coordination of growth and development of the plant. Here, we describe a novel recessive mutant, mildly insensitive to ethylene (mine), which displayed insensitivity to the ethylene precursor, ACC (1-aminocyclopropane-1-carboxylic acid), in the root under the dark-grown conditions. By contrast, mine roots exhibited a normal growth response to exogenous IAA (indole-3-acetic acid). Thus, it appears that the growth responses of mine to ACC and IAA resemble those of weak ethylene insensitive (wei) mutants. To understand the molecular events underlying the crosstalk between ethylene and auxin in the root, we identified the MINE locus and found that the MINE gene encodes the pyridoxine 5′-phosphate (PNP)/pyridoxamine 5′-phosphate (PMP) oxidase, PDX3. Our results revealed that MINE/PDX3 likely plays a role in the conversion of the auxin precursor tryptophan to indole-3-pyruvic acid in the auxin biosynthesis pathway, in which TAA1 (TRYPTOPHAN AMINOTRANSFERASE OF ARABIDOPSIS 1) and its related genes (TRYPTOPHAN AMINOTRANSFERASE RELATED 1 and 2; TAR1 and TAR2) are involved. Considering that TAA1 and TARs belong to a subgroup of PLP (pyridoxal-5′-phosphate)-dependent enzymes, we propose that PLP produced by MINE/PDX3 acts as a cofactor in TAA1/TAR-dependent auxin biosynthesis induced by ethylene, which in turn influences the crosstalk between ethylene and auxin in the Arabidopsis root.

  • Incident investigation is regarded as a means to improve safety performance. For the prevention of industrial accidents, measures such as providing safety education, enhancing management interest and participation, establishing a safety management system, and conducting inspection of the work site are necessary. In particular, accident investigation activities, which are an important element of safety management, help to prevent similar accidents, thereby minimizing damage and enhancing work safety. They are critical for understanding business-related incidents and the vulnerabilities and opportunities associated with them. Therefore, it is clear that accident investigation activities are important for accident prevention. The primary focus of many incident investigation processes is on identifying the cause of an event. While considerable research has been conducted on potential accident investigation tools there has been little research on including the views and experiences of practitioners in the accident investigation process. In this study, a questionnaire survey was conducted among safety managers in the domestic manufacturing/construction industry to understand the practice of accident investigation. The investigation pertained to companies' accident investigation systems, the competence of investigators, and the identification and recommendations of the cause of accidents. From the analysis results of accident investigations, investigators' competence, the difficulty level of investigations, and the root causes of accidents were identified from the viewpoint of the participants of the accident investigations. In particular, the development of standardized and simple accident investigation methods and their dissemination to companies were found to be necessary for activating the root cause of accidents. Based on this, it can be used as basic data for the development of root cause analysis investigation techniques that are easily applicable to organizations.

Changes in Growth Characteristics of Seven Foliage Plants Grown in an Indoor Bio-Wall System Depending on Irrigation Cycle

  • In order to increase the indoor air purification effect of plants, plants need to be placed on 5-10% of indoor spaces. To increase the density and utilization of plants in indoor spaces, studies on bio-wall, a vertical green wall system, have been recently conducted. The purpose of this study was to investigate the growth characteristics of 7 indoor plants introduced to the system and their rooting zones at different irrigation cycles. This study was conducted to investigate a proper irrigation cycle for the continuous maintenance of bio-wall systems. The conditions of their growth environment were maintained as follows: light intensity, 20-50 μmol·m-2·s-1 PPFD; and temperature, 20 - 25℃. For fertilization, Hyponex diluted with water at the ratio of 1:1,000 was supplied to plants. Irrigation was treated at intervals of 1, 3, 5, and 7 days for 1 hour at a time. As a result, there was no significant difference in the growth of plants between different irrigation cycles. Dieffenbachia 'Marianne' showed a significant decrease in the number of leaves at the irrigation cycle of 7 days. In addition, the chlorophyll content was relatively low at the irrigation cycle of 7 days. In terms of the color of leaves, a decrease in L value and b value and an increase in a value were observed, resulting in changes in brightness and color. Ardisia pusilla 'Variegata' showed a slightly higher photosynthetic activity and stomatal conductance when it was watered every day and once per 5 days, while Epipremnum aureum showed a relatively higher photosynthetic activity and stomatal conductance at the irrigation cycle of 3 days. In the case of root activity, it was found that the longer irrigation cycle, the higher root activity compared to daily irrigation. The development of roots of Peperomia clusiifolia was promoted by watering at long intervals. However, in the case of Aglaonema 'Siam-Aurora', the total number of roots decreased at the interval of 7 days. In conclusion, a proper irrigation cycle for the sustainable maintenance of vertical bio-wall systems seems to be 3 days.

Development of transgenic disease-resistance root stock for growth of watermelon.(oral)

  • To protect the plant against several soil-borne pathogens, we are currently constructing disease-resistant transgenic root stock for the growth of cucurbitaceae vegetable plants, watermelon and gourd. We made a watermelon cDNA library from Cladosporium cucumerinum-Infected leaves for substractive hybriazation and differential screening. We isolated the several pathogen inducible cDNA clones, such as caffeoyl-CoA-methyltransferase, LAA induced protein, receptor-like kinase homolog, hydroxyproline-rich glycoprotein, catalase, calmodulin binding protein, mitochondrial ATPase beta subunit, methyl tRNA synthetase and WRKY transcription factors. We previously obtained CaMADS in pepper and galactinol synthase ( CsGolS) in cucumber that were confirmed to be related with disease-resistance. CaMADS and CsGolS2 were transformed into the inbred line 'GO701-2' gourd, the inbred line '6-2-2' watermelon and the Kong-dye watermelon by Agrobacterium tumerfaciens LBA4404. Plant growth regulators (zeatin, BAP and IAA) were used for shoot regeneration and root induction for optimal condition. Putative transgenic plants were selected in medium containing 100mg/L kanamycin and integration of the CaMADS and CsGO/S2 into the genomic DNA were demonstrated by the PCR analysis. We isolated major soil-borne pathogens, such as Monosporascus cannonballus, Didymella bryoniae, Cladosporium cuvumerinum from the cultivation area of watermelon or root stock, and successfully established artificial inoculation method for each pathogen. Screening of Insecticide to Control Rhophalosiphum nymphaeae for Alisma plantago as Second Crop in Paddy Field

  • The purpose of this study was to evaluate the effect of pesticides on insect pest control of Rhophalosiphum nymphaeae, growth characteristies and dry root yield from the cultivated after early maturing rice cropping. All pesticides treated had no effect on the growh period, flowering rate of Alisma plantago. The major pesticides were methomyl-Lf, 24.1 %, imidacloprid-Wp, 10%, carbosulfan-Wp, 20%, and methomyl-Wp, 45%. Dry yield of root were increased largely with imidacloprid-Wp, 10% (10g/20$\ell$), pesticide than the other pesticides and control. All pesticides were had no injury with recommended concentration. On the other hand all pesticides were slightly harmful in the double dosage level for the Alisma plantago.

  • The need for this study must be considerable, as being activate of green roofs research that the organization and construction obtain access to more development technologies. Nevertheless, the green roofs system has begun to apply since 1980's, the green roofs technology was restricted to develop without verification of technologies such as a load or water leakage. There is a limit as urethane waterproofing to almost domestic waterproofing materials and methods of construction for general green roofs. The introduction of materials and methods of construction which are appropriated to property of green roofs could be a decisive factor in a long-range durability and economical maintenance cost, moreover. it support to variety construction system and organization. This present paper describes a necessity of waterproofing and root barrier system is one of the sub-organization based on green roofs construction. which have enormously large impact on the durability.

  • The author has studied on 910 roentgenograms of lower second molars, which were taken by intraoral technic, and obtained the following results. 1. The development of crown of lower second molars was completed 8.25 years. 2. The formation of mesial and distal roots in full length on lower second molars was completed as follows: a. mesial roots 15. 07 years b. distal roots 15. 53 years 3. The formation of apical foramina of mesial and distal roots of lower second molars was closured as follows: a. apical foramen of mesial root 18.00 years b. apical foramen of distal root 18.79 years 4. As a general rule, the mesial roots were developed more earlier than distal roots.

Chemical Control of Gray Mold in Scutellaria baicalensis Georg

  • This study was conducted to evaluate the control effect of fungicides on control of Gray mold, growth characteristics, and root yield in the cultivation of Scutellaria baicalensis after barley cropping. All fungicides treated had no effect on the growth and flowering rate of Scutellaria baicalensis. All seed disinfectants had no effect on the growth and flowering date of Scutellaria baicalensis Georg. The major seed disinfectants were Benomyl Wp, $20\%$, Captan Wp, $50\%$, Hymexazol Ec, $30\%$, Carboxin D, $37.5\%$. Dry root yield were increased largely with Benomyl Wp, $20\%$, seed disinfectant than the other seed disinfectants had no injury with standard dosage. On the other hand, all seed disinfectants had slight injury in the double dosage level for the Scutellaria baicalensis Georg.

In vitro Micropropagation of Rosa hybrid L.

  • To determine the appropriate concentrations of nutrients and growth regulators for shoot proliferation and root initiation, several rose hybrid tea cultivars were cultured. Cultured shoot tips and lateral buds from different cultivars proliferated multiple shoots on Murashige and Skoog (MS) medium supplemented with 0 to 4 mg/L BA and 0 to 0.05 mg/L NAA. The ability of the explants to proliferate shoots and initiate roots was affected by genotype, the nodal position of explant, the strength of MS basal medium and growth regulators used. The buds nearest the apex exhibited the slowest rate of development. Most cultivars had the highest shoot proliferation when cultured on MS medium with 2 mg/L BA and 0.01 mg/L NAA, but the degree varied by cultivars. Root development was enhanced by lowering the concentration of MS salts.
