Xanthomonas oryzae, a bacterial pathogen causing leaf blight disease (BLB) in rice, can cause widespread disease and has caused epidemics globally, resulting in severe crop losses of 50% in Asia. The pathogen is seed-borne and is transmitted through seeds. Thus, control of BLB requires the elimination of the pathogen from seeds. Concern about environment-friendly organic production has spurred improvements in a variety of biological disease control methods, including the use of bacteriophages, against bacterial plant pathogens. The present study explored the potential of bacteriophages isolated from diseased plant leaves and soil samples in killing the bacterial pathogen in rice seeds. Eight different phages were isolated and evaluated for their bacteriolytic activity against different pathogenic X. oryzae strains. Of these, a phage designated ${\varphi}XOF4$ killed all the pathogenic X. oryzae strains and showed the broadest host range. Transmission electron microscopy of ${\varphi}XOF4$ revealed it to be a tailed phage with an icosahedral head. The virus was assigned to the family Siphoviridae, order Caudovirales. Seedlings raised from the seeds treated with $1{\times}10^8pfu/ml$ of ${\varphi}XOF4$ phage displayed reduced incidence of BLB disease and complete bacterial growth inhibition. The findings indicate the potential of the ${\varphi}XOF4$ phage as a potential biological control agent against BLB disease in rice.
BACKGROUND: The disease resistant (OsCK1) rice was generated by inserting choline kinase (CK1) and phosphinothricin acetyltransferase (PAT) genes isolated from Oriza sativa and Streptomyces hygroscopicus into the genome of rice (Nakdongbyeo). With the potential problems of safety, the non-target organism evaluation is required as an essential element for the environmental risk assessment of genetically modified (GM) crops. In present study, we studied the effects on survival of Misgurnus anguillicaudatus and Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. METHODS AND RESULTS: The M. anguillicaudatus and C. carpio were fed on disease resistant (OsCK1) rice and non-genetically modified (non-GM) rice (Nakdongbyeo) to 0, 10, 100, 1,000 and 5,000 mg/L, as treatment concentration respectively. The OsCK1 rice used for the test was confirmed to have the OsCK1/PAT gene expression by the PCR and ELISA analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of M. anguillicaudatus and C. carpio fed on between OsCK1 rice and non-GM rice. The 96hr-$LC_{50}$ values showed no difference between OsCK1 rice (>5,000 mg/L) and non-GM rice (>5,000 mg/L). CONCLUSION(S): The results of this study suggested that there was no significant difference in toxicity for M. anguillicaudatus and C. carpio between OsCK1 rice and non-GM counterparts.
In double cropping fields, there has been a risk of increase rice stripe viral disease outbreaking as the vector, small brown planthopper that overwinter in barley fields increases. Especially rice cultivation in Honam region has two types of cropping system such as rice only cropping in a season and double cropping both of rice and barley. In double cropping system, harvesting of barley could induce migration of the vector from barley to rice field. We found that the vector's density was different in rice fields by distance from barley field. The vector's density in rice field was highest 63 numbers within one meter from the neighbor barley field, and the numbers reduced greatly as the distance from barley field increases. The disease incidence according to rice cultivation methods was different, Donjin 1 was appeared to be a susceptible variety showing 2.4% and 2.5% of disease incidence in direct and transplanting cultivation, respectively. The small brown planthopper in a nearby rice field reach peak 3 days after the barley field was harvested.

Fusarium proliferatum is the causal agent of stalk and root rot disease of maize, foot rot disease of rice, basal and root rot disease of onion and knife cut disease of sugarcane in Iran. In recent years, incidence and severity of these diseases have been increased in Iran. Fifty seven F. proliferatum single-spore isolates collected from diseased maize, rice, onion and sugarcane plants at different areas were used to study genetic diversity by determination of vegetative compatibility groups (VCGs). Chlorate-resistant nitrate non-utilizing (nit) mutants were recovered from selected isolates of F. proliferatum and used in complementation tests. All isolates in which both nit1 and NitM (or nit3) mutants were recovered, demonstrated self-compatibility. Vegetative compatibility tests by pairing nit mutants identified 30 VCGs among 57 isolates. Twenty-three isolates belonged to singlemember VCGs and the remaining 34 isolates, belonged to other seven multimember VCGs. Segregation of F. proliferatum isolates obtained from various area and host plants into different VCGs in Iran is reported for the first time. In this study, none of isolates obtained from rice complemented with any other isolates from onion and sugarcane and, non complementation occurred between onion and sugarcane isolates. Also, only one complementation occurred between one isolate of maize and one isolate of sugarcane and rice. Thus, a correlation between VCGs grouping and host preferences was founded. It is concluded that natural populations of F. proliferatum in Iran are probably genetically divergent and include isolates representing a potential risk for disease development.

For application of sodium hypochlorite as a seed disinfectant to the control of bakanae disease caused by Gibberella fujikuroi in rice, we investigated the effects of sodium hypochlorite for antifungal activity, eliminating fungus from seeds and reducing disease occurrence in vitro and greenhouse. The viability of the pathogen was significantly reduced at $80{\mu}l/l$ concentration of sodium hypochlorite, and the pathogens did not grow at over $100{\mu}l/l$ concentration of sodium hypochlorite. The effect of eliminating fungus was 90% at treatment of 0.3% sodium hypochlorite solution to infected rice seeds for eight hours. When the rice seeds were soaked into 0.5% and 0.3% sodium hypochlorite solutions for twelve hours, the disease incidences of rice seedling were remarkably reduced to 4.3% and 4.7%, respectively, compared to 97.3% of non-treatment control. The rates of seedling stand were 29.1% and 26.9% higher with the sodium hypochlorite treatment than that of non-treatment control. When prochloraz and sodium hypochlorite was treated to naturally severely infested rice seeds with bakanae disease, the disinfection effect was higher than that of prochloraz alone treatment. When the seeds were soaked in sodium hypochlorite before or after prochloraz, the rate of seed contamination was low as 4.0% or 6.3%, respectively, compared to prochloraz alone as 13.7%. The disease incidence was low as 3.7% or 8.3%, respectively, compared to prochloraz alone as 14.3%. The disinfection effect of treatment with prochloraz after sodium hypochlorite was higher than that of treatment with prochloraz before sodium hypochlorite.

Bakanae disease, caused by Gibberella fujikuroi, is one of the most devastating diseases threatening rice production in Korea. In recent years, the incidence of bakanae disease became alarming due to the mechanical transplanting practice where the spread of bakanae can be amplified during accelerating seeds growth, due to the use of seeding boxes. The development of resistant rice cultivars could be the primary and effective method for controlling bakanae disease. However, the effects of individual resistance genes are relatively small. Therefore, pyramiding of bakane R genes in rice breeding is a promising strategy having a high potential to mitigate the advert effects of bakanae disease. This study employed a gene pyramiding approach to develop bakanae disease resistant rice lines carrying qBK1, qFfR1 introduced from rice line MY299BK and cv. Nampyeong, respectively. The MY299BK carries qBK1 introduced from cv. Shingwang, which was found to have a high resistance compare to Nampyeong. In addition, the pyramiding effect of the qBK1 and qFfR1 resistance genes were investigated, and the presence or absence of these genes helped us investigate their interaction through bioassay method and MAS. Furthermore, the distribution of resistance in the population showed a biased distribution toward resistance in the F6:7 populutions. However, we could not confirm the accumulation effect of the resistance gene, but the difference between the two genes by the SN2 marker was confirmed. Therefore, the qBK1 gene harbored by MY299BK appears to be different from the qFfR1 carried by Nampyeong, suspected to possess a different bakanae disease resistant gene different from those found in MY299BK and Nampyeong.

This study investigated the effect of Lactiplantibacillus plantarum JSA22-fermented rice drinks on dextran sodium sulfate (DSS)-induced colitis in mice. Twenty-four mice were randomly assigned; No colitis (Con), colitis with tap water (DSS-only), colitis with unfermented rice (DSS-UFR), and colitis with fermented rice (DSS-FR). After inducing colitis with 2% DSS for 5 days, they were given Tap water, UFR drink, or FR drink for an additional 6 days. The DSS-FR group had significantly lower Disease Activity Index (DAI) scores compared to the DSS-only group, but no significant difference with the DSS-UFR group. Colon length was reduced in the DSS-only group. The DSS-only group had significantly higher IL-6 mRNA levels compared to the Con group, while the DSS-FR groups showed significantly lower IL-6 mRNA levels compared to the DSS-only group. These results suggest that rice drinks fermented with Lactiplantibacillus Plantarum JSA22 ameliorate the severity of DSS-colitis, by potentially reducing proinflammatory cytokines.
This study was carried out to investigate factors affecting disease development of rice brown spot caused by Cochliobolus miyabeanus in the field and under the greenhouse conditions. In normal paddy fields, the pathogen was more frequently isolated from dried rice straws than wet straws after overwintering. The pathogen was also readily isolated from wet rice straws of salty paddy fields although infection rate of the fields was low. The disease symptom was first observed at 2 to 3days after inoculation when the seedlings were kept for 6 hours in dew chamber. The number of lesions peaked at 14 hr incubation in cultivars Chucheongbyeo and 24 hr incubation in cultivar Daesanbyeo and Tetep. Sixty three-d-old seedlings were more susceptible than 21 to 49-d-old seedlings regardless of cultivar. There was some difference in number of lesions observed, but the lesions were evenly spread regardless of leaf position. Light on the rice seedlings at 3,000 lux or 6,000 lux showed more severe symptoms than those in the dark.
Rice contaminated with fungal species during storage is not only of poor quality and low economic value, but may also have harmful effects on human and animal health. The predominant fungal species isolated from rice grains during storage belong to the genera Aspergillus and Penicillium. Some of these fungal species produce mycotoxins; they are responsible for adverse health effects in humans and animals, particularly Aspergillus flavus, which produces the extremely carcinogenic aflatoxins. Not surprisingly, there have been numerous attempts to devise safety procedure for the control of such harmful fungi and production of mycotoxins, including aflatoxins. This review provides information about fungal and mycotoxin contamination of stored rice grains, and microbe-based (biological) strategies to control grain fungi and mycotoxins. The latter will include information regarding attempts undertaken for mycotoxin (especially aflatoxin) bio-detoxification and microbial interference with the aflatoxin-biosynthetic pathway in the toxin-producing fungi.
To predict rice blast, many machine learning methods have been proposed. As the quality and quantity of input data are essential for machine learning techniques, this study develops three artificial neural network (ANN)-based rice blast prediction models by combining two ANN models, the feed-forward neural network (FFNN) and long short-term memory, with diverse input datasets, and compares their performance. The Blast_Weathe long short-term memory r_FFNN model had the highest recall score (66.3%) for rice blast prediction. This model requires two types of input data: blast occurrence data for the last 3 years and weather data (daily maximum temperature, relative humidity, and precipitation) between January and July of the prediction year. This study showed that the performance of an ANN-based disease prediction model was improved by applying suitable machine learning techniques together with the optimization of hyperparameter tuning involving input data. Moreover, we highlight the importance of the systematic collection of long-term disease data.
