Fine mapping of rice bacterial leaf blight resistance loci on K1 and K2 of Korean races of Xoo (Xanthomonas oryzae) using GWAS analysis

  • Bacterial leaf blight(BLB), caused by X. oryzae pv. oryzae(Xoo), is one of the most destructive diseases of rice due to its high epidemic potential. Understanding BLB resistance at a genetic level is important to further improve the rice breeding that provides one of the best approaches to control BLB disease. In the present investigation, a collection of 192 accessions was used in the genome-wide association study (GWAS) for BLB resistance loci against four Korean races of Xoo that were represented by the prevailing BLB isolates under Xoo differential system. A total of 192 accessions of rice germplasm were selected on the basis of the bioassay using four isolated races of Xoo such as K1 and K2. The selected accessions was used to prepare 384-plex genotyping by sequencing (GBS) libraries and Illumina HiSeq 2000 pairedend read was used for GBS sequencing. GWAS was conducted using TASSEL 5.0. The TASSEL program uses a mixed linear model (MLM). The results of the bioassay using a selected set of 192 accessions showed that a large number of accessions (93.75%) were resistant to K1 race and K2 resistant germplasm proportion remained between 66.67. The genotypic data produced SNP matrix for a total of 293,379 SNPs. After imputation the missing data was removed, which exhibited 34,724 SNPs for association analysis. GWAS results showed strong signals of association at a threshold of [-log10(P-value)] more than 5 (K1 and K2) for nine of the 39 SNPs, which are plausible candidate loci of resistance genes. These SNP loci were positioned on rice chromosome 2, 9, and 11 for K1 and K2 races. The significant loci detected have also been illustrated and make the CPAS markers for NBS-LRR type disease resistance protein, SNARE domain containing protein, Histone deacetylase 19, NADP-dependent oxidoreductase, and other expressed and unknown proteins. Our results provide a better understanding of the distribution of genetic variation of BLB resistance to Korean pathogen races and breeding of resistant rice.

비트잎가루를 첨가한 설기떡의 품질특성 연구 (Quality Characteristics of Sulgidduk with Beet Leaf Powder)

  • The purpose of this study is to propose the optimum adding rate of freeze-drying beet leaf powder, which has antioxidant components, that have superior DPPH radical extinction effects, in the Sulgidduk, a representative of steamed rice cake, which improve its functionality. According to the measured results on moisture contents and pH levels of Sulgidduk, with added freeze-drying beet leaf powder, the moisture contents have been significantly declined with increasing rates of added beet leaf powder. The pH levels of sample groups are within the range of 6.26-6.13. From the chromatography of measured results, lightness and yellowness have declined and redness have increased along with increasing rates of added beat leaf powder. According to the texture of measured results, the hardness, and chewiness have declined by the increasing the rate of added beet leaf powder. For the storage period, hardness, and chewiness have been inclined, but cohesiveness declines, with increased the storage period, while there are no changes from elasticity. The DPPH contents of Sulgidduk, with added freeze-drying beet leaf powder, inclined by increasing the rates of added beet leaf powder. The sensory test results of color, aroma and fragrance incline with increasing rates of added beet leaf powder. From acceptance test results, the sample group added with 3% receive the highest appraisals.

몇가지 낙엽에서의 궐련벌레, Lasioderma serricorne F.(Coleoptera : Anobiidae), 생태적 특성에 관한 연구 (Ecological characteristics of cigarette beetle, Lasioderma serricorne F., on several leaf litters)

  • The feeding preference of cigarette beetles, Lasioderma serricorne F., was tested using various leaf litters. The number of the trapped L. serricome was 45.25$\pm$10.44 at flue-cured leaf tobacco, Nicotiana tabacum L., 23.50$\pm$6.0 at chinese juniper, Juniperus chinensis L., 1l.75$\pm$4.99 at oak, Qqercus acutissiuma C., and 1l.50$\pm$2.52 at rice-straw, (Oryza. sativa L.). The response of oviposition was 93.20$\pm$26.22 at flue-cured leaf tobacco, 53.60$\pm$11.82 at chinese juniper, 48.20$\pm$20.90 at oriental arborvitae, Thuja orientalis L., 31.80$\pm$18.10 at cherry-tree, Prunus serrulata var. spontanea M., and 29.40$\pm$13.7 at rice-straw. However, the oviposition was respectively low at gingko, Ginkgo biloba L.,(5.40$\pm$2.97), turf grass, Zoysia japonica S., (5.20$\pm$13.7), and oak (3.00$\pm$l.41). The augmentation was maximum at chinese juniper (27.33$\pm$19.44 of emerged adults) followed by Magnolia obovata (8.50$\pm$9.33). Fifty percent of the tested species leaf litters including cherry-tree did not show any augmentation. The adult activities after hibernation were primarily found in May and June at Kwangju and Suwon, and in April at Chungju. The field activity of L. serricome at Suwon was mostly lower than that at other places, except in August at Chungju. The first appearance of L. serricome was observed earlier at Chungju and Kwangju than at Suwon, and the frequency of insect appearance was high in July, August, and September. L. serricome could hibernate by feeding on many kinds of plant leaf litters and it's population could be maintained in the open field in Korea.

벼의 생산력(生産力) 분석(分析) II 생산구조(生産構造) 평가(評價)와 군낙평점(群落評點) (Analysis of Productivity in Rice Plant II Evaluation of Canopy Structure and Canopy Score)

  • 새로 육종(育種)된 다수성(多收性) 품종인 IR677계통(수원 213 및 214)과 진흥(振興) 및 팔달(八達)을 사용 포장상태(圃場狀態)에서의 군란구조(群落構造)를 비교분석(比較分析)하여 다음과 같은 결과(結果)를 얻었다. 1) IR667계통 (IR8${\times}$Taichung Native 1${\times}$Yukara)은 군락(群落)높이가 낮고 개벽도(開蘖度)가 크며 개엽도(開葉度)가 적고 엽장비율(葉長比率)(지엽장(止葉長)/3엽장(葉長))이 적고 4+5절간장(節稈長)이 짧으며 5절간(節幹) 직경(直俓)이 커서 결과적으로 엽면적지수(葉面積指數)가 크며 수중형(穗重型)이고 엽중비율(葉重比率)(엽중(葉重)/엽초십간중)이 큰 장점을 갖는 반면 엽폭(葉幅)이 크고 엽수(葉數)(잎수/$m^2$)가 적고 비엽면적(比葉面積)( $cm^2/g$)이 적고 노화(老化)에 의한 군락파양율(군낙파양율)이 큰 단점(短點)을 갖고있다. 2) IR667 계통은 군락구조(群落構造)의 10개(個) 구성요소(構成要素)에 의한 구조(構造)의 평가방식(評價方法)으로 제안(提案) 한 "군락평점(群落評點)"이 훨씬 높으며 결과적으로 수량(收量)도 높았다. 3) 개별엽(個別葉)의 질(質)은 장려품종이 좋은때문에 보다 높은 수량을 위한 조합(組合)은 장려품종의 엽(葉)으로 IR667의 구조(構造)를 갖는 것이다.

Relationship between RADARSAT Backscattering Coefficient and Rice Growth

  • This study was carried out to assess the use of RADARSAT data which is C-band with HH polarization for the rice growth monitoring in Korea. Nine time-series data were taken by shallow incidence angle (standard beam mode 5 or 6) during rice growing season. And then, backscattering coefficients ($\sigma$$^{\circ}$) were extracted by calibration process for comparing with rice growth parameters such as plant height, leaf area index(LAI), and fresh and dry biomass. Field experimental data concerned with rice growth were collected 8 times for the ground truth at the study area, Tangjin, Chungnam, Korea. At the beginning of rice growth, backscattering coefficients were ranged from -l6~-l3dB when rice fields were not covered with rice canopy and flooded. At the maximum vegetative stage of rice, backscattering coefficients of the rice field were the highest ranging from -4.4dB~-3.1dB. The temporal variation of backscattering coefficient($\sigma$$^{\circ}$) in rice field was significant in this study. Backscattering coefficient ($\sigma$$^{\circ}$) of rice field was a little bit lower again after heading stage than before. This results show RADARSAT data is promising for rice monitoring.

우리나라 재래벼의 작물학적 특성 (Agronomic Characteristics of Korean Landrace in Rice)

  • 우리나라의 재래벼 192품종을 공시하여 다양한 작물학적 특성을 평가하고 양적 형질간의 상근관계를 검토하여 유기농업 및 친환경농업에 적합한 품종을 선발하고 추후 이를 품종육성의 소재로 활용하고자 실험을 수행한 결과는 다음과 같다. 1. 재래벼는 파종에서 출수까지 평균 111.1일이 소요되어 중생종에 속하는 품종이 많았고 초장과 간장은 일반 재배 품종에 비하여 대부분 20∼40cm 정도 컸다. 2. 엽폭과 이삭의 평균 길이는 각각 1.3cm, 22.4cm이었고 주당 이삭수는 평균 10개로 소얼성의 특성을 나타내었으며, 현미의 장폭비는 평균 1.7이었고 장립종도 일부 존재하였으며 천립중은 평균 21.6g이었다. 3. 재래벼의 양적 형질간 상관관졔는 간장과 수장, 엽폭, 출수기간에는 고도로 유의한 정의 상관이 있었으며, 출수기와 천립중 간에는 고도로 유의한 부의 상관이 존재하였다. 4. 간강도는 27.1%가 약한 특성을 나타내었고 엽신과 엽초의 색은 3.6%가 자주줄무늬를 나타냈으며 엽각와 지엽각도는 직립형이 각각 46.9%, 10.9%이었고 엽노화도는 지연형이 24.0%이었다. 5. 재래벼의 74.5%가 까락이 있었고 찰벼는 20.3%, 갈색종피는 4.2%가 존재하였으며, 주두색, 영색, 부선색 및 망색은 백색, 갈색, 적색 및 자색 등 다양한 색이 존재하였다.

인삼의 부산물을 이용한 식의성 단백질의 효율 향상을 위한 연구 (A Study on the Improvement of Dietary Protein-efficiency by Supplement of the Panax Ginseng-by-products.)

  • Our nation is confronted with the situation that the rice, a principal food, short of some essential amino acids, leads to imbalanced meals insufficient in the nutrient of Protein, to bring many difficulties in the elevation of nutritional state in our nation. While. our country has been produced much amounts of Panax Ginseng roots which has a stimulating effects on the metabolism of protein, lipid and nucleic acids in the body. And the leaf and trunk of Panax Ginseng were also produced a considerable amounts as the by-products. Author believe that these by-products (leaf and trunk) of Panax Ginseng might have some components possessing simillar activity with Panax Ginseng root although the quantity and qualify of the functional components may more or less be different. Therefore, this study was demised to observe the supplemental effect of the Panax Ginseng-by-Products on the dietary protein efficiency and nutritional state of rats. The feeds used for this experiment were rice containing 30% barely, fish four, and the leaf, trunk and small root of the Panax Ginseng, and the contents of the general nutrients including protein, lipid and carbohydrate etc. in each feed were analyzed for the combination of each feed. And, being based on analytical values of Protein in food. fish Pour as Protein source was added were rice containing 30% barely to be include 8.6 to 8.7%, 12%, 15% and 18% of protein. Then 2% of the leaf, trunk or small reef of Panax Ginseng was supplemented into each of above protein diet group, ton 16 kinds of diets were Prepared. The male albino rats from a Pure strain, weighing 70g to 80g. were used for experimental animals. They were maintained with coresponding fist for f and 8 weeks, and the growth rate, consumption of diets and protein, efficiency of feed and Protein in animals were determined. The lipids, proteins and cholesterols in serum and liver were also determined quantitatively after they were sacrificed in coresponding term. The results obtained are summarized as follows: 1. Body weigh of diet group containing 8.6 to 8.7%,12%, and 15% of protein are increased remarkably by supplement of 2% of the leaf or small root of Panax Ginseng in comparison with each of controls. But this tendency could not observed in diet group containing 18eA Proteins. 2. Feed efficiency showed same tendency in comparison with changes of gained body weight. Specially, in each of diets containing 8.7%, 12%, 15% and 18% of Proteins, supplement of the leaf of Panax Ginseng showed the better feed efficiency than supplement of the trunk or small root. 3. In feeding group for 8 weeks, protein efficiency showed worst efficiency in diet containing 18% proteins and showed the best efficiency was the diet group containing 12% Proteins. And the efficiency was improved according to supplement of the leaf of Panax Ginseng. 4. Nitrogen contents in serum and liver did not show large differences each other in all diet groups. But contexts of total cholesterol and 1ipid were decreased markedly in diet groups containing 12%, 15% and 18% of proteins in comparison with diet group containing 8.6% to 8.8% of proteins.

뽕잎 분말 첨가 쌀 파스타 제조의 최적화에 관한 연구 (A Study on the Optimization of Rice Pasta with Addition of Mulberry Leaf Powder)

  • 본 연구는 쌀로 만든 생 파스타 제조시 약이성 재료인 뽕잎 분말을 첨가하여 기능성 파스타로써 이용 가능성을 알아보고자 하였다. 예비 실험을 거쳐 강력분 40 g과 쌀가루 60 g에 0.5, 1.0, 1.5, 2.0%의 뽕잎 분말을 쌀 파스타에 첨가하여 제조한 후 기계적 검사(수분 함량, 색도, 텍스쳐, 인장)와 관능검사(정량적 묘사 분석, 기호도 검사)를 실시하였다. 뽕잎가루 첨가군은 첨가량이 증가할수록 생면 쌀 파스타의 수분 함량이 유의적(p<0.01)으로 낮아지는 경향을 보였고, 색도는 뽕잎 첨가량이 증가할수록 명도와 적색도는 유의적(p<0.001)으로 낮아졌고, 황색도는 유의적인 차이가 없었다. 뽕잎 분말을 첨가한 쌀 파스타의 숙면 경도(hardness)는 시료 간에 유의적인 차이를 보였고, 부착성(adhesiveness), 씹힘성(chewiness)은 유의적인 차이가 없었다. 생면의 인장 길이 (tension distance)는 MRP 0.5(4.73)가 가장 높았는데, 이는 뽕잎 분말의 수분 함량이 적기 때문에 뽕잎가루 첨가량이 증가할수록 인장 길이가 감소되는 것으로 사료된다. 인장 강도(tension force)는 MRP 0.5(34.47)로 가장 컸으며, 뽕잎가루 첨가량이 증가할수록 인장 강도가 감소하였다. 조리면의 인장 길이(tension distance)는 뽕잎 분말은 첨가하지 않은 CON이 가장 높은 값을 나타내었으나, 유의적인 차이를 보이지 않았으며, 인장 강도(tension force)는 유의적(p<0.001)인 차이를 보였다. 정량적 묘사 분석 결과, 뽕잎가루의 첨가량이 증가할수록 뽕잎의 색, 풋내, 고소한 냄새, 쓴맛, 깔깔한 맛은 강하게 윤기와 쫀득한 정도는 약하게 평가되었고, 기호도 검사 결과, 색, 냄새, 텍스쳐, 전체적인 기호도에서 MRP 1.5가 가장 기호도가 높았다. 따라서 적절한 뽕잎의 첨가는 전체적인 기호도에 긍정적인 영향을 미치며, 뽕잎가루 1.5%, 첨가시 쌀 파스타의 관능적 및 기계적인 품질 특성의 향상이 가능한 가장 적절한 첨가 수준이라고 사료된다.

Mapping Paddy Rice Varieties Using Multi-temporal RADARSAT SAR Images

  • This study classified paddy fields according to rice varieties and monitored temporal changes in rice growth using SAR backscatter coefficients (${\sigma}^{\circ}$). A growing period time-series of backscatter coefficients was set up for nine fine-beam mode RADARSAT-1 SAR images from April to October 2005. The images were compared with field-measured rice growth parameters such as leaf area index (LAI), plant height, fresh and dry biomass, and water content in grain and plants for 45 parcels in Dangjin-gun, Chungnam Province, South Korea. The average backscatter coefficients for early-maturing rice varieties (13 parcels) ranged from -18.17 dB to -6.06 dB and were lower than those for medium-late maturing rice varieties during most of the growing season. Both crops showed the highest backscatter coefficient values at the heading stage (late July) for early-maturing rice, and the difference was greatest before harvest for early-maturing rice. The temporal difference in backscatter coefficients between rice varieties may play a key role in identifying early-maturing rice fields. On the other hand, comparisons with field-measured parameters of rice growth showed that backscatter coefficients decreased or remained on a plateau after the heading stage, even though the growth of the rice canopy had advanced.

Hymexazole과 Metalaxyl의 혼합제처리가 수도 유묘의 생육 및 저온장해에 미치는 영향 (Effect of Mixture of Hymexazole and Metalaxyl on Growth and Low Temperature Injury in Rice Seedlings)

  • 본 시험은 입고병 방지제로 알려진 hymexazole과 metalaxyl의 혼합제(Tachigarace)가 수도 유묘의 생육 및 저온장해 경감에 미치는 영향을 살펴보고 도 체내 저온 대응의 생리적 기작을 밝히고자 작물시험장 인공기상실에서 통일형인 용문벼를 공시하여 hymexazole과 metalaxyl의 혼합제(Tachigarace)를 pot(5$\times$15$\times$10cm)당 0, 9, 18, 36mg를 토양혼화 처리하고 저온(12/12$^{\circ}C$) 처리한 후 저온반응 생리를 파악하였으며 일본형인 팔공벼를 공시하여 노화묘의 경엽을 절제한 후 hymexaazole과 metalaxyl의 혼합제(Tachigarace)를 상자(30$\times$60$\times$3cm)당 0.36g 시용하고 0, 7, 14, 21일후에 생육 양상을 검토한 결과를 요약하면 다음과 같다. 1. hymexazole과 metalaxyl의 혼합제(Tachigarace) 처리묘는 무처리묘에 비하여 지상부와 지하부 생육, 건물중, 굴기력 및 발근력이 많았으며 18mg 처리구에서 가장 효과적이었다. 2. 저온(12/12$^{\circ}C$) 처리시 유묘는 hymexazole과 metalaxyl의 혼합제(Tachigarace) 처리로 적고 및 엽고사 정도가 현저히 감소하였으며 지상부와 지하부의 저온 신장성은 36mg/pot 까지는 처리양이 증가할수록 컸다. 3. 저온하에서 hymexazole과 metalaxyl의 혼합제(Tachigarace) 처리묘는 엽면적, 뿌리 용적, 엽록소 함량 및 뿌리의 산소 소모량 등 생리활성의 감소폭이 작으며 기공저항성이 크고 증산량은 작아서 유묘의 저온에 대한 내성이 강하였다. 4. 육묘일수 연장에 따라 초장, 건물중 및 엽면적이 증가하였으며 경엽 절제와 hymexazole과 metalaxyl의 혼합제(Tachigarace) 처리에서 그 증가폭이 크게 나타났다. 5. 발근력은 육묘일수 연장으로 방임구에서는 점감하였으나 경엽 절제구에서는 점증하였고 hymexazole과 metalaxyl의 혼합제(Tachigarace) 처리에서는 그 효과가 더욱 뚜렷하였다.
