Plants have evolved physiological, biochemical and metabolic mechanisms to increase their survival under the adverse conditions. This present study has been performed to select salt-tolerant rice mutant lines through in vivo and in vitro mutagenesis with gamma-rays. For the selection of the salt-tolerant rice mutants, we conducted three times of selection procedure using 1,500 gamma ray mutant lines resulted from an embryo culture of the original rice cv. Dongan (wild-type, WT): first, selection in the a nutrient solution with 171 mM NaCl; second, selection under in vitro condition with 171 mM NaCl; and third, selection in a reclaimed saline land. Based on a growth comparison of the entries, out of the mutant lines, two putative 2 salt tolerant (ST) rice mutant lines, ST-87 and ST-301, were finally selected. The survival rate of the WT, ST-87 and ST-301 were 36.6%, 60% and 66.3% after 7 days in 171 mM NaCl treatment, respectively. The WT and two salt tolerant mutant lines were used to analyze their genetic variations. A total of 21 EcoRI and Msel primer combinations were used to analyze the genetic relationship of among the two salt-tolerant lines and the WT using the ABI3130 capillary electrophoresis system. In the AFLP analysis, a total of 1469 bands were produced by the 21 primer combinations, and 700 (47.6%) of them were identified as having polymorphism. The genetic similarity coefficients were ranged from 0.52 between the ST-87 and WT to 0.24 between the ST-301 and the WT. These rice mutant lines will be used as a control plot for physiological analysis and genetic research on salt tolerance.
Rice consumption per capta, in South Korea, has been decreased dramatically, owing to the changes of living patterns. Because of not only the major energy source of Korean people but also major income source of Korean farmers, diversifying end-use-quality of rice has been demanded. To the context, 'Suweon 472', a high yielding and early mature japonica line and released as 'Namilbyeo' to framers in 2002, was treated with a chemical mutagen, Sodium Azide to find endosperm mutant types. A total of nine endosperm mutat lines, including five waxy, one dull, two floury, and one white core type, were identified from the 3,542 mutatagen treated lines. Amylose contents, iodine reaction, disintegration in alkali solution, gelatinization in urea solution and amylogram properties of those nine endosperm mutant lines were evaluated to address the possibility as new genetic materials for diversifying rice quality of Korean japonica cultivars. All embryo mutants were clearly differentiated from their wild type, 'Suweon 472', in terms of physic-chemical properties evaluated. The endosperm mutant lines would be very useful in expanding untiliztation of rice through opening new rice markets of processed foods from Korean japonica rice.

Genetic diversity is one of fundamental parameters for rice cultivar improvement. Rice mutants are also a new source for rice breeding innovation. In this study, ninety-three SSR markers were applied to evaluate the genetic variation among nineteen rice mutant lines. The results showed that a total of 169 alleles from 56 polymorphism markers was recorded with an average of 3.02 alleles per locus. The values of polymorphism information content (PIC) varied from 0.09 to 0.79. The maximum number of alleles was 7, whereas the minimum number of alleles was 2. The heterozygosity values ranged from 0.10 to 0.81. Four clusters were generated using the unweighted pair group method with arithmetic mean (UPGMA) clustering. Fourteen phenotype characteristics were also evaluated. The correlation coefficient values among these phenotye characteristics were obtained in this study. Genetic diversity information of rice mutant lines can support rice breeders in releasing new rice varieties with elite characterisitics.
To investigate dose-effect of a chemical mutagen, sodium azide on a rice elite line, Suweon472, seed aliquots were treated with five different concentrations of sodium azide. The degree of mutation levels of each aizde concentration were estimated by using DNA fingerprinting techniques such as RAPD and AFLP. Some selected mutant lines ($M_4$) were also subjected for DNA fingerprinting to estimate their mutation levels by comparing the banding patterns of the wild type, Suweon 472. RAPD and AFLP fingerprinting patterns indicated that dose-effect of different azide concentrations was not clear. With allele description of detected AFLPs among favorable mutant lines, it was possible to discriminate each mutant line from others which have similar phenotypes and reactions against pathogens. AFLP fingerprinting patterns of waxy mutant lines, otherwise, were highly homogeneous as well as their phenotypic and agronomic characters.

The Korean Rice Ds-tagging lines Database (KRDD) is designed to provide information about Ac/Ds insertion lines and activation tagging lines using japonica rice. This database has provided information on 18,158 Ds lines, which includes the ID, description, photo image, sequence information, and gene characteristics. The KRDD is visualized using a web-based graphical view, and anonymous users can query and browse the data using the search function. It has four major menus of web pages: (i) a Blast Search menu of a mutant line; Blast from rice Ds-tagging mutant lines; (ii) a primer design tool to identify genotypes of Ds insertion lines; (iii) a Phenotype menu for Ds lines, searching by identification name and phenotype characteristics; and (iv) a Management menu for Ds lines.
To investigate the mutagenecity of sodium azide (SA) and to select mutants having various agronomic characteristics in rice (Oryza sativa L. ssp. japonica), dry seeds of rice variety 'Suweon 472' were treated with 0, 0.5, 1.0, 2.0, and 4.0 mM SA solutions prepared in 0.1 M phosphate buffer (pH 3.0). Germination rate, seedling height and sterility were investigated in $M_1$ generation and chlorophyll mutations were observed in $M_3$ generation. Germination rate and seedling height decreased as the increase of SA concentration in $M_1$ generation, the maximum seed sterility (40.8%) was found at 4.0 mM SA concentration. Chlorophyll mutants were occurred in $M_3$ generation and the frequency calculated on a line basis was 13.5% at the same treatment. Many kinds of mutations for morphological and agronomic characters were observed and mutations with short culm and glabrous leaf were frequently found in $M_3$ generation. Interestingly, five mutant lines resistant to blast or bacterial blight (BB) were selected and evaluated with several isolates in $M_3$ generation although Suweon 472 has been known to be susceptible to blast and BB. These mutants showed all resistance to seven isolates of blast and a total of 76 lines among 2,567 lines evaluated showed resistant to race K1 of bacterial blight. Two mutant lines (440172 and 41272) showed different reaction to BB isolates from the other resistant mutants. A few kinds of endosperm mutants were also identified and most of them were waxy mutants.
This study was carried out to select randomly amplified polymorphic DNA (RAPD) markers associated with grain weight of a large-grain mutant, Hyacp 39-26-1, derived from anther culture of a rice cultivar, 'Hwayeongbyeo'. The segregation mode for grain weight in an F$_2$ population from a cross, 'Hwayeongbyeo/Hyacp 39-26-1', showed a nearly normal distribution. One hundred and ninety-one F$_2$plants ranged from 21.8 g to 34.7 g in 1,000-grain weight with a mean of 26.8 g. Five hundred and twenty primers were used to detect the RAPD markers associated with the grain weight of the large-grain mutant. Of these primers, 54 primers showed polymorphism between 'Hwayeongbyeo' and 'Hyacp 39-26-1'. Four RAPD markers (OPB18, OPH07, OPT20, and OPX20) were significantly related to the grain weight of twenty one F$_3$ lines derived from the cross, 'Hwayeongbyeo/Hyacp 39-26-1'. This RAPD marker could facilitate the early and efficient selection of high-yield lines through improvement of grain weight in rice.
Recently, giant embryonic rice and functional rice food are preferred by more consumers, which are attributed to the fact that the embryo has high concentrations of essential amino acids, fatty acids, and vitamins relative to other parts of rice grains. In this report, the heredity and stability of giant embryo mutations in successive generations were analyzed regarding a giant embryonic line, 'P47', induced by T-DNA insertion and a $F_2$ population from a cross between 'P47' and 'Junam'. The mutant lines with increases of 1.5, 1.7 and 1.8 times on embryo length, width and 100-embryo weight to those of the control showed stable inheritance across three generations. The continuous frequency distributions of embryo size in the $F_2$ population showed that the embryo size is a quantitative trait of polygene controlled. In addition, wide range of transgressive segregations of six traits affecting embryo size confirmed exchange of genetic materials and recombination between genes controlling embryo size. Five giant embryo mutant lines selected from the $F_2$ population will be used for artificial selection and improvement of giant embryonic varieties.
Proteomic analysis was performed in order to identify proteomic changes by salt stress between the Japonica cv. Donganbyeo (WT) and two salt-tolerant (ST) mutant lines by using the SDS-PAGE and 2-DE. Two salt tolerant rice mutant lines, ST-87 and ST-301, were selected by in vitro mutagenesis with gamma-ray. Three-week-old seedlings were treated with 171 mM NaCl for 7 days. In the SDS-PAGE, three proteins with molecular weights of 27, 46 and 58 kDa were highly increased under salt treatment. Total proteins from shoots of both WT and ST-lines were separated by two-dimensional gel electrophoresis. In 2-DE, 201, 226, 217 and 213 protein spots were detected in the untreated-or treated-WT and untreated- or treated-ST-87, respectively. Of theses, 17 and 10 protein spots were up- and down-regulated under salt stress in the WT, respectively. While, 16 and 8 protein spots were up- and down-regulated under salt stress in the ST-87, respectively, compared with the untreated plants. High intensity or de novo synthesized proteins were analyzed by MALDI-TOF/MS analysis.

When plants undergo stress, Reactive oxygen species (ROS) which remove bad elements such as mildew and virus is activated in plant body. However, if ROS is excessively increased, plant will be harmed itself by destruction of cell and signal system and phenomenon of lipid peroxidation. In order to identify content of lipid peroxidation and activity of some enzymes scavenging ROS, phenotypical and physiological analysis was performed with two mutant lines, Till-II-877 and Till-II-894, comparing with cv. Dongan (WT). In phenotype analysis, two mutant lines give to well-conditioned growth better than WT in since 5 days after salt treatment. In enzyme activities, there was a modest difference in the content of catalase (CAT) and peroxidase (POD) between Till-II-877 and Till-II-894, two mutant lines showed high levels in CAT contents than WT. However, they express low levels in POD contents. In MDA analysis, the content of Till-II-877 was higher than that of WT, but Till-II-894 was lower. This result indicates that two mutants have different mechanism against salt stress.
