Inheritance of Scentedness of Rice Leaf in a Scented Breeding Line 'P-33-C-19' (향도의 방향성 유전)

  • The inheritance of scent in rice leaf and its genetic association with base color, hull color and resistance to bacterial leaf blight were studied in F$_2$ population of two rice crosses, P-33-C-19 (scented)/Zhu-Lian-Ai and P-33-C-19/Kataktara DA2. The scent was identified by leaf analysis at tillering stage. The scented and non-scented plants segregated in the ratio of 1scented: 3non-scented showing that a recessive gene responsible for the expression of scentedness. The scentedness was independently segregated with base color. hull color and resistance to bacterial leaf blight.

  • This experiment was conducted to study the multiplication and spread of bacterial population in water exuded through the hydathode of infected leaf of Xanthomonas campestris pv. oryzae in resistant and susceptible rice cultivars to bacterial leaf blight. The results obtained are summarized as follows. The bacterial multiplication in resistant cultivars was almost constant from three days to twelve days after inoculation with population of 10$^3$-10$^4$cfu/$\textrm{cm}^2$, but the multiplication was increased as days after inoculation extended in susceptible cultivars. The speed of bacterial multiplication and the number of bacteria spread above and below the inoculated position were closely related with the resistance of rice cultivars to bacterial leaf blight. The bacterial multiplication and spread were observed throughout the all growing stages including seedling, maximum tillering and flag leaf stages. The bacterial populations in water exuded through the hydathode were dependent on the multiplication and spread, and the populations were also closely related with the resistance of rice cultivars.

Effects of Cassava Leaf Meal on the Rumen Environment of Local Yellow Cattle Fed Urea-Treated Paddy Straw

  • An experiment was conducted as a Latin square design with four rumen fistulated local yellow cattle with a mean live weight of 230 kg. The treatments were: $(CLM_0)$ urea-treated rice straw ad libitum plus 1 kg cassava root meal (basal diet), $(CLM_{500})$ basal diet plus 500 g cassava leaf meal, $(CLM_{1000})$ basal diet plus 1,000 g cassava leaf meal, and $(CLM_{1500})$ basal diet plus 1,500 g cassava leaf meal. The results showed that there were differences in dry matter intake of urea-treated rice straw between treatments (p<0.05). The highest total dry matter intake was observed for treatment $CLM_{1500}$, with 2.62 kg DM/100 kg LWt/day, followed by treatments $CLM_{1000}$, $CLM_{500}$ and $CLM_0$, with 2.42, 2.00 and 1.86 kg DM/100 kg LWt/day, respectively. The ruminal ammonia concentration on treatment $CLM_{1500}$ was greater than on treatments $CLM_{1000}$, $CLM_{500}$ and $CLM_0$. There were non-significant differences in the ruminal pH among the treatments. The in sacco degradability of cassava leaf meal and cassava root meal was high, and on average 75 and 85% respectively of the DM had disappeared after 24 h of incubation. Degradation rate of urea treated rice straw was 64% after 72 h of incubation.

The Effects of Silicate Nitrogen, Phosphorus and Potassium Fertilizers on the Chemical Components of Rice Plants and on the Incidence of Blast Disease of Rice Caused by Pyricularia oryzae Cavara (규산 및 삼요소 시비수준이 도체내 성분함량과 도열병 발생에 미치는 영향)

  • In an attempt to develop an effective integrated system of controlling blast disease of rice caused by Pyricularia oryzae Cav., the possibility of minimizing the disease incidence by proper application of fertilizers has been investigated. Thus the effect of silicate, nitrogen, phosphorus and potassium fertilizers on the development of blast disease as well as the correlation between the rice varieties an4 strains of P. oryzae were studied. The experiments were made in 1971 and 1973 by artificial inoculation and under natural development of the blast disease on rice plants. The results obtained are summarized as follows. 1. Application of silicate fertilizer resulted in the increase of silicate as well as total sugar and potassium content but decrease of total nitrogen and phosphorus in tile leaf blades of rice plants. 2. The ratios of total C/total N. $ SiO_2/total$ N, and $K_2O/total$ N in leaf blades of rice plants increased by the application of silicate fertilizers. There was high level of negative correlation between the ratios mentioned above and the incidence of rice blast disease. 3. Application of silicate fertilizer reduced the incidence of rice blast disease. 4. The over dressing of nitrogen fertilizer resulted in the increase of total nitrogen and decrease of silicate and total sugar content in leaf blades, thus disposing the rice plants more susceptible to blast disease. 5. Over dressing of phosphorus fertilizer resulted in the increase of both total nitrogen and Phosphorus, and decrease of silicate content in the leaf blades inducing the rice plants to become more susceptible to blast disease. 6. Increased dressing of potash resulted in the increase of silicate content and $K_2O/total$ N ratio but decrease of total nitrogen content in leaf blades. When potassium content is low in the leaf blades of rice plants, the additional dressing of potash to rice plant contributed to the increase of resistance to blast disease. However, there was no significant correlation between additional potassium application and the resistance to blast disease when the potassium content is already high in the leaf blades. 7. When four rice varieties were artificially inoculated with three strains of P. oryzae, the incidence of blast disease was most severe on Pungok, least severe on Jinheung and moderate on Pungkwang and Paltal varieties. 8. Disease incidence was most severe on the second leaf from top and less sever on top and there leaf regardless of the fertilizer application when 5-6 leaf stage rice seedlings of four rice varieties were artificially inoculated with three strains of P. oryzae. 9. The pathogenicity of three strains of P. oryzae was in the order of $P_1,\;P_2,\;and\;P_3$ in their virulence when inoculated to Jinheung, Paltal, Pungkwang varieties but not with Pungok. The interaction between strains of P. oryzae and rice varieties was significant.

Heading date and final Leaf Number as Affected by Sowing Date and Prediction of Heading Date Based on Leaf Appearance Model in Rice (벼 파종기에 따른 출수기 및 최종 엽수 변화와 출엽 모델에 의한 출수기 예측)

  • Sowing date experiments were carried out by employing a rice variety "Kwanganbyeo" in both field and phytotron with natural daylength. In phytotron, temperatures were controlled at daily mean of 21$^{\circ}C$ and 24$^{\circ}C$. The responses of final leaf number and beading date were analyzed in relation to daylength during photo-sensitive period (PSP). Based on the component models predicting the final leaf number and leaf appearance rate, a rice phenology model was established and verified. Days from sowing to flowering (DSF) were shortened and final number of leaves (FNL) increased as sowing dates were delayed from 25 April to 5 June in field and phytotron. The increased leaf appearance rate (LAR) and the reduced FNL, respectively, due to the higher temperature and the shorter daylength in delayed sowings in the field brought about greater shortening of DSF than in the phytotron where only FNL was reduced by shorter daylength in delayed sewings. FNL showed very close relationship with the average daylength during PSP of six-leaf stage to panicle initiation, being well fitted to the following rational function ($R^2$=0.98):(equation omitted) where D is daylength and a, b, and c are the constants that were estimated as 14.694, -0.992, and -0.068 in Kwanganbyeo, respectively. The rice phonology model, which was composed of two component models for LAR and FNL, predicted DSF very accurately. The differences between the observed and predicted DSF was less than two days in the sewing date field experiments in 1999 and 2000 of which data were not used for the model construction.struction.

Nitrogen Mineralization of Cereal Straws and Vetch in Paddy Soil by Test Tube Analysis

  • Mineralization of organic N is an important factor in determining the appropriate rate of organic matter application to paddy fields. A kinetic analysis was conducted for nitrogen mineralization of rice, barley, Chinese milk Ovetch (Astragalus sinicus L.; MV) and narrow leaf vetch straw in paddy soil. Nitrogen immobilization occurred rapidly and its rate increased in straw with high C/N ratio. The amount of nitrogen mineralization was rapid in the first year of rice-vetch cropping system. The rate constant (K) depended on the C/N ratio of organic matter. Mineralization of straw increased at high temperature. The amount of available N increment resulted in fast mineralization of straw, especially in rice and barley straw. Chinese milk vetch had the greatest mineralization rate at all temperatures and fertilization levels followed by narrow-leaf vetch. However, rice and barley straws with high C/N ratio immobilized the soil N at the initial incubation duration. Chinese milk vetch or narrow leaf vetch was not effectively mineralized in mixed treatments with rice or barley straw. The mineralization rate of organic matter was mostly affected by the C/N ratio of straw and temperature of incubation. Organic matter with low C/N ratio should be recommended to avoid the immobilization of soil N and the increasing mineralization rate of straw.

Different expression levels of OsPLS1 control leaf senescence period between indica and japonica-type rice

  • Leaf senescence is the process of aging in plants. Chlorophyll degradation during leaf senescence has the important role translocating nutrients from leaves to storage organs. The functional stay-green with slow leaf yellowing and photosynthesis activity maintenance has been considered one of strategy for increasing crop productivity. Here, we have identified two QTLs on chromosome 9 and 10 for leaf senescence with chlorophyll content of RIL population derived from a cross between Hanareum 2, early leaf senescence Indica-type variety, and Unkwang, delayed leaf senescence Japonica variety. Among these QTLs, we chose qPLS1 QTL on chromosome 9 for further study. qPLS1 was found to explain 14.4% of the total phenotypic variation with 11.2 of LOD score. Through fine-mapping approach, qPLS1 QTL locus was narrowed down to about 25kb in the marker interval between In/del-4-7-9 and In/del-5-9-4. There are 3 genes existed within 25kb of qPLS1 locus: LOC_Os09g36200, LOC_Os09g36210, and LOC_Os09g36220. Among these genes, transcript level of LOC_Os09g36200 was increased during the leaf senescence stage and the expression level of LOC_Os09g36200 in Indica was higher than in Japonica. Finally, we chose LOC_Os09g36200 as candidate gene and renamed it as OsPLS1-In and OsPLS1-Jp from Indica- and Japonica-type rice, respectively. OsPLS1-In and OsPLS1-Jp overexpressing transgenic plants showed both early leaf senescence phenotype. These results indicate that OsPLS1 functions in chlorophyll degradation and the difference of expression level of OsPLS1 cause the difference of leaf senescence between Indica and Japonica in rice.

  • We investigated the systemic effects of leaf folder (Cnaphalocrocis medinalis) resistant rice (transgenic rice, Nakdongbyeb) in Sprague-Dawley rats for 13 weeks. Leaf folder resistant rice was added to the diet at percentage levels of 0, 5 and 20 percentage/feeder and was administered for 13 weeks. The results did not show any changes in food and water intake. There were also no biologically significant changes in both body and organ weights, hematological and blood biochemical parameters, autopsy and histopathology between the treatment and control groups. Based on these results, no observed adverse effect level (NOAEL) of transgenic rice was considered to be more than 10,000 mg/kg b.w. under the conditions of the present study.

  • Agriculture is the most primitive civilized Activities of mankind but also the propellant of civilization development. Because it is the most basic material goods source of mankind. Among these materials rice is one of the most important part of these, we call them the substance of survival. From the beginning of the agricultural activities to the present we have experienced three industrial revolutions and are experiencing the Fourth Industrial Revolution. With the development of science and technology makes the efficiency of agricultural production is higher and higher, but compared with the original we are facing the same problem: natural disasters; pests and diseases; now also face the depletion of resources, environmental degradation and other issues. Therefore, improve and cultivate new crop varieties to make it better resistance and more production for better develop modern agriculture. It's very helpful for human social development. And also it is the responsibility and task of modern molecular breeding. In this study, I used bacterial leaf blight to find a better resistance gene to improve the resistance of rice. Frist Cultivate k3 of bacterial leaf blight, than inoculation by leaf clipping method (Kauffman,1973) in CNDH and SNDH population at 40days after rice transplanting. Check the lesion length by inoculation plants at 14days after inoculation, and record data for QTL analysis program. Than I get 4 intervals in 3 different chromosomal regions. I found these defense genes in the 4 intervals. So I used NCBI Justbio, Rapdb, etc. to finding these genes in physical map, than design primer for map base cloning. At last these defense genes will be employed in further research for introduction of the gene to the parental plant and rice breeding for solving food crisis.
