Studies on the Relationship Between Chemical Contents of Rice Plants and Resistance to Rice Blast Disease (수도체의 함유성분과 도열병 저항성에 관한 연구)

  • For the investigations on the mechanism of blast disease resistance in the rice variety 'Toilg-il', Ihe chemical contents such as silicate, nitrogen and flavonoid in the leaf blades of tile variety were determined as compared to moderately susceptible variety 'Pung-kwang' A correlation between disease severity of rice blast and the chemical contents in the variety 'Pung-kwang' were also studied. The results showed that silicate and flavonoid contents were decreased inversely with application of nitrogen fertilizer, and negative correlation between silicate and flavonoid contents and disease severity in the field were observed. In the resistant variety 'Tong-il', these chemical contents and silicified cells were significantly higher than in the variety 'Pung-kwang' From these results, it seemed that the high contents of silicate and flavonoid in the leaf blades were related closely to the resistance of the variety 'Tong-il'.

Genomic Differentiation Among Oyster Mushroom Cultivars Released in Korea by URP-PCR Fingerprinting

  • URP primers of 20 mer derived from repetitive sequence of rice were used to assess genetic variation of oyster mushroom consisting of 10 cultivars of Pleurotus ostreatus, two cultivars of P. florida and two cultivars of P. sajor-caju which were registered in Korea. URP2F and URP38F primers produced cultivar-specific PCR polymorphic bands in the Pleurotus species. UPGMA cluster analysis using the URP-PCR data showed that 14 Pleurotus cultivars are genetically clustered into large three groups. The URP-PCR data provided important information for more efficient breeding strategies of Pleurotus cultivars.

Effect of Water Extract of Germinated Brown Rice on Adiposity and Obesity Indices in Mice Fed a High Fat Diet

  • An extract obtained from germinated brown rice was evaluated for protective effects against the adverse effects of a chronic high fat diet. C57BL6/J mice were divided into three groups; normal diet + water group (ND), high fat diet + water group (HD), high fat diet + brown rice water extract group (HD + BE) and fed for 8 weeks (6 day/week). The ND group diet was 11 kcal fat $\%$. The HD group and HD + BE group diet were 42 kcal fat $\%$. Weight gains were not significantly different between groups. However, abdominal fat $\%$ was 1.6-fold higher in the HD group than HD + BE group. Feed consumption was significantly higher in the ND group than HD group. Serum triglyceride, total cholesterol and LDL-cholesterol increased with a high fat diet, but decreased by brown rice water extract administration to the high fat diet group. Serum HDL-cholesterol decreased by the high fat diet, but increased by the brown rice wate extract administration. Also, serum HDL-cholesterol/total cholesterol ratio was 2.3-fold higher in the HD + BE than the HD. Liver triglyceride and total cholesterol were not significantly different among groups. These data suggest that brown rice water extract administration improves the serum lipid profiles of C57BL/6J mice.

Possibility of breeding super rice cultivars using gene linkage

  • The rapid growth of world population, reduction of cultivated areas for crop production, and detrimental effects of pests, diseases, and climate changes have required to breed new rice cultivars with high yield, accepted quality, but strong resistance to abiotic and biotic stresses. However, traditional breeding needs much time to breed a new cultivar, whereas the successful use of molecular breeding is still questionable. We have developed a novel mutation which allow to cross many rice cultivars together with low segregation, that allow to breed a new cultivar in only several cropping. The mechanism has been unknown, but we suggest that gene linkage may play a crucial role, of which the semi dwarf gene might be the center gene for gene linkage occurrence. The phenomenon of this possible gene linkage is contrary to Mendel rules, but it is promising to breed new rice cultivars, of which, the most elite genes in rice might be able to gather in a targeted rice variety.

  • Ac/Ds mutant lines of this study were transgenic rice plants, each of which harbored the maize transposable element Ds together with a GUS coding sequence under the control of a promoterless(Ds-GUS). We selected the mutants that were GUS expressed lines, because the GUS positive lines will be useful for identifying gene function in rice. One of these mutants was identified knock-out at Oszinc626(NP_001049991) gene, encoding a RING-H2 zinc-finger protein, by Ds insertion. In this mutant, while primary root development is normal, secondary root development from lateral root was very poor and seed development was incomplete compare with normal plant. RING zinc-finger proteins play important roles in the regulation of development in a variety of organisms. In the plant kingdom, a few genes encoding RING zinc-finger proteins have been documented with visible effects on plant growth and development. The consensus of the RING-H2(C3-H2-C3 type) domain for this group of protein is $Cys-X_2-Cys-X_{28}-Cys-X-His-X_2-His-X_2-Cys-X_{14}-Cys-X_2-Cys$. Oszinc626 encodes a predicted protein product of 445 amino acids residues with a molecular mass of 49 kDa, with a RING-zinc-finger motif located at the extreme end of the C-terminus. RT-PCR analysis indicated that the expression of Oszinc626 gene was induced by IAA, cold, dehydration, high-salinity and abscisic acid, but not by 2,4-D, and the transcription of Oszinc626 gene accumulated primarily in rice immature seeds, root meristem and shoots. The gene accumulation patterns were corresponded with GUS expression.

Indigestible Carbohydrate Contents and Physical Properties of Goami2 harvested at the Maximized Milling Quality

  • Milling qualities and indigestible carbohydrate fractions (ICF) depending on harvesting time of Goami2 (G2), mutant of Ilpum (IP) rice, was examined. Fifty days after heading (DAH) maximized head rice milling quality (57.69%) and ICF content ($5.09{\pm}0.36\;g/100\;g$). ICF contents and physical properties of G2 and IP at 50 DAH were compared. ICF of G2 was three times higher than that of IP ($1.61{\pm}0.09\;g/100\;g$). Parboiling treatment increased ICF of G2 to $7.18{\pm}0.16\;g/100\;g$. G2 showed lower water absorption index, which could lower pasting properties, but higher water solubility index, implying it contains more soluble components. Texture properties of G2 were different from those of IP, showing higher hardness, and lower adhesiveness and cohesiveness. Positive correlation was observed between ICF and hardness, but reverse correlation between ICF and cohesiveness.

Physico-chemical Properties of Giant Embryo Brown Rice (Keunnunbyeo)

  • Brown rice with a giant embryo (GE) was observed on the quality parameters of the enlargement of embryo, nutritional components, and physical properties, in comparison to normal embryo brown rice (NE). Also, the effects of germination on the quality parameters were examined. GE embryo was approximately 2.68 times larger than of NE rice. Total free sugars were significantly higher in GE rice (71.96 vs. 41.17 mg/100 g), and germinated rice increased in fructose, but decreased in sucrose and maltose. No significant difference in mineral contents was found in GE and NE rice and their germinated rice, whereas a significant increment was observed on reducing sugars and gamma-amino butyric acid (GABA) contents in GE rice. The lower water absorption index (WAI) of GE rice resulted in relatively lower pasting viscosity, whereas the increased WSI in germinated rice might be attributable to the significant increment of soluble components in GE rice.

Functional properties of an alternative, tissue-specific promoter for rice NADPH-dependent dihydroflavonol reductase

  • A deletion analysis of the Oryza sativa dihydroflavonol reductase (DFR) promoter defined a 25 bp region (-386 to -362) sufficient to confer pericarp-specific expression of ${\beta}$ -glucuronidase(GUS) reporter gene in transgenic rice. Site-specific mutagenesis of these conserved sequences and subsequent expression analysis in calli which transiently expressed the mutated promoter::GUS gene showed that both bHLH (-386 to -381) and Myb (-368 to -362) binding sites in the DEL3 (-440 to 70) promoter were necessary for complete expression of the GUS gene including the tissue-specific expression of DFR::GUS gene. The GUS gene was expressed well in the mutated Myb (-368 to -362) binding site, but not as strong as in normal condition, implying that the Myb is also necessary to express GUS gene fully. Also, we found the non-epistatic relation between Rc and DFR. There were no changes of expression patterns GUS under the Rc and rc genotypes. Thus, DFR expression might be independent of the presence of functional Rc gene and suggested that Rc and Rd (DFR) share the same pathway controlling the regulation of flavonoid synthesis but not a direct positive transcriptional regulator of DFR gene.

Characteristics of Grain Quality at Different Transplanting Times among Rice Cultivars (벼의 품종별 이앙시기가 미질 특성에 미치는 영향 IV. 지경위치별 미립의 외관특성과 미질특성의 변화)

  • This experiment was conducted to investigated the characteristics of grain appearance, chemical components. gelatinization, and palatability of cooked rice on each rachis branches as harvested from the transplanting plot of May 20 and June 5 in each two varieties of early-maturing and late-maturing types. In evaluation of rachis branches, the percentage of ripended grain and 1,000 grain weight were found to be high and heavy at upper part of panicle. The factors influencing palatability such as maximum viscosity and breakdown as gelatinization characteristics, and hardness, apringiness, cohesiveness and chewiness as texture characteristics of cooked rice were found to be high at upper part of panicle , indicating the parlability was favored more at upper part than at lower part of panicle due to the genetics and physiological metabolism of rice plants.
