브로콜리(Brassica oleracea var. italica) 잎에서 sulforaphane의 함량 변화 (Content changes of Sulforaphane in Leaves of Brassica oleracea var. italica)

  • 브로콜리 13종의 잎을 대상으로 sulforaphane의 함량을 분석한 결과, 2종에서는 검출되지 않았으나 11종에서는 검출되었다. "필그림" 품종이 가장 많은 sulforaphane 함량을 나타냈다. 브로콜리 잎을 blinder로 브로콜리의 잎을 균질화한 후에 sulforaphane의 생합성능은 30분 후에 급격히 증가하여 60분까지 함량이 유지되었다. 또한 sulforaphane은 동결건조를 할 경우 브로콜리 잎의 분말에서 검출되었으나 열풍건조를 할 경우 휘산 되어 분말에서 검출되지 않았다. 브로콜리의 부위별로(꽃 봉우리, 잎, 줄기 및 뿌리) sulforaphane의 함량을 분석한 결과, 뿌리에서 가장 많은 sulforaphane이 검출 되었고 다음으로 잎에서 높았다. 브로콜리 잎에서 생합성된 sulforaphane의 경시변화를 분석한 결과, 상온보관 보다 냉동보관 상태에서 오래 지속되었으나 냉동보관 조건에서도 3주 후에 sulforaphane의 함량이 1/2 이하로 감소하였다.

  • Chemical properties of the different parts of watercress(Oenanthe javanica D.C.) grown and harvested from the culture fields under different cultivating methods were studied. In proximate analyses of watercress in parts, moisture contained more in stem than in root or in leaf, but crude protein and crude lipid contents were lower in stem than in root or in leaf. Crude ash in root contained up to about double amount in leaf or stem. Major mineral elements detected in watercress were Fe, Mg, Ca, and K, and their contents in root was higher than those in stem or leaf. Especially, Fe in root was significantly higher than that in stem or in leaf. Total free sugar composed mainly with fructose and glucose, was the highest in watercress from Hwasoon and followed the watercress from Jeonju and Donggok in order. Free sugar content was highest in leaf and the lowest in root. Major water-soluble vitamins were vitamin C, thiamin and biotin and the content of vitamin C was higher than others. These vitamins contained more in leaf than in stem or root. Niacin contained 6.09 mg/100 g in leaf of watercress from Hwasoon, which was much higher than others, but it was not detected in stem of watercress from Jeonju. Organic acids detected were oxalic acid, citric acid and malic acid and other 12 organic acids were not detected. In fatty acid composition, there were significant differences among watercresses from different parts and different culture fields. Linoleic acid, linolenic acid and palmitic acid were major fatty acids contained in watercress and it took about 80% of the total content. Amino acid content in leaf was higher than that in root and in stem. Glutamic acid and proline were major amino acids in stem of watercress from Jeonju and in stem of watercresses from Hwasoon and Donggok, respectively. In leaves of all three watercresses glutamic acid content was the highest.

Growth, yield and mineral element responses to temperature in Codonopsis lanceolata

  • Temperature has an adverse effect for potential growth and development of Codonopsis lanceolata. The present study examined the responses of growth, yield and mineral element contents under temperature variables in C. lanceolata. Plant height exhibited the best result as being 2 m or higher the open field culture, while the growth is the most restricted in the $30^{\circ}C$. The open field culture also had the largest number of leaves in the early stage of growth. The number of leaves and the height tended to be increased with the temperature of $20^{\circ}C$ and $25^{\circ}C$. The number of branches showed no significance as 32 branches and 26 branches in open field culture and $20^{\circ}C$, respectively. The smallest number of branches was observed in the $30^{\circ}C$ with 15 branches. The leaf color recorded the lowest value in the open field culture unlike the growth above the ground. But all other temperature had no significance with the leaf color value between 50.0 and 52.6. The elevated temperature induced the thinner and shorter diameter and length of root. However, the root diameter was not significantly changed with the change of 1.8 cm and 2.1 cm at open field culture, with $20^{\circ}C$ and $25^{\circ}C$, respectively. The fresh weights of the primary root and lateral roots were 6.3 g and 3.1 g, respectively, at the high temperature. The result showed the thickening of the lowest part under the ground as restricted. The fresh weights of the main root and lateral root were the highest on the open field culture, 20.6 g and 12.2 g, respectively. The contents of cellulose were the highest as 4.41% in the $20^{\circ}C$, followed by the open field culture, $25^{\circ}C$ and $30^{\circ}C$. The higher temperature reduced the contents of cellulose. Moreover, P showed higher ratio in the high temperature while Ca and Mg exhibited the higher ratio towards the low temperature. On the contrary, minor quantity (less than 1%) of mineral nutrients was observed towards all temperature variables.

Induction of systemic resistance in Panax ginseng against Phytophthora cactorum by native Bacillus amyloliquefaciens HK34

  • Background: Korean ginseng (Panax ginseng Meyer) is a perennial herb prone to various root diseases, with Phytophthora cactorum being considered one of the most dreaded pathogens. P. cactorum causes foliar blight and root rot. Although chemical pesticides are available for disease control, attention has been shifted to viable, eco-friendly, and cost-effective biological means such as plant growth-promoting rhizobacteria (PGPR) for control of diseases. Methods: Native Bacillus amyloliquefaciens strain HK34 was isolated from wild ginseng and assessed as a biological control agent for ginseng. Leaves from plants treated with HK34 were analyzed for induced systemic resistance (ISR) against P. cactorum in square plate assay. Treated plants were verified for differential expression of defense-related marker genes using quantitative reverse transcription polymerase chain reaction. Results: A total of 78 native rhizosphere bacilli from wild P. ginseng were isolated. One of the root-associated bacteria identified as B. amyloliquefaciens strain HK34 effectively induced resistance against P. cactorum when applied as soil drench once (99.1% disease control) and as a priming treatment two times in the early stages (83.9% disease control). A similar result was observed in the leaf samples of plants under field conditions, where the percentage of disease control was 85.6%. Significant upregulation of the genes PgPR10, PgPR5, and PgCAT in the leaves of plants treated with HK34 was observed against P. cactorum compared with untreated controls and only pathogen-treated plants. Conclusion: The results of this study indicate HK34 as a potential biocontrol agent eliciting ISR in ginseng against P. cactorum.

Phytotoxic Effect of Xanthium occidentale Leaf Extract on Seed Germination and Early Seedling Growth of Alfalfa and Barnyard Grass

  • Compositae plants are known to contain biologically active substances that are allelopathic to agricultural crops as well as weed species. Aqueous extracts from leaves of Xanthium occidentale were assayed against alfalfa (Medicago sativa) to determine their allelopathic effects, and the result showed that the extracts applied onto filter paper significantly inhibited seed germination as well as root growth of alfalfa. Untreated seeds germinated in 60h, but extract concentrations greater than 30g $\textrm{L}^{-1}$ delayed seed germination. The extracts significantly inhibited seed germination of alfalfa, and $\beta$-amylase activity of alfalfa and barley seeds during 24-36 hours after treatment. Aqueous extracts of 40 g $\textrm{L}^{-1}$ from X. occidentale were completely inhibited the hypocotyl and root growth of alfalfa. Aqueous leaf extracts showed the highest inhibitory effect and followed by root and stem extracts. Early seedling growth of both alfalfa and barnyard grass (Echinochloa crus-galli) was significantly reduced by methanol extracts. By means of high-performance liquid chromatography, chlorogenic acid and trans-cinnamic acid were quantified as the highest amounts from water and EtOAc fractions, respectively. BuOH and EtOAc fractions of X. occidentale reduced alfalfa root growth more than did hexane and water fractions. The findings of the bioassays for aqueous or methanol extracts reflected that the inhibitory effect of extract was closely related to the level of responsible allelochemicals found in plant extracts.

The Effects of Thinning on Fine Root Distribution and Litterfall in a Pinus koraiensis Plantation

  • The purpose of this study was to investigate the effects of thinning on fine root biomass and vertical distribution. and litterfall amount in a 50 year old Pinus koraiensis plantation in Chuncheon, Kangwon Province. Fine root (< 2 mm in diameter) biomass ($367\;g/m^2$) in the site 'OC_75', thinning once in 1975, was 68% of those in the site 'CON', no thinning after planting, and in the site 'TC_00', thinning twice in 1975 and 2000. There were no significant differences of dead roots among treatments. Diameter $0{\sim}1\;mm$ roots were vertically decreased only in the TC_00 site. The litterfall was very similar between OC_75 ($5.2\;Mg\;ha^{-1}\;yr^{-1}$) and TC_00 ($4.7\;Mg\;ha^{-1}\;yr^{-1}$), but the composition of litterfall was different: The proportion of leaves and branches was 80% and 13% in OC_75 and 56% and 36% in TC_00, respectively. Reduction of P. koraiensis density by thinning decreased leaf litter as well as fine roots of P. koraiensis, but increased fine roots production by neighboring understory plants offset the reduction of fine roots of P. koraiensis. We suggest that belowground as well as aboveground responses, including both over- and understory vegetation, should be considered to measure the responses of trees in thinned forest ecosystems.

Sodium nitroprusside mediates seedling development and attenuation of oxidative stresses in Chinese cabbage

  • Nitric oxide (NO) has been shown to be involved in diverse physiological processes in microbes, animals and plants. In this study, the involvement of NO in the development and possible roles in oxidative stress protection of Chinese cabbage (Brassica rapa subsp. pekinensis cv. Samrack-ulgari) seedlings were investigated. Exogenous application of sodium nitroprusside (SNP) retarded root elongation, while increasing lateral root formation of Chinese cabbage. Plants showed no signs of external stress due to SNP application in true leaves. Cotyledons of 3-week-old Chinese cabbage plants were found to be highly sensitive to SNP application. Treated cotyledons displayed rapid tissue collapse and associated cell death. Although SNP application reduced root growth under normal growth conditions, it also enhanced methyl viologen (MV)-mediated oxidative stress tolerance. Analysis of SNP application to Chinese cabbage leaf disks, revealed SNP-induced tolerance against oxidative stresses by MV and $H_2O_2$, and evidence includes prevention of chlorophyll loss, superoxide anion (${O_2}^-$) accumulation and lipid peroxidation. This report supports a role for nitric oxide in modulating early seedling development, programmed cell death and stress tolerance in Chinese cabbage.

Effect of seeding depth on seedling growth and dry matter partitioning in American ginseng

  • Greenhouse and field experiments with American ginseng (Panax quinquefolius L.) stratified seed sown at depths of 10 to 100 mm were carried out to determine effects of seeding depth on seedling emergence, growth and development and to calculate optimum seeding depth. The time to 50% seedling emergence ($E_{50}$) in the field increased linearly from 17 d at 20 mm seeding depth to 42.5 d at 80 mm. Seedling emergence and root weight (economic yield) at the end of the first year each increased quadratically with the increase of seeding depth. Maximum emergence and root yields were produced at sowing depths of 26.9 and 30.6 mm respectively. In a greenhouse pot experiment, increasing seeding depth from 10 to 100 mm increased partitioning of dry matter to leaves from 23.6% to 26.1%, to stems from 6.9% to 14.2%, and decreased dry matter to roots from 69.5% to 59.7%. Optimum seeding depth was 31.1 mm for a corresponding maximum root weight of 119.9 mg. A predictor equation [X (seeding depth, mm)=Y (seed weight, mg)/9.1+20.96] for seeding depth for ginseng, based on data for ten vegetable crops, their seed weights and suggested seeding depths, predicted a seeding depth of 28.3 mm for ginseng similar to that reported above for most pot and field experiments.

Identification and Transcriptional Analysis of Priming Genes in Arabidopsis thaliana Induced by Root Colonization with Pseudomonas chlororaphis O6

  • Root colonization of Arabidopsis thaliana with Pseudomonas chlororaphis O6 induces systemic tolerance against diverse pathogens, as well as drought and salt stresses. In this study, we demonstrated that 11 genes in the leaves were up-regulated, and 5 genes were down-regulated as the result of three- to five-days root colonization by P. chlororaphis O6. The identified priming genes were involved in cell signaling, transcription, protein synthesis, and degradation. In addition, expression of selected priming genes were induced in P. chlororaphis O6-colonized plants subjected to water withholding. Genes encoding defense proteins in signaling pathways regulated by jasmonic acid and ethylene, such as VSP1 and PDF1.2, were additional genes with enhanced expression in the P. chlororaphis O6-colonized plants. This study indicated that the expression of priming genes, as well as genes involved in jasmonic acid- and ethylene-regulated genes may play an important role in the systemic induction of both abiotic and biotic stress due to root colonization by P. chlororaphis O6.

검은점뿌리썩음병 감염이 참외의 광합성 및 생육에 미치는 영향 (Effect of Monosporascus Root Rot Infection on Photosynthetic Activity and Plant Growth of Oriental Melon)

  • 검은점뿌리썩음병의 병원균에 감염된 참외의 광합성 및 생육 반응을 측정한 결과, 수확기 참외의 광합성 속도는 이병주가 건전주에 비해 현저하게 낮았고, 기공저항은 이병주에서 큰 것으로 나타났다. 목부일비액은 이병주에서 전혀 분비되지 않았다. 수확기 참외의 잎과 줄기에서 무기양분함량은 이병주와 건전주 간에 차이를 보여주지 않았다. 참외의 생육특성은 이병주가 건전주에 비해 엽면적이나 생체중및 건물중에서 건전주에 비해 낮았으며, 평균 과중도 같은 경향을 보였다.
